Description

Yian Ning pill for treating coronary heart disease and preparation method thereof
Technical Field
The invention belongs to the technical field of medicine preparation, and relates to Yian Ning pills for treating coronary heart disease and a preparation method thereof.
Background
The honeyed pill is prepared from honey serving as an adhesive, is a dosage form which is most widely applied in clinical application of traditional Chinese medicine, is compared with decoction, is slowly decomposed in intestinal tracts after being taken by patients, gradually releases drug effects, has lasting effect, and can be slowly absorbed by patients aiming at drugs with high toxicity or high irritation, so that adverse reactions can be effectively reduced. Although the production equipment and the process of the honeyed pill are relatively simple, the quality of the honeyed pill is difficult to control in the preparation process.
The Chinese patent CN100339111C discloses a medicine for curing coronary heart disease and a preparation method thereof, which is prepared by taking American ginseng, dendrobium, cordyceps, schisandra chinensis, chicken's gizzard-skin, tortoise plastron, turtle shell, pilose antler, saffron, pseudo-ginseng, red sage root, sea horse, ganoderma lucidum and snake gall as raw materials, has obvious curing effect on coronary heart disease and can obviously relieve or eliminate other complications. However, the medicine (Yi an Ning pill) has insufficient medicine powder viscosity, the extract amount obtained by decocting the medicine materials in water and concentrating the decoction is also small, and the pill preparation process has great difficulty, so that the yield is required to be further improved.
In the preparation of traditional Chinese medicine pill, refined honey is required to be added as adhesive when the medicine powder is not enough in viscosity. Except selecting honey, the refined honey is refined, the water content of the refined honey is controlled, so that the refined honey has moderate viscosity, and the method is a key link for preparing the traditional Chinese medicine pill. Because honey contains about 25% moisture and contains impurities such as waxes and starches. These impurities are not removed and the wax floats on the surface of the pellet, forming an extremely thin wax layer which can lead to cracking of the pellet surface after storage for a period of time. When the honey contains more starch impurities, the finished product of the pill can be dry and hard. Therefore, honey must be refined before use to remove impurities, destroy biological enzymes, kill microorganisms, and properly reduce water content to enhance adhesion. If the honey refining link is not well controlled, the finished pill products can be mildewed, dried and hard, cracked and the like during storage, and the quality of the pills is seriously affected.
The refined honey generally used in the preparation of traditional Chinese medicine pills is divided into three types of tender honey (with the water content of 18-20%), medium honey (with the water content of 14-16%) and old honey (with the water content of less than 10%) according to different refining degrees, wherein the refining degree of honey is selected according to the adhesive property of traditional Chinese medicine powder in a traditional Chinese medicine prescription, the water content of the powder, the viscosity of an extract and the proportion of the powder to the extract, and the refining temperature and time are used for refining the tender honey, the medium honey or the old honey.
In order to improve the quality and the yield of the Yi-an-ning pill, the preparation formability of the Yi-an-ning pill and the yield of the pill are evaluated and selected to select a proper honey refining scheme, and the dissolution condition of the finished product are further verified.

Preparation of test solution: taking three parts of Yianning pills prepared in each example and respectively measuring the contents of ginsenoside Rb 1 and notoginsenoside R 1, precisely weighing 18 pills each, respectively placing the pills in three different small cups, respectively placing 18 granules in each cup, and carrying out ultrasonic degassing for 30 minutes before using the distilled water, wherein the volume of a dissolution medium is 250mL of purified water, and the temperature of the dissolution medium is as follows: the rotation speed is 100 rpm at 37 ℃. The sampling time is set to 15, 30, 60, 120, 180, 240 and 360 minutes, the sampling amount is 2mL each time, and the same temperature and the same amount of release medium are added immediately after sampling. The sample solution is centrifuged at high speed at different sampling times, the supernatant is taken and filtered by a microporous filter membrane of 0.45 microns, and the subsequent filtrate is taken as the sample solution.
Assay: respectively precisely sucking 20 μl of blank sample (purified water), reference solution and test solution, and injecting into liquid chromatograph for measurement (wherein chromatographic results of sample solution 240 and 360 min of YIANNING pill prepared in examples 3 and 4 are shown in fig. 3 and 4), wherein the liquid chromatograph conditions are: venusil XBP C18 chromatographic column (4.6X105 mm,5 μm) was used, with octadecylsilane chemically bonded silica as filler; acetonitrile as mobile phase A and water as mobile phase B, and performing gradient elution according to the procedure of table 1; the detection wavelength is 203nm; the theoretical plate number is not lower than 4000 calculated according to the peak of notoginsenoside R 1, and the ginsenoside Rb 1 peak is not lower than 5000.
TABLE 1 elution procedure
Calculating according to an external standard method to obtain the contents (mg/mL) of ginsenoside Rb1 and notoginsenoside R 1 in each 1mL of the sample solution sampled at each time point; the dissolution rate of ginsenoside Rb 1 and the dissolution rate of notoginsenoside R 1 at each time point of the Yianning pill are respectively calculated according to the following formula:
Ginsenoside Rb 1 dissolution rate= (ginsenoside Rb 1 content×250 in each 1mL of sample solution sampled at each time point) ×100/(18 pill Yi Anning pill weight×Yian ning pill ginsenoside Rb 1 content (mg/g));
The dissolution rate of notoginsenoside R 1 = (the content of notoginsenoside R 1 in each 1mL of sample solution sampled at each time point is multiplied by 250) multiplied by 100/(18 pills of Yianning pills weight multiplied by the content of notoginsenoside R 1 in Yianning pills (mg/g)).
The dissolution curve was drawn with the average value of the dissolution rates measured in triplicate as ordinate and time as abscissa.
The moisture content of the refined honey prepared in examples 1-6, and the pill-making conditions and pill quality conditions of the Yianning pills are shown in Table 2.
Table 2 results of the pelleting conditions and pellet quality measurements of the examples
As can be seen from the results in Table 2, for the preparation forming process of YI AN NING WAN, when the honey refining temperature is 115-120deg.C, that is, the water content of the refined honey is 10-14%, the key process conditions such as soft material preparation and pill rubbing are good, and the qualification rate of pill finished products is high. If the water content of the refined honey is too high, the viscosity is insufficient, the soft material is too soft, so that the extrusion is not smooth, the extrusion is easy to break, and even the extrusion cannot be performed for preparing pills; the water content of the refined honey is too low, the viscosity is increased, the viscosity and hardness of the soft material are too high, and the pill cannot be rubbed. In addition, when the water content is 10.0%, although the pill making condition meets the requirement, the dissolving time of the prepared Yianning pill is close to the upper limit of 120 minutes specified under the four-part general rule 0108 pill item of the Chinese pharmacopoeia 2020, and after the pill is stored for a period of time, the risk of unqualified dissolving time limit exists, so that in the preparation and forming process of the Yianning pill, the honey refining temperature is preferably 115-117 ℃, and the honey refining water content is preferably 12-14%.
As can also be seen from the dissolution curves of fig. 5 and 6, the produced pellets dissolve slowly when the moisture content of refined honey is 10.0%, which is related to the low moisture content of refined honey and the strong adhesiveness; the greater the viscosity of the refined honey, the greater the hardness of the prepared pill, the longer the dissolution time and the slower the dissolution speed. The water content of refined honey is high, the dissolving time is short, and the dissolution of pills is quickened.
