技术领域

背景

使用高度特异性且有效的HIV Rev依赖性指示细胞来筛选某些天然植物和中药(TCM)的抗HIV活性。本文已鉴定出多种植物提取物和中药成分具有抗HIV活性。同时发现这些TCM成分以剂量依赖的方式通过不同的机制来抑制HIV感染。鉴于HIV的高突变率,阻断HIV在多个步骤复制以预防病毒耐药性的发展变得非常重要。本文描述了用于治疗HIV感染的天然草本成分和TCM的组合、制备方法和使用方法。

甘薯提取物(0.5g/mL,在H2O中)也用细胞培养基进行连续稀释(例如1:10、1:50和1:1250),并用于处理Rev-A3R5-GFP细胞以抑制HIV-1感染。我们观察到了甘薯的剂量依赖性地抑制了HIV-1(参见图4D-4F)。甘薯在高剂量(1:10和1:50)下抑制了HIV-1感染,但在低剂量(1:1250稀释)下未能抑制。
图5A-5L展示了在使用HIV-1Rev依赖型指示细胞筛选中,草本中药抗HIV成分的结果。植物中药成分从黄芪(Radix Astragali)、甘草(Radix Glycyrrhizae)、灵芝(Ganoderma Lucidum Seu Japonicum)、山楂(Fructus Crataegi)、金银花(FlosLonicerae)、莲子心(Plumula Nelumbinis)、陈皮(Pericarpium Citri Reticulatae)、菊花(Flos Chrysanthemi)、竹叶(Folium Bambusae)或荷叶(Folium Nelumbinis)中用水提取,然后用于处理Rev-A3R5-GFP细胞,处理时间为持续1小时。随后,细胞在每种提取物的存在下,感染HIV-1(NL4-3)2小时之久。感染后,细胞经过洗涤并在持续存在每种提取物的条件下培养3天。在细胞感染2天后对GFP表达进行定量。在流式细胞术期间添加碘化丙啶(P.I.)以染色死细胞。GFP表达仅对活细胞种群进行定量分析,结果如表7所总结。

图6A-6F展示了来自不同植物中草本中药提取物的不同抗HIV机制,显示了天然植物和中药提取物对HIV-1进入和早期感染步骤的影响。Rev-A3R5-GFP细胞用自莲子心、酸樱桃的提取物或包含这两种提取物的组合("Comb")处理1小时后,在这些提取物的存在下感染HIV-1 2小时。感染后,细胞被洗涤,然后在没有提取物的情况下培养3天。在感染后的第2天,使用碘化丙啶(P.I.)进行流式细胞仪染色以标记死细胞。GFP表达仅针对存活细胞群体进行量化,结果如表8所总结。

使用Rev依赖指示细胞筛选抗HIV化合物的示例协议
材料:
A.莲子心提取物:将1克溶解在40毫升蒸馏水中。用沸水浸泡过夜,自然冷却。
B.新鲜酸樱桃(来自COSTCO,由Exporte par Farm Co.S.A,智利进口)。将25克搅拌均匀在50毫升蒸馏水中。
C.美国甘薯根:将25克搅拌均匀在50毫升蒸馏水中。
D.HIV-1Rev依赖性细胞Rev-A3R5-GFP。
E.RPMI-1640细胞培养基+10%FCS+Pen/Strep。
F.HIV-1(NL4-3)病毒。
实验程序:
样品稀释的准备:将1微量离心机中,以最大速度(14,000转/分钟)离心1分钟以沉淀残渣。上清液被转移到一个新的1.5毫升管中。
A=上述制备的莲子心原始溶液
A1=1:5稀释(200微升A+800无血清的RPMI1640=1000微升)
A2=1:25稀释(200微升A1+800无血清的RPMI1640=1000微升)
A3=1:125稀释(200微升A2+800无血清的RPMI1640=1000微升)
对B(新鲜酸樱桃提取物的原始溶液)和C(甘薯根的原始溶液)进行类似的稀释。
细胞的预处理:计数细胞,重新悬浮在2x106/毫升。按照下表(表1)的顺序,将90微升分别加入每个试管中。根据表1的列表,向A3加入10微升的A,向B3加入10微升的B,或向C3加入10微升的C。
在CO2细胞培养孵育箱中在37℃下孵育30分钟。
通过加入200微升NL4-3病毒来感染细胞。向A至C3添加额外的20微升药物,感染细胞2小时。
洗涤,然后重悬于1mL培养基中,每管中加入药物。
在37℃下培养感染的细胞72小时。
在48小时和72小时时进行流式细胞术分析。在流式细胞术中添加碘化丙啶(P.I.)(2μg/毫升)以染色死细胞。只分析活细胞的GFP表达。

Claims (11)
