Background
Type 2 diabetes mellitus is a common chronic disease of modern people, mainly because the sensitivity of cells to insulin is reduced, impedance is generated, blood sugar cannot enter the cells, pancreas can enter the cells in order to allow blood sugar to enter the cells, and more insulin is secreted, but insulin resistance is gradually generated over time, a large amount of glucose remains in blood to cause hyperglycemia, diabetes mellitus is changed, common pathogenic factors comprise obesity, diet and insufficient exercise amount besides inheritance, particularly the phenomenon of obesity of modern people is more and more popular, and metabolic syndrome such as hypertension and hyperlipidemia is also possible besides the possibility of the Type-two diabetes mellitus.
There is no cure for type ii diabetes, but weight loss, diet and exercise can help control the disease, but if diet and exercise still cannot control blood glucose, there is a fear that treatment with diabetes medication or insulin is still needed.
The balsam pear school name Momordica charantia, also known as the melon, the brocade litchi, the melon and the grape, belongs to the Cucurbitaceae (cuurbitaceae) and the Momordica (Momordica), is a daily edible vegetable for Asians, and the producing area comprises Taiwan, the south of the mainland of China, southeast Asia and the Japanese Chongman. The annual output of the Taiwan balsam pear in China is about 3,000 metric tons, the variety is various, and particularly the Taiwan balsam pear is one of the most common fruits and vegetables in Taiwan, and the Taiwan balsam pear is rich in nutrition and contains a plurality of beneficial organic components such as amino acids and the like required by human bodies. Compared with common bitter gourd, the bitter gourd has higher nutrient content, especially 2-3 times higher saponin content and 2-3 times higher vitamin content than common bitter gourd. Therefore, researches show that the Momordica Charantia has the medicinal and health-care functions of reducing blood sugar, reducing blood fat, resisting tumor, preventing osteoporosis, regulating endocrine, resisting oxidation, resisting bacteria, improving human immunity and the like.
Therefore, development and application research of Momordica charantia are more and more important, and various industries are not urgent in need of developing health-care food containing Momordica charantia, which can be used for assisting type II diabetics with medicines so as to make blood sugar control of patients more effective.
Disclosure of Invention
In view of the above, the present invention aims to provide a blood sugar regulating health care composition, which can help a patient with type II diabetes to stably reduce blood sugar by using the combination of the effective components in the mountain balsam pear pulp and mountain balsam pear seed and the effective components of Chinese herbal medicines, and can also regulate the comprehensive indexes of the patient with hyperlipidemia, high cholesterol, abnormal liver metabolic enzymes and abnormal uric acid.
Specifically, the invention provides a blood sugar regulating health care composition, which is characterized by at least comprising the following components: 20-60 wt% of a mountain balsam pear component; 5-20wt% of mountain balsam pear seed component; 1-20wt% of cinnamon component; 1-15 wt% of a momordica grosvenori component; 1-10wt% of licorice component; wherein the content of the momordica saponins in the mountain balsam pear component is 10 weight percent; the content of mogroside V in the momordica grosvenori component is 5wt%; the liquorice flavonoid content in the liquorice component is 2wt%; and the cinnamaldehyde content in the cinnamon component is 2% by weight.
In an embodiment of the present invention, the mountain balsam pear component at least includes mountain balsam pear extract, and the mountain balsam pear extract is prepared as follows: a. cleaning fresh Momordica Charantia, removing seeds, slicing, and oven drying until the water content is below 5wt%; b. crushing the dried Momordica Charantia slices with a 60-mesh sieve by using a crusher to obtain Momordica Charantia powder, and mixing the Momordica Charantia powder with 50-95 vt% ethanol water solution for extraction for 60-180 minutes to obtain Momordica Charantia extract; c. concentrating the Momordica Charantia extract under reduced pressure at 50deg.C and under-0.06 Mpa to obtain Momordica Charantia concentrate; d. mixing the mountain balsam pear concentrated solution and embedding materials for embedding, drying after embedding, and crushing by using a 100-mesh screen by using a crusher to obtain the mountain balsam pear extract.
In an embodiment of the present invention, in the step b, the weight ratio of the mountain balsam pear powder to the ethanol aqueous solution is 1:10 to 1: 30.
In one embodiment of the present invention, in the step c, the weight ratio of the mountain balsam pear concentrate to the embedding material is 1:1 to 1: between 10.
In an embodiment of the present invention, the mountain bitter gourd ingredient further comprises the mountain bitter gourd powder obtained by the step b.
In an embodiment of the present invention, the mountain balsam pear seed component at least includes mountain balsam pear seed extract, and the manufacturing method of the mountain balsam pear seed extract is as follows: a. crushing the mountain kusnezoff seed by using a 60-mesh sieve by using a crusher to obtain mountain kusnezoff seed powder, and performing supercritical extraction under the conditions that the extraction pressure is 35Mpa and the extraction temperature is 50 ℃, and removing oil content of the mountain kusnezoff seed powder to ensure that the oil content of the mountain kusnezoff seed powder is below 1wt%; b. extracting the mountain bitter gourd seed powder with pure water for 60-120 minutes after oil removal to obtain mountain bitter gourd seed extract; c. concentrating under reduced pressure at 50deg.C under-0.06 Mpa, and drying to obtain dried extract.
In one embodiment of the present invention, in the step b, the weight ratio of the mountain kusnezoff seed powder after removing the oil component to the pure water is 1:10 to 1: 30.
In an embodiment of the present invention, the fructus momordicae component at least includes a fructus momordicae extract, and the method for preparing the fructus momordicae extract includes: a. crushing fructus Siraitiae Grosvenorii with 60 mesh sieve by using a crusher to obtain fructus Siraitiae Grosvenorii powder, mixing the fructus Siraitiae Grosvenorii powder with pure water, and extracting for 60-120 min to obtain fructus Siraitiae Grosvenorii extractive solution; b. concentrating the fructus Siraitiae Grosvenorii extractive solution under reduced pressure at 50deg.C and pressure of-0.06 Mpa, and drying to obtain dried fructus Siraitiae Grosvenorii extract; in the step a, the weight ratio of the momordica grosvenori powder to the pure water is 1:10 to 1: 30.
In an embodiment of the invention, the fructus momordicae component further includes the fructus momordicae powder obtained in the step a.
In an embodiment of the present invention, the licorice component at least includes licorice extract, and the method for preparing the licorice extract is as follows: a. crushing liquorice with a 60-mesh screen by using a crusher to obtain liquorice powder, and then mixing the liquorice powder with an ethanol aqueous solution with the concentration of 50-95 vt percent for extraction for 60-120 minutes to obtain liquorice extract; b. mixing the licorice extract with embedding material, embedding, drying after embedding, and crushing with 100 mesh sieve with crusher to obtain licorice extract.
In an embodiment of the present invention, in the step a, a weight ratio of the licorice powder to the aqueous ethanol solution is 1:10 to 1: 30; in step b, the weight ratio of the licorice extract to the embedding material is 1:1 to 1: between 10.

In this context, the values and parameters used to define the scope of the invention essentially inevitably contain standard deviations due to the individual test methods, and are therefore most often represented by approximate numerical values, which, in the specific embodiments, are as precisely as possible. According to one embodiment of the present invention, the blood glucose regulating health composition of the present invention may be administered with or without an excipient. The blood glucose-regulating health composition of the present invention may be formulated into a tablet containing various adjuvants, various disintegrants, granule binders or lubricants to form a solid dosage form, or may be filled into soft or hard gelatin capsules or may be encapsulated in a biodegradable carrier.
Excipients suitable for use in the present invention may be, for example, one or more of starch, dextrin, microcrystalline cellulose, sodium carboxymethyl starch, croscarmellose sodium, gum acacia, magnesium stearate, lactose. In addition, the blood glucose regulating health care composition of the invention can further comprise at least one of a sweetener, a carrier and an antioxidant which are compatible with other components in the composition.

The blood sugar regulating health care composition comprises a mountain balsam pear component, a mountain balsam pear seed component, a fructus momordicae component, a liquorice component and a cinnamon component; wherein the weight percentage of the mountain balsam pear component is generally between 20 and 60 percent, preferably between 25 and 60 percent, more preferably between 30 and 60 percent, and most preferably between 35 and 60 percent; the weight percentage of the mountain balsam pear seed component is generally between 5 and 20 percent, preferably between 5 and 15 percent, and most preferably between 8 and 14 percent; the weight percentage of the momordica grosvenori component is generally between 1 and 15wt%, preferably between 3 and 10wt%, and most preferably between 5 and 10wt%; the weight percentage of the licorice component is generally between 1 and 10wt%, preferably between 1 and 8wt%, and most preferably between 3 and 6wt%; the weight percentage of the cinnamon components is generally between 1 and 20wt%, preferably between 5 and 15wt%, most preferably between 8 and 13 wt%.
According to the technical ideas of the invention, the effective components in the mountain balsam pear component, the fructus momordicae component, the liquorice component and the cinnamon component in the blood sugar regulating health-care composition need to be quantitatively analyzed, for example, the content of the balsam pear saponin in the mountain balsam pear component is 10wt%, the content of the mogroside V in the fructus momordicae component is 5wt% and the content of the liquorice flavonoid in the liquorice component is 2wt%; and the cinnamaldehyde content in the cinnamon component is 2% by weight.
The mountain balsam pear component generally comprises mountain balsam pear extract extracted from mountain balsam pear pulp, is obtained by embedding mountain balsam pear concentrated solution extracted and concentrated by ethanol water solution with concentration of 50-95 vt% by embedding the mountain balsam pear concentrated solution by embedding materials, and the finished product obtained by embedding the mountain balsam pear concentrated solution by the embedding materials can increase water solubility and human body utilization rate.
According to the technical idea of the invention, the weight ratio of the mountain balsam pear powder relative to the ethanol aqueous solution is 1:10 to 1: 30; preferably in the range of 1:10 to 1: 25; more preferably at 1:10 to 1: 20.
According to the technical idea of the present invention, the embedding material may be any one of whey protein, carboxymethyl cellulose, maltodextrin, egg yolk lecithin, acacia, chitosan, cyclodextrin, and gelatin; preferably any one of whey protein, maltodextrin, egg yolk lecithin, acacia, chitosan, cyclodextrin, and gelatin; more preferably any one of whey protein, gum arabic, chitosan, cyclodextrin, and gelatin; most preferred is any one of whey protein, chitosan, cyclodextrin, and gelatin. The weight ratio of the mountain balsam pear concentrate to the embedding material is generally 1:1 to 1: between 10, preferably between 1:1 to 1: 8; more preferably at 1:1 to 1: 5; most preferably in the range of 1:1 to 1: 3.
In order to avoid that the content of the momordica saponins in the momordica charantia extract exceeds a quantitative standard (10 wt%), the content of the momordica saponins can be adjusted by appropriately adding momordica charantia powder to the momordica charantia component, wherein the source of the momordica charantia powder is generally a dry raw material used for preparing the momordica charantia extract.
The mountain balsam pear seed component is generally composed of mountain balsam pear seed extract, and the mountain balsam pear seed extract is obtained by removing oil content from mountain balsam pear seeds by supercritical extraction, extracting by pure water and concentrating under reduced pressure; the supercritical extraction is used to remove oil in the seed of Momordica charantia so as to avoid the risk of residual solvent caused by deoiling with organic solvent (n-hexane), and pure water extraction is used to obtain high-content Momordica Charantia protein from the seed of Momordica Charantia.
According to the technical idea of the invention, the weight ratio of the mountain bitter melon seed powder with oil removed relative to the pure water is 1:10 to 1: 30; preferably in the range of 1:10 to 1: 25; more preferably at 1:10 to 1: 15.
The fructus momordicae component is generally composed of fructus momordicae extract, and is obtained by extracting fructus momordicae with pure water, concentrating under reduced pressure and drying. According to the technical idea of the invention, the weight ratio of the momordica grosvenori to the water is 1:10 to 1: 30; preferably in the range of 1:10 to 1: 25; more preferably at 1:10 to 1: 15.
Likewise, in order to avoid that the mogroside V content in the momordica grosvenori extract exceeds a quantitative standard (5 wt%), a momordica grosvenori powder may be moderately added to the momordica grosvenori component to adjust the mogroside V content, and the source of the momordica grosvenori powder is generally a dry raw material used in preparing the momordica grosvenori extract.

Hereinafter, preferred embodiments of the present invention will be described in further detail.
Preparation example 1-mountain balsam pear component
Firstly, cleaning fresh mountain balsam pear, removing seeds, slicing and drying to ensure that the moisture content of the mountain balsam pear slices is below 5wt%, crushing the dried mountain balsam pear slices by a 60-mesh sieve by using a crusher to obtain mountain balsam pear powder, and extracting the mountain balsam pear powder for 60 to 180 minutes by using an ethanol aqueous solution with the concentration of 50 to 95vt percent (the weight ratio of the mountain balsam pear powder to the ethanol aqueous solution is 1:20), thereby obtaining mountain balsam pear extract.
Concentrating the Momordica Charantia extract under reduced pressure at 50deg.C under-0.06 Mpa, and mixing the concentrated Momordica Charantia solution with cyclodextrin at weight ratio of 1:2, stirring until the solution is uniform and is not layered, drying at 50 ℃ for 18-24 hours, and finally crushing by using a crusher through a 100-mesh screen to obtain the momordica grosvenori extract.
Quantitatively analyzing the components of the Momordica Charantia extract to confirm that the content of the momordica saponins in the Momordica Charantia extract is 15wt%; then, in order to adjust the saponin content of the mountain bitter gourd component, the mountain bitter gourd powder obtained by drying and crushing the mountain bitter gourd was added and mixed with the mountain bitter gourd extract, and the content of the bitter gourd saponin in the mountain bitter gourd component was adjusted to 10wt%.
Preparation example 2-mountain balsam pear seed component
Crushing mature mountain kusnezoff seed with 60 mesh sieve to obtain mountain kusnezoff seed powder, and supercritical fluid extracting at 35MPa and 50 deg.c to remove oil content in mountain kusnezoff seed powder to make the oil content below 1wt%.
Extracting the oil-removed mountain bitter gourd seed powder with pure water for 60-120 min (the weight ratio of mountain bitter gourd seed powder to pure water is 1:10) to obtain mountain bitter gourd seed extract, concentrating the mountain bitter gourd seed extract under reduced pressure at 50 ℃ and under the pressure of-0.06 Mpa, and spray drying to obtain the dried mountain bitter gourd seed extract, wherein the mountain bitter gourd seed extract can be directly used as mountain bitter gourd seed component of the blood sugar regulating health care composition in the following embodiments.
Preparation example 3-Momordica grosvenori component
Crushing fructus Siraitiae Grosvenorii with 60 mesh sieve by using pulverizer to obtain fructus Siraitiae Grosvenorii powder, adding pure water, extracting for 60-120 min (weight ratio of fructus Siraitiae Grosvenorii powder to pure water is 1:10), obtaining fructus Siraitiae Grosvenorii extract, concentrating the fructus Siraitiae Grosvenorii extract under reduced pressure at 50deg.C under-0.06 Mpa, and spray drying to obtain dried fructus Siraitiae Grosvenorii extract.
Performing component quantitative analysis on the momordica grosvenori extract to confirm that the mogroside V content in the momordica grosvenori extract is 7.1wt%; next, in order to adjust the saponin content of the momordica grosvenori component, the aforementioned dried and crushed momordica grosvenori powder was added to be mixed with the momordica grosvenori extract, and the mogroside V content in the momordica grosvenori component was adjusted to 5wt%.
Preparation example 4-Glycyrrhiza component
Crushing liquorice with a 60-mesh sieve by using a crusher to obtain liquorice powder, and extracting with 50-95 vt percent ethanol water solution for 60-120 minutes (the weight ratio of the liquorice powder to the ethanol water solution is 1:10) to obtain liquorice extract.
Then mixing the licorice extract with cyclodextrin in a weight ratio of 1:2, stirring until the solution is uniformly layered, drying at 50 ℃ for 18-24 hours, and crushing by using a 100-mesh screen by using a crusher to obtain the liquorice extract.
Quantitatively analyzing the components of the licorice extract to confirm that the content of licorice flavonoids in the licorice extract is 3.2wt%; next, in order to adjust the flavonoid content in the licorice component, the above-mentioned licorice powder obtained by drying and crushing licorice was added and mixed with the licorice extract, and the licorice flavonoid content in the licorice component was adjusted to 2wt%.
PREPARATION EXAMPLE 5-Cinnamomum cassia Components
Crushing the cinnamon by using a 60-mesh sieve by using a crusher, and extracting the cinnamon by using an ethanol water solution with the concentration of 50-95 vt percent for 60-120 minutes (the weight ratio of the cinnamon to the ethanol water solution is 1:10), thereby obtaining the cinnamon extract.
Then, the cinnamon extract and cyclodextrin are mixed according to a weight ratio of 1:2, stirring until the solution is uniformly layered, drying at 50 ℃ for 18-24 hours, and crushing by using a 100-mesh screen by using a crusher to obtain the cinnamon extract.
Quantitatively analyzing the components of the cinnamon extract, and confirming that the cinnamaldehyde content in the cinnamon extract is 2.6wt%; then, in order to adjust the cinnamaldehyde content in the cinnamon component, the aforementioned cinnamon powder obtained by drying and crushing cinnamon was added and mixed with the cinnamon extract, and the cinnamaldehyde content in the cinnamon component was adjusted to 2% by weight.
Example 1
First, the mountain bitter gourd components, mountain bitter gourd seed components, momordica grosvenori components, licorice components, and cinnamon components obtained in the foregoing preparation examples 1 to 5 were mixed, the weight percentages of the components being as follows:
35-60 wt% of mountain balsam pear extract,
8-14wt% of mountain balsam pear seed extract,
5-10wt% of Siraitia grosvenorii extract,
3-6% of licorice extract,
8-15 wt% of cinnamon extract,
Then adding sweetener dc101 and excipient magnesium stearate for mixing, wherein the weight percentage of dc101 in the blood sugar regulating health care composition is 32.9wt%, and the weight percentage of magnesium stearate in the blood sugar regulating health care composition is 0.1wt%.
The mixed blood sugar regulating health care composition is pressed into blood sugar regulating health care oral tablets with the weight of 500mg by a tablet press.
The prepared blood glucose-regulating health-care oral tablet was administered to 29 subjects suffering from diabetes, hyperglycemia or hyperlipidemia, one tablet was administered after three meals each day, and the ages, physical symptoms and administration days of the subjects are shown in Table 1 below.

The subjects were examined for pre-meal blood glucose, glycosylated hemoglobin, triglycerides, total cholesterol, low-density cholesterol, high-density cholesterol, and uric acid before the start of the administration and after the end of the administration, respectively, and the results obtained are recorded in table 2.

From the results of table 2, it is found that most subjects have a tendency to lower the blood glucose level, glycosylated hemoglobin, triglyceride, total cholesterol, low-density cholesterol, high-density cholesterol, and uric acid.
In addition, the blood sugar change and glycosylated hemoglobin index change of each subject during the taking period are observed, the blood sugar before meal of most subjects can be obviously reduced about 5 days after taking the blood sugar regulating health care oral tablet, and the glycosylated hemoglobin index of the subjects can be reduced about 1% after taking the blood sugar regulating health care oral tablet for about 15 days. Therefore, the blood sugar regulating health care composition has obvious auxiliary blood sugar reducing effect, and if the composition can be taken for more than 3 months, the medicine stopping of a type 2 pre-diabetes patient can be realized under the condition of regulating the life style; the patients with type 2 diabetes mellitus for more than 20 years can realize needle and medicine reduction.

1-15 wt% of a momordica grosvenori component; and
1-10wt% of licorice component; wherein the method comprises the steps of
The content of the momordica saponins in the mountain balsam pear component is 10wt%;
the content of mogroside V in the momordica grosvenori component is 5wt%;
the liquorice flavonoid content in the liquorice component is 2wt%;
the cinnamaldehyde content in the cinnamon component is 2wt%; and
the mountain balsam pear seed component at least comprises mountain balsam pear seed extract, and the manufacturing method of the mountain balsam pear seed extract comprises the following steps:
a. crushing the mountain bitter gourd seeds by using a crusher to obtain mountain bitter gourd seed powder, and performing supercritical extraction under the conditions that the extraction pressure is 35Mpa and the extraction temperature is 50 ℃, and removing oil content of the mountain bitter gourd seed powder to enable the oil content of the mountain bitter gourd seed powder to be below 1wt%;
b. extracting the mountain bitter gourd seed powder with pure water after oil removal to obtain mountain bitter gourd seed extract;
c. concentrating the mountain balsam pear seed extract under reduced pressure and drying to obtain a dried mountain balsam pear seed extract; wherein the method comprises the steps of

a. cleaning fresh Momordica Charantia, removing seeds, slicing, and oven drying until the water content is below 5wt%;
b. crushing the dried mountain balsam pear slices by using a crusher to obtain mountain balsam pear powder, and then mixing the mountain balsam pear powder with an ethanol aqueous solution with the concentration of 50-95 vt percent for extraction to obtain mountain balsam pear extract;
c. concentrating the mountain balsam pear extract under reduced pressure to obtain mountain balsam pear concentrate;
d. mixing the mountain balsam pear concentrated solution with embedding materials for embedding, drying after embedding, and crushing by a crusher to obtain the mountain balsam pear extract;

a. crushing fructus Siraitiae Grosvenorii with a pulverizer to obtain fructus Siraitiae Grosvenorii powder, mixing the fructus Siraitiae Grosvenorii powder with pure water, and extracting to obtain fructus Siraitiae Grosvenorii extractive solution;
