Vynález se týká způsobu provedení konfirmačních testů pomocí fluorochromních substrátů u indikátorů fekálního znečištění, či potenciálně patogenních bakterií, jehož účelem je potvrzení taxonomické kategorie mikroorganismů, a tak přesnější specifikace hygienicky významného bakteriálního znečištění.The invention relates to a method for performing confirmatory tests using fluorochrome substrates for faecal contamination indicators or potentially pathogenic bacteria, the purpose of which is to confirm the taxonomic category of microorganisms and thus to specify more precisely hygienically significant bacterial contamination.
Dosavadní stav technikyBACKGROUND OF THE INVENTION
Dosud běžný způsob identifikace mikroorganismů je založen na sledování jejich morfologických, kultivačních, fyziologických, biochemických, ekologických, popřípadě dalších významných vlastností.The current method of identification of microorganisms is based on monitoring their morphological, cultivation, physiological, biochemical, ecological, or other important properties.
V případě baktérií, které jsou morfologicky obtížně rozeznatelné nebo zcela nerozeznatelné, je při jejich identifikaci kladen důraz pouze na specifakaci jejich fyziologických a biochemických vlastností, při tom je třeba pracovat s čistými kulturami mikroorganismů, jejichž příprava bývá obtížná.In the case of bacteria which are morphologically difficult to discern or completely indistinguishable, their identification focuses only on the specification of their physiological and biochemical properties, while working with pure cultures of microorganisms, which are difficult to prepare.
Tento způsob identifikace má řadu nevýhod. Je značně zdlouhavý, nebot trvá déle než týden, takže hrozí nebezpečí z prodlení při eventuálních hygienických závadách. Navíc je pracovně velmi náročný, nebot k přesnému provedení identifikace jednoho bakteriálního kmene, podle současných vědeckých poznatků, je třeba provést až 70 i více testů na různých kultivačních mediích. Mimo to vyžaduje i vysoce kvalifikovaný a dobře zapracovaný personál.This method of identification has a number of disadvantages. Pro běžnou praxi není třeba, ke klasifikaci hygienických či jiných závad, provádět přesnou identifikaci jednotlivých kultur mikroorganismů, nebot plně postačí provést konfirmační testy s kulturami izolovanými ze selektivních médií. Na základě jejich výsledků lze zařadit mikroorganismy s vysokou spolehlivostí do příslušné taxonomické kategorie (druh,rod), nebo pouze do určité skupiny mikroorganismů (např. koliformní bakterie”).For routine practice, it is not necessary to accurately identify individual cultures of microorganisms to classify hygiene or other defects, since it is fully sufficient to perform confirmatory tests with cultures isolated from selective media. Podstata vynálezuSUMMARY OF THE INVENTION
Tyto nedostatky řeší způsob dle vynálezu, jehož podstata spočívá v tom, že se předem připraví průmyslově vhodná absorpční podložka a nasytí se pro příslušný mikroorganismus specifickým fluorochromním substrátem. Takto připravená podložka se uchovává v suchém stavu, zatavená do nepropustné folie, při teplotě do (menší než) 4° C, za nepřístupu světla.These drawbacks are solved by the process according to the invention, which consists in preparing an industrially suitable absorbent pad and saturating it with a specific fluorochrome substrate. The substrate thus prepared is stored in a dry state, sealed in an impermeable sheet, at a temperature of up to (less than) 4 ° C, in the absence of light.

Ve vzorku pitné vody ae atanoví počet kolonií termotolerantních koloformních bakterií na ” m PC ” mediu metodou membránových filtrů. Po kultivaci ae určí počet laktózo-pozitivních kolonií.In the drinking water sample, the number of colonies of thermotolerant coloform bacteria on the "m PC" medium is determined by the membrane filter method. Snterokoky se stanoví metodou membránových filtrů, které jsou kultivovány na absorpčních podložkách nasycených tekutým mediem (SP mediem). Po kultivaci se membránový filtr přenese na novou absorpční podložku nasycenou kultivačním mediem obsahujícím fluorochromní substrát, t. j. 4-methyl-umbelliferyl -B- glukosid.Snterococci are determined by the method of membrane filters, which are cultured on absorbent pads saturated with liquid medium (SP medium).
