Abstract
Die Erfindung ist fuer die Medizin und fuer die pharmazeutische Industrie von Bedeutung. Ziel ist es, durch Bereitstellung neuer stabiler Ergopeptine eine Verbesserung bekannter Therapiemoeglichkeiten bzw. eine Ausdehnung auf neue Indikationen zu erreichen. Der Erfindung liegt daher die Aufgabe zugrunde, durch gelenkte Fermentation Ergopeptine herzustellen, in denen der Prolinteil des Peptidfragmentes durch ein Analogon dieser Aminosaeure ersetzt ist. Erfindungsgemaesz wird einem submers Ergopeptine bildenden Stamm in der Wachstumsphase des Mycels eine dem Prolin analoge Aminosaeure zugesetzt und der Stamm in Gegenwart dieser Aminosaeure kultiviert; die modifizierten Ergopeptine werden aus dem Naehrmedium isoliert. Beispielsweise werden Ergosin bildende Staemme von Claviceps purpurea verwendet. Zur Herstellung von 1'sz-Methyl-5'Alpha-isobutyl-9'thiaergopeptin (Thiaergosin) wird als dem Prolin analoge Aminosaeure 1,3-Thiazolidin-4-carbonsaeure eingesetzt.The invention is important for medicine and for the pharmaceutical industry.
