Abstract
Der Differenzierungstest fuer Staphylokokken und Staphylokokkenspezies kann in der Human- und Veterinaermedizin sowie Militaermedizin und Hygiene bei mikrobiologischen Routinelaborbestimmungen Anwendung finden. Das Ziel der Erfindung besteht darin, einen Differenzierungstest fuer Staphylokokken zu entwickeln, der einfach, hochsensitiv und in kuerzester Zeit Staphylokokkenspezies differenziert. Die Erfindung hat die Aufgabe, einen Differenzierungstest zu entwickeln, der die Nachteile der bekannten Tests, wie lange Reaktionszeit, vermeidet und gleichzeitig eine Anbindung des Reagenz an Erythrozyten negiert. Erfindungsgemaess wird die Aufgabe dadurch geloest, indem Fibrinogen an Latex-Partikel gebunden und nachfolgend in an sich bekannter Weise mittels Objekttraegeragglutination mit den Staphylokokken geprueft wird. Das verwendete Fibrinogen muss hoch gereinigt sein. Die verwendeten Latex-Partikel sollten vorzugsweise aus hydrophilen oder hydrophoben Polysterol-Latex-Partikeln in der Groessenordnung von 0,5...1,5 mm bestehen. Dadurch wird eine Sensivitaet des Tests erreicht, die noch 160 des Kolonietrockengewichtes von Staphylococcus aureus ermittelt. Bei Verwendung von Vollplasma werden dagegen bis zu 5 Einzelkolonien von Staphylococcus aureus fuer ein positives Testergebnis benoetigt. Die Herstellung einer latexgekoppelten Fibrinogenpraeparation ist ohne groesseren Aufwand moeglich und ist nicht mit den Nachteilen behaftet, die mit der Konservierung und Stabilisierung von Erythrozyten verbunden sind.The differentiation test for staphylococci and staphylococcal species can be used in human and veterinary medicine as well as military medicine and hygiene in routine microbiological laboratory tests. Als Differenzierungskriterien für Staphylococcus aureus können die Phosphataseaktivität, Lysisbildner im Rahmen der Phagentypisierung und die Dnase-Aktivität herangezogen werden, wobei die Reaktionen nach Menzies (J. elin. Path. 30 [1977] 606) und Shanholzer et al. (Am. J. elin. Path. 77 [1982] 587) zeitaufwendig und mit einem hohen technischen Aufwand verbunden sind. Bekannt ist nach Jacherts (Z. f. Hyg. 142 [1956] 502), daß Staphylococcus aureus über eine lösliche und eine gebundene Plamakoagulase verfügt.
