Anwendungsgebiet der ErfindungField of application of the invention
The invention relates to a method for stabilizing a live virus ir.-vaccine against the effects of temperature. The method is also applicable to the production of measles living virus vaccine in vaccine production.
Charakteristik des bekannten Standes der TechnikCharacteristic of the known state of the art
Aqueous solutions of live virus vaccines are known to be unstable. The usual technology to reduce instability is to remove the water by lyophilization. But even when dried, this vaccine loses part of its infectivity when stored. The speed of this loss of infectivity is the higher, the higher the storage and handling temperature. It is therefore important to maximize stability for the vaccine by choosing an optimal environment for the virus. As chemical components of this environment, a variety of chemical compounds are known. None of these compounds alone or their mixtures previously used resulted in the production of a measles vaccine, the virus content C is less than a logarithmic titer decreases after thermal exposure for 7 days at 37 0, as required by the WHO for this vaccine.

Ziel der ErfindungObject of the invention
The aim of the invention is therefore to develop a method for stabilizing z. To comply with WHO's request for the measles vaccine.
Darlegung des Weses der Erfindung 'Explanation of the essence of the invention
It has surprisingly been found that the thermostability of virus vaccines in solutions of amino acids, polyhydroxy compounds and polysaccharides increases considerably.
As a vaccine virus z. As the measles virus strain L16 / SSW used. The virus-containing cell culture medium, adjusted to 10 3 to 10 ° plaque-forming units / ml, is mixed with a sterile-filtered stabilizer mixture in a ratio of 2: 3 parts by volume. The stabilizer mixture contains the amino acid L-arginine (eg as hydrochloride), the polyhydroxy compound sucrose, sorbitol or trehalose and the polysaccharide dextran with an average molecular weight of 8 kD to 14OkD, preferably 40 ~ 70kD, in the mass ratio 5: 2: 3 , The mass of the stabilizer mixture per one vaccine dose is set at 20 to 40 mg. The mixture of virus and stabilizer is optionally filled under sterile conditions into one (0.5 ml) or ten (10 ml) vaccine doses and then frozen. In the final stage of vaccine production, the water in the vaccine is removed by lyophilization. Lyophilization conditions should be such that the freeze-dried vaccine has a residual moisture content of no more than 0.4% as measured by the WHO method. The process according to the invention surprisingly leads to success by addition of 50% of the amino acid component and in pure form. The live virus vaccine exhibits only a decrease in virus titer of less than one logarithmic titer level by the method of the invention after 7 days' temperature treatment at 37 ° C., which meets the WhO requirements. The measles vaccine may be administered alone or in combination with other viral vaccines. For example, mumps and / or rubella vaccine) Due to the higher thermostability, it is possible to use the vaccine not only in the temperate but also in the subtropical and tropical climates.
