本发明涉及一种葡萄球菌快速分离鉴定培养基。主要解决现有的用于葡萄球菌分离培养的血平板易生杂菌、检验程序复杂的问题。技术方案:一种葡萄球菌快速分离鉴定培养基,含有的成分及重量百分比为,蛋白胨0.85—1.50、酵母浸膏0.10—0.25、氯化钠7.30—7.60、甘露醇0.70—1.80、明胶2.80—3.20、磷酸氢二钾0.40—0.60、琼脂1.50—2.00、蒸馏水100。本发明不易生长杂菌、检验程序简单。
Description
本发明涉及葡萄球菌快速分离鉴定培养基。
现在我国临床、预防、药检及商检等机构对金黄色葡萄球菌的检验方法为先培菌、再分离培养于血平板,然后做生化反应和血浆凝固酶等繁锁复杂的试验。用血平板培养葡萄球菌容易生长杂菌,发生变形菌弥漫生长而无法观察的现象,并且用血平板做鉴定试验程序复杂,检测不便,尤其不适应基层检验工作的需要。
本发明的目的在于提供一种培养葡萄球菌不易生长杂菌、检验程序简单的葡萄球菌快速分离鉴定培养基。
本发明的技术方案;一种葡萄球菌快速分离鉴定培养基,其含有的成分及重量百分比为,蛋白胨0.85-1.50、酵母浸膏0.10-0.25、氯化钠7.30-7.60、甘露醇0.70-1.80、明胶2.80-3.20、磷酸氢二钾0.40-0.60、琼脂1.50-2.00、蒸馏水100。
本发明的有益效果:用本发明分离培养葡萄球菌与血平板培养葡萄球菌相比,不易生杂菌。做实验,将105株金黄色葡萄球菌分别接种于本发明和血平板,经培养有32株葡萄球菌在血平板上因变形菌弥漫生长而无法观察,而在本发明生长的葡萄球菌状况良好,无任何杂菌生长。对66株葡萄球菌重复性试验结果表明,本发明对葡萄球菌分离鉴定重复性好,本发明于4℃冰箱中保存,6个月后质量稳定,使用效果良好。用本发明从接种到初步鉴定报告葡萄球菌检查结果在43小时内即可完成,有助于临床感染及食物中毒病原菌的快速诊断,应用方便价格低廉,改变了过去分离鉴定需时间长、试剂消耗高、程序繁杂、结果不够准确的弊端。
下面叙述本发明的实施例。
制备本发明时,蛋白胨可选用牛肉胨或多胨。还可加入重量百分比不大于0.2的乳糖。取牛肉胨10克、酵母浸膏2.5克、氯化钠75克、明胶30克、甘露醇10克、磷酸氢二钾5克、乳糖2克、琼脂15克,加蒸馏水1000ml,将前述物质溶于水,溶解后调PH7.0±0.1,分装三角烧瓶中121℃灭菌150分钟,冷却后冰箱保存备用。
当需用本发明检验时,将本发明表面充分干燥,以白金耳划线接种置37℃,36-43小时培养后观察菌落形态、耐盐性色素形成、甘露醇发酵和明胶液化。观察甘露醇发酵时,直接滴0.04%BTB于菌落,立即观察结果菌落及周围呈黄色为阳性;观察明胶液化时,需滴加20%磺胺硫酸水溶液或硫酸饱和溶液,2分钟内,在白浊培养基上的菌落周围呈现明显透明环为阳性。

1、一种葡萄球菌快速分离鉴定培养基,其含有的成分及重量百分比为,蛋白胨0.85-1.50、酵母浸膏0.10-0.25、氯化钠7.30-7.60、甘露醇0.70-1.80、明胶2.80-3.20、磷酸氢二钾0.40-0.60、琼脂1.50-2.00、蒸馏水100。
2、根据权利要求1所述的葡萄球菌快速分离鉴定培养基,其特征在于:蛋白胨可以为牛肉胨或多胨。
3、根据权利要求1或2所述的葡萄球菌快速分离鉴定培养基,其特征在于:还可加入重量百分比不大于0.2的乳糖。
