Abstract

技术领域
本发明涉及生物医药技术,具体涉及一种外泌体及cricRNA在糖尿病肾病足细胞损伤中的应用。
背景技术
糖尿病肾病是糖尿病最常见的并发症之一。目前,糖尿病肾病发病机制还未探明,并且缺乏有效治疗手段。糖尿病肾病的特点是肾小球滤过屏障被破坏,出现进行性蛋白尿。足细胞对于维持肾小球滤过屏障的结构和功能的完整性至关重要。有研究表明足细胞损伤和丢失是进行性慢性肾脏病的预测因子,与尿蛋白量直接相关。在糖尿病的情况下,足细胞可能会在许多致病信号(包括高血糖症、氧化应激、血管活性介质、细胞因子、和生长因子)的作用下发生功能障碍或损伤。
CircRNA是在真核细胞中广泛存在的、多样的内源性非编码RNA。其形成共价闭合、连续稳定的环状结构,不具有5’至3’极性以及3’polyA末端。核酸外切酶RNaseR不容易降解CircRNA,较线性RNA更加稳定。CircRNA一般无蛋白编码能力,但可发挥miRNA分子海绵作用,调控基因转录和选择性剪接。竞争性内源RNA(ceRNA)是CircRNA最经典的作用模式,即CircRNA通过碱基互补配对吸附miRNA,使得miRNA不能调控靶基因mRNA,从而使目的基因高表达。因此,CircRNA可执行重要的调控功能,且与众多疾病的发病机制有关,包括肾脏疾病。由于CircRNA可以影响疾病的发生和发展,也是于各种疾病潜在的诊断标记物和治疗靶点。
CircRNA通常以游离RNA分子的形式存在于细胞内外。这些RNA分子的大小从几百个核苷酸到数千个核苷酸不等。在细胞外,CircRNA大多装载于细胞外囊泡(EV)内,如外泌体。外泌体CircRNA参与了细胞间的信息交换,介导了一些疾病的发生发展。例如:癌细胞来源外泌体circUSP7通过调节NSCLC中的miR-934/SHP2轴诱导CD8+T细胞功能障碍。外泌体CircRTN4通过激活狼疮性肾炎中的miR-513a-5p/Fibronectin通路导致系膜细胞功能障碍。缺氧预处理的周细胞来源外泌体circEhmt1通过调节NFIA/NLRP3通路诱导微血管功能障碍。
GSK3β是调节Nrf2的许多信号通路的节点。GSK3β在活化时可以直接进入细胞核,调控核内Nrf2,使其出核并降解。GSK-3β也可使蛋白Fyn在苏氨酸残基处磷酸化,导致其核转移。进入细胞核后的Fyn可磷酸化Nrf2的Tyr568位点,其与Crm1结合后会从细胞核转移到细胞质并降解。此外,GSK3β能够直接磷酸化Nrf2的许多丝氨酸残基(包括Neh6结构域在内的与SCF/b-TrCP破坏基序重叠的残基),导致Nrf2出核和蛋白酶体降解。Nrf2是机体抗氧化应激和抗炎的主要保护因子。在众多疾病模型中,Nrf2活性及核聚集均会被抑制,导致其下游蛋白NQO1、γ-GCS、HO-1的表达减少。Nrf2基因敲除的动物出现患各种疾病的概率显著增加,如糖尿病肾病、肾小球肾炎、心脏病、骨骼肌丧失、癌症、动脉粥样硬化、肝损伤,视力缺陷。
目前尚未有研究报道外泌体circRNA是否参与通过调控GSK3β诱导糖尿病肾病足细胞损伤的作用机制。为此,本发明利用高糖环境的小鼠肾小管上皮细胞外泌体刺激足细胞,以研究外泌体及circRNA在糖尿病肾病足细胞损伤中的作用机制,为糖尿病肾病的诊断与治疗提供新的思路。
发明内容
(一)解决的技术问题
针对现有技术的不足,本发明提供了一种外泌体及cricRNA在糖尿病肾病足细胞损伤中的应用。
(二)技术方案
为实现以上目的,本发明通过以下技术方案予以实现:
第一方面,本发明提供了高糖环境的肾小管上皮细胞(Mouse kidney proximaltubular epithelial,TKPT)来源外泌体在制备诱导足细胞发生损伤的药物或制剂中的用途。
第二方面,本发明提供了高糖环境的肾小管上皮细胞来源外泌体在糖尿病肾病足细胞损伤中的调控应用。
进一步地,高糖环境的肾小管上皮细胞来源外泌体上调足细胞的GSK3β表达,抑制Nrf2表达,诱导足细胞的凋亡和损伤。
第三方面,本发明提供了circRNA-RALYL在制备诱导足细胞发生损伤的药物或制剂中的用途。

(三)有益效果
本发明提供了一种外泌体及cricRNA在糖尿病肾病足细胞损伤中的应用。我们研究了高糖环境的肾小管上皮细胞来源外泌体对足细胞的影响,首次发现高糖环境TKPT细胞来源外泌体可激活足细胞GSK3β蛋白表达水平,抑制Nrf2表达水平,诱导足细胞的凋亡和损伤;同时首次发现了高糖环境TKPT细胞来源外泌体中有53个差异表达的circRNA,其中在HG-exo组中有4个circRNA表达上调,49个circRNA表达下调,并证实了mmu_circ_0010491(circRNA-RALYL)在高糖环境TKPT细胞来源外泌体中表达上调;为进一步研究circRNA-RALYL在糖尿病肾病足细胞损伤中的调控机制,我们发现了circRNA-RALYL通过ceRNA机制,竞争性地与miR-378c结合,上调足细胞的GSK3β表达,抑制Nrf2表达,诱导足细胞的凋亡和损伤。以上结果表明了高糖环境TKPT细胞来源外泌体及circRNA-RALYL具有诱导足细胞的凋亡和损伤的调控作用,为制备糖尿病肾病足细胞损伤模型提供了新的构建方法,为糖尿病肾病的治疗开拓了新思路。

为使本发明实施例的目的、技术方案和优点更加清楚,下面将结合本发明实施例,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
1细胞实验分组
1.1TKPT细胞来源外泌体对足细胞的影响
DMEM培养基培养小鼠肾小管上皮细胞(TKPT细胞),将2瓶生长状态良好且一致,融合度一致约为40%-50%的肾小管上皮细胞随机分为2组,分别更换新鲜的5.6mM正常糖浓度培养基(NG组)、30mM高糖培养基(HG组)。放入37.0℃培养箱中培养48小时。收集细胞培养基并提取外泌体。再用5.6mmol/L葡萄糖浓度的1640培养基培养小鼠足细胞(MPC细胞),当细胞生长达培养皿70%-80%融合时,用不含FBS的1640培养基饥饿12小时使细胞同步化,再分别加入两组肾小管上皮细胞来源外泌体,对照组加入PBS溶液,根据以上不同处理,将足细胞分为:Ctrl组、NG-exo组、HG-exo组。处理时间点为48小时。
1.2circ_mmu_0010491对足细胞的影响
使用5.6mmol/L葡萄糖浓度的1640培养基培养足细胞,当足细胞生长达培养皿70%-80%融合时,将细胞随机分为四组,用不含FBS及双抗的1640培养基饥饿12小时使细胞同步化,更换新鲜的不含双抗的葡萄糖浓度5.6mmol/L的1640完全培养基,随机挑选两组细胞分别加入与lipo3000共孵育的miRNA-378c的mimics以及其阴性对照。以上两组细胞刺激6h后,更换新鲜的5.6mmol/L葡萄糖浓度的1640完全培养基,并加入circ_mmu_0010491过表达腺病毒(MOI=300),再分别将剩下两组暂时未做处理的细胞分别加入circ_mmu_0010491过表达腺病毒及其空载对照。转染16h后,更换新鲜培养基。根据不同以上不同处理,将足细胞分为:circ_mmu_0010491Vehicle组,circ_mmu_0010491OE组,circ_mmu_0010491OE+miRNA-378c NC组,以及circ_mmu_0010491OE+miRNA-378c mimics组。过表达病毒及其对照处理时间点为42h,Mimics及其对照处理时间点为48h。
2结果
2.1高糖环境下的TKPT细胞来源外泌体对足细胞的影响
2.1.1提取TKPT细胞来源外泌体并鉴定
为了进一步探索肾小管上皮细胞来源外泌体对足细胞的调控作用机制,我们进行了体外试验。分别在正常糖浓度及高糖浓度下培养小鼠肾小管上皮细胞(TKPT细胞),分为NG组、HG组,提取各组培养液中的外泌体并鉴定。使用透射电镜呈现外泌体的表征,包括:形状和大小。镜下可见圆形的外泌体囊泡,其尺寸范围为53至152nm(图1A)。Western blot示:NG组、HG组外泌体标志物CD9、ALIX和TSG101的表达均为阳性(图1B)。NTA结果示:NG组外泌体的平均尺寸为113.6nm,HG组外泌体的平均尺寸为117.1nm(图1C)。鉴定结果表明:我们提取的正常糖浓度及高糖浓度TKPT细胞的胞外囊泡均符合外泌体的表征。
2.1.2芯片筛选高糖刺激的TKPT细胞来源外泌体中差异表达circRNA
通过Affymetrix U133 plus 2.0表达谱芯片筛选出HG-exo组与NG-exo组外泌体差异表达的circRNA谱(图2)。在两组中共筛选出53个差异表达的circRNA(P<0.05,且FC≥2)。在HG-exo组中有4个circRNA表达上调,49个circRNA表达下调。差异表达的circRNA在两组中的聚类效果较好,HG-exo组NG-exo组组间差异较小。其中,HG-exo组mmu_circ_0010491(circRNA-RALYL)表达上调。
2.1.3环状RNA鉴定
常规的引物设计围绕目的片段上下游分别设计引物,称为convergent primer。而环状RNA与普通线性RNA相比,引物设计需要在其环化位点两侧,确保PCR扩增产物包含其环化位点,而这种引物称之为背对背引物,即divergent primer,以便区分与其相对应的线性RNA。因此环状RNA使用divergent primer和convergent primer进行扩增,均能成功扩增,而与其对应的线性RNA,则需要convergent primer进行PCR反应。
在Circbase、Circbank、Circular RNA Interactome数据库均显示,RALYL基因外显子可环化形成mmu_circ_0010491(图3A)。在TKPT细胞提取的RNA中成功表达出正确的mmu_circ_0010491。对mmu_circ_0010491的qRT-PCR扩增产物(图3B)进行Sanger测序(图3C),发现成环拼接位点与预期一致。背对背引物验证试验(图3D)结果可见:在TKPT细胞中,cDNA使用mmu_circ_0010491的divergent primer和convergent primer均能扩增到目的mmu_circ_0010491条带;而gDNA的divergent primer不能扩增到mmu_circ_0010491条带,convergent primer扩增到明显的目的条带;cDNA、gDNA使用GAPDH的convergent primer均能扩增到目的条带,但divergent primer不能扩增到目的条带。这说明与线性RNA相比,mmu_circ_0010491明显呈现环状结构。
2.1.4人鼠环状RNA同源性分析
我们将高糖刺激的TKPT细胞来源外泌体芯片分析筛选出来的mmu_circ_0010491,与糖尿病肾病肾穿刺活检组织筛选出来的has_circ_0137193进行同源性分析(图4)。分析发现:mmu_circ_0010491与has_circ_0137193具有高度同源性,同源度达到87.1%。
2.1.5预测mmu_circ_0010491可能靶向的miRNAs
为了进一步探索mmu_circ_0010491(circRNA-RALYL)在糖尿病肾病足细胞损伤中的作用,验证circRNA-RALYL是否通过ceRNA机制,竞争性地结合miRNA,进而调控下游蛋白的表达。我们通过生信分析预测了circRNA-RALYL可能结合的miRNAs。我们应用公共数据库miRanda、PITA和RNAhybrid,分别筛选circRNA-RALYL可能靶向的miRNA(图5A)。miRanda数据库筛选出circRNA-RALYL可能靶向的miRNA为53个,PITA数据库筛选出circRNA-RALYL可能靶向的miRNA为30个,RNAhybrid数据库筛选出circRNA-RALYL可能靶向的miRNA为360个。三个数据库分析结果取交集,得到13个可能靶向的miRNA,包括miR-378c(图5B)。
2.1.6验证mmu_circ_0010491可结合miR-378c
为了进一步验证mmu_circ_0010491(circRNA-RALYL)是否可以结合miR-378c,我们进行了双荧光素酶基因报告试验(图6)。试验结果显示:与NC mimics组相比,mmu-miR-378c可以显著下调mmu_circ_0010491-wt组的luciferase的表达(P<0.001)。而mmu_circ_0010491突变后,与NC mimics组相比,mmu-miR-378c未能下调mmu_circ_0010491-mu组的luciferase的表达(P>0.05)。说明miR-378c和mmu_circ_0010491确实存在结合作用。
2.1.7预测并验证miR-378c结合的mRNA
为了进一步探索与miR-378c存在结合位点的mRNA,利用公共生物生物信息数据库Targetscan分析发现,GSK3β与miR-378c有结合位点(图7A)。随后我们进行了双荧光素酶基因报告试验(图7B)。试验结果显示:与miR-NC组相比,miR-378c可以显著下调m-GSK3β-3UTR-wt组的luciferase的表达(P<0.001)。而突变后,与miR-NC组相比,miR-378c未能下调m-GSK3β-3UTR-mut组的luciferase的表达(P>0.05)。说明miR-378c和GSK3β确实存在结合作用,并且抑制了GSK3β的表达。
2.1.8TKPT细胞来源外泌体可上调MPC细胞circRNA-RALYL表达水平
为了检测TKPT细胞来源外泌体是否可被小鼠足细胞(MPC细胞)摄取,使用荧光染料PKH26标记TKPT细胞外泌体,并将外泌体加入至MPC细胞培养液中。24小时后在荧光显微镜下观察发现MPC细胞胞浆中出现了PKH26荧光(图8A)。由此可知,NG组、HG组的TKPT细胞来源外泌体均可被MPC细胞摄取。qRT-PCR检测(图8B)可见:HG组外泌体的circRNA-RALYL表达水平显著高于NG组外泌体,差异有统计学意义(P<0.05)。进一步检测各组外泌体对MPC细胞circRNA-RALYL表达水平的影响,qRT-PCR检测(图8C)可见,与Ctrl组相比,HG-exo组的circRNA-RALYL表达水平显著上调,差异有统计学意义(P<0.05)。由此可见,高糖刺激可使TKPT细胞来源外泌体的circRNA-RALYL表达水平增高,外泌体被足细胞摄取后,又可以增高足细胞circRNA-RALYL的表达水平。
2.1.9高糖环境TKPT细胞来源外泌体可激活足细胞GSK3β
Western blot结果显示(图9):HG-exo组足细胞中GSK3β蛋白表达水平明显高于NG-exo组、Ctrl组。HG-exo组足细胞中Nrf2蛋白表达水平明显低于NG-exo组、Ctrl组。由此可见TKPT细胞来源外泌体可激活足细胞的GSK3β,抑制Nrf2表达水平。另外一方面,HG-exo组足细胞标记物Podocin蛋白表达水平明显低于NG-exo组、Ctrl组。HG-exo组细胞凋亡标记蛋白表达水平明显高于NG-exo组、Ctrl组。由此可见TKPT细胞来源外泌体可诱导足细胞凋亡。
2.1.10高糖环境TKPT细胞来源外泌体可诱导足细胞损伤
各组足细胞的免疫荧光染色结果可见(图10):HG-exo组足细胞标记蛋Podocin表达水平明显低于NG-exo组、Ctrl组。由此可见高糖环境TKPT细胞来源外泌体可诱导足细损伤。
2.2mmu_circ_0010491对足细胞的影响
2.2.1mmu_circ_0010491对足细胞的调控作用
为了进一步探索mmu_circ_0010491(circRNA-RALYL)在足细胞中对GSK3β调控作用,我们将mmu_circ_0010491腺病毒(circ_0010491OE组)以及vehicle(circ_0010491vehicle组)转染足细胞。再向足细胞中转染入mimics-NC(circ_0010491OE+miR-378c NC组)以及miR-378c mimics(circ_0010491OE+miR-378c mimics组),观察不同处理条件下GSK3β、Nrf2、Podocin的表达水平的变化情况。Western blot结果显示(图11):与circ_0010491vehicle组相比,circ_0010491OE组的GSK3β表达水平上调,Nrf2表达水平下调。与circ_0010491OE+miR-378c NC组相比,circ_0010491OE+miR-378c mimics组GSK3β表达水平下调,Nrf2表达水平上调。由此可见,mmu_circ_0010491可以激活GSK3β、抑制Nrf2,但是这种作用会被miR-378c所消除。此外,与circ_0010491vehicle组相比,circ_0010491OE组的Podocin表达水平下调。与circ_0010491OE+miR-378c NC组相比,circ_0010491OE+miR-378c mimics组Podocin表达水平上调。由此可见,mmu_circ_0010491可以诱导足细胞损伤,但是这种作用会被miR-378c所消除。
2.2.2mmu_circ_0010491对GSK3β的调控作用
对各组细胞进行GSK3β免疫荧光染色,结果与Western blot一致。与circ_0010491vehicle组相比,circ_0010491OE组的GSK3β表达增强,与circ_0010491OE+miR-378c NC组相比,circ_0010491OE+miR-378c mimics组GSK3β表达水平减弱。由此可见,mmu_circ_0010491可以激活GSK3β,但是这种作用会被miR-378c所消除(图12)。
3结论
我们研究了高糖环境的肾小管上皮细胞来源外泌体对足细胞的影响,首次发现高糖环境TKPT细胞来源外泌体可激活足细胞GSK3β蛋白表达水平,抑制Nrf2表达水平,诱导足细胞的凋亡和损伤;同时首次发现了高糖环境TKPT细胞来源外泌体中有53个差异表达的circRNA,其中在HG-exo组中有4个circRNA表达上调,49个circRNA表达下调,并证实了mmu_circ_0010491(circRNA-RALYL)在高糖环境TKPT细胞来源外泌体中表达上调;为进一步研究circRNA-RALYL在糖尿病肾病足细胞损伤中的调控机制,我们发现了circRNA-RALYL通过ceRNA机制,竞争性地与miR-378c结合,上调足细胞的GSK3β表达,抑制Nrf2表达,诱导足细胞的凋亡和损伤。以上结果表明了高糖环境TKPT细胞来源外泌体及circRNA-RALYL具有诱导足细胞的凋亡和损伤的调控作用,为制备糖尿病肾病足细胞损伤模型提供了新的构建方法,为糖尿病肾病的治疗开拓了新思路。

3.如权利要求2所述的应用,其特征在于,高糖环境的肾小管上皮细胞来源外泌体上调足细胞的GSK3β表达,抑制Nrf2表达,诱导足细胞的凋亡和损伤。
4.外泌体circRNA在制备诱导足细胞发生损伤的药物或制剂中的用途。
5.外泌体cricRNA在糖尿病肾病足细胞损伤中的调控应用。

9.根据权利要求5所述的外泌体cricRNA在糖尿病肾病足细胞损伤中的调控应用,其特征在于,所述circRNA-RALYL通过ceRNA机制,竞争性地与miR-378c结合,上调足细胞的GSK3β表达,抑制Nrf2表达,诱导足细胞的凋亡和损伤。
10.高糖环境的肾小管上皮细胞来源外泌体和circRNA-RALYL用于构建糖尿病肾病足细胞损伤模型的应用。
