Abstract
还原青蒿素的生产工艺属天然化合物衍生物制备的生产工艺的发明。本发明的目的在于进一步提高抗疟新药青蒿素的效价,减低用药剂量,亦为肺吸虫患者提供用量及价格均低于现有药物吡喹酮的有效新药。本发明的另一目的是为了生产制作简便程度及价格均明显优于青蒿素其他衍生物,又便于口服或直肠给药的新药。
Description
本发明是关于天然化合物衍生物制备的生产工艺的发明。
疟疾为世界性主要流行病之一,六十年代起由于恶性疟原虫对氯喹等现有抗疟药物产生了抗药性,国际上在疟疾防治上遇到很大困难,迫切要求寻找新的抗疟药物。中医研究院中药研究所于1971年发现中药青蒿的提取物对疟疾有突出疗效,并于1972年从中分离得到新结构类型的抗疟有效单体-青蒿素。经科学实验及临床实践证明青蒿素为一具有“速效、低毒”特点的抗疟新药,对各型疟疾包括抗氯喹株恶性疟及抢救脑型疟均有很好疗效。
为了进一步提高青蒿素的效价,减低用药剂量,发明人对青蒿素衍生物进行了研究,并研制成功还原青蒿素。还原青蒿素临床抗疟效价高于青蒿素6~8倍。而且对肺吸虫亦有良好效价,较现有药物吡喹酮剂量小,价格便宜。
本发明的生产工艺:以青蒿素为原料溶于甲醇后,降温至0℃~5℃,分次加入为原料十分之四量的钠硼氢(或钾硼氢)还原至反应完全,酸中和后冰水析,过滤,干燥得还原青蒿素。
本发明生产工艺的简便程度及价格均明显优于青蒿素其他衍生物,且便于口服及直肠给药。

- 还原青蒿素的生产工艺,其特征是以青蒿素为原料,溶于甲醇后,以钠硼氢(或钾硼氢)为还原剂,酸中和后冰水析。
