一种尿亚硝酸盐生化检测液其包含0.35-0.45w/v%对氨基苯磺酸、0.2-0.3mgw/v%α-萘胺、20-40v/v%冰醋酸、1-2v/v%甲醇和蒸馏水。本发明提供了一种群众性自我动态跟踪检测的尿亚硝酸盐生化检测液与尿亚硝酸盐自我检测方法。根据连续检测显色结果的记录曲线图形显示能自我判断:1.提示健康预警信号及致病因子,2.提示菌尿的早期检出率,3.菌尿治疗过程中疗效的同步跟踪监测。本尿亚硝酸盐生化检测液为实现上述功能提供了可能。

本发明涉及一种以尿亚硝酸盐为主的尿亚硝酸生化检测液及其用途
                      背景技术
泌尿系统的细菌感染是一种多发疾病,特别是已婚妇女呈高发态势,为慢性发病过程,菌尿早期发现是极易根治的。
泌尿系统的细菌感染在治疗过程中的疗效同步监控。目前医院难以做到药物治疗的疗效同步跟踪动态检测。
另一方面,随着人们对于健康的关注和自觉意识的不断提高,希望能在临床以外诸如家庭、社区、旅游地等寻常条件下,对早期菌尿感染就能自我动态跟踪监控和检测,实现简便的自我筛选。提供预警信号,提高菌尿的早期检出率,继而通过积极的早期治疗提高治愈率。但是,迄今为止,对于菌尿自我检测,尚未有过适合临床外“民间使用”的方法和产品的相关报道。
因此,不论是临床还是家庭自我预防监测,需要一种快速、简便,准确的尿亚硝酸生化检测液和尿亚硝酸自我检测方法。本发明就满足了这一需求,为实现菌尿的家庭自我预防监测的临床预检提供了可能。

本发明提供了一种尿亚硝酸盐生化检测液,其包含0.35-0.45w/v%对氨基苯磺酸、0.2-0.3mgw/v%α-萘胺、20-40v/v%冰醋酸、1-2v/v%甲醇和蒸馏水。
所述尿亚硝酸盐生化检测液的配方如下所示

根据上述配方所配制的尿亚硝酸盐生化检测液与新鲜尿液产生的显色反应,若呈深樱红、樱红、浅樱红、浅粉红、微粉红、痕迹显示亚硝酸为0.005%、0.0025%、0.0003%、0.0002%、0.0001%、0.00005%的浓度。
根据尿亚硝酸盐生化检测液与新鲜尿液含不同浓度的亚硝酸盐反应所呈现的不同颜色,提供了渐变量化色谱以供检测者对照使用。提供尿亚硝酸盐生化检测显色结果的动态记录图,以及菌尿治疗过程中疗效的同步监控。
所述新鲜尿液—立即及时自测带有体温的尿液标本是新鲜的。
本说明书还提供传统经典的尿酸碱生化检测液,其配方如下

  蒸馏水  加至     100毫升
尿酸碱生化检测液与新鲜尿液检测之比为1∶1,可以在于玻片上进行反应。碱性尿呈红色,中性尿呈橙色,酸性尿呈黄色。并提供了尿酸碱生化检测液与新鲜尿液反应显色结果的渐变量化色谱以供检测者对照使用。提示鉴别酸性尿液形成结晶以及碱性尿液形成结晶的疾病。
本发明还提供检测尿亚硝酸盐浓度的尿亚硝酸盐生化检测液的说明书,其中包括尿亚硝酸盐自我检测方法、尿酸碱生化检测液、尿酸碱自我检测方法、试管和玻璃片。所用滴管的滴量为20±2滴/ml,“一滴”相当于约0.045-0.05ml。

与现有技术相比,本发明提供了快速、简便、可靠的尿亚硝酸盐生化检测液和尿亚硝酸盐自我检测方法。给菌尿临床前预检提供了可能。并与经典的尿亚硝酸盐生化定性检测结果实现了良好的吻合,因而具有很强的通用性和社会普及意义。
                      具体实施方式
                        实施例1
               尿亚硝酸盐生化检测液的制备
                        工艺要求:
α-萘胺及甲醇溶于冰醋酸之溶液中至全溶后对氨基苯磺酸溶于>60℃蒸馏水400毫升中至全溶冷却后倾入上液中,再加蒸馏水至需配量。
所得产品呈朱色溶液,PH为2±0.3,560nm吸光度为0.03~0.5A用棕色瓶避光密封常温保存。
                        实施例2
             尿亚硝酸盐生化检测液的灵敏度
将亚硝酸盐的水溶液制备成0.005%、0.0025%、0.0003%、0.0002%、0.0001%、0.00005%的浓度,容量为0.5毫升于6个标明浓度的试管中再滴加尿亚硝酸盐生化检测液各1滴观察:

以上实验显示,本发明检测液的灵敏度可达到0.0002-0.00005%左右。
                              实施例3
                          尿亚硝酸盐检测
于玻片上滴加新鲜尿液1滴,再于尿液上滴加尿亚硝酸盐生化检测液1滴观察:
呈深樱红、樱红、浅樱红、浅粉红、微粉红、痕迹显示亚硝酸为0.005%、0.0025%、0.0003%、0.0002%、0.0001%、0.00005%的浓度。均为阳性结果,提示菌尿感染的程度。
以上显色结果显示与经典方法检验结果呈现良好吻合。

Claims (5)

1.一种尿亚硝酸盐生化检测液其包含0.35-0.45w/v%对氨基苯磺酸、0.2-0.3mgw/v%α-萘胺、20-40v/v%冰醋酸、1-2v/v%甲醇和蒸馏水。
2.说明书的内容包括含尿亚硝酸盐的尿液与权利要求1所述的尿亚硝酸盐生化检测液用量之比为1∶1。
盛尿亚硝酸盐生化检测液与尿之检测反应观察:呈深樱红、樱红、浅樱红、浅粉红、微粉红、痕迹显示亚硝酸为0.005%、0.0025%、0.0003%、0.0002%、0.0001%、0.00005%的不同浓度。
3.根据权利要求2所述说明书内容还包括:渐变量化色谱;连续不同检测时段检测显色结果的动态曲线记录图。
4.根据权利要求2所述的说明书内容还包括尿酸碱生化检测液及尿酸碱自我检测技术、试管及玻片,塑瓶凹槽剪口处其滴量为20±2滴/毫升。
5.根据权利要求1所述尿亚硝酸盐生化检测液用于尿亚硝酸盐显色反应的定性测定的用途。主要能在家庭中进行一日多次地自我跟踪动态检测,显示尿路感染的预警信号及鉴别致病细菌;提高菌尿早期检出率,以及菌尿患者的治疗过程中疗效的同步监控。
