Abstract
本发明公开了亚油酰胺在调节树突状细胞活化及银屑病治疗中的应用。本发明以小鼠原代树突状细胞(DC)为研究对象,考察了亚油酰胺对抗原蛋白模拟物Eα‑GFP刺激下DC抗原加工呈递能力、LPS刺激下DC成熟度以及DC与T细胞共培养条件下对T细胞活化的影响,实验结果显示,Eα‑GFP刺激DC后,亚油酰胺能够明显抑制DC对抗原的呈递能力;LPS刺激DC后,亚油酰胺影响DC的成熟度,明显抑制DC的共刺激分子的表达;DC与T细胞共培养条件下,亚油酰胺明显抑制DC活化介导的T细胞增殖和细胞因子的分泌。同时,本发明通过咪喹莫特诱导的银屑病模型,发现亚油酰胺在缓解和治疗银屑病这类自身免疫性疾病中的作用,提供了亚油酰胺的新用途,且具有重要的经济效益和社会效益。

技术领域
本发明涉及亚油酰胺在调节树突状细胞活化及银屑病治疗中的应用,属于免疫调节技术领域。
背景技术
树突状细胞(DC)因其细胞形状如树突状而得名,负责启动适应性免疫反应,因此起着免疫系统“哨兵”的作用。DC是骨髓(BM)来源的白细胞,是专职性的也是最有效的抗原呈递细胞类型,负责启动所有抗原特异性免疫反应。因此,它们通过将先天免疫系统的微生物感知特征与适应性反应的精细特异性联系起来,是免疫反应的主要调节者。
银屑病是一种慢性炎症介导的免疫性疾病,全球患病率约为2~3%,主要影响皮肤和关节。银屑病的病理特征是角质细胞异常增殖和免疫细胞过度浸润,涉及先天和适应性免疫系统。目前临床已观察到银屑病的免疫异常包括树突状细胞的抗原呈递能力增强、NF-κB信号通路的过度激活、DC通过产生IL-12和IL-23等细胞因子促进特定TH细胞群的扩增(特别是TH17细胞,它是IL-17的主要来源)和IL-17细胞因子分泌的增强,从而促进宿主的免疫反应和免疫细胞的浸润。因此树突状细胞和T细胞过度活化在银屑病的进展中发挥重要作用。
目前临床上对银屑病的治疗方法主要包括局部和全身治疗,其中局部治疗被认为是轻中度银屑病的主要治疗方法。局部治疗相对安全、有效,患者耐受性较好,避免了系统用药对内脏系统的不利影响。然而,长时间局部外用可能会发生皮肤萎缩、毛细血管扩张、色素沉着,甚至出现刺激性皮炎等不良反应,若突然停药还能诱发重型银屑病。针对银屑病的全身治疗主要是口服和静脉注射药物。目前常用的口服药物有甲氨蝶呤(减少上皮层肥大,增加T细胞凋亡)、阿维A(干扰角质形成细胞的增殖),环孢霉素(减少IL-2和IFNγ生成)等,全身口服药物治疗根据药物的不同作用原理会一定程度缓解临床症状,但往往会也带来一定副作用,如胃肠道刺激、肝功能损伤、肾功能异常以及骨髓抑制等。除口服用药外,生物制剂亦可以通过静脉注射产生全身效应。生物制剂是从生物体中提取的糖蛋白,它们具有与免疫系统的特定目标相互作用的能力。已知的生物制剂包括单克隆抗体、细胞因子、反义寡核苷酸、融合蛋白和RNA,其中最常用于临床的生物制剂是TNFα和IL-17的抑制剂。这些生物制剂的药效相对更高,但价格昂贵。截至目前,银屑病的治疗已经有了很大的进展,但临床上仍然面临一定的主要挑战,如不同患者群体对治疗的不同反应、各种治疗方式所带来的副作用及不同患者对药物价格的可接受程度等。因此,亟需探索更高效、更安全且性价比高的策略以满足银屑病的临床需求。
亚油酰胺(Linoleamide)又称9,12-十八烯酰胺(9,12-Octadecadienamide)或亚油酸酰胺(Linoleic acid amide),是机体内源代谢物-初级脂肪酸酰胺家族中的成员。亚油酰胺于1989年首次在女性黄体期血浆中被发现并鉴定。1995年,亚油酰胺从睡眠剥夺的猫、大鼠以及人的脑脊髓液中分离出来,并被证明其作为内源睡眠诱导剂可诱导猫、大鼠和人类的睡眠。已有研究表明亚油酰胺可增加胞质中Ca2+水平,这可能是由于亚油酰胺对内质网Ca2+-ATP酶的抑制引起的。目前没有关于亚油酰胺在树突状细胞功能调控及银屑病应用中的报道。
发明内容
本发明的目的是提供一种亚油酰胺在调节树突状细胞活化及银屑病治疗中的应用,扩展了亚油酰胺的应用范围。
亚油酰胺能够用于抑制树突状细胞过度活化及银屑病治疗。
具体地,所述应用表现为下述1)-3)中任一种:
1)抑制DC的抗原加工和呈递能力;
2)抑制LPS诱导的DC相关的共刺激因子的表达;
3)抑制DC活化T细胞的能力。
本发明以小鼠原代树突状细胞(DC)为研究对象,考察了亚油酰胺对抗原蛋白模拟物Eα-GFP刺激下DC抗原加工呈递能力、LPS刺激下DC成熟度以及DC与T细胞共培养条件下对T细胞活化的影响。实验结果显示,Eα-GFP刺激DC后,亚油酰胺能够明显抑制DC对抗原呈递能力;LPS刺激DC后,亚油酰胺对DC的成熟度有影响,能够明显抑制DC的共刺激分子的表达,共刺激因子为CD80、CD86、MHC I和MHC II;DC与T细胞共培养条件下,亚油酰胺对DC活化T细胞的能力有影响,能够明显抑制T细胞的增殖和细胞因子的分泌,细胞因子为IFNγ、IL-17A、IL-4。这些结果表明亚油酰胺对DC相关的先天免疫和适应性免疫均具有显著抑制效果。
本发明通过咪喹莫特(IMQ)诱导的银屑病模型,考察了亚油酰胺对IMQ诱导的银屑病模型的影响,实验结果显示,与野生型小鼠相比,IMQ诱导的小鼠皮肤浸润更多的DC和T细胞,同时炎症因子表达更高,DC的成熟度更高,然而,使用50mg/kg的亚油酰胺对小鼠腹腔给药可以显著抑制免疫细胞的浸润、DC的成熟度以及炎症因子的表达。
本发明公开了亚油酰胺在调节树突状细胞活化及银屑病治疗中的作用,提供了亚油酰胺的新用途,扩展了亚油酰胺的应用范围,且具有重要的经济效益和社会效益。

实施例1、小鼠原代树突状细胞BMDC细胞的分离与分化
1、分离小鼠股骨
1)泡沫板放置一张吸水纸或铺一层保鲜膜。
2)小鼠颈椎脱臼处死,放在铺有泡沫板的吸水纸或保鲜膜上,小鼠全身酒精消毒、腹侧向上固定。
3)用镊子夹住小鼠后腿表皮,用剪刀向上剪至小鼠大腿根部,向下剪至小鼠的膝盖下1厘米左右处剪断。用镊子夹住剪断的小腿向上剥离,直至小鼠股关节处,露出白色球型的骨关节,从此处将整个股骨取下,剔除多余的组织,将股骨浸泡入PBS中。
4)取另一侧股骨,方法同上。
2、从小鼠股骨中分离单核细胞
1)物品准备:酒精灯、50ml离心管(用于放置培养基和PBS)、消毒止血钳、消毒纱布、1ml注射器、1640+10%血清+1%双抗;
2)用消毒的止血钳从双抗中夹取一根股骨,置于消毒纱布上,用纱布折叠包裹,双手固定,两个拇指放在膝关节后侧向上用力折断,断面呈现出梅花状,将吸有培养基的注射器针尖插入梅花状的中心,向下推动针尖直至不能继续为止。注射器连同股骨斜抵在50ml离心管内壁,缓缓向下推动注射器将培养基注入骨髓腔,将骨髓中的细胞冲出,骨髓颜色逐渐变白。(取另一侧骨髓中的单核细胞,方法同上)
3)取出的细胞计数后按2×106个/ml细胞量接种于6孔板中分化,每个孔溶液体积2mL,加10ng/ml的GM-CSF和5ng/ml的IL-4,37℃培养箱分化6天。培养至第三天每孔补充2mL含10ng/ml的GM-CSF和5ng/ml的IL-4的1640培养基。
实施例2、亚油酰胺对BMDC抗原加工呈递的效应检测
1、细胞接种
1)将6孔板中的上清收集至50mL离心管中,离心(1500rpm,5min);并使用PBS清洗细胞,离心(1500rpm,5min)。
2)离心结束,弃去上清,用新的培养基重悬混匀细胞,用移液枪吸取10ul进行计数,最终以每先孔1×105细胞量接种在24孔板中。
2、药物预处理:将亚油酰胺溶解于DMSO中配制1M的母液,再稀释成10、20mM工作液体。向24孔板中加入对应浓度的亚油酰胺,终浓度为50μM、100μM,各孔中DMSO的浓度保持一致。37℃培养箱,静置2小时。
3、EαEGFP刺激BMDC
亚油酰胺对BMDC细胞预处理后加入50μg/ml的EαEGFP,37℃培养箱,刺激24h。
4、BMDC抗原加工呈递能力的流式检测:将刺激后的BMDC收集至1.5ml离心管中,PBS进行清洗收集,1500rpm离心5min收集细胞沉淀,再用PBS清洗一遍后,使用100μl重悬沉淀,按照说明书比例添加流式抗体,冰上或4℃孵育30min,流式上机检测,结合EGFP和YAe的荧光强度评价抗原呈递能力。
实施例3、亚油酰胺对BMDC成熟度的效应检测
1、细胞接种
1)将6孔板中的上清收集至50mL离心管中,离心(1500rpm,5min);并使用PBS清洗细胞,离心(1500rpm,5min)。
2)离心结束,弃去上清,用新的培养基重悬混匀细胞,用移液枪吸取10μl进行计数,最终以每孔1×105细胞量接种在24孔板中,每孔溶液体积为500μl。
2、药物预处理:将亚油酰胺溶解于DMSO中配制1M的母液,再稀释成10、20mM工作液体。向24孔板中加入对应浓度的亚油酰胺,终浓度为50μM、100μM,各孔中DMSO的浓度保持一致。37℃培养箱,静置2小时。
3、LPS刺激BMDC
亚油酰胺对BMDC细胞预处理后加入1μg/ml的LPS,37℃培养箱,刺激24h。
4、BMDC成熟度的流式检测:将刺激后的BMDC收集至1.5ml离心管中,PBS进行清洗收集,1500rpm离心5min收集细胞沉淀,再用PBS清洗一遍后,使用100μl重悬沉淀,按照说明书比例添加流式抗体,冰上或4℃孵育30min,流式上机检测CD80、CD86、MHC I和MHC II表达量。
实施例4、亚油酰胺对BMDC活化T细胞的效应检测
1、BMDC细胞接种
1)将6孔板中的上清收集至50mL离心管中,离心(1500rpm,5min);并使用PBS清洗细胞,离心(1500rpm,5min)。
2)离心结束,弃去上清,用新的培养基重悬混匀细胞,用移液枪吸取10ul进行计数,最终以每孔1×105细胞量接种在24孔板中。
2、药物预处理:将亚油酰胺溶解于DMSO中配制1M的母液,再稀释成10、20mM工作液体。向24孔板中加入对应浓度的亚油酰胺,终浓度为50μM、100μM,各孔中DMSO的浓度保持一致。37℃培养箱,静置2小时。
3、LPS刺激BMDC
亚油酰胺对BMDC细胞预处理后加入1μg/ml的LPS,37℃培养箱,刺激过夜。
4、OVA323-339肽段刺激BMDC
次日,向24孔板的BMDC细胞中加入10μg/ml的OVA323-339肽段,37℃培养箱,刺激2h。
5、OTII T细胞分离
1)分离OTII小鼠脾脏细胞:小鼠颈椎脱臼处死,放在铺有泡沫板的吸水纸或保鲜膜上,小鼠全身酒精消毒、腹侧向上固定。左手用镊子夹起腹部的皮毛,右手用剪刀以中线位置由下而上剪开,左右手持镊子,在腹部约右下角找到脾脏(约1cm呈血红色,易分离);
2)取出脾脏放置配有70μm滤网的50ml离心管上,并加少许MACS auto buffer浸湿,用注射器的活塞端在滤网上进行研磨,直到看不到红色位置(期间加MACS auto buffer冲洗),收集滤液,1500rpm,离心5min;
3)离心后弃上清,使用1ml gibco ACK lysing buffer悬浮细胞沉淀用于裂解红细胞,室温裂解3~5min;
4)加入10mL MACS auto buffer终止裂解,混匀后,用新的70μm滤网(置于新的50mL离心管上)过滤,1500rpm,5min离心,吸弃上清,用1mL MACS auto buffer重悬细胞沉淀。
5)细胞计数:取10μl悬液混合10μl台盼蓝混匀后10μl取加到细胞计数板中,使用细胞计数仪进行计数。
6)孵育分离CD4 T细胞的抗体:小鼠CD4 T细胞分离磁珠试剂盒的抗体标准:2~3×107个/ml细胞,溶液总体积为100μl,对应添加抗体体积为10μl,按上述比例添加抗体体积,混匀,4℃旋转孵育5~10min;
7)孵育分离CD4 T细胞的磁珠:分别取20μl/管加入含抗体和细胞的悬液里(抗体:磁珠体积比=1:2),4℃旋转孵育10~15min;
拿出磁力架放置无菌台,将LS分选柱放置磁力架上,使用buffer(3ml/个柱子)对柱子进行清洗,之后下面放置50ml离心管(为T细胞分离做准备);
8)孵育磁珠之后分别向每管加500μl MACS auto buffer混匀后过柱子,之后再加3mL buffer清洗柱子,清洗3次,每次3ml,50ml离心管收集T细胞液,1500rpm,3~5min离心,吸弃上清;加1ml buffer重悬沉淀,取10μl悬液混合10μl台盼蓝混匀后10μl取加到细胞计数板中,使用细胞计数仪进行计数。
6、T细胞标记CFSE:
分选后的OTII T细胞使用室温的PBS洗涤一次,1500rpm,3~5min离心,吸弃上清,去除MACS auto buffer中的血清;使用1ml PBS重悬细胞,添加5μM CFSE 37℃避光孵育20min;向细胞中加入5倍于原始染色体积的培养基(至少含有1%的FBS)并孵育5min;
通过离心沉淀细胞并将它们重新悬浮在新鲜的预热完全培养基中。
以1×105cells/well,100μl/well,即1×106cells/ml,接种至96wells。
7、DC与T细胞共培养:
将预处理的DC细胞收集至1.5ml离心管中,DC细胞和CD4 T细胞共培养比例按细胞量比例DC:CD4+T细胞=1:2接种至96孔板中,即DC细胞2.5×104cell,100μl/well:T细胞5×104cell,100μl/well,37℃,72h。收集细胞前5h左右,每孔收集50μl上清冻存于-80℃用于后续ELISA检测;之后每孔添加一定比例的蛋白质转运抑制剂和PMA/ion混合刺激剂,使胞内可检测到的细胞因子信号放大。孵育4-5h后收集细胞进行后续流式检测分析。
8、流式检测T细胞增殖能力:CFSE标记后的T细胞会自发488激发光,随着细胞增殖代数增加,荧光强度会逐渐减弱,分析荧光的减弱程度可以分析细胞增殖;
9、流式检测T细胞分泌细胞因子能力:共培养后收集细胞,PBS清洗一遍,对其进行表面CD4流式抗体标记,标记后使用200μl/管的4%多聚甲醛对细胞进行固定,室温15~20min,之后使用破膜液进行清洗,使用破膜液配制胞内染色需要的流式抗体,按比例添加抗体后室温放置胞内染色45min~1h,之后使用破膜液进行清洗,流式上机检测细胞因子IFNγ、IL-4和IL-17A表达。
10、ELISA检查共培养上清中的细胞因子:
将冻存于-80℃的细胞上清液放置冰上融化,融化后使用ELISA kit分别检测细胞因子IFNγ、IL-4和IL-17A的含量,具体过程参考kit说明书。
实施例5、亚油酰胺对咪喹莫特(IMQ)诱导银屑病小鼠模型的影响
1、咪喹莫特(IMQ)诱导银屑病小鼠模型的建立
实验前一天使用脱毛器和脱毛膏对小鼠进行背部剃毛处理,背部脱毛面积2.5cm×2.5cm。从实验第一次给药开始算第0天,从第0天开始,每天在剃毛区域涂抹62.5mg咪喹莫特乳膏,同时每天对小鼠背部皮肤进行观察和疾病评分,并测量背部皮肤厚度。连续涂抹乳膏6天,第6天麻醉拍照,记录皮肤病变情况。脱颈处死后,分离小鼠脾脏和腋窝处的引流淋巴结放置在PBS中,脾脏用于拍照记录质量和大小变化,淋巴结用于分析T细胞数量变化以及DC数量和成熟度变化。分离小时背部皮肤,分2份,一份浸泡于4%多聚甲醛固定用于免疫组化分析,一份浸泡于Trizol中用于提取RNA分析炎症因子水平。
2、亚油酰胺对咪喹莫特(IMQ)诱导银屑病小鼠模型的影响:实验共分为三组,①对照组,每天腹腔注射100μl PBS,每天涂抹62.5mg凡士林乳膏,②模型组,每天腹腔注射100μl PBS,每天涂抹62.5mg咪喹莫特乳膏,③给药组,每天腹腔注射100μl亚油酰胺(50mg/kg),每天涂抹62.5mg咪喹莫特乳膏,连续给药6天。
3、Trizol法提取皮肤总RNA
1)取新鲜动物皮肤组织剪碎置于组织匀浆管中(同时加入2-3颗研磨珠),加入预冷的Trizol液800μl-1ml于匀浆管中,在组织研磨器中迅速匀浆,每30s暂停查看是否裂解好重复3次左右以充分研碎组织直到彻底裂解。此步骤可冻存于-80℃保存直到提取RNA。
2)裂解后的组织混合物3000rpm离心5min,取上清500μl,加入100μl氯仿,氯仿:Trizol=1:5;
3)使用振荡器剧烈震荡15-30s,(为紫红色)
4)静置5min后,4度15000rpm,10min或12000rpm,15min(上层Trziol+RNA,下层为蛋白质与残渣)(注:离心时间越长可使分层更加明显,易分离)。
5)分两次吸取上层透明液体至新的干净tubes(高压灭菌,无RNA酶),每管+80ul/100ul异丙醇(异丙醇:上清=1:1),上下颠倒(沉淀RNA);
6)静置10min沉淀RNA,4度,15000rpm,10min;
7)快速吸弃上清;
8)每管+750μl DEPC水配置-75%乙醇,上下颠倒(洗去DNA与蛋白质杂质)。
可冻于-80度;
9)4度,15000rpm,3min,8~9步骤重复一次;
10)吸弃上层清液;慢吸
11)余下的tubes在保鲜膜上置于细胞台上晾干;
12)加65度预热DEPC H2O,16-20ul/tubes;
13)使RNA溶解后,测RNA浓度,冻存于-20度。
4、mRNA水平检测皮肤细胞因子表达

qPCR体系:10μl,一个样品3个重复,加内参GAPDH

引物F+R 0.5μl
H2O 4.2μl
2×SYBR 5μl
引物/水/mix+SYBR/DNATem mix封口,混合-离心-程序-保存-开始-编名字。
以提取的RNA反转录为cDNA为模板检测小鼠皮肤的细胞因子IL-17A、IL-17F、IL-22、IL-23、IFNγ、TNF-α的表达。
实验结果:
1、亚油酰胺对EαEGFP刺激下BMDC抗原加工呈递的影响
本发明实施例2测定了小鼠原代树突状细胞中,亚油酰胺对EαEGFP蛋白刺激后,BMDC处理和呈递抗原能力的影响,结果如图1所示。
由图1可以看出,BMDC细胞在EαEGFP蛋白刺激24h后,可将EαEGFP蛋白进行加工并呈递,DC细胞呈递抗原能力与对照组相比显著增加。与此同时,100μM亚油酰胺处理可显著抑制DC细胞呈递Eα能力(p<0.01),提示亚油酰胺对BMDC的抗原加工和呈递能力具有显著抑制效果。
2、亚油酰胺对LPS刺激下BMDC成熟度的影响
为进一步探究亚油酰胺对BMDC后续活化成熟能力的影响,本发明实施例3测定了小鼠原代树突状细胞中,亚油酰胺对脂多糖LPS刺激后,BMDC表达共刺激因子CD80、CD86、MHC I和MHC II产生的影响,结果如图2所示。
由图2可以看出,BMDC在LPS刺激后,流式细胞术方法检测到细胞表面表达的CD80,CD86,MHC I和MHC II与未刺激组相比显著增加。与此同时,100μM亚油酰胺可显著抑制这些共刺激因子的表达(p<0.01),提示亚油酰胺对LPS诱导的BMDC激活具有抑制效果。
3、亚油酰胺对DC与T细胞共培养条件下T细胞活化的影响、:
在前期DC抗原呈递和成熟度检测的结果下,为了进一步明确亚油酰胺对DC功能的影响,本发明实施例4使用DC与OT II共培养体系研究亚油酰胺对DC后续活化CD4 T细胞的影响。结果如图3所示。
由图3可以看出,与未处理的BMDC相比,OVA肽段处理后的DC与T细胞共培养可以促进T细胞的增殖和相关的细胞因子如IFNγ,IL-17A,IL-4分泌,使用LPS和OVA肽段处理后的DC与T细胞共培养可以更显著的促进T细胞的增殖和相关的细胞因子分泌。相比使用100μM亚油酰胺处理后,BMDC对于T细胞的激活能力显著下降,具体表现在增殖能力和细胞因子分泌能力都显著下降。
4、亚油酰胺对IMQ诱导的小鼠银屑病的影响
本发明实施例5使用咪喹莫特(IMQ)诱导的银屑病模型作为体内模型探索亚油酰胺对自身免疫性疾病的影响。结果如图4所示。
由图4可以看出,与野生型小鼠相比,IMQ诱导的小鼠背部皮肤明显增厚,同时伴有鳞屑样皮屑和红肿,使用qPCR分析背部皮肤的炎症因子(IL-17A、IL-17F、IL-23、IFNγ和TNF-α)表达明显增加,然而,使用亚油酰胺对小鼠进行给药后,相比IMQ模型组小鼠,银屑病症状明显减轻,厚度降低,鳞屑样和红肿的现象明显减弱,使用qPCR分析背部皮肤的炎症因子(IL-17A、IL-17F、IL-23、IFNγ和TNF-α)表达明显降低。提示亚油酰胺对IMQ诱导的银屑病具有明显缓解作用。
Claims (9)
1.亚油酰胺在制备预防和/或治疗银屑病的药物中的应用。
2.根据权利要求1所述的应用,其特征在于:所述应用表现为下述1)-3)中任一种:
1)抑制DC的抗原加工和呈递能力;
2)抑制LPS诱导的DC相关的共刺激因子的表达;
3)抑制DC活化T细胞的能力。
3.根据权利要求2所述的应用,其特征在于:所述DC的抗原加工和呈递能力为Eα抗原和MHC复合体的表达。
4.根据权利要求2所述的应用,其特征在于:所述共刺激因子为CD80、CD86、MHC I和MHCII。
5.根据权利要求2所述的应用,其特征在于:所述DC活化T细胞的能力为T细胞的增殖和细胞因子IFNγ、IL-17A、IL-4的表达。
6.亚油酰胺在制备具有下述任一种功能的产品中的应用:
1)抑制DC的抗原加工和呈递能力;
2)抑制LPS诱导的DC相关的共刺激因子的表达;
3)抑制DC活化T细胞的能力。
7.根据权利要求6所述的应用,其特征在于:所述DC的抗原加工和呈递能力为Eα抗原和MHC复合体的表达。
8.根据权利要求6所述的应用,其特征在于:所述共刺激因子为CD80、CD86、MHC I和MHCII。
9.根据权利要求6所述的应用,其特征在于:所述DC活化T细胞的能力为T细胞的增殖和细胞因子IFNγ、IL-17A、IL-4的表达。
