Abstract
本发明公开了双光治疗的白蛋白载药纳米粒在制备用于治疗宫颈癌药物中的应用,属于医药技术领域。本发明双光治疗的白蛋白载药纳米粒由白蛋白负载吲哚菁绿和羟基茜草素形成;纳米粒体系中同时含有ICG和purpurin;ICG光热剂不仅可以通过光热作用将光能转化为热能,同时还能通过光动力治疗产生活性氧来抑制细胞活性,而purpurin则通过抑制GDH1活性,破坏细胞内氧化还原稳态的同时切断癌细胞营养供应;基于上述的三种抗肿瘤机制,解决单一性化疗毒性大、治疗效果低等缺点,通过纳米体系联合光动光热治疗提高羟基茜草素对宫颈癌细胞的治疗效果,为解决宫颈癌的易复发、死亡率高等技术难题提供了新的解决方案。
Description
技术领域
本发明涉及双光治疗的白蛋白载药纳米粒在制备用于治疗宫颈癌药物中的应用,属于医药技术领域。
背景技术
目前,癌症(恶性肿瘤)作为全球最致命的疾病之一,正在严重威胁着人类的健康,而且其发病率正在呈现年轻化的特点。当涉及恶性肿瘤治疗时,化疗一直是肿瘤的主要治疗手段,但单一化疗药物的使用治疗效果有限,因此迫切需要安全性高、效果显著的新方法用于肿瘤治疗。近年来借助光动力(PDT)、光热(PTT)、联合疗法等诸多潜在治疗策略用于肿瘤治疗广泛引起科研人员的兴趣,其中PTT、PDT因具备无创、副作用小等特点使之成为研究的热点。然而肿瘤细胞内高水平的谷胱甘肽(GSH)严重影响了PDT效果,单一的PTT又依赖过高热的产生,进而造成治疗顺应性差和组织损伤,所以研究人员致力于使用不同方法改善PDT和PTT效果。吲哚菁绿(ICG)因被FDA批准使用、并同时能够产生光热-光动一体化作用而备受广大研究人员的青睐;可是ICG半衰期短、稳定性差等缺点限制了其疗效。
大量研究表明,白蛋白作为机体内源性物质在药物递送中是一种理想的载体材料,基于白蛋白设计的联合治疗策略不仅能够克服单一疗法的局限性,还能提高各组分药物的稳定性;但在实际应用过程中,由白蛋白作为载体负载吲哚菁绿(ICG)在用于肿瘤细胞时,仍然会存在ICG光动力不足的问题,进而会造成治疗顺应性差和组织损伤。
发明内容
针对现有技术存在的缺陷与不足,本发明提供了双光治疗的白蛋白载药纳米粒在制备用于治疗宫颈癌药物中的应用;本发明设计并成功优化合成了基于双光治疗的功能白蛋白纳米载药(ICG&purpurin@BSA,BIP)颗粒,其中,羟基茜草素purpurin可通过抑制谷氨酸脱氢酶GDH1功能、限制谷氨酰胺分解进而下调GPx1水平,进而降低癌细胞内GSH含量实现细胞损伤;将其作为纳米药物载体,结合光动力疗法、光热疗法、抑制谷氨酸脱氢酶GDH1来破坏细胞稳态并切断营养供应等机制,构建包含该纳米药物的载药体系,通过增强的PDT协同温和的PTT一定程度上提高了宫颈癌的治疗效果,最终使联合疗法的效果得以充分发挥。
本发明的设计的优点主要包括两个方面,一方面在提高药物稳定性的基础上,避免了单一PTT导致温度过高引起的局部组织高热现象,进而实现温和的光热治疗,进一步改善了患者的依从性;另一方面,purpurin通过减少ROS的消耗实现协同增强的PDT效果;综上所述,利用白蛋白作为药物载体,在提高ICG、purpurin稳定性的同时,以实现温和的PTT和协同增强的PDT,将为高效低毒的肿瘤治疗提供潜在新策略。
本发明的第一个目的是提供了双光治疗的白蛋白载药纳米粒在制备用于治疗宫颈癌药物中的应用,所述双光治疗的白蛋白载药纳米粒由白蛋白负载吲哚菁绿和羟基茜草素形成。
在一种实施方式中,所述白蛋白为牛血清白蛋白、人血清白蛋白、卵清蛋白、重组蛋白中的任一种。
在一种实施方式中,所述白蛋白载药纳米粒中吲哚菁绿终浓度为30μg/mL,羟基茜草素终浓度为2.3~6.9μg/mL。
在一种实施方式中,所述白蛋白载药纳米粒中吲哚菁绿终浓度为30μg/mL,羟基茜草素终浓度为6.9μg/mL。
在一种实施方式中,所述双光治疗的白蛋白载药纳米粒的制备具体包括如下步骤:
(1)A液的配制:将吲哚菁绿(ICG)和羟基茜草素(purpurin)溶解于二甲亚砜中,避光搅拌均匀后得到A液;
(2)B液的配制:将白蛋白溶解于超纯水中,搅拌至完全溶解,得到B液;
(3)将B液缓慢的逐滴加入A液中,常温避光搅拌反应;反应结束后将反应液2~6℃搅拌过夜,100KD超滤管纯化,超纯水洗涤,重复超滤,得到白蛋白载药纳米粒。
在一种实施方式中,步骤(1)吲哚菁绿(ICG)和羟基茜草素(purpurin)摩尔比为(0.5~2):1;优选为1:1。
在一种实施方式中,步骤(2)所述B液中白蛋白的浓度为2~5mg/mL;优选为4mg/mL。
在一种实施方式中,步骤(3)所述A液与B液的混合体积比为1:5。
在一种实施方式中,当吲哚菁绿和羟基茜草素摩尔比为1:1时,此时A液中的吲哚菁绿浓度为6mg/mL,羟基茜草素浓度为2mg/mL;B液中白蛋白的浓度为4mg/mL;A液与B液的混合体积比为1:5。
本发明的第二个目的是提供一种用于治疗宫颈癌的药物,所述药物包含白蛋白载药纳米粒活性成分,所述双光治疗的白蛋白载药纳米粒由白蛋白负载吲哚菁绿和羟基茜草素形成。
本发明的有益效果:
(1)本发明白蛋白载药纳米颗粒通过以牛血清白蛋白为载体提高负载药物的生物相容性和治疗作用;并且纳米体系含有ICG(光热剂)和purpurin;ICG光热剂不仅可以通过光热作用将光能转化为热能,同时还能通过光动力治疗产生活性氧(ROS)来抑制细胞活性,而羟基茜草素(purpurin)则通过抑制GDH1活性,破坏细胞内氧化还原稳态的同时切断癌细胞营养供应;基于上述三种抗肿瘤机制(光动力疗法、光热疗法、抑制谷氨酸脱氢酶1来破坏细胞稳态并切断营养供应),解决单一性化疗毒性大、治疗效果低等缺点,设计基于联合疗法的策略和技术方案,通过纳米体系联合光动光热治疗提高化疗药物(羟基茜草素)对宫颈癌细胞的治疗效果,为解决宫颈癌的易复发、死亡率高等技术难题提供了新的解决方案;
(2)本发明白蛋白的载药纳米颗粒的制备,合成操作简单便捷,具有合适的粒径大小(电镜下尺寸200nm左右),作为药物载体具有良好的药物负载能力,能通过疏水作用负载吲哚菁绿与羟基茜草素,具有较高的生物相容性;
在细胞毒性试验中,在808nm激光照射下,与单独装载吲哚菁绿或单独装载羟基茜草素的载药系统(ICG@BSA,BI或purpurin@BSA,BP)对HeLa细胞的存活率相比,吲哚菁绿与羟基茜草素共同装载在纳米体系后(ICG&purpurin@BSA,BIP)对HeLa细胞的存活率大大减少,证明了此纳米系统在ICG和purpurin的协同作用下,减少ICG产生ROS消耗的同时,能提高宫颈癌的治疗效果;通过测定吲哚菁绿的光热疗法诱发温度变化、活性氧产生能力、载药系统生物相容性等,确定载药纳米体系的抗肿瘤效果;
(3)本发明的白蛋白载药纳米颗粒,能够降低化疗药物剂量的同时,通过协同光热疗法和光动力疗法,提高药物对宫颈癌细胞的治疗效果,对探索利用纳米材料作为载体的联合疗法用于递送抗癌药物等方面具有一定的指导意义,并为提高PDT效果提供一种潜在策略。

具体实施方式
下面结合附图和实施例,对本发明进行具体描述。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1
一种制备白蛋白载药纳米粒的方法,所述方法包括如下步骤:
(1)A液的制备:将6mg吲哚菁绿(ICG)和2mg羟基茜草素(purpurin)(ICG与purpurin的摩尔比1:1)溶解于4mL的二甲亚砜中,避光搅拌均匀后得到A液;
(2)B液的制备:将80mg的牛血清白蛋白(BSA)溶解于20mL的超纯水中,搅拌至完全溶解,得到4mg/mL的B液;
(3)将B液缓慢的逐滴加入A液中,每2秒1滴,此过程为常温搅拌反应,要求避光,缓慢,其后,将所得反应液在4℃避光搅拌过夜,100KD超滤管纯化,3000RCF离心后取上液,超纯水洗涤多次,重复超滤,合并收集,即得到白蛋白载药纳米粒,即ICG&purpurin@BSA,缩写为BIP载药纳米粒。
实施例2
与实施例1的区别仅在于,调整步骤(1)吲哚菁绿和羟基茜草素的摩尔比分别为2:1和1:2;其它参数和条件与实施例1相同。
对比例1
与实施例1的区别仅在于,步骤(1)省去吲哚菁绿或省去羟基茜草素的添加;其它参数和条件与实施例1相同,分别制备白蛋白载药纳米粒BI和白蛋白载药纳米粒BP。
白蛋白载药纳米粒性能表征
1、对实施例1和2分别制备的白蛋白载药纳米粒进行粒径测定,结果如表1所示:

2、对实施例1和对比例1分别制备的白蛋白载药纳米粒进行表征
使用透射电镜(TEM)、动态光散射(DLS)对制得的白蛋白载药纳米粒的平均粒径进行测量:
图1为透射电镜(TEM)对制得的白蛋白载药纳米粒的平均粒径进行测量,结果表明白蛋白载药纳米粒BP、BI粒径均小于200nm,BIP粒径在200nm左右,载药纳米粒粒径较均一,分散性良好。
图2为动态光散射(DLS)对制得的白蛋白载药纳米粒的平均粒径进行测量,结果显示白蛋白载药纳米粒BP、BI、BIP粒径分别为190nm、220nm、359nm;图2D不同载药纳米粒的Zeta电位显示药物的成功负载。
对白蛋白纳米粒BIP进行稳定性测试,结果如图3A所示,随着时间的延长,BIP纳米粒粒径变化较小,且分散性显示良好,图3B显示样品溶液底部并未发现沉淀现象,7天稳定性实验表明BIP溶液具有良好的物理稳定性。
利用FTIR和488nm特征荧光验证purpurin的成功负载,结果如图4A所示,FTIR结果显示BIP和purpurin在1049、880cm-1处有相同吸收峰,且将BIP冻干粉末在488nm处显示出purpurin的特征荧光(图4C)。
利用UV-vis验证ICG的成功负载,图4B表征结果显示BIP与ICG在780nm处有相同特征峰,且FTIR结果显示在667cm-1处有ICG的特征峰(图4A),以上结果均表明BIP能够成功制备。
如图5所示,利用外分光光度计分别在778nm和486nm处绘制ICG(图5A)和purpurin(图5B)的标准曲线,然后利用各自标曲测量纯化后游离的ICG、purpurin的紫外吸收,并计算其在纳米粒中的含量。最终,BIP中purpurin的封装效率跟载药水平分别为61.2%、3.4%,ICG则为88.7%、14.7%。
为研究BIP纳米粒中药物的释放特性,将BIP放置于不同环境的PBS中进行释放研究,不同pH条件下研究BIP中药物的释放情况发现,结果如图5C所示,弱酸性环境更有利于BIP中药物的释放。
3、白蛋白载药纳米粒BIP光热能力研究
按实施例1制备的BIP,对比例1制备的BI,将5个含有1mL不同样品(BSA,purpurin,ICG,BI,BIP)塑料管离心管置于808nm(其中BSA浓度为400μg/mL,ICG浓度为30μg/mL,purpurin浓度为6.9μg/mL,BI与BIP中ICG浓度均为30μg/mL),功率强度为1Wcm-2的激光下,照射下4min;然后,使用近红外热成像仪(实时温度记录仪)每30秒记录每组的温度,持续4min。
如图6(A)所示,BSA跟游离的purpurin并无热量产生,而热量主要来自于光热剂ICG及含有ICG的载药系统BI和BIP。
根据图6(B)温度曲线图可知,随着光照时间的延长,载药体系所产生的热量越多,这是载药系统产生良好细胞抑制效果的主要原因。
图6(C)显示与游离ICG比较,BIP经过连续4个光照-冷却循环后仍然具有良好的光热稳定性,图6(D)所示,ICG的光热效应可随浓度的增加而增加,具有浓度依赖性,4分钟光照后,温度最高能达到48.5℃,更重要的是此温度在发挥光热治疗的同时不会损伤周围其他组织,这对宫颈癌患者非常有利,以上结果表明本发明制备的BIP能够通过温和的光热疗法进行癌症治疗。
4、白蛋白载药系统的细胞摄取情况研究
通过激光共聚焦显微镜拍摄不同发射波长下细胞内药物的分布情况。在激光共聚焦专用的培养皿中加入1mL密度为1×105个/mL的HeLa细胞,在培养箱中孵育直至细胞充分贴壁。将实施例1制得的ICG&purpurin@BSA(BIP)载药系统分散液以其包裹的ICG的浓度为计量单位,当ICG在BIP中的浓度为30μg/mL时,其与HeLa细胞分别孵育2、4、6小时后,用PBS清洗共聚焦培养皿3次后,充分除去残留在孔板中的药物,然后加入1mL 4%的多聚甲醛溶液固定细胞。随后加入500μL DAPI染色液,10min后完成细胞核的染色,并用1mL PBS置于的摇床上洗五次,每次10min以除去残留的DAPI染色液。洗涤完成后每皿加入1mL PBS放于4℃冰箱中避光保存。样品使用激光扫描共聚焦显微镜(CLSM)观察HeLa细胞对载药系统的吞噬效果以及不同发射波长下细胞内药物的分布情况。
图13结果显示,孵育时间为2小时时,有较少的荧光信号(ICG的绿色荧光和purpurin的红色荧光)被观察到,然而孵育时间延长至6小时时,荧光信号能被清楚的观察到。这就证明了ICG&purpurin@BSA(BIP)载药系统可以被肿瘤细胞快速摄取,成功进入细胞内,由此证实了本发明的ICG&purpurin@BSA(BIP)载药系统用于治疗宫颈癌的可行性。
5、特定波长光照刺激下细胞内产生ROS的检测
首先,向24孔板中加入1mL密度为1×104个/mL的HeLa细胞,放入培养箱孵育48h。然后加入无血清DMEM分散的ICG&purpurin@BSA(BIP)(CICG,0,10,20,30μg/mL),继续孵育6h。然后用ROS检测试剂盒检测其产生ROS的能力,详细操作如下:用PBS洗净孔板里残留的药物和培养基,置于200r/min摇床上洗涤3次,每次10min,洗涤时注意避光。随后用无血清DMEM稀释DCFH-DA使其浓度为10μM,将其与细胞放于培养箱中共孵育30min后用无血清培养基避光洗涤,置于200r/min的摇床上洗涤3次,每次10min,再用特定波长的光照射24孔板,照射参数条件为808nm,4min,1w/cm2,随后用荧光显微镜FITC通道观察ROS的产生情况。
如图14A所示,实施例1制得的白蛋白载药系统顺利地在特定波长照射下产生大量的ROS,ROS试剂盒中产生的DCF结合而显示出强烈的绿色荧光。而且,与BI组比较发现BIP组在光照后能够产生更多的ROS,图14B表明BIP在光照后产生的ROS具有浓度依赖性。图15是对图14产生的ROS进行流式定量分析所得数据,图15A显示,相同ICG浓度且光照下,BIP比BI能够产生更多的ROS,图15B表明且随着BIP浓度增加,ROS成浓度依赖性。综上,图14、图15证明ICG&purpurin@BSA(BIP)载药系统具有产生ROS的能力,并且随着ICG浓度的增加,产生的ROS也随之增加,在ICG浓度为30μg/mL时,BIP比BI经光照后产生的ROS更多,表明BIP能够通过增强的光动力疗法进行癌症治疗。
实施例3白蛋白载药纳米粒BIP在小鼠体内的稳定性和治疗有效性研究
选择生长状态良好的HeLa细胞用于肿瘤模型的构建,选用重量为18-22g的雌性(4-6周)健康裸鼠(Balb/c nude),裸鼠购买于斯贝福(北京)生物技术有限公司;所有动物实验过程均依据江南大学伦理委员会要求进行。
首先将生长状态良好的HeLa细胞用胰酶消化,PBS重悬后调整细胞浓度至1×106/mL,于裸鼠右侧腋下处皮下注射重悬的HeLa细胞100μL以建立肿瘤实验模型,借助数显游标卡尺衡量肿瘤生长大小;等待肿瘤体积达到一定体积(100mm3)时,肿瘤模型建立完成;
设定两个实验组(游离ICG组、BIP组),每组1只;分别采用尾静脉注射100μL含有相同浓度的ICG(70μg)的药物,注射药物8小时后,使用小动物活体成像系统(激发745nm/发射840nm)检测药物(ICG、BIP)在小鼠体内的分布情况。
结果如图7所示,活体成像和组织成像分析结果显示游离ICG进入小鼠体内后,主要分布在小鼠肝脏和肾脏,肿瘤部位只能摄取较少的ICG,说明游离ICG在体内半衰期可能比较短且很容易被机体代谢掉,不利于治疗的进行。而BIP组虽然在肝肾显示有较强的ICG荧光,但在肿瘤部位也有一定富集,说明BIP可以被肿瘤组织摄取,这可能归功于白蛋白具有一定的靶向能力而易被饥饿的肿瘤细胞摄入;另外,从BIP的器官分布还能看出,BIP主要经肝脏代谢,相比于游离ICG而言,经BSA递送的药物肾脏清除率较低、稳定性更好。因此,以上结果表明BIP能够被肿瘤组织摄取并表现出良好的体内稳定性。
为了进一步分析白蛋白纳米载药纳米粒在体内的光热转换效果。建模两周后,设置两组实验组(PBS-NIR组、BIP-NIR组),分别尾静脉注射PBS和BIP(含ICG 70μg)各100μL,8小时后将小鼠麻醉进行肿瘤部位激光照射(808nm,1W cm-2,4min),随后利用近红外成像仪记录光照后肿瘤部位的温度变化情况,最后绘制温度变化曲线。
结果如图8所示,图8A表明BIP在体内能实现良好的光热转换效果,图8B温度变化结果显示,BIP经过4分钟光照,最高温度可达45.1℃,说明BIP能够被肿瘤组织成功摄取并发挥温和的光热作用,而PBS组光照后温度接近正常体温,并不能产生PTT作用。经过二者体内光热效果对比发现,BIP在体内能够实现温和的光热治疗,且具备两大特点,一方面,温和的光热在杀死肿瘤细胞的同时不会影响正常组织,这很大程度上能够提高光热治疗的顺应性,另一方面也验证BIP在体内的成功递送,这为基于干扰氧化还原稳态设计的一体化白蛋白药物递送系统提供基础。
为了进一步研究BIP递药系统对肿瘤组织的影响,采用苏木精&伊红(H&E)染色法对肿瘤组织切片进行研究。苏木精为天然碱性染料,能使细胞核呈蓝色,而伊红可使细胞质染成粉色。从图9可以看出,PBS组几乎无组织损伤,随着光照的进行,各加药组均展现出不同程度的组织损伤,其中ICG-NIR、BI-NIR、BIP-NIR现象最为明显,猜测这可能与细胞中出现相应的细胞核溶解、组织坏死、纤维增生、炎性浸润及空泡化现象有关,BIP-NIR组中空泡化现象比较严重,进一步说明BIP治疗的有效性。
实施例4
BIP载药纳米粒在用于治疗宫颈癌中的应用
选择生长状态良好的HeLa细胞用于肿瘤模型的构建,选用重量为18-22g的雌性(4-6周)健康裸鼠(Balb/c nude),裸鼠购买于斯贝福(北京)生物技术有限公司;所有动物实验过程均依据江南大学伦理委员会要求进行。
首先将生长状态良好的HeLa细胞用胰酶消化,PBS重悬后调整细胞浓度至1×106/mL,于裸鼠右侧腋下处皮下注射重悬的HeLa细胞100μL以建立肿瘤实验模型,借助数显游标卡尺衡量肿瘤生长大小;等待肿瘤体积达到一定体积(100mm3)时,肿瘤模型建立完成;
分别设定7个实验组(PBS-NIR、Pur-NIR、ICG-NIR、BI、BIP、BI-NIR、BIP-NIR组),每组各4只小鼠,其中NIR为Near-infrared irradiation缩写,表示光照处理组;给药时间设为每两天给药一次,尾静脉注射PBS或药物组(含ICG 70μg,purpurin 161μg)各100μL,共给药6次(1、3、5、7、9、11),光照组(NIR组)在每次给药后的8小时进行光照,非光照组不进行光照,(808nm,1W cm-2,4min),在给药过程中每两天记录小鼠的体重和肿瘤体积大小,用于后续实验分析;
在治疗结束后处死所有小鼠并取出各鼠的肿瘤和组织用于实验研究,比较各实验组肿瘤体积大小,判断抗肿瘤的效果。
结果如图10所示:图10(A)为体内抗肿瘤实验示意图;图10(B)和(C)为治疗结束后各组肿瘤体积大小统计图和直观图,由图可知,BIP-NIR组肿瘤抑制效果最为明显,对照组(PBS-NIR组)肿瘤体积大约是BIP-NIR组的10倍,表明BIP良好的抑制肿瘤生长的能力。与游离药物组(purpurin-NIR、ICG-NIR)相比,BIP-NIR也表现出增强的抗肿瘤效果。与游离ICG-NIR对比,BIP-NIR肿瘤生长抑制率提高了32.6%,与游离purpurin-NIR对比,肿瘤生长抑制率提高约47.6%。
另外,BIP-NIR组与BI-NIR组比较,BI-NIR组虽然也有一定的治疗效果,但是经过11天的治疗后BIP-NIR组肿瘤体积更小,与空白组对比效果更为明显(肿瘤生长抑制率为87%),说明BIP-NIR总体治疗效果大于BI-NIR组,这是因为与BI-NIR比较,BIP-NIR组多负载了purpurin,BIP中搭载的purpurin作为氧化还原稳态调节剂能够降低肿瘤细胞内GSH含量,进而减少ICG光动力治疗时所产生ROS的消耗,增强ICG光动力效果,与单独BI-NIR组比较,最终实现优于BI-NIR的抗肿瘤效果。
图10(D)显示了各实验组肿瘤的重量,结果显示BIP-NIR组肿瘤重量最低,与肿瘤大小相对应,进一步说明BIP-NIR组治疗效果最为理想。图10(E)显示在治疗期间小鼠的体重并未发生明显变化,侧面体现治疗策略的安全性。
对比例2
参照实施例4的应用方法,采用MTT法(噻唑蓝)将BIP应用于A431(人皮肤鳞癌细胞)、PATU-8988(人胰腺癌细胞)、MDA-MB-231(人乳腺癌细胞)的抑制效果;结果如图11所示,当BIP中ICG浓度为30μg/mL,purpurin浓度为6.9μg/mL时,在光照条件下(808nm,1W cm-2,4min),HeLa细胞的存活率最低(19.4%),治疗效果最为明显。
实施例5白蛋白递药系统使用量探究
通过MTT(噻唑蓝)法研究了实施例1中载药白蛋白载药系统对HeLa细胞的体外细胞毒性。
首先按实施例1制备BIP纳米粒、对比例1制备BI、BP纳米粒备用,然后将HeLa细胞以1×104细胞/孔的密度接种到96孔板中,孵育36h(37℃,5% CO2)。
因为BIP是一体化合成,为更好表述浓度关系,另外为探究BIP中实际负载的药物对细胞的抑制效果,以负载药物的浓度进行表示浓度关系。用移液枪除去96孔板中的培养基,再将用培养基稀释白蛋白载药系统分散液(使得BIP中ICG浓度依次为,0μg/mL、10μg/mL、20μg/mL、30μg/mL、40μg/mL,此时purpurin浓度对应为,0μg/mL、2.3μg/mL、4.6μg/mL、6.9μg/mL、9.2μg/mL)添加到孔板中,继续孵育6h。孵育完成后移除载药的载药系统分散液,加入新鲜培养液后,用808nm激光照射4分钟,其功率强度为1Wcm-2,光照后继续培养24小时,然后移除培养液,加入100μL新鲜培养基和10μL MTT溶液(5mg/mL),继续孵育4h。随后,添加100μL MTT Buffer(三联溶解液),过夜读板。在酶标仪上记录每个孔在570nm的光密度值(Optical density,OD)。每次实验重复三次。细胞活力计算公式如下:细胞存活率Cellviability=OD实验/OD对照。
如图12(A)所示高剂量的BSA也不会抑制HeLa细胞,在BSA浓度为0-800μg/mL范围内,HeLa细胞的存活率均在90%以上,表明BSA作为载体具有良好的安全性。
如图12(B)探究了不同浓度BIP对HeLa细胞的抑制作用,用MTT法检测了不同浓度下白蛋白载药系统对细胞的抑制率。可见,当无光照时,且载药系统中ICG的浓度为30μg/mL,HeLa细胞的存活率仍能达到80%,说明BIP在此浓度下安全性较好。
因此,当白蛋白载药系统中ICG的浓度低于30μg/mL时均为合理的使用范围,考虑到后续实验药物的承载量,以选择载药系统中ICG的浓度为30μg/mL为例进行的应用实验,当ICG浓度为30μg/mL时,BIP经光照后细胞抑制率可为19.4%。另外,在808nm激光照射下,同时载有ICG和purpurin的载药系统BIP比单独载药的BP或BI表现出更优秀的细胞抑制效果,而且细胞抑制效果具有浓度依赖性。
实施例6白蛋白载药系统血液相容性研究
利用裸鼠鼠血中的红细胞与游离药物和BIP共孵育以研究纳米粒子的血液相容性。将取好的鼠全血置于抗凝管内防止凝结,用离心法获取全血中的红细胞,分别设置两组溶血实验组(游离ICG组,BIP组),阳性对照组为含Triton X-100(10%)的PBS,空白对照为PBS组,然后分别加入800μL不同浓度的药物(所含ICG浓度相同,分别为0、10、20、30、40μg/mL),再向EP管内补充200μL新制备含2%红细胞的PBS混悬液,最后将各样品于恒温培养箱中孵育3小时,温度设置为37℃,孵育完毕后使用离心机离心(3000rpm,5min)并取200μL上清液于酶标仪测定541nm处的吸光度,使用以下公式测定样品的溶血率。通过图9所示,与游离药物相比,BIP具有良好的生物相容性。
图16A和B为游离药物血液相容性实验结果,图16A显示随着游离药物ICG浓度的增加,液层溶血现象越明显,图16B为游离药物对应溶血率变化,结果显示其溶血率随药物浓度增加而增加,表明游离药物浓度越高其安全性越差,当游离ICG浓度为30μg/mL时(此时purpurin浓度为6.9μg/mL),其溶血率为48.33%。由图16CD BIP的血液相容性实验结果可知,当BIP中ICG浓度为30μg/mL时(purpurin浓度为6.9μg/mL)其溶血率仅为4.8%。经对比可知,在此浓度下BSA将ICG和purpurin的安全性提高了10倍左右。当BIP中ICG浓度为40μg/mL时,其溶血率由4.8%增加到了6.3%,过高的HR值不利于治疗的展开(一般低于5%),以上表明BIP递药系统具有较好的血液相容性且当粒子中ICG浓度为30μg/mL时既可以满足安全性要求又可以达到良好的治疗效果。
以上所提供的实施例并非用以限制本发明所涵盖的范围,所描述的步骤也不是用以限制其执行顺序。本领域技术人员结合现有公知常识对本发明做显而易见的改进,亦落入本发明权利要求书所界定的保护范围之内。
Claims (10)
1.双光治疗的白蛋白载药纳米粒在制备用于治疗宫颈癌药物中的应用,其特征在于,所述双光治疗的白蛋白载药纳米粒由白蛋白负载吲哚菁绿和羟基茜草素形成。
2.根据权利要求1所述的应用,其特征在于,所述白蛋白为牛血清白蛋白、人血清白蛋白、卵清蛋白和重组蛋白中的任一种。
3.根据权利要求1所述的应用,其特征在于,所述白蛋白载药纳米粒中吲哚菁绿终浓度为30μg/mL,羟基茜草素终浓度为2.3~6.9μg/mL。
4.根据权利要求1所述的应用,其特征在于,所述白蛋白载药纳米颗粒中吲哚菁绿终浓度为30μg/mL,羟基茜草素终浓度为6.9μg/mL。
5.根据权利要求1所述的应用,其特征在于,所述双光治疗的白蛋白载药纳米粒的制备具体包括如下步骤:
(1)A液的配制:将吲哚菁绿ICG和羟基茜草素purpurin溶解于二甲亚砜中,避光搅拌均匀后得到A液;
(2)B液的配制:将白蛋白溶解于超纯水中,搅拌至完全溶解,得到B液;
(3)将B液缓慢的逐滴加入A液中,常温避光搅拌反应;反应结束后将反应液2~6℃搅拌过夜,100KD超滤管纯化,超纯水洗涤,重复超滤,得到白蛋白载药纳米粒。
6.根据权利要求1所述的应用,其特征在于,步骤(1)吲哚菁绿ICG和羟基茜草素purpurin摩尔比为(0.5~2):1。
7.根据权利要求1所述的应用,其特征在于,步骤(2)所述B液中白蛋白的浓度为2~5mg/mL。
8.根据权利要求1所述的应用,其特征在于,步骤(3)所述A液与B液的混合体积比为1:5。
9.根据权利要求1所述的应用,其特征在于,当吲哚菁绿和羟基茜草素摩尔比为1:1时,此时A液中的吲哚菁绿浓度为6mg/mL,羟基茜草素浓度为2mg/mL;B液中白蛋白的浓度为4mg/mL;A液与B液的混合体积比为1:5。
10.一种用于治疗宫颈癌的药物,其特征在于,所述药物包含白蛋白载药纳米粒活性成分,所述双光治疗的白蛋白载药纳米粒由白蛋白负载吲哚菁绿和羟基茜草素形成。
