Abstract
本发明提供了一种缓释抗菌的口气清新剂及其制备方法。所述缓释抗菌口气清新剂,以质量份数计,包括:0.05‑0.3份聚合物抑菌胶囊、5‑20份保湿稳定剂、0.1‑0.5份表面活性剂、0.05‑2份甜味剂、0.06‑0.1份pH值调节剂和50‑59份去离子水;其中,所述聚合物抑菌胶囊的壳材为2‑甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸‑乙醇酸共聚物;芯材为抑菌植物精华。通过2‑甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸‑乙醇酸共聚物外壳,不仅起到保护、缓释的作用,还能增强聚合物抑菌胶囊的生物相容性,附着在口腔黏膜上,增强抗菌效果和抗菌时间,并且2‑甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸‑乙醇酸共聚物外壳与金银花协同抑制口臭的效果更好,具备技术进步。
Description
技术领域
本发明涉及口腔气雾剂技术领域,特别涉及一种缓释抗菌口气清新剂及其制备方法。
背景技术
口腔环境适合微生物生长繁殖,当机体的自我口腔清洁功能下降,口腔致病菌大规模传播导致口腔微生态破坏,造成口腔卫生不洁、口腔炎症、异味等口腔疾病。通过选用相应的口腔护理方式清洁口腔,可以保持口腔和牙齿清洁,避免牙齿发黄、口臭等口腔疾病。进行口腔保健最有效和最方便的方法是使用口气清新剂,口气清新剂容易携带,还可以抑制细菌生长、快速清新口气,由于其易于吸收,效果快,无明显的毒副作用,作为口腔日常护理的一种补充产品,受到了年轻消费者喜爱,市场增长潜力巨大。
出于安全因素考虑,现在市场上销售的口气清新剂很多打着中草药概念,宣称具有安全杀菌、抑菌、治疗口臭作用。但实际上,单一的中草药对杀菌和抑菌的作用有限,主要还是由于添加的杀菌剂或抗生素起到的效果,并且通常需要加入表面活性剂或其他化学物质维持体系稳定。但是添加的杀菌剂或者抗生素可能会对使用者产生刺激,并不十分安全。
中国发明专利CN107714530A公开了一种用于去除口腔异味、抑菌、清新口气的口腔卫生喷剂,能够高效抑菌,对有害菌的抑菌率达99%,有效维护口腔菌群平衡,但其使用二氧化氯作为抗菌剂,虽然可以起到功效作用,但其安全性仍不能完全保证。中国发明专利CN101292945B公开了一种具有清除口腔异味、清新口气功效的组合物,通过多种途径达到清除口腔异味、清新口气的功效,完全以天然草本为主要原料,虽然不用担心安全问题,但是抗菌效果并不明显,而且持久性差,无法长期保持口腔清新的效果。
因此,有必要研究一种安全有效、无刺激的口腔喷雾,改善口腔微环境,缓解口腔气味问题,长期保持口腔健康。
发明内容
发明目的:本发明的目的是提供一种缓释抗菌口气清新剂及其制备方法,安全有效的延长喷雾在口腔的抑菌时间,改善口腔环境,适用于人和宠物使用。
本发明的技术方案:
一种缓释抗菌口气清新剂,以质量份数计,包括:
0.05-0.3份聚合物抑菌胶囊;
5-20份保湿稳定剂;
0.1-0.5份表面活性剂;
0.05-2份甜味剂;
0.06-0.1份pH值调节剂;
50-59份去离子水;
其中,所述聚合物抑菌胶囊的壳材为2-甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸-乙醇酸共聚物;芯材为抑菌植物精华。
通过采用上述技术方案,采用水作为主要溶剂来对喷雾中的组分进行溶解,并通过乳化剂的设置,使得难溶于水的组分于水中形成水包油型的乳化液体系,从而使得气雾剂在贮存和使用过程中以分散均匀且稳定的乳化液形式存在,一定程度上能够提高口腔气雾剂的作用效果。
优选地,所述抑菌植物精华的原料选自冬青、薄荷脑、迷迭香、天竺葵、绿茶、金银花、厚朴酚、三七中的一种或更多种组合。
优选地,所述抑菌植物成分包括冬青5-10份,薄荷脑20-40份,迷迭香5-10份,天竺葵6-15份,绿茶9-18份,金银花5-10份,厚朴酚9-15份和三七3-9份。
优选地,所述抑菌植物精华的制备方法包括,按照相应比例分别选取植物成分,混合搅拌后倒入粉碎机内粉碎;在粉碎后的混合物中加入清水,并进行蒸煮、过滤,得到混合液;采用蒸馏方式对混合液进行浓缩,得到浓缩液,萃取,得到所述抑菌植物精华。
优选地,所述聚合物抑菌胶囊的制备方法包括,
步骤1:将聚乳酸-乙醇酸共聚物溶于溶剂,完全溶解后加入二苯甲酮溶液中,室温避光浸泡,待置换成膜后取出,然后加入到2-甲基丙烯酰氧乙基磷酸胆碱的去离子水溶液中,用紫外光源照射反应,反应结束后,超声清洗,得到所述壳材;
步骤2:将所述抑菌植物精华溶于去离子水中形成水相,将壳材溶于有机溶剂中形成油相;抽取油相,加入到水相中,震荡超声获得油包水型乳液;抽取所述乳液,注入到聚乙烯醇/水混合溶剂中,升温搅拌,使溶剂挥发,得到产品微球,即为所述聚合物抑菌胶囊。
优选地,所述聚乳酸-乙醇酸共聚物与二苯甲酮的质量比为1:10-20。
优选地,所述2-甲基丙烯酰氧乙基磷酸胆碱的去离子水溶液的浓度为0.01-0.1mol/L。
优选地,所述紫外光的功率为200-300W,中心波长350±50nm。
优选地,所述抽取油相,加入到水相中,油相与水相的体积比为1:20-40mL。
优选地,所述升温搅拌,温度为80-100℃,搅拌转速为500-800rpm,搅拌时间为4-6h。
优选地,所述缓释抗菌口气清新剂还包括食用色素、酒精和维生素E中的一种或更多种组合。
优选地,所述保湿稳定剂为丙二醇或甘油;所述表面活性剂为Span20、Span40、Span60、Span80、Tween85和月桂酸单甘油酯中的一种或几种;所述甜味剂为糖精钠、安赛蜜、甜叶菊糖苷、木糖醇和山梨糖醇中的一种或几种;所述pH值调节剂为柠檬酸和碳酸氢钠的混合物。
通过采用上述技术方案,保湿稳定剂和表面活性剂起到增溶的作用,从而使得口腔气雾剂从喷雾瓶中喷出时呈分散均匀的液滴;而甜味剂能够增强口腔气雾剂的口感风味。
在另一方面,本发明还提供如上所述缓释抗菌口气清新剂的制备方法,具体包括,
将聚合物抑菌胶囊与表面活性剂混合,形成油相O;将其他原料溶解于水中形成水相W1,
将油相O加入至水相W中,高速分散后得到W/O型乳状液;
将W/O型乳状液进行灌装,获得口腔清新剂。
有益效果:
通过改性聚合物包覆植物抗菌精华,能够安全有效的延长喷雾在口腔的抑菌时间,改善口腔环境,缓解口腔气味问题,长期保持口腔健康。2-甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸-乙醇酸共聚物外壳,不仅起到保护、缓释的作用,还能增强聚合物抑菌胶囊的生物相容性,附着在口腔黏膜上,增强抗菌效果和抗菌时间。并且发现,2-甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸-乙醇酸共聚物外壳与金银花协同抑制口臭的效果更好,具备技术进步。
具体实施方式
以下将结合具体实施方案来说明本发明。需要说明的是,下面的实施例为本发明的示例,仅用来说明本发明,而不用来限制本发明。在不偏离本发明主旨或范围的情况下,可进行本发明构思内的其他组合和各种改良。
本发明所使用的化学试剂若无特殊说明,均为普通市售分析纯。实施例所使用聚乳酸-羟基乙酸共聚物(PLGA)的分子量为85000,LA∶GA=80∶20。
制备2-甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸-乙醇酸共聚物
称取5g PLGA,溶于50mL的N-甲基吡咯烷酮(NMP)中,得到PLGA/NMP溶液。待其完全溶解后,将所述PLGA/NMP溶液倒入1L、0.5mol/L的二苯甲酮/乙醇溶液中,于常温下避光浸泡3h,待置换成膜后取出表面吸附二苯甲酮的PLGA膜,以去离子水清洗后干燥。再将所述吸附二苯甲酮的PLGA膜置于浓度为0.05mol/L的2-甲基丙烯酰氧乙基磷酸胆碱的水溶液中,在300W,中心波长(350±50)nm的条件下进行紫外光照射,照射时间3h。反应结束后,取出2-甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸-乙醇酸共聚物,经去离子水超声清洗后,冷冻干燥,备用。
制备抑菌植物精华
按照相应比例取出天然植物共1000g,搅拌后倒入粉碎机内粉碎;在粉碎后的混合物中再加入清水3000克,蒸煮4h,过1500目滤筛,得到混合液。
对混合液进行浓缩,得到浓缩液,向浓缩液中加入乙酸乙酯,加热搅拌后静置萃取,得到抑菌植物精华。
制备聚合物抑菌胶囊
将芯材溶于去离子水中形成水相,将壳材溶于有机溶剂中形成油相;抽取3mL油相,加入到100mL水相中,200W震荡超声3min获得油包水型乳液;抽取所述乳液,注入到500mL聚乙烯醇/水(体积比2:1)的混合溶剂中,升温搅拌,直至溶剂挥发,得到产品微球,即为所述聚合物抑菌胶囊。
实施例1
将0.05份聚合物抑菌胶囊与0.5份表面活性剂(月桂酸单甘油酯)混合,形成油相O;将5份保湿稳定剂(丙二醇)、2份甜味剂(木糖醇)、0.1份pH值调节剂(柠檬酸)和50份去离子水混合形成水相W,
将油相O加入至水相W中,8000r/min分散10min后得到W/O型乳状液;
将W/O型乳状液进行灌装,获得口气清新剂1。
其中,所述聚合物抑菌胶囊的壳材为2-甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸-乙醇酸共聚物;芯材为5份冬青,40份薄荷脑,5份迷迭香为原料制备得到的抑菌植物精华。
实施例2
口气清新剂2的制备方法参照实施例1,区别仅在于,所述芯材为10份天竺葵,15份绿茶,5份金银花为原料制备得到的抑菌植物精华,记为口气清新剂2。
实施例3
口气清新剂3的制备方法参照实施例1,区别仅在于,所述芯材为10份天竺葵,15份绿茶,15份厚朴酚为原料制备得到的抑菌植物精华,记为口气清新剂3。
实施例4
口气清新剂4的制备方法参照实施例1,区别仅在于,所述芯材为绿茶为原料制备得到的抑菌植物精华,记为口气清新剂4。
实施例5
口气清新剂5的制备方法参照实施例1,区别仅在于,所述芯材为厚朴酚为原料制备得到的抑菌植物精华,记为口气清新剂5。
实施例6
口气清新剂6的制备方法参照实施例1,区别仅在于,所述芯材为金银花为原料制备得到的抑菌植物精华,记为口气清新剂6。
对比例1
口气清新剂7的制备方法参照实施例6,区别仅在于,所述壳材为聚乳酸-乙醇酸共聚物。
对比例2
口气清新剂8的制备方法参照实施例6,区别仅在于,将以金银花为原料制备得到的抑菌植物精华替换聚合物抑菌胶囊直接添加。
针对上述口气清新剂1-8进行性能测试:
实验1抑菌性能测试:采用上述口气清新剂1-8,分别选取琼脂并加入至无菌培养皿中,然后在无菌培养皿上接种培养基,根据GB/T20944.1-2007中的方法制备试验菌液,并接种到培养基上,将各组分口腔气雾剂喷雾至培养基上,在培养8h和24h时,观察培养基上菌种的面积变化,计算抑菌率。
实验2乳液稳定性测试:采用上述口气清新剂1-8,分别将各组分进行离心振荡,在3000r/min的转速下离心20min后静置,分别记录乳液的分层情况。
实验3口腔(人)挥发性硫化物测试:选择有口源性口臭或口气清新需求的人士,共有50人纳入研究,男25人,女25人,年龄20-50岁。50人分别使用美国Interscan公司生产的RH-17型口气测量仪测量口腔挥发性硫化物,作为对照,再使用上述口气清新剂1-8进行喷雾清新,洗脱期1天,记录重复喷雾3次后的口腔挥发性硫化物的数值,并计算每款口气清新剂使硫化物下降平均值。
实验4口腔(宠)挥发性硫化物测试:从宠物店选择50只宠进行实验,猫25只,狗25只,年龄品种均随机,实验步骤与实验3步骤相同,计算硫化物下降平均值。
上述实验数据如表1所示。

由表1可以看出在使用口气清新剂8h时,由于时间较短,口气清新剂1-6的抑菌率虽然略优于口气清新剂7-8,但不是特别明显。但是随着时间的推移,抑菌率差距开始变大:口气清新剂7中聚合物抑菌胶囊的壳材为聚乳酸-乙醇酸共聚物,和口气清新剂1-6相比,随着时间的延长抑菌率下降显著,说明2-甲基丙烯酰氧乙基磷酸胆碱接枝壳材可以延长口气清新剂抑菌的持久性和高效性,延长其在口腔中的作用时间。由口气清新剂1-6可知,金银花在口腔喷雾中发挥重要的抑菌作用,与-甲基丙烯酰氧乙基磷酸胆碱接枝壳材协同提高产品抑菌的高效性和持久性。
80-90%的口气是由口腔内的微生物引起的,特别是革兰氏阴性厌氧菌分解消化口腔内的滞留物质,产生挥发性硫化物及其他异味物质从而导致口气。通过对使用口气清新剂1-8的前后受试者的测试值变化情况,说明口气清新剂1-6可以减少挥发性硫化物的产生,因此对口腔炎症、清除口腔异味有特殊效果,安全无刺激,能够适合人与宠物使用。
本发明还可以由其它多种实施例,在不背离本发明精神及其实质的情况下,熟悉本领域的技术人员当可根据本发明作出各种相应的改变和变形,但这些相应的改变和变形都应属于本发明所附的权利要求的保护范围。
Claims (3)
1.一种缓释抗菌口气清新剂,其特征在于,以质量份数计,包括:
0.05-0.3份 聚合物抑菌胶囊;
5-20份 丙二醇;
0.1-0.5份 月桂酸单甘油酯;
0.05-2份 甜味剂;
0.06-0.1份 pH值调节剂;
50-59份 去离子水;
其中,所述聚合物抑菌胶囊的壳材为2-甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸-乙醇酸共聚物;芯材为抑菌植物精华;
所述抑菌植物精华的原料选自冬青、薄荷脑、迷迭香、天竺葵、绿茶、金银花、厚朴酚、三七中的一种或更多种组合;
所述抑菌植物精华的制备方法包括,按照配方选取植物成分,混合搅拌后倒入粉碎机内粉碎;在粉碎后的混合物中加入清水,并进行蒸煮、过滤,得到混合液;采用蒸馏方式对混合液进行浓缩,得到浓缩液,萃取,得到所述抑菌植物精华;
所述聚合物抑菌胶囊的制备方法包括,
步骤1:称取5g PLGA,溶于50mL的N-甲基吡咯烷酮中,得到PLGA/NMP溶液;待其完全溶解后,将所述PLGA/NMP溶液倒入1L、0.5mol/L的二苯甲酮/乙醇溶液中,于常温下避光浸泡3h,待置换成膜后取出表面吸附二苯甲酮的PLGA膜,以去离子水清洗后干燥;再将所述吸附二苯甲酮的PLGA膜置于浓度为0.05mol/L的2-甲基丙烯酰氧乙基磷酸胆碱的水溶液中,在300W、中心波长350±50nm的条件下进行紫外光照射,照射时间3h;反应结束后,取出2-甲基丙烯酰氧乙基磷酸胆碱接枝的聚乳酸-乙醇酸共聚物,经去离子水超声清洗后,冷冻干燥,备用,得到所述壳材;
步骤2:将芯材溶于去离子水中形成水相,将壳材溶于有机溶剂中形成油相;抽取3mL油相,加入到100mL水相中,200W震荡超声3min获得油包水型乳液;抽取所述乳液,注入到500mL聚乙烯醇/水体积比2:1的混合溶剂中,升温搅拌,直至溶剂挥发,得到产品微球,即为所述聚合物抑菌胶囊。
2.根据权利要求1所述缓释抗菌口气清新剂,其特征在于,所述缓释抗菌口气清新剂还包括食用色素、酒精和维生素E中的一种或更多种组合。
3.根据权利要求1-2任一所述缓释抗菌口气清新剂的制备方法,其特征在于,包括,
将聚合物抑菌胶囊与月桂酸单甘油酯混合,形成油相O;将其他原料溶解于水中形成水相W,
将油相O加入至水相W中,高速分散后得到W/O型乳状液;
将W/O型乳状液进行灌装,获得口腔清新剂。
