现有技术说明

发明内容
这里揭示的本发明提供一种应用NIR谱测量的无创伤性体内方法,按照化学与结构特性对组织分类。组织分类模型利用一群示例个体的NIR谱吸收测量建立。谱测量经评估,可识别最能代表组织类型差别的有关特征。为了增强有关特征并提取这些代表组织内变化的特征,应用了统计分析技术。分类例行程序确定的最佳模型,根据组织类型的差别在示例群体内规定等级,使等级内的变化比等级间的变化小。判断规则根据各个体组织的结构与化学特性,对代表性群体中各成员指定等级成员资格。

生物组织的各种特征可用NIR光谱法测量,因为这些特征通常在NIR波长区(700~2500nm)内具有其吸收与散射特性造成的唯一特征谱。这些特征中许多按组织类型而变化,因而便于将组织分为独立的类型。可用NIR吸收与散射图形测量的有用特征,包括但不限于脂肪组织厚度、组织水合作用、蛋白质吸收幅值、组织散射特性、皮肤厚度、温度相关效应、老化相关效应、性别相关谱特性、光径长度估值、组织中血液容量分数、环境影响相关谱特性与血球比容度。按组织类型变化的特征可利用统计技术同组织样本谱隔离开来,然后相应地对样本分类。
组织分类模型的研制

首先获取一群示例受验者的NIR测量值(11)。运用众所周知所述的方法为本方法准备好个体。在动物受验者情况下,可能要对受验者作麻醉或使其固定不动。还希望制备被观测的样本表面,以尽量减小表面不规则引起的谱干扰,如要求除掉动物受验者的表面毛发。使用检测近红外波长区(700~2500nm)光的分光仪收集NIR测量值。NIR测量值可用各种单位表达,其中有反射度单位和负的基数为10的对数反射度单位。本发明方法可以应用各种市售的NIR分光仪,如Nicolet Magna-IR 760或Perstorp Analytical NIRS5000,但为了更全面地描述适合本发明实施的仪器,可参照本申请的原始申请中的图19,该申请是S.Malin,T.Ruchti于1999年7月22日提交的美国专利申请连续号09/359,191,题为“An Intelligent System for NoninvasiveBlood Analyte Prediction”。样本测量值或组织吸收谱是吸收值矢量m∈RN,从属于一组跨越近红外波长区(700~2500nm)的N个波长λ∈RN。
特征选择
谱特征选择(12)可以包括定性评估样本谱,以识别从中可找出随组织类型变化的谱区或有关特征。在波长区边界处截去光谱,这里呈现的组织特定变化可减少数据组的复杂性与维数。通过减少数据组的维数,实现的数据减少可提高分类模型的预测能力。通过把样本谱与例如蛋白质或脂肪的已知谱吸收图形比较,有助于特征选择。在识别出一个或多个含组织特定信息的波长区之后,分类模型只使用这些选择的区域。
特征增强
一旦识别出有关特征,就对谱测量值应用特征增强法(13)。虽然NIR反射测量与组织样本的化学和结构特征相关,但是相关是非线性的,大部分原因在于谱测量值被光散射冲淡了(见P.Geladi,D.MacDougall,H.Martens,Linearization and Scatter Correction for Near-infrared ReflectanceSpectra of Meat,Applied Spectroscopy.Vol.39,pp.491-500(1985))。应用诸如多重散射校正(MSC)技术的特征增强,可校正目标组织溶剂的散射特性变化而造成的干涉谱变化。在本申请的原始申请中(S.Malin,T.Ruchti于1999年7月22日提交的题为“An Intelligent System for NoninvasiveBlood Analyte Prediction”的美国专利申请连续号09/359,191),揭示了一种应用MSC的特征增强法。各样本的散射评估方法是转动样本谱,使之按下式与参考谱

其中a与b是斜率和截距,e代表拟合误差。然后,通过下式校正该光谱:
其中x是处理过的吸收谱。这样,各样本的谱均实现了标准化,所有样本都相对于参考谱归一化(还可见B.Wise and N.Gallagher,PLS Toolbox2.0,Eigenvector Research,Inc.Manson(1998)):当有关特征是组织中吸收物种造成的或者组织特定参数具有不同谱形状时,也可对有关特征附近的波长区应用MSC。校正邻区散射会使不校正波长区中的特性形状被放大,这样就有效增强了目标化学与结构特征。
特征提取
特征增强后,用特征提取法提取那些有关分类的特征。特征提取可用能增强供说明样本测量的质量或状态的任何数学变换实现,旨在简明地表示组织测量部位的化学与结构特性和生理状态。
对特征增强的数据应用较佳的特征提取法,即主成分分析法(PCA)(14)。PCA把多维数据分解成排序的因数,表示该数据组内潜在的变化(见R.Johnsonand D.Wichern,Applied Multivariate Statistical Analysis,3rd.ed.;Prentice-Hall,New Jersey(1992))。这些因素称为主成分。第一主成分代表数据中最主要的变化,第二主成分代表第二重要的变化,第三主成分代表第三重要的变化,依次类推。主成分或主因数代表数据中抽象或简明特征。由因数表示的抽象特征不一定具有与物理系统相关的特定说明。具体而言,主成分分析记分是有用的特征,虽然并不总是明白其物理解释。然而,简明特征可通过事先了解该样本而得出,能与物理现象直接相关。例如,蛋白质与脂肪的已知吸收特征谱可用来测定它们对组织谱吸收度的贡献。测得的贡献一般用作某一特征,通过一单值代表潜在的变量。
事先了解组织样本有助于选择与该组织的物理现象直接相关的特征。特征可通过下式直接用散射校正的测量值导出的矢量z∈RM表示:
其中f:Rn→Rm是测量空间到特征空间的映射。分解f(·)得出特定的变化,fi(·):Rn→Rm i,用于测定特定的特征。维数Mi指明第i个特征是标量还是矢量,而所有特征和起来就是矢量z。通过减少复杂谱变化的维数而得出的特征,很有利于组织分类处理。因此,特征提取允许特征标识代表直接归因于物理现象的变化并排除所有其它变化,从而把高维数的难题进一步减为少量便于管理的维数。
因数选择
通过目察因数分析得出的因数负载和记分,选择与分类模型有关的因数(15)。如上所述,对表示为数据矩阵的散射校正的数据组作PCA,把数据变化纳入因数或主成分。对于代表组织特定特征的因数或主成分,把对应的因数记分按组织类型分组,而对应的因数负载指示对组织特定变化负责的特征。在因数记分表示成散射曲线时,经简单目察就能看出有关的因数和主成分。
分类校正
对组织特定因数的记分应用有判断规则的分类程序(即Fisher线性判别式分析)(16)。分类程序基于某种判别函数,在最大化时,找出代表群体分类的最佳模型(见R.Duda与P.Hart,Pattern Classification and Scene Analysis,JohnWiley and Sons,New York(1973))。Fisher线性判别式分析(LDA)是用于该分类模型的较佳程序。LDA在两群之间找出使等级间变化最大而等级内变化最小的线。LDA判别函数为
其中w是定向单元矢量,Sb是等级间散射矩阵,Sw为等级内散射矩阵。矢量w选成使等级间变化与等级内变化之比最大。把样本投射到w上,进一步减小该问题的维数(见R.Duda与P.Hart,Pattern Classification and SceneAnalysis,John Wiley and Sons,New York(1973))。应用该判别函数,根据前M个记分对样本分类。在下述较佳实现方式中,M=3。本领域技术人员知道还可用其它分类法。
判断规则
建立的判断规则(17)用于确定样本的所属等级。判断规则用来确定样本的等级成员资格的判别式是,样本在w上的投射(标量),是大于还是小于这两群均值的平均值(见R.Johnson and D.Wichern,Applied MultivariateStatistical Analysis,3rd.ed.;Prentice-Hall,New Jersey(1992))。标量L与等级界限I作比较,若L>
L,样本定为群1(18),否则定为群2(18)。
组织样本的分类
在本申请的原始申请中(S.Malin,T.Ruchti在1999年7月22日提交的题为“An Intelligent System for Noninvasive Blood Analyte Prediction”的美国专利申请序号09/359,191),详述了揭示的分类模型对实际组织样本分类的实施情况。一般而言,组织分类的操作步骤是:
1.NIR测量
2.特征提取
3.图形分类
4.等级成员资格指定。
对新样本获取一组吸收度值,这些值属于一组跨越近红外(700~2500nm)的波长。为了削弱光散射、噪声与仪器变化引起的误差而不影响有关信号,对光谱测量值作预处理。前述的特征提取包括应用任一种能隔离有利于解释的样本特定状态或质量的数学变换。通过应用一种或多种多重分析法,如主成分分析、局部最小平方或人工神经网络,完成特征提取。提取的特征与一组预定等级数据比较并测出相似性。判断规则应用判别函数指定等级成员资格。
实验结果
引言
在本发明一较佳实施例中,这里揭示的算法适用于把一群生长激素转基因鼠与一群非转基因控制鼠分离开来。所描述的本实施例专用于转基因鼠。然而,本发明适用于对可作精确光谱测量的其它物种的其它组织分类。
生长激素转基因鼠具有在启动基因控制下插入其基因组的外来生长激素基因。控制鼠和转基因鼠是同窝出生的,因而除了转基因个体里存在生长激素基因外,其它与生长激素鼠在遗传上相同。外来生长激素基因的存在使受验动物产生过量的生长激素。与非转基因鼠相比,转基因生长激素鼠呈现众多从属于过量产生生长激素的显型效应,如骨架庞大、内脏器官过大、体重增加和各种特性的组织异常(见Wolf等人,还可见R.Wanke,E.Wolf,W.Hermanns,S.Folger,T.Buchmüller,G.Brem:The GH-Transgenic Mouseas an Experimental model for Growth Research:Clinical and PathologicalStudies,Hormone Research.vol.37,pp.74-87(1992))。
方法
运用漫反射光谱法对一群14只鼠作NIR测量,这群鼠包括8只生长激素鼠和6只控制鼠。从每只鼠腹部收集四次同样的观测结果,每天一次,历时三天。测量前,用可注射的麻醉剂麻醉鼠,并用动物剪刀修整,以防受验体毛发影响光谱测量。三天内,每天从各群动物中选择动物,按随机顺序作观测。该测量应用前述的分光仪装置。
应用众所周知的统计方法,根据原始光谱数据计算两群受验体中每一群的平均光谱。图2示出生长激素鼠(21)与控制鼠(22)的平均谱。目察光谱(21和22)示出两波长区中有发散。第一个有关特征(23)表明控制鼠在1100~1350nm波长区中有一吸收带。第二个有关特征(24)表明在控制鼠光谱中的1600~1880nm波长区内有一吸收带。与控制鼠不同,转基因受验体没有这种吸收带。
图3示出在两个标识的波长区内(有关特征),两群鼠的平均谱(31与32)与纯动物脂肪谱样本(33)的比较结果。动物脂肪谱清楚地示出1100~1350nm波长区的第一吸收带(34)和1600~1880nm波长区的第二(35)与第三(36)吸收带。控制群的平均谱的第一吸收带(37)和第二与第三吸收带(38、39)反映了脂肪谱的诸吸收带(34、35、36)。同时,转基因GH群的平均谱没有类似的吸收带,说明这两群鼠的光谱差异是组织的脂肪成分差异引起的。
为增强有关特征,应用了前述的多重散射校正法(MSC)。图4示出散射校正的谱数据曲线。由该散射校正谱可见,把观察的吸收带表示成该群样本内的变化度。大部分样本不呈现明显的吸收带,而其它样本有显著的吸收带。
在1100~1350nm和1600~1850nm波长区内,对散射校正的数据作主成分分析。目察得出的因数记分示出第二主成分很重要。如图5所示,第二主成分的因数记分整齐地分成对应于两群鼠的两组(51、52)。图6曲线清楚地示出动物脂肪谱的吸收带与主成分2负载的相关性,显然,两鼠群间的分离是组织脂肪成分。
为定义一合适的分类模型,对前三个主要成分记分作Fisher线性判别分析,使等级内变化最小,等级间变化最大。
在证实分类校准中使用“略去一”交叉证实法,即尤其适用于少量数据组的校准程序。对这种分析,应用了迭代处理,其中把一个样本排除出数据组,用其余样本作校准,并用得到的校准预测排除在外的样本。该过程重复进行,直到所有的样本都被略去和预测过。
结果
校准程序的结果列于表1。

共检查171个光谱样本,GH转基因个体谱样本99个,控制样本72个。在这些样本中,99个或100%转基因样本分类正确;54个或72%控制样本分类正确,因而该算法对每群样本正确分类的精度为90%。而鼠群按其组织的化学与结构差别分类,不凭借入侵活组织检查法。数据便于一个人收集,可以发展在线分类,与常规法要求数天或几星期相比,只稍几分钟。
讨论
虽然该实施例针对按组织结构与化学差异对鼠进行分类,但是本领域技术人员显然明白,利用NIR测量指示组织中的化学、结构与生理变化可应用于其它领域,如无创伤性血液分析物预测、伤口愈合研究与皮肤病治疗。
这里描述的等级定义限于一群特定的受验体,不能通用于所有的受验体。适合所有受验体的分类模型包括有足够数量的样本,对特定组织参数模拟成群变化的总范围。虽然本实施例应用了特定数量的特征、等级、判断规则和分类模型,但是本发明可在每一结构中用任意数量的分类组织样本。实验结果证明了本发明的有效性与有益性,但是得出的精度直接取决于谱测量精度。通过改进噪声电平与分光仪的分辨度,可进一步提高结果精度。
虽然这里参照某些实施例描述了本发明,但是本领域技术人员显然明白,可用其它应用代替这里提出的应用而不违背本发明的精神与范围,因此本发明只受制于下面所包括的诸权利要求。
Claims (49)
1.一种为组织样本分类研制分类模型的方法,其特征在于包括以下步骤:
提供一群示例受验体的一组光谱吸收测量值;
在所述光谱测量值内选择一个或多个有关特征,其中可发现按组织类型的变化;
增强所述有关特征;
提取至少一个与分类有关的有关特征;
选择与所述样本的结构与化学特性相关的所述有关特征的因数;
根据结构与状态相似性定义所述组织样本的等级,其中级内变化比级间变化小;和
指定等级成员资格。
2.如权利要求1所述的方法,其特征在于把每个所述测得的吸收谱表示为吸收值矢量m∈RN,它从属于一组跨越700~2500nm波长区的N个波长λ∈RN。
3.如权利要求1所述的方法,其特征在于所述特征选择步骤还包括步骤:
目察所述样本谱;
把所述样本谱与已知谱吸收图形作比较;
根据所述样本谱与所述已知谱吸收图形之间观察到的相似形,选择所述有关特征;和
在所述选择的有关特征的边界处截去所述样本谱。
4.如权利要求1所述的方法,其特征在于所述特征增强步骤应用对光散射校正所述样本谱的手段。
5.如权利要求4所述的方法,其特征在于所述校正所述谱的手段包括多重散射校正。
6.如权利要求5所述的方法,其特征在于评估每个所述样本谱的散射的方法是按下式将所述样本谱转到参考谱

应用判断规则指定等级成员资格。
18.如权利要求17所述的方法,其特征在于所述特征提取步骤包括任一种为解释而增强样本测量质量或状态的数学变换,以简明地表示组织测量部位的结构特性与生理状态,其中用得到的一组特征分类受验体。
19.如权利要求17所述的方法,其特征在于所述分类模型包括以预定等级确定一组相似性测量的手段。
20.如权利要求17所述的方法,其特征在于所述判断规则包括根据一组判断引擎计算的测量值指定等级成员资格的手段。
21.如权利要求19所述的方法,其特征在于还包括步骤:
提供一分类系统,所述系统假定所述等级都各不相同,并迫使各测量值被定为单一等级。
22.如权利要求21所述的方法,其特征在于所述特征表示为矢量z∈RM,并由下式确定:

增强所述有关特征;
提取至少一个与分类有关的有关特征;
选择与所述样本的结构与化学特性相关的所述有关特征的因数;
按结构与状态相似性定义所述组织样本的等级,其中级内变化比级间变化小;和

27.如权利要求26所述的方法,其特征在于所述转基因鼠是外来生长激素转基因鼠。

目察所述样本谱;
将所述样本谱与动物脂肪谱吸收图作比较;

提取对应于组织特定变化的特征;
图形分类,其中分类模型将所述提取的特征与一组示例测量值作比较;和
应用判断规则指定等级成员资格。
39.如权利要求38所述的方法,其特征在于所述转基因鼠是外来生长激素转基因鼠。
