Abstract
The invention relates to a reagent for erythocyte measurements, in particular for erythocyte counting. Their goal is to ensure a sufficient accuracy of Erythozytenmessungen and a high long-term stability of the measuring apparatus and the method with low equipment and process costs. The invention had the object of finding a reagent which greatly reduces the adsorbability of erythrocytes without altering their shape. As the reagent, a propylene oxide-ethylene oxide copolymer was found. As expedient has been found: the weight ratio of the propylene oxide to the ethylene oxide = 6: 4, the total molecular weight 3000g per mole and the concentration of 5-50 mg of reagent per liter of dilute blood.

Anwendungsgebiet der Erfindung:Field of application of the invention:
Derartige Messungen werden üblicherweise in medizinischen Labors für diagnostische Zwecke durchgeführt,,The invention relates to a reagent for erythrocyte measurements, in particular for the determination of the erythrocyte count and the packed cell volume. Such measurements are usually performed in medical laboratories for diagnostic purposes,
Charakteristik der bekannten technischen Lösungen:Characteristic of the known technical solutions:
Auch- kontinuierlich arbeitendeThe assurance of an accuracy of at least 3 % of the erythrocyte count and the determination of the packed cell volume is a prerequisite for a reliable diagnosis. This accuracy is difficult to achieve with particle counters and automatic analyzers that work with multiply used reaction vessels. Also - continuously working
Die fehlerreduzierende Wirkung· dieser Maßnahme ist geringοDevices and devices with disposable containers are not always reliable in the specified range of accuracy. The causes of the large measurement errors (up to 3o .%) Have hitherto been regarded as microhemolytic and aggregation of the erythrocytes. To reduce the microhemolytic reaction vessels can be made of a special material, eg «special polyethylenes. Such materials are expensive; a use of the vessels produced from them must be multiple, so that he economically. is acceptable. The achievable reduction of measurement errors is considerable, but not sufficient. In addition, the effect weakens after frequent use. To limit aggregation, it makes sense to take the measurement shortly after dilution of the blood sample. But even here, a limit is set by the necessary reaction times of the blood particles with added reagents. The error-reducing effect of this measure is small
Applicant's investigations have identified as the main cause adsorption processes between the erythrocytes and the walls of the reaction vessels.
This cause is known as a source of error for the count of platelets. According to US Pat. No. 4,160,644 this can be. Errors greatly reduced by the addition of a polyethylene glycol solution either to the platelet suspension or to its solvent. "A small amount should suffice. Attempts to use this solution also for erythrocyte measurements, brought no positive result, even with the addition of larger solution concentrations.

Darlegung des Wesens der Erfindung:Explanation of the essence of the invention:
It was an object of the present invention to find a reagent which greatly reduces the adsorbability of the erythrocytes without changing their shape and thus permits an exact erythrocyte count and determination of the packed cell volume.
Erfindungsgemäß wird als Reagens ein Propylenoxid-Ethylenoxid-Copolymer verwendet.According to the invention, a propylene oxide-ethylene oxide copolymer is used as the reagent.
The effect according to the invention is dependent on the ratio of the constituent parts of the reagent and on their solution concentration. häng'ig. Most pronounced it is at a weight ratio of the propylene oxide to the ethylene oxide content of about 6: 4 and a total molecular weight of about 3ooo g each as low has a concentration within the range 5 ~ 5o mg / 1 proved.
The reagent is added either to the suspending medium of the blood, to a specific saline solution, or to the diluted blood. It causes a lowering of the original interfacial tension of the dilute blood liquid components, thereby improving their wetting, emulsifying and dispersing properties. Due to these changes, the adsorption capacity of the erythrocytes decreases, so that no or only very few of them are retained in the vicinity of the vessel walls and do not enter into the measurement.
