Description
Vynález se Lýká nového kmene produkčního mikroorganismu Agrobacterium rudiobucLur (Beierinck ct van Dcldcn 1902, Conn 1942) s antagonistickým účinkem vůči Fytotoxickým houbám označeným jako M 11. Kmen produkuje antibiotika a této schopnosti lze využít např. k odstranění či zmírnění vzniku jabloňové půdní únavy. Vzorek kultury tohoto kmene je uložen v Československé sbírce kultur mikroorganismů University 3. E. Purkyně v Brně pod číslem CCM 3996.
Z dosavadních literárních údajů je známo, že některých půdních a rizosférních mikroorganismů lze použít k inokulaci rostlin s cílem stimulovat růst at již přímým účinkem, tj. např. produkcí růstových látek (Hussain A. et al. - Folia Microbiol. 15; 468, 1970; Brown Μ. E. - 3. appl. Bacteriol. 35: 443, 1972; Přikryl Z. et al. - Biologia Plantarum 27: 159, 1985), či nepřímým účinkem, tj. např. antagonistickým účinkem vůči fytopatogenním mikroorganismům ke zlepšení zdravotního stavu rostlin (Cook R. 3. a Rovira A. D. - Soil Biol, Biochem. 8: 275, 1976; Staněk M. - Soil Biol. Biochem. 8: 269, 1976; Staněk M. - Schippers B. a Gamus W. (ed): 247, Academie Press, London 1979).
Nyní bylo zjištěno, že některé bakterie izolované s povrchu kořenů jabloní mohou potlačovat rozvoj fytotoxických hub v rizosféře, podílejících se na vzniku půdní únavy.
Na základě selektivního výběru a účinku UV záření byl vyprodukován nový kmen produkčního mikroorganismu Agrobacterium radiobacter s antagonistickým účinkem vůči Pytotoxickým houbám zejména houbám rodu Penicillíum a Alternaria. Uvedený kmen -bakterií byl izolován metodou omyvů kořenů (Harley 3. L. a Waid 3. S. - Trans. Brit. mycol. Soc. 38: 104, 1955).
g kořenů jabloňových semenáčů bylo třepáno 10 minut ve 100 ml sterilní vodovodní vody. Voda slita a znovu třepáno ve 100 ml vody. Po desetinásobném omytí byly potom 1 mm úseky nařezaných kořínků kladeny na agarové živné médium se streptomycinem (modifikované médium Smith N. R, a Dawson V. T. - Soil Sci. 58: 467, 1944).
Izolace byla provedena z kolonií bakterií obrůstajících kořenové úseky. Potom následovala jejich selekce a testy ke stanovení antibiotické aktivity vůči fytotoxickým houbám, též vlivu na růst jabloňových semenáčů.
Ke kultivaci izolovaných kmenů bakterií byl použit mnsopeptonový agar nebo bujón. Kultivace probíhala při 28 °C na třepačce.
Morfologie kmene: G- tyčinky vyskytující se nepravidelně, kolonie - kulaté, hladké, lesklé, bez pigmentu, pohyblivost - peritrichální bičíky.
Fyziologie a biochemie kmene;
pozitivní reakce - kataláza, 3-ketolaktoza, nitráty, eskulin, ONPG, Simmons citrát, ureáza, glycerol, trehalóza, manitol, sorbitol, laktóza, ínositol, celobióza, sacharóza, růst na Endo agaru, růst při 37 °C.
slabČ pozitivní reakce - glukóza aerobně, fosfatáza negativní reakce - oxidáza, King B, želatina, Tween 80, indol, sirovodík, lysin, ornitin, fenylalanin, acetoin, malonát, glukóza anaerobně, růst na 7% NaCl, růst při 41 °C.
Kmen byl identifikován v Čs. sbírce mikroorganismů Univerzity 3. E. Purkyně v Brně.
V laboratorních podmínkách metodou titrace antibiotik byla zjištěna až dvojnásobná antibiotické aktivita u bakterie Agrobacterium radiobacter vůči fytotoxické houbě Penicillium a ve skleníkových podmínkách stimulace růstu jabloňových semenáčků.
Vynález lze použít v ovocných školkách a k ozdravění půdy jabloňové výsadby (řízkovanci, semenáče).
