Herpes simplex typ 1 (HSV-1) orální a typ 2 (HSV-2) genitální jsou geneticky příbuzné a většina jejich antigenů křížově reaguje. Současně používané metody pro určování přítomnosti specifických protilátek typu HSV-2 v lidských sérech prozatím užívají bu3 metod nepřímých, např. neutralizační test (Rawls a spol., 1970, J. Immunol. 104, 599 až 606), kdy na základě výpočtu poměru hodnot titrů protilátek proti HSV-1 a proti HSV-2 (II/I index) se stanovuje přítomnost HSV-2 protilátek, vysyoováním typově společných protilátek buněčným extraktem (Porghani a spol., 1975, J. Clin. Microbiol. 2, 410 až 418), blokováním křížově reagujících antigenů heterotypickými protilátkami (Vestergaard a spol., 1979, Acta Path.
