Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung eines antithrombotischen Wirkstoffpflasters mit ausreichender Penetrationsfaehigkeit und guter, ueber laengere Zeit anhaltender Wirkstoffdosierbarkeit. Das erfindungsgemaesse Wirkstoffpflaster enthaelt Heparin und bzw. oder Heparinoide als Wirkstoffkomponente. Die gemaess der Erfindung hergestellten Wirkstoffpflaster weisen bei entsprechenden in-vitro-Versuchen deutlich verlaengerte Thrombinzeiten bei Rinderfibrinogen und entsprechende therapeutische Effekte bei Thrombophlebitis und Thrombosen auf. Der genannte antithrombotische Wirkstoff wird in einem gefuellten Kautschuk-Haftklebstoff-Praemix mit einem Gehalt von 15% bis 25% an Weichmachern, der mindestens zu 70% aus Lanolin besteht, in reiner Form oder in Form eines Adduktes mit Zeolithen und bzw. oder Schichtsilikaten in die Haftklebstoffmischung eingebracht. Die Zeolithe und bzw. oder Schichtsilikate weisen einen Wirkstoffbeladungsgrad von 0,5% bis 50% auf. Die wirkstoffhaltige Masse wird bei Temperaturen von 20C hergestellt und nachfolgend auf ein Traegermaterial mit einer Masseauflage von 150 gm 2 bis 400 gm 2 im Direkt- oder Transferbeschichtungsverfahren aufgetragen und bei Temperaturen von 20C bis 110C getrocknet.The invention relates to a method for producing an antithrombotic active substance patch having sufficient penetrability and good, over a long time persistent drug dosing. Heparinoide können sowohl in reiner Form als auch in Form von Zeolith- und bzw. oder Schichtsilikat-Addukten eingesetzt werden. Unter einem Zeolith werden synthetische Zeolithe der Typen A, X, Y und Mordenit verstanden, unter einem Schichtsilikat solche des Magadiit-Typs sowohl in reiner als auch in gebundener Form. Besonders geeignet sind Zeolithe vom Typ 13 X. Die Zeolithe bzw. Schichtsilikate werden mit Wirkstoffmengen im Bereich von 0,5% bis 50%, vorzugsweise 10% bis 40%, beladen. Die Beschichtungsdichte, die Zusammensetzung der Haftklebstoffmasse und die Art des verwendeten Zeolithes bestimmen dann im wesentlichen die Freisetzungsgeschwindigkeit des Wirkstoffs unter anderem durch den Weg des Wassers nach der Aufnahme aus der Oberfläche der Haut, dem Transport des Wassers zum Wirkstoff bzw. zum Komplex Wirkstoff-Matrix durch Zerstörung des Wirkstoff-Matrix-Komplexes und dem daraus resultierenden Gehalt an „freiem" Heparin bzw. Heparinoid in der Haftklebstoffmasse.
