Výroba piva spočívá v kontrolovaném zkvašování cukrů obsažených v mladině pivovarskými kvasinkami. Jako pivovarské kvasinky se přitom označují kulturní kvasinky několika druhů a rodů: k produkci spodně kvašených piv se používá druh Saccharomyces pastorianus, k produkci svrchně kvašených piv druh Saccharomyces cerevisiae, k produkci speciálních druhů piv např. rody Brettanomyces nebo Torulaspora (viz např. T. Kochláňová a kol.: „Kvasinky nonSaccharomyces a jejich význam v pivovarském průmyslu. I. část - Brettanomyces (Dekkera)“ a „Kvasinky non-Saccharomyces a jejich význam v pivovarském průmyslu. II. část“, obojí Kvasný Průmysl 62/2016 (7-8)). Pivovarské kvasinky přitom při své činnosti přeměňují sacharidy na ethanol, oxid uhličitý a energii potřebnou k pomnožování kvasinek, a také přibližně dva tisíce dalších látek, zejména vyšších alkoholů, esterů, aldehydů, atd., které ovlivňují výsledné senzorické vlastnosti hotového piva. Pro celkový senzorický dojem je pak velmi důležitý celkový poměr těchto všech vyšších alkoholů a esterů, neboť jednotlivé sloučeniny jsou samy o sobě velmi často pod prahovou koncentrací senzorického vnímání. Obecně je přitom např. známo, že kmeny svrchních kvasinek Saccharomyces cerevisiae produkují větší množství a šiřší spektrum senzoricky aktivních látek než kmeny spodních kvasinek Saccharomyces pastorianus (viz např. Lodolo a kol.: „The yeast Saccharomyces cerevisiae - the main character in beer brewing.“ FEMS Yeast Res 8(7):1018-36, 2008) a také to, že „lehčí“ chuť spodně kvašených piv způsobuje, kromě nižší teploty hlavního kvašení, také změněná exprese genů zodpovědných za produkci esterů u druhu Saccharomyces pastorianus (viz např. FEMS Yeast Res 8 (7): 1018-36, 2008) and that the "lighter" taste of bottom-fermented beers causes, in addition to the lower main fermentation temperature, an altered expression of the genes responsible for ester production in Saccharomyces pastorianus (see e.g. Procopio et al .: "Function and regulation of yeast genes involved in higher alcohol and ester metabolism during beverage fermentation", European Food Research and Technology 233 (5): 721-729, November 2011).

Podstata technického řešeníThe essence of the technical solution
Podstatou technického řešení je pivo připravené zkvašováním pivní mladiny kvasinkami kmene Saccharomyces cerevisiae CCM 8714, který se izoloval ze spontánního kvašení hroznového moštu. Pro další použití se připravil selekcí a opakovaným přeočkováváním na sladinovém agaru s tetracyklinem. Kultivace pak probíhala aerobně v mikrobiologickém inkubátoru při teplotě 26 °C po dobu 3 až 5 dní. Po nárůstu kultury kvasinek na povrchu agaru se tato kultura přenesla sterilizovanou očkovací kličkou na novou Petriho misku s agarem a rozočkovala se tzv. křížovým roztěrem, při kterém se ředí koncentrace kvasinek na agaru. Tento postup se opakoval do doby, než kultura kvasinek narostla do podoby viditelně oddělených kolonií. Následná izolace kmene kvasinek Saccharomyces cerevisiae CCM 8714 se pak provedla naočkováním vybrané kolonie na Petriho misku obsahující sladinový agar.The essence of the technical solution is a beer prepared by fermentation of beer wort by yeast strain Saccharomyces cerevisiae CCM 8714, which was isolated from spontaneous fermentation of grape must.
