一种脉冲输出可正、可负、也可正负交替出现的
细胞电穿孔融合仪,它是在已有的细胞电穿孔融合仪
的输出端加装一个倒相开关,这样就可以方便地实现
脉冲极性变化输出。
Description
本实用新型涉及一种可实现细胞外源基因转移和细胞电融合的装置。该装置尤其能提供正负交替的脉冲,提高基因转移和细胞融合的效率。
目前,公知的细胞电穿孔融合仪是由正弦波发生器和脉冲发生器(矩型脉冲或RC尖脉冲)组成。细胞电融合的实现过程为,在正弦交变电场的作用下,细胞可经电介质电泳形成珠串排列,造成细胞间的紧密接触,然后施加数个脉冲诱导相邻细胞的细胞膜在接触区穿孔而融合。细胞外源基因转移的实现过程为,细胞在脉冲电场的作用下,细胞膜可逆电穿孔,外源基因可以进入细胞内部。但是,目前已商品化的一般电融合仪的输出脉冲极性固定,在实验中连续输出的几个脉冲都是同方向的,由于细胞在电穿孔时有不对称击穿现象存在,同方向脉冲影响了基因转移和细胞融合效率的充分发挥。
本实用新型的目的是提供一种细胞电穿孔融合仪,它的脉冲输出极性可正、可负,也可正负交替。正负交替的脉冲可以克服细胞的不对称击穿现象,提高细胞融合率和外源基因转移效率。
本实用新型的目的是这样实现的:该仪器由正弦波发生器和矩型脉冲发生器组成,在仪器的输出端加装一个倒相开关由它来控制脉冲的输出极性。该倒相开关采用双刀双投机械开关,它的两个刀位接输出插孔,两组各两个触点交叉连接后,相对应的两个触点接原输出线路,另两个对应触点悬空。
由于采用上述方案,可以方便地控制脉冲的输出极性,且结构简单。
下面结合附图对本实用新型进一步说明。

在图1和图2中,在仪器的输出端装有倒相开关(4),它采用一个双刀双投机械开关,可以控制脉冲波的输出极性。该开关的触点a与c’连接,a’与c连接,刀位b和b’接输出插孔,触点a接3,a’接地。假设拨向左边为输出正脉冲,则拨向右边就可输出负脉冲,连续输出脉冲时若左右交替拨动该开关,则可输出正负交替脉冲。

1、一种细胞电穿孔融合仪,它是由正弦波发生装置和脉冲波发生装置组成,其特征是:在输出端之前电连接倒相开关,该倒相开关采用双刀双投机械开关,它的两个刀位接输出插孔,两组各两个触点交叉连接后相对应的两个触点接原输出线路,另两个对应触点悬空。
