Abstract
Die Erfindung bezieht sich auf ein Herstellungsverfahren von Thrombozytenpraeparaten (PRP, PC) unter Anwendung der Differentialzentrifugation bzw. von Zellseparatoren. Diese Praeparate werden in der Medizin angewendet. Erfindungsgemaess wird ein Vollblut-Blutstabilisator-Gemisch mit N-(2-Mercaptopropionyl)-glycin in einer Konzentration von vorzugsweise 5 bis 10 mmol/l einer mehrstufigen Differentialzentrifugation unterzogen, oder dem PRP nach der ersten Zentrifugation wird vor der weiteren Zentrifugation N-(2-Mercaptopropionyl)-glycin in solcher Menge zugesetzt, dass die Konzentration 5 bis 15 mmol/l betraegt. Dieses Herstellungsverfahren kann bei physiologischem p H-Wert durchgefuehrt werden. Als Blutstabilisator werden vorteilhaft in diesem Verfahren ACD (Citronensäure, Citrat, Dextrose) oder CPD (Citrat, Phosphat, Dextrose) eingesetzt. Sie entsprechen mit ihren Eigenschaften einer Präparation nahe dem physiologischen pH-Wert. Erfindungsgemäß ist es ebenfalls möglich, 0,5 mol/l N-(2-Mercaptopropionyl)-glycin in isotoner NaCI-Lösung direkt dem Vollblut-Blutstabilisator-Gemisch in einer Menge zuzusetzen, daß die Konzentration im vorgenannten Gemisch ca. 7 mmol/l beträgt. Danach erfolgt in jedem Fall eine Inkubation von 1 Stunde. Mittels Differentialzentrifugation des vorgenannten Gemisches, z. B. 250 ml für 10 min bei 250 g, wird ein PRP und danach aus diesem durch weiteres Zentrifugieren für 8min bei 1000g ein PC hergestellt. Der Überstand, plättchenarmes Plasma, kann sofort abgehoben werden. Es folgt die Resuspension im verbliebenen Plasmarest bzw. in N-(2-Mercaptopropionyl)-glycin freiem Plasma. DieThrombozytenfunktionen sind ca. 1,5 Stunden nach der Resuspension in N-(2-Mercaptopropionyl)-glycin freiem Plasma voll restituiert.1. Nach einer Inkubationszeit von ca. 1 Stunde erfolgt die Zentrifugation für 8 min bei 1 000g. Der Überstand, plättchenarmes Plasma, wird abgehoben. Das Thrombozytensediment kann sofort im verbliebenen Plasmarest bzw. in N-(2-Mercaptopropionyl!-glycin freiem Plasma resuspendiert werden.
