Anwendungsgebiet der Erfindung Applicant Gebet the invention
The invention relates to a process for the preparation of a pharmaceutical excipient with special properties which are valuable in particular for the production of solid drug forms.
Charakteristik der bekannten technischen'Lösungen' Characteristic of the known technical ' solutions '
Bei der Produktion von Tabletten und Drageekernen werden an das Material zwei wichtige Anforderungen gestellt; Die Substanzen sollen gut preßbar sein, d. h. sie sollenIn the production of tablets and dragee cores, two important requirements are placed on the material; The substances should be well compressible, d. H. You should
. 1. gute Fließeigenschaften besitzen und unter einem Druck zwischen 5 und 30 kH/cm relativ feste Komprimate bilden und sie sollen, 1. have good flow properties and form under a pressure between 5 and 30 kH / cm relatively firm compressed and they should
Deliver the active substance to the organism at a certain speed, ie disintegrate more or less rapidly in water or dissolve in solution and be extractable.
Therefore, for the processing of active ingredients into tablets, dragées and other solid dosage forms basically several pharmaceutical HHfsstoffe needed that not only increase the mass, but auoh. in the heterogeneous fixed-off system, a required cbls
It is particularly advantageous for the same adjuvant to influence several factors in the intended manner, for example as a good explosive extract and at the same time as a good binder.
So far, only a few substances are known, which prove in this sense as ideal Tabletiderhilfsstoffe.
Cellulose powders which are commercially available as so-called microcrystalline celluloses are preferred.
These substances are good free-flowing, so that they can be pressed directly without granulation.
In addition, they provide mechanically resistant compresses even at relatively low pressing pressure, which easily disintegrate in the water.
The use of cellulose has therefore been studied extensively and introduced internationally in the pharmaceutical industry.
A disadvantage of the so-called. Microcrystalline cellulose is that they are only suitable for those tablets that are to disintegrate rapidly in water.
Ziel der Erfindung, Aim of the invention
The aim of the invention is the preparation of a pharmaceutical excipient which, like special cellulose powders, can be well and directly processed, but which makes it possible selectively to selectively adjust the disintegration and liberation properties of solid neurospores and thereby to optimize the efficacy of medicaments.
Darlegung des; Wesens der Erfindung Presentation of the; We sens the invention
Derartige Cellulose-Kunststoff-Kombinationen konnten je nach CharakterThe essence of the invention is to take advantage of the excellent conditions that cellulose has due to its chemical structure and fiber structure and to combine it with the properties of plastics. Such cellulose-plastic combinations could vary depending on the character
For the chemical change of the cellulose, the graft copolymerization was considered. As a result, the basic structure was retained, in contrast to a derivative formation on the cellulose molecule.
The grafting of the cellulose is in principle sufficiently known (1, 2). So far, however, it has only been used for the finishing of cotton and for the preparation of wooden objects. As a result, a material was obtained which has no relation to the pharmaceutical production and is unsuitable for it. The processes of the textile industry and the refinement of the wood can be taken over by the pharmaceutical industry.
It has been found that a free-flowing cellulose graft copolymer which is excellently suitable for the preparation of solid dosage forms is obtained when so-called microcrystalline cellulose or generally collulose powder is used as a novel starting material. The graft polymerization can be directed so that either only the surface or all of the cellulose particles are continuously subjected to the reaction. The resulting products behave differently accordingly. Suitable monomers are u. a. Methacrylic acid esters, acrylonitrile, acrylic esters, styrene, vinyl derivatives and silicones. The reaction is induced by energy-rich radiation. In the polymerization, in addition to the graft copolymers, especially homopolymers which are incorporated into the pores of the cellulose matrix and contribute to the stimulation of the properties. The degree of grafting and the extent of homopolymerization are u. a. depending on the radiation dose.
The loose powders prove themselves in the production of granules, tablets, dragee cores and the filling of gelatin steck caps and in other solid dosage forms. In most cases, it is sufficient to mix the active ingredients with the cellulose graft copolymers.
