In damp and warm places, a number of macroscopic villous, flocculent or arachnoid colonies, i.e. molds, can grow on the articles. The mould is wide in distribution and strong in reproductive capacity, can not be found by naked eyes when being in a spore state, has long dormancy period and strong stress resistance, can not be reproduced temporarily when the conditions are not suitable, can preserve activity for a long time, and is full of spores of the mould in the air. The bacteria are a type of prokaryotic microorganisms, are a type of prokaryotic organisms which are thin and short in shape and simple in structure and are propagated mostly in a binary fission mode, have the diameter of 0.5-5 mu m, and are organisms which are most widely distributed in the nature and have the largest individual number. Spores are dormant bodies of bacteria and have extremely strong resistance to adverse environments. The small and light spores can also drift around with the wind, fall into a proper environment and can germinate into bacteria. The rapid proliferation and sporulation properties of bacteria make them almost ubiquitous. The paper harms the growth of microorganisms and the formation of bacterial plaque, not only can influence the visual readability effect of the paper, but also can degrade paper components to cause irreversible lesion, so that the strength of the paper is reduced, the service life of the paper is shortened, and even a large number of metabolites are bonded to form a paper brick which cannot be repaired. The ancient books can not be stored in vacuum, so that the ancient books face the risk of being corroded by microorganisms such as mold and the like all the time, the initial pollution is invisible to the naked eyes and cannot be found easily, and once the ancient books are found, the ancient books are damaged. To prevent and treat the damage of microorganisms such as mold and the like to ancient books, the cause of scientific cognitive damage is firstly required to reveal which kind or kinds of microorganisms influence the ancient books, so that microorganisms or/and nucleic acid samples can be scientifically, effectively and safely collected from the ancient books, and a sample is provided for scientifically analyzing the invasion of the ancient books by the microorganisms, so that the method has important practical significance.
The current conventional acquisition process is as follows: after the sampling position is determined, wetting the wetted polyester fiber swab by using a wetting solution (0.15mol/L NaCl and Tween with the volume fraction of 0.1%), and then performing unidirectional soft wiping for 20 times within the range of 2cm multiplied by 2cm in a target area, so as not to damage the surface of the paper; the method for collecting the air-settled sample as a control comprises the following steps: when sampling is started, the polyester fiber swab is dipped in wetting liquid and then stands on a sampling worktable, and the swab is collected in other sample processing modes after sampling is finished. It can be seen that the traditional sampling mode has the following defects that the sampling process has a friction process on ancient books, chemical reagents are used, the sampling efficiency is low, and the human error in the sampling process is large.

Detailed Description
Example 1
As shown in FIG. 1, a sampling device for biological samples in ancient scripts and paintings of ancient book archives is composed of an exhaust fan 100, a collecting device 200 and a sampling device 300.
Exhaust fan 100 is used to provide power to draw out the biological samples in the ancient calligraphy and painting of the ancient book file by suction. The air exhauster can be provided with a plurality of different gears so as to meet the requirements of different drawing forces.
The collecting device 200 is a filter membrane type collecting device, and comprises a conical upper pressing body 203, a filter membrane 204 and a conical lower pressing body 205, wherein an exhaust pipe 201 is connected to the conical upper pressing body 203, a collecting pipe 202 is connected to the conical lower pressing body 205, the filter membrane 204 is arranged between the conical upper pressing body 203 and the conical lower pressing body 205, and the conical upper pressing body 203, the filter membrane 204 and the conical lower pressing body 205 are fixed together through bolts. In order to ensure the sealing effect, a sealing ring 208 is arranged between the conical upper pressing body 203 and the conical lower pressing body 205.
In order to obtain a comprehensive biological sample of ancient scripts and paintings of the ancient book file, the sampling device 300 adopts an all-directional sampling device. As shown in FIG. 1, the omnibearing sampling device comprises a sampling body 303 and a supporting body 308, wherein the sampling body 303 is provided with a sampling port I304 and a pin hole 305 and is connected with a sampling pipe 301, the supporting body 308 is provided with an air inlet 306 and a pin 307 and is connected with an air inlet pipe 302, the sampling body 303 and the supporting body 308 are fixed together by inserting the pin 307 into the pin hole 305, and an ancient calligraphy and painting sample 500 of an ancient book file to be sampled is arranged between the sampling body 303 and the supporting body 308. Preferably, the outer edges of the sampling port I304 and the air inlet 306 are smaller than the outer edge of the ancient calligraphy and painting sample 500 of the ancient book file to be sampled.
Suction fan 100 is connected to suction line 201 of collection device 200 via hose 400, and collection pipe 202 of collection device 200 is connected to sampling pipe 301 of sampling device 300 via hose 400.
The working process is as follows: the ancient calligraphy and painting 500 of the ancient book file is arranged between the sampling body 303 and the supporting body 308, the sampling port I304 and the air inlet 306 correspond to the ancient calligraphy and painting 500 of the ancient book file, and the sampling body 303, the ancient calligraphy and painting sample 500 of the ancient book file and the supporting body 308 are fixed together by inserting the pin 307 into the pin hole 305. The conical upper pressing body 203, the filter membrane 204 and the conical lower pressing body 205 are fixed together by bolts. The exhaust fan 100 is started, biological samples on the ancient calligraphy and painting samples 500 of the ancient book archives are sucked out through suction, and are adsorbed on the filter membrane after passing through the sampling port I304, the sampling body 303, the sampling pipe 301, the hose 400, the collecting pipe 202 and the conical pressing body 205 in sequence. The exhaust blower was turned off and the filter was removed for analysis.
Preferably, in one embodiment, the sampling device 300 is a point sampling device for obtaining a biological sample from a point on the ancient painting of the ancient book file. As shown in FIG. 3, the point sampling device comprises a main body I309, wherein the upper end of the main body I309 is connected with a sampling tube 301, and the lower end of the main body I309 is provided with a sampling port II 310. During sampling, the sampling port II 310 is aligned with the sampling point.
Preferably, in one embodiment, in order to obtain a biological sample from a part of the ancient painting of the ancient book file, the sampling device 300 is a facial sampling device, as shown in fig. 4, the facial sampling device comprises a main body II 311, one end of the main body II 311 is connected with the sampling tube 301, and the other end is a sampling port III 312. When sampling, the sampling port III 312 is placed in the sampling part.
Preferably, in one embodiment, the sampling device 300 is an in-line sampling device for obtaining biological samples from the locations of the lines in the ancient book archives. As shown in FIG. 5, the wire sampling device comprises a main body III 313, one end of the main body III 313 is connected with a sampling tube 301, and the other end is provided with a sampling port IV 314. When sampling, the sampling port IV 314 is aligned with the binding part.
Example 2
As shown in FIG. 2, a sampling device for biological samples in ancient scripts and paintings of ancient book archives is composed of an exhaust fan 100, a collecting device 200 and a sampling device 300.
Exhaust fan 100 is used to provide power to draw out the biological samples in the ancient calligraphy and painting of the ancient book file by suction. The air exhauster can be provided with a plurality of different gears so as to meet the requirements of different drawing forces.
The collection device 200 is a culture medium type collection device, as shown in fig. 2, the culture medium type collection device includes a bottle 206 and a plurality of support frames 207 loaded with solid culture medium, and the support frames 207 are fixed on the inner wall of the bottle 206 in an interpenetration manner. The two ends of the bottle body 206 are respectively connected with an exhaust pipe 201 and a collecting pipe 202.
In order to obtain a comprehensive biological sample of ancient scripts and paintings of the ancient book file, the sampling device 300 adopts an all-directional sampling device. As shown in FIG. 1, the omnibearing sampling device comprises a sampling body 303 and a supporting body 308, wherein the sampling body 303 is provided with a sampling port I304 and a pin hole 305 and is connected with a sampling pipe 301, the supporting body 308 is provided with an air inlet 306 and a pin 307 and is connected with an air inlet pipe 302, the sampling body 303 and the supporting body 308 are fixed together by inserting the pin 307 into the pin hole 305, and an ancient calligraphy and painting sample 500 of an ancient book file to be sampled is arranged between the sampling body 303 and the supporting body 308. Preferably, the outer edges of the sampling port I304 and the air inlet 306 are smaller than the outer edge of the ancient calligraphy and painting sample 500 of the ancient book file to be sampled.
Suction fan 100 is connected to suction line 201 of collection device 200 via hose 400, and collection pipe 202 of collection device 200 is connected to sampling pipe 301 of sampling device 300 via hose 400.
The working process is as follows: the ancient calligraphy and painting 500 of the ancient book file is arranged between the sampling body 303 and the supporting body 308, the sampling port I304 and the air inlet 306 correspond to the ancient calligraphy and painting 500 of the ancient book file, and the sampling body 303, the ancient calligraphy and painting sample 500 of the ancient book file and the supporting body 308 are fixed together by inserting the pin 307 into the pin hole 305. The exhaust fan 100 is started, the biological samples on the ancient calligraphy and painting samples 500 of the ancient book archives are sucked out through suction, and enter the bottle body 206 through the sampling port I304, the sampling body 303, the sampling pipe 301, the hose 400 and the collecting pipe 202 in sequence, and are adsorbed on the solid culture medium. The exhaust blower is turned off and the rack 207 is removed for analysis.
Preferably, in one embodiment, the sampling device 300 is a point sampling device for obtaining a biological sample from a point on the ancient painting of the ancient book file. As shown in FIG. 3, the point sampling device comprises a main body I309, wherein the upper end of the main body I309 is connected with a sampling tube 301, and the lower end of the main body I309 is provided with a sampling port II 310. During sampling, the sampling port II 310 is aligned with the sampling point.
Preferably, in one embodiment, in order to obtain a biological sample from a part of the ancient painting of the ancient book file, the sampling device 300 is a facial sampling device, as shown in fig. 4, the facial sampling device comprises a main body II 311, one end of the main body II 311 is connected with the sampling tube 301, and the other end is a sampling port III 312. When sampling, the sampling port III 312 is placed in the sampling part.
