The invention relates to a process for the preparation of 2-bromoethylamine hydrobromide, which is a valuable intermediate for the preparation of medicaments.
Zur Darstellung von 2-Bromethylaminhydrobromid sind folgende Methoden bekannt:For the preparation of 2-bromoethylamine hydrobromide, the following methods are known:
1. Die Spaltung von N-(2-Bromethyl)phthalimid mit Bromwasserstoffsäure und Eisessig bei Siedetemperatur (S.Gabriel, Ber. dtsch. ehem. Ges. 50, 826 [1917]).1. The cleavage of N- (2-bromoethyl) phthalimide with hydrobromic acid and glacial acetic acid at boiling temperature (S.Gabriel, Ber. Dtsch. Ehem. Ges. 50, 826 [1917]).
Weiterhin muß N-(2-Bromethyl)phthalimid gesondert, z. B. aus Phthalimid und 1,2-Dibromethan, hergestellt werden. In this case, however, only an impure, hygroscopic product is obtained. Furthermore, N- (2-bromoethyl) phthalimide must be separated, for. B. of phthalimide and 1,2-dibromoethane.
Bogen, J. Amer. ehem. Soc. 64,2709 [1942]).2. The reaction of ethyleneimine with 48% hydrobromic acid at 0 to 5 0 C and then concentrating the reaction mixture in vacuo (EJMasters and MT sheet, J. Amer. Former. Soc. 64.2709 [1942]).
The use of ethyleneimine requires special technical equipment and safety measures because of its high toxicity and reactivity.
Leffler, R.Adams, J. Amer. ehem. Soc. 59, 2252 [1937]).3. The reaction of ethanolamine hydrobromide with excess phosphorus tribromide. After combining the reactants is heated, using a vigorous reaction that is difficult to control even with a batch size of 1 mole. The washing of the crude product with dry ether is another disadvantage of this method (M.T. Leffler, R.Adams, J. Amer., Soc., 59, 2252 [1937]).
4. Ausgehend von Ethanolamin wurde 2-Brommethylaminhydrobromid folgendermaßen erhalten:4. Starting from ethanolamine, 2-bromomethylamine hydrobromide was obtained as follows:
Ges. 50,826 [1917]).4.1. By saturation of a solution of ethanolamine and fuming hydrobromic acid at 0 ° C with hydrogen bromide and heating the mixture to 170 0 C (S.Gabriel, Ber. Dtsch. Former. Ges. 50.826 [1917]).
By heating ethanolamine and concentrated hydrobromic acid and slowly distilling off the water formed in the reaction and the excess hydrogen bromide acid via a well-separating column or distilling off and refluxing.
Die Ausbeuten betragen bei dieser Verfahrensweise ca. 70% ohne Aufarbeitung der Mutterlauge (Houben-Weyl, Bd.5/4, S.375).After the reaction has ended, the hot reaction mixture is poured into acetone and, after cooling, the 2-bromoethylamine hydrobromide is filtered off with suction. The yields are in this procedure about 70% without workup of the mother liquor (Houben-Weyl, Bd.5 / 4, p.375).
Nachteile der Darstellungsmethoden von 2-Bromethylaminhydrobromid, die vom Ethanolaminausgehen, sind bei 4.1. die hohen Temperaturen, die ein Arbeiten unter Druck erfordern,Disadvantages of the methods of preparation of 2-bromoethylamine hydrobromide starting from the ethanolamine are 4.1. the high temperatures that require working under pressure,
Bei niedrigen Temperaturen erstarrt das Reaktionsgemisch, so daß es sich nicht mehr in Aceton suspendieren läßt.at 4.2. the need for a well-separating column, the high operating effort in distilling off and refluxing at intervals, the pouring of the hot reaction solution in acetone, because it vaporized large amounts of acetone. At low temperatures, the reaction mixture solidifies, so that it can no longer be suspended in acetone.
Another disadvantage of 4.1. and 4.2. listed procedures is that large amounts of dilute hydrobromic acid incurred as a byproduct.
In summary, it must be stated that the technical preparation of 2-Bromethylaminhydrobromit is not solved.
The aim of the invention is to develop a process for the simple industrial production of 2-bromoethylamine hydrobromide in high yield and purity.
The invention has the object of finding a technically simple method for the preparation of 2-Bromoethylaminhydrobromid - starting.von ethanolamine and hydrobromic acid - to find that the disadvantages of the known methods of presentation, such as working under pressure, the use of a column, the intervals Abdestiilieren and Refluxing, avoiding the pouring of the hot reaction solution in acetone and the attack of large amounts of dilute hydrobromic acid.
According to the invention, the solution of the problem is achieved in that initially only as much hydrobromic acid is used, as is necessary for salt formation of ethanolamine. The water formed in the salt formation is distilled off to an internal temperature of about 13O 0 C without column, added to the residue concentrated hydrobromic acid and slowly distilled off (about 10 hours) hydrobromic acid until the internal temperature has risen again to about 13O 0 C. ,
Die Lösung wird mit Aktivkohle behandelt, filtriert, abgekühlt und das kristalline 2-BromethylaminhydrobromidThen, in a vacuum, preferably at temperatures of 115 to 125 0 C, further hydrobromic acid is removed. After completion of the distillation, the residue is cooled to about 80 0 C and dissolved in an aliphatic alcohol, preferably isopropanol. The solution is treated with charcoal, filtered, cooled and the crystalline 2-bromoethylamine hydrobromide
abgesaugt. · . .aspirated. ·. ,
For salt formation, hydrobromic acid of any concentration can be used. Conveniently, the acid recovered in the esterification is used.
A particular embodiment of the invention is that the reaction in the presence of an azeotropic entraining agent, preferably xylene, is carried out only at atmospheric pressure, water or hydrobromic acid separated using a water separator and the entrainer is distilled off before recrystallization.
Although the entire amount of concentrated hydrobromic acid needed for the esterification may be added all at once, another embodiment of the invention is to add the concentrated hydrobromic acid in several portions.
Das erfindungsgemäße Verfahren hat gegenüber den bekannten Darstellungsmethoden folgende Vorteile:The method according to the invention has the following advantages over the known presentation methods:
The hydrobromic acid recovered in the esterification can be used for the salt formation of the ethanolamine to the ethanolamine hydrobromide, so that no waste hydrobromic acid must be eliminated.
Working under pressure and the use of a column are not necessary.
The intervals Abdestiilieren and reflux cooking, 'requires a high operating effort, eliminated.
The use of isopropanol instead of acetone, the crude reaction product can be recrystallized with the addition of activated carbon, which has a favorable effect on the color of 2-Bromoethylaminhydrobromids.
Furthermore, the isopropanol can be added to the cooled reaction mixture; this eliminates the difficulties encountered in pouring the hot reaction solution into acetone.
The yields of 2-bromoethylamine hydrobromide are 10% higher without working up the mother liquor.
