Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von acetylierten (6-Desoxy-D-heptopyranosid)uronimidsaeure- bzw. thioimidsaeure-alkylester-hydrochloriden. Ziel der Erfindung ist es, ein Verfahren fuer die Herstellung dieser Uronsaeurederivate zu entwickeln. Die Uronsaeurederivate der allgemeinen Formel III, in der R fuer einen Alkylrest, R1, R2 fuer ein Wasserstoffatom oder eine Acetoxygruppe, R3 fuer einen Alkylrest und X fuer ein Sauerstoff- oder ein Schwefelatom stehen, koennen durch Umsetzung der Nitrile der allgemeinen Formel I, in der R, R1 und R2 die obige Bedeutung besitzen, mit Alkoholen oder Thioalkoholen der allgemeinen Formel II, in der R3 und X wie oben definiert sind, in Gegenwart von Chlorwasserstoff hergestellt werden. Diese Uronsaeurederivate koennen als organische Zwischenprodukte fuer weitere Synthesen verwendet werden. Sie sind insbesondere zur Herstellung von C-Nucleosidanaloga mit potentiell biologisch aktiven Eigenschaften geeignet.The invention relates to a process for the preparation of acetylated (6-deoxy-D-heptopyranoside) uronimidesaeure- or thioimidsÄure alkyl ester hydrochlorides. Die Uronsäurederivate der allgemeinen Formel III, in der R für einen Alkylrest, R1, R2 für ein Wasserstoffatom oder eine Acetoxygruppe, R3 für einen Alkylrest und X für ein Sauerstoff-oder Schwefelatom stehen, können durch Umsetzung der Nitrile der allgemeinen Formel I, in der R, R1 und R2 die obige Bedeutung besitzen, mit Alkoholen oder Thioalkoholen der allgemeinen Formel II, in der R3 und X wie oben definiert sind, in Gegenwart von Chlorwasserstoff hergestellt werden. Die Umsetzungen werden in einem absoluten aprotischen Lösungsmittel, vorzugsweise in Diethylether, durchgeführt. Es ist zweckmäßig, die Nitrile I und die Alkohole bzw. Thioalkohole Il im Molverhäitnis 1:1 umzusetzen. Es sollte ein Überschuß an Chlorwasserstoff verwendet werden. Die Reaktionstemperaturen liegen um 00C. Die Reaktionszeiten betragen einige Tage. Im einzelnen wird folgendermaßen verfahren: Die Nitrile I werden in absolutem Diethylether suspendiert. Nach Zugabe der Alkohole bzw. Thioalkohole Il leitet man in diese Suspension bei O0C trockenen Chlorwasserstoff bis zur Sättigung ein. Man fügt 0,01 mol absolutes Ethanol hinzu und leitet in diese Suspension bei O0C bis zur Sättigung trockenen Chlorwasserstoff ein. Die erhaltene Lösung wird 3 Tage bei 50C aufbewahrt. Man engt die Reaktionsmischung im Vakuum ein, wäscht den zurückbleibenden kristallinen Niederschlag mit absolutem Ether und trocknet ihn über Phosphorpentoxid im Exsikkator. Ausbeute: 70%d.Th. Schmelzpunkt: 118-120°C(Methyl) - tri-O-acetyl-e-deoxy-aD-gluco-heptopyranosidyluronimic acid ethyl ester hydrochloride 0.01 mol methyl-2,3,4-tri-O-acetyl-6-cyano-6 deoxy-D-gluco-pyranosidwird suspended in 20 ml of absolute ether. one adds 0.01 mol absolute ethanol and forwards in said suspension at 0 ° C until saturation, a dry hydrogen chloride. Schmelzpunkt: 50-700C(Methyl 2,3,3-tri-O-acetyl-6-deoxy-aD-gluco-heptopyranoside) -uronothioimidic acid benzyl ester hydrochloride 0.01 mole methyl-2,3,4-tri-0-acetyl-6 -cyano-6-deoxy-aD-glucopyranoside and 0.01 mol of benzyl mercaptan are reacted in 50 ml of ether, as described in Example 1.
