Abstract
The invention relates to a method for producing a drug-like patch with a targeted release rate of pharmacologically active substances. Depending on their molecular structure, the active compounds are adsorptively bound to zeolites and / or layer silicates and incorporated into a medical pressure-sensitive adhesive based on a solution polymer or a rubber-resin adhesive mixture. The rate of drug delivery is substantially controlled by water infiltration and the resulting rate of desorption, the rate of polymer-dependent diffusion, the concentration of drug in the microcompartments, the layered nature of the system, and the drug's ability to dissolve. The active substance-containing pressure-sensitive adhesive mass may contain substances which influence a transcutaneous penetration. Tracer materials used are drug impermeable films or metal-polymer laminates.
Description
The invention relates to a method for producing a medicinal patch for use in human medicine.
The use of pharmaceutical preparations for percutaneous therapy are known, these are essentially preparations in the form of ointments or sprays. The disadvantage of this percutaneous administration is the poor dosing of the active substance and, due to the individual and locally different skin permeation, fluctuations in the therapeutically required drug concentration.
Transdermal systems which release a drug more or less continuously over a relatively long period of time and thus serve to maintain a therapeutically desired drug supply are known.
So z. As dosing membranes in the documents DE-OS 2135533, US-PS 3598122 and US-PS 3797494 used. There are also oversaturated Wirkstofflösungsmittelkomplsxe from DD-WP 217989 known or have been proposed in the DD-WP application 273884.4 for use.
Disadvantages of these technical solutions consist in the high cost of controlling the drug release especially in the dosing membranes as well as in problems of production-moderate production of multi-layer combinations.
Another Lösu: ig is that is used as Dosiermembran the pressure-sensitive adhesive itself, such. As in EP-PS 33615, in which case the vote of the Liberationsgeschwindigkeit of the drug by the adhesive is particularly difficult and thus to readjust for each Wirks'ibstanz.
Thus, high expenditures are required for the production of drug patches and some drugs have not yet been developed for transdermal systems due to the complexity in controlling drug release.
The object of the invention is to develop a process for the preparation of an active substance patch with a targeted release rate of pharmacologically active substances, disadvantages of known processes, which are particularly in the field of complicated production and the complicated control of the release mechanism, to be avoided.
The invention has for its object to develop a process for the preparation of an active substance patch, which has a targeted release rate of transdermal active substances over a longer period of therapy.
The object is achieved in that active ingredients are adsorbed bound to zeolites and / or phyllosilicates and incorporated in a medical pressure-sensitive adhesive premix.
It has surprisingly been found that zeolite or phyllosilicate-active substance complexes are suitable for keeping the rate of active ingredient release constant over longer periods of time. From the complex, the drug is desorptively released, diffused in the transpiration of the skin in the pressure-sensitive adhesive. Subsequently, the active ingredient is dissolved and diffused depending on the composition of the pressure-sensitive adhesive to the surface to the skin.
The Da system has the advantage that a variety of pharmacologically active substances that meet certain criteria for transdermal applications can be bound according to their molecular structure to a suitable zeolite and / or layered silicate.

The pressure-sensitive adhesive can furthermore contain up to 5% of solubilizer in pure form or bonded to zeolite or phyllosilicate. Lipophilic or hydrophilic agents improve the distribution and liberation or migration of the active substance into and out of the pressure-sensitive adhesive layer. schrittweise in Mischeinrichtungen innig miteinander verrührt. The solution polymer pressure-sensitive adhesive mixture is prepared by mixing the substances into the solvent polymer either as solid or pasted by means of polymer or solvent.
The active substance-containing pressure-sensitive adhesive is applied to a carrier material in either the direct or transfer coating process with a weight applied of 50 g / m 2 to 400g / m 2 and dried at temperatures of 2O 0 C to 110 0 C. When using the transfer coating method, silicone paper is used as the subcarrier.
The active substance layer can also be constructed in a multi-layered manner, wherein the reservoir layers have different compositions and concentrations of constituents. With this system, a concentration gradient towards the skin can be achieved, which makes it possible to control the therapeutically desired active substance liberation.
Permeation. The carrier materials used are polymer-impermeable metal-coated polymer films or metal-polymer laminates. In addition to protection against drug losses, the occlusion of the skin area has an increasing effect on the penetration or permeation.

The preparation of the antithrombotic active substance patch described below was carried out in a laboratory technicum. In a laboratory kneader, the following ingredients were processed to a homogeneous mass:
The zeolite adduct contains as active ingredient sodium pentosan polysulfate having a molecular weight <6000 g / mol, which is adsorptively bound to 50% by weight of a synthetic, thermally activated type 13X zeolite. This is processed with wool wax to a premix and mixed at a temperature of 40 0 C in the mass.
In a transfer coating method, the mass is applied to silicone paper, dried at 50 0 C and laminated to an aluminum composite foil. This is followed by drying at 30 ° C. The pressure-sensitive adhesive coating is 200 g / m 2 . In further process steps, packaging takes place up to the finished product.
The drug patch prepared in this way was tested for release of active ingredient from the patch mass in vitro via a thrombin time determination.
For this purpose, defined patches were incubated in a Tris buffer solution. Depending on the incubation period, the time to coagulation of bovine fibrinogen was measured from the respective experimental batch (tick method).
It has been demonstrated that in a test period of 24 hours, the drug was continuously delivered to the incubation medium.
Clinical studies have shown a significant therapeutic success in the diagnosis of thrombophlebitis and thrombosis compared to conventional treatment methods with externally applied antithrombotics.
