本发明涉及疫苗学领域和免疫学领域,具体而言,涉及包含纳米颗粒的疫苗。
技术背景
SARS冠状病毒1(SARS-CoV-1)、新型冠状病毒(SARS冠状病毒2, SARS-CoV-2)和MERS冠状病毒(MERS-CoV)因人类感染蝙蝠或骆驼 CoVs5–8而出现。在灵猫、蝙蝠和马来亚穿山甲中循环的Beta冠状病毒(BetaCoVs)在遗传上与SARS-CoV-1和SARS-CoV-2相似,使用人ACE2 作为受体(Zhou P et al.A pneumonia outbreak associated with a newcoronavirus of probable bat origin.Nature 579, 270–273(2020);Li W et al.,Batsare natural reservoirs of SARS-like coronaviruses.Science 310,676–679(2005);Ge XY et al.,Nature 503,535–538(2013);Guan Y et al.,Science 302, 276–278(2003);11.Xiao K et al.Isolation of SARS-CoV-2-related coronavirus fromMalayan pangolins.Nature 583,286–289, doi:10.1038/s41586-020-2313-x(2020).[PubMed] [CrossRef][Google Scholar])。这些SARS相关动物冠状病毒有可能传播给人类。能够中和多种betaCoVs且能够预防或治疗betaCoVs感染的交叉中和抗体(cross-nAbs)已经从感染了SARS-CoV-1的人中分离出来,为开发betaCoV疫苗提供了概念验证。
目前已经开发或者进入临床应用的预防新冠疫苗感染的疫苗种类有mRNA疫苗、蛋白疫苗、铁蛋白纳米颗粒疫苗等。mRNA疫苗的基本原理是将编码一种或多种免疫原的转录本送入宿主细胞的细胞质中,在那里翻译产生免疫原蛋白,随后被封存在细胞内,纳入细胞膜,或分泌出来。刺激机体产生针对免疫原蛋白的中和抗体,从而中和入侵体内的病毒。蛋白疫苗,例如新冠疫苗重组亚单位疫苗,采用的是基因工程技术,在真核细胞(例如CHO细胞)内表达病原体抗原如RBD(receptor binding domain,受体结合域)蛋白,经过纯化加入氢氧化铝佐剂从而来制成。铁蛋白纳米颗粒疫苗(Saunders,K.O.et al.Neutralizingantibody vaccine for pandemic and pre-emergent coronaviruses.Nature (2021).https://doi.org/10.1038/s41586-021-03594-0)使用纳米颗粒技术,将RBD蛋白“展示”在由铁蛋白24聚体(ferritin 24-mer) 构成的纳米颗粒上,供免疫系统识别。同时使用一种叫做3M-052/明矾 (3M-052/Alum)的佐剂,进一步促进免疫反应,以实现更好的免疫效果。蛋白疫苗半衰期短,在体内容易降解;mRNA疫苗需要在体内翻译成蛋白,刺激产生抗体的过程比蛋白疫苗多了一个翻译步骤,推迟了抗体产生的时间。将免疫原蛋白偶联在铁蛋白纳米颗粒上,能够延长半衰期,预期可以取得更好的免疫效果。但是将铁蛋白作为免疫原蛋白的载体,铁蛋白本身就是一个免疫原,会刺激机体产生中和抗体,从而被清除出体内,大大影响纳米颗粒疫苗的免疫效果。另外制备铁蛋白颗粒疫苗需要同时制备生产两种蛋白——载体蛋白(铁蛋白)和免疫原抗原,从而导致铁蛋白纳米颗粒疫苗的工业化制备和生产成本高昂、工艺复杂,不容易生产出稳定的产品。本领域仍需要新的疫苗能够解决上述的缺陷。
发明内容
针对现有技术疫苗存在的缺陷,本发明人经过大量的研究,获得了一种新的疫苗制备方法,用无免疫原性的生物可降解材料例如PLGA制成纳米颗粒,其上偶联抗原蛋白,颗粒内部也可以包裹病毒抗原或mRNA, 制成PLGA纳米颗粒疫苗,颗粒表面的抗原蛋白首先在体内产生高滴度的中和抗体,颗粒内部的抗原或mRNA(如果有的话)持续被释放出来, mRNA表达成蛋白,刺激机体长时间持续产生抗体,并且刺激产生CD4+ 和CD8+T细胞免疫反应。在预防病毒感染和清除体内病毒方面效率更高,并且能够中和各种病毒突变株。
因而,在第一方面,本申请提供一种疫苗,其包含免疫原性组合物,所述免疫原性组合物包含生物可降解纳米颗粒和结合于所述纳米颗粒表面上的一种或更多种病原体特异性抗原,其中:
所述抗原是新型冠状病毒(SARS-CoV-2)的全长S蛋白(刺突蛋白) 和/或其受体结合域(RBD),或者可以包含所述的全长S蛋白和/或其RBD。
所述生物可降解纳米颗粒可以为生物可降解聚合物纳米颗粒,包括但不限于:聚乳酸-羟基乙酸、聚乳酸、聚己内酯、聚丁二醇丁二酸酯、聚苯胺、聚碳酸酯、乙丙交酯共聚物或乙交酯己内酯共聚物纳米颗粒中的任意一种或至少两种的混合物;优选为选自聚乳酸-羟基乙酸(PLGA)、聚乳酸(PLA)和聚己内酯(PCL)的纳米颗粒,更优选为PLGA纳米颗粒。
所述抗原可以通过静电作用、疏水作用、氢键在内的物理吸附、化学共价偶联或特异性结合而结合到所述纳米颗粒上。优选所述抗原通过化学共价偶联结合到所述纳米颗粒上。
所述疫苗可以包含佐剂。所述佐剂可以选自包括以下的组:氢氧化铝、磷酸铝、单磷酰脂质、寡核苷酸CpG、明矾(alum)和3M-052,例如可以为氢氧化铝、明矾或明矾与3M-052的混合物。
该方面的疫苗可以用于预防或治疗新型冠状病毒感染或新型冠状病毒肺炎。
本申请第二方面提供第一方面所述的免疫原性组合物在制备疫苗中的用途。所述疫苗可以用于预防或治疗新型冠状病毒感染或新型冠状病毒肺炎。
第三方面,本申请提供一种预防或治疗新型冠状病毒感染或新型冠状病毒肺炎的方法,包括给有需要的人士施用第一方面的疫苗。
本申请的所述疫苗中所使用的抗原也可以来自其他病原体,例如来自其他病毒、细菌、寄生虫。
因此,本申请的第四方面提供一种疫苗,其包含免疫原性组合物,所述免疫原性组合物包含生物可降解纳米颗粒和结合于所述纳米颗粒表面上的一种或更多种病原体特异性抗原。
所述抗原可以为蛋白或多肽。所述病原体可以是病毒、细菌、寄生虫,例如可以是病毒,例如HIV、HPV、HCV、HBV、EB、水痘带状疱疹病毒、寨卡病毒、狂犬病、流感病毒、巨细胞病毒、埃博拉病毒等。所述抗原可以是适合用作预防或治疗所述病原体感染的病原体特异性免疫原。

在本申请的上述方面的技术方案中,所述纳米颗粒内部可以进一步包裹所述抗原和/或其他抗原或者编码所述抗原和/或所述其他抗原的 mRNA。在这种情况下,纳米颗粒表面的抗原蛋白首先在体内产生抗体,纳米颗粒内部的抗原或mRNA持续被释放出来,mRNA表达成蛋白,刺激机体长时间持续产生抗体,并且刺激产生CD4+和CD8+T细胞免疫反应。在预防细菌、病毒等感染和清除体内细菌、病毒等方面效率更高,并且能够中和各种突变株。

冠状病毒是一种带有RNA的正向包膜病毒,其基因组大小约为26~ 32kb。基因组RNA和磷酸化核衣壳(N)蛋白被埋在磷脂双层中并被刺突糖蛋白(S)三聚体覆盖,膜(M)蛋白(III型跨膜糖蛋白)和包膜(E)蛋白位于病毒包膜的S蛋白之间。冠状病毒是在自然界中广泛存在的一类病毒,可引起包括呼吸道、消化道及神经系统在内的多系统疾病。
新型冠状病毒(SARS-CoV-2)的S蛋白位于病毒最外层,在膜上规则排列成冠状结构,参与病毒与宿主细胞表面的病毒受体结合并介导病毒通过膜融合进入细胞的过程,在诱导宿主产生中和抗体过程中起重要作用。目前COVID-19疫苗的研发是以全长S蛋白或部分S蛋白(S蛋白中的受体结域(RBD))作为目标抗原。

本申请所述的新型冠状病毒的S蛋白或其RBD蛋白可以是新型冠状病毒任何变体或谱系的S蛋白或其RBD蛋白。
生物可降解纳米颗粒
生物可降解纳米颗粒已经用于药物、疫苗和各种其他生物分子的递送。在本申请中,生物可降解纳米颗粒可以为生物可降解聚合物纳米颗粒,包括但不限于:聚乳酸-羟基乙酸、聚乳酸、聚己内酯、聚丁二醇丁二酸酯、聚苯胺、聚碳酸酯、乙丙交酯共聚物或乙交酯己内酯共聚物纳米颗粒中的任意一种或至少两种的混合物。优选选自聚乳酸-羟基乙酸(PLGA)、聚乳酸(PLA)和聚己内酯(PCL)纳米颗粒。这些聚合物纳米颗粒在体内经历水解,产生生物可降解代谢物单体,如乳酸、羟基乙酸等,最终降解为CO2和水。
这些纳米颗粒(NPs,nanoparticles)的制备方法是本领域已知的。例如,PLGA NPs常常通过乳化-扩散、溶剂蒸发和纳米沉淀法来制备。 PLA NPs通常通过溶剂蒸发、溶剂替代、盐析和溶剂扩散法来制备。PCL NPs通常通过纳米沉淀、溶剂替代和溶剂蒸发法来制备。这些纳米颗粒也可以从市场上购得。
所述纳米颗粒的平均粒径可以例如为大约10-990nm,例如为约 900nm、约850nm、约800nm、约750nm、约700nm、约650nm、约600nm、约550nm、约500nm、约450nm、约400nm、约350nm、约300nm、约250nm、约200nm、约150nm、约100nm或小于100nm,例如大小为约90nm、约80nm、约70nm、约60nm、约50nm、约40nm、约30nm、约20nm/或约10nm。优选纳米颗粒的平均粒径在10-500nm范围内,更优选在10-300nm范围内。
免疫原性组合物
本申请所述的免疫原性组合物包含生物可降解聚合物纳米颗粒和结合于所述纳米颗粒(表面)上的抗原(免疫原),其中所述的抗原和生物可降解聚合物纳米颗粒如上所述。
所述抗原(免疫原)可以包含或是新型冠状病毒的S蛋白或其RBD 蛋白。
所述抗原也可以是其他病原体特异性的抗原。所述病原体例如可以是病毒,例如HIV、HPV、HCV、HBV、EB、如寨卡病毒、狂犬病、流感病毒、巨细胞病毒、埃博拉病毒等。所述病原体也可以细菌或寄生虫。所述抗原也可以是适用于制备预防或治疗癌症的疫苗中的癌症特异性抗原。
制备所述免疫原性组合物的方法是本领域已知的。所述抗原可以通过静电作用、疏水作用、氢键在内的物理吸附、化学共价偶联或特异性结合而结合到所述纳米颗粒上,优选通过化学共价偶联结合到所述纳米颗粒上。例如,蛋白或多肽-PLA纳米颗粒免疫原性组合物可以通过将市购的、带有表面羧基的聚乳酸(PLA-COOH)用MES缓冲液洗涤,并在该缓冲液中与抗原混合并孵育来制得。蛋白或多肽-PLGA纳米颗粒免疫原性组合物例如可以按照本申请实施例中所述的方法来制备。
所述纳米颗粒内部还可以包裹所述抗原或其他抗原或者编码抗原的mRNA。在这种情况下,纳米颗粒表面的抗原蛋白首先在体内产生抗体,纳米颗粒内部的抗原或mRNA持续被释放出来,mRNA表达成蛋白,刺激机体长时间持续产生抗体,并且刺激产生CD4+和CD8+T细胞免疫反应。在预防细菌、病毒等感染和清除体内细菌、病毒等方面效率更高,并且能够中和各种突变株。
本申请的免疫原性组合物用于疫苗,不是人工抗原呈递细胞 (artificialantigen-presenting cell,aAPC),不包含肽-MHC复合物。
本申请所述的抗原不是基于新型冠状病毒SARS-CoV-2RBD-SD1蛋白设计的自组装蛋白。所述SARS-CoV-2SARS-CoV-2RBD-SD1蛋白的氨基酸序列如图5所示。
疫苗
本申请还涉及疫苗,其包含本文所述的免疫原性组合物。
本申请的疫苗还可以包含额外的佐剂。所述佐剂包括但不限于选自以下的一种或多种:氢氧化铝、磷酸铝、单磷酰脂质、寡核苷酸CpG、明矾(alum)和3M-052,例如可以为氢氧化铝、明矾或明矾与3M-052 的混合物。
所述佐剂的用量为本领域技术人员所熟知。例如,每剂所述疫苗中佐剂的含量可以为1μg~1000μg,优选10μg~500μg,更优选10~ 100μg,最优选每剂量10~50μg。
本申请所述的疫苗可以为任何药学上可接受的剂型,例如可以为针剂或雾化剂。所述疫苗免疫方式可以为肌肉注射、皮下注射、皮内注射、微针注射、经鼻喷雾给药中至少之一种。
预防或治疗方法
本申请还涉及一种预防或治疗病原体感染或由其引起的疾病、或者预防或治疗癌症的方法,包括向受试者施用前面所述的疫苗。
在本文中所使用的术语“施用”指将预定量的物质通过某种适合的方式引入受试者或有需要的个体,包括人类或动物。本申请的疫苗可以通过任何常见的途径给药,只要它可以到达预期的组织。给药的各种方式是可以预期的,包括腹膜、静脉、肌肉、皮下、皮层、口服、局部、鼻腔、肺部和直肠,但是本发明不限于这些已举例的给药方式。然而,由于口服给药时,口服给药的疫苗的活性成分应该被包被或被配制以防止其在胃部被降解。优选地,本发明的疫苗可以注射制剂被给药。此外,本发明的疫苗可以使用将活性成分传送到靶细胞的特定器械来给药。
本发明实施例中的疫苗的给药频率和剂量可以通过多个相关因素被确定,该因素包括要被治疗的疾病类型、给药途径、病人年龄、性别、体重和疾病的严重程度以及作为活性成分的药物类型。
本申请的技术方案与以往技术相比有以下有益效果:
(1)本申请的包含聚合物(如PLGA)纳米颗粒的疫苗中所采用的聚合物如PLGA、PLA是生物可降解的合成高分子材料,由C、H、O 构成,没有免疫原性,不会刺激机体产生抗体,不会被抗体清除。降解后PLGA和PLA纳米颗粒被分解成为乳酸和羟基乙酸、最终分解为 H2O和CO2,乳酸和羟基乙酸都是机体内天然存在的,因此不会对机体产生任何毒害作用。PLGA具有优良的生物相容性、生物降解性以及低免疫原性、低毒性和较好的机械强度,经美国食品与药品管理局批准可以用于药物控释载体。铁蛋白纳米颗粒疫苗即蛋白质微球制剂与传统化学合成的药物相比,存在分子大、稳定性差、容易被胃肠道中蛋白质水解酶降解以及生物半哀期短、利用度不高、很难通过生物屏障等缺点。而PLGA聚合物可以被用来构造成各种疫苗、蛋白质、大分子物质药物的长效和缓释载药系统,能有效解决上述问题,是近年来备受重视的蛋白质类药物的缓释材料之一。开发聚合物如PLGA纳米颗粒疫苗的方法之一是开发含有抗原的控制释放纳米微球,这类微球在注射后能够缓慢释放出抗原,用控制释放技术制成的疫苗能够达到长时间持续产生抗体,终身抵抗病毒感染的目的。PLGA类微球不仅在生物降解、释放的适配率等方面具有优异的特性,PLGA类微球更吸引人的方面是其除了能够引起抗体应答外,还可以引起T细胞免疫应答,这使PLGA纳米颗粒疫苗对于抵抗由细菌、病毒和寄生虫引起的细胞内感染以及彻底清除体内病毒具有重要的意义。
(2)铁蛋白纳米颗粒疫苗的制备需要同时生成两种蛋白—铁蛋白和抗原蛋白。聚合物如PLGA纳米颗粒疫苗制备过程简便易行(见具体实施例),只需将抗原蛋白连接在PLGA纳米颗粒上即可,工艺较易放大,成本低廉。
(3)本申请实施例表明,将抗原(免疫原)蛋白偶联在PLGA 纳米颗粒表面所制成的聚合物如PLGA纳米颗粒疫苗,所产生的总抗体滴度远远高于未偶联纳米颗粒的蛋白疫苗,并且刺激机体产生了很强的T细胞免疫应答。
实施例
提供以下实施例用于说明本发明,而非限制本发明的权利要求。
实施例1.纳米颗粒疫苗的制备
按照以下步骤制备纳米颗粒疫苗。
1制备RBD或S蛋白纳米颗粒疫苗
1.1.将PLGA(Resomer,货号:5050DLG 2.5A,PS5117)用丙酮溶解制成PLGA NPs乳液,PLGA NPs经电镜检查,粒径为约100nm(参见图1)。
1.2.根据需求取一定量3.5mg/mL的PLGA NPs乳液,
1.3.加入MES,磁力搅拌10min,充分溶解,取样,测pH(pH范围:5.3-5.8)。
1.4.加入EDC和NHS,磁力搅拌10min;
1.5.用HEPES,总体积12.5倍体积置换清洗PLGA NPs乳液,0.22 μm过滤器过滤。
1.6.加入RBD蛋白或S蛋白(购自广东华南疫苗股份有限公司),并充分混合均匀。在磁力搅拌器上室温搅拌1h,4℃静置过夜(≥18h), 0.22μm过滤装置过滤。
1.7.通过切向流装置用WFI(最低10L,若有需要可提高用量)清洗连接了蛋白的PLGA NPs乳液,检测蛋白浓度,4℃保存。
实施例2免疫小鼠
将以RBD蛋白或S蛋白作为免疫原蛋白、如上所述制备的连接了蛋白的PLGA NPs乳液,作为PLGA纳米颗粒疫苗(不含佐剂),对照组为 RBD蛋白或S蛋白疫苗(没有偶联在PLGA纳米颗粒上,直接使用),同时比较了30μg/只和10μg/只的不同剂量,通过肌肉注射的方式免疫 BalB/C小鼠(4-5周,18-22克/只),每组8只小鼠,一共免疫三次,每次间隔3周,免疫后三周取血,检测其中的总抗体滴度以及取脾脏检测T细胞免疫应答。
此外,将上述制备的以S蛋白作为免疫原蛋白的PLGA纳米颗粒疫苗置于雾化器中,对照组为S蛋白疫苗(没有偶联在PLGA纳米颗粒上,直接使用),通过经鼻喷雾的方式免疫BalB/C小鼠(4-5周,18-22克/ 只),每组8只小鼠,一共免疫两次,每次间隔3周,免疫后三周取血,检测其中的总抗体滴度。
实施例3.体液免疫应答——血清总IgG抗体滴度检测
用0.05M碳酸盐缓冲液(pH 9.6)将纯化后蛋白稀释至0.5μg/mL,包被96孔板(100μL/孔),4℃孵育过夜;使用10%NBSC封闭(100μ L/孔),37℃孵育2h;用PBS倍比稀释待测血清,加入96孔板,37℃孵育1h;加入HRP标记的羊抗鼠IgG(1:10000),37℃孵育1h;OPD底物显色(100μL/孔),37℃反应15min;2mol/L硫酸(50μL/孔)终止反应;检测A450。A450数值大于0.1且大于阴性对照的2.1倍最大血清稀释度视为特异性IgG抗体滴度。
结果分别示于图2和图3。
由图2和图3可以看出,将抗原蛋白(RBD蛋白或S蛋白)偶联于 PLGA上,用所述抗原偶联的PLGA进行免疫,相对于未经偶联的抗原而言,可以显著提高血清总IgG抗体滴度。例如,图2显示,偶联于PLGA 的免疫原RBD蛋白——2-针剂(30μg+PLGA)、甚至1-针剂(10μg+PLGA) 的终点效价都显著高于未偶联的免疫原RBD蛋白——3-针剂(30μg);偶联于PLGA的免疫原S蛋白——8-针剂(5μg+PLGA)的终点效价显著高于未偶联的免疫原S蛋白——9-针剂(5μg)。
实施例4细胞免疫应答
T细胞免疫应答检测是采用EliSpot方法检测脾脏T细胞分泌的 IFN-Y和IL-4。取出试剂盒中的96孔包被板条,每孔加入200μl无菌的PBS(1X)洗涤4次,每孔加入200μl含有10%FBS的RPMI-1640培养基,室温静置30分钟后将其液体扣出;根据细胞计数结果,将细胞浓度调整到2.5×106个/ml,96孔板每孔铺细胞数为2.5×105个/100μ l,每个样品铺2个孔,检测样品每孔加入含1μg蛋白的纳米颗粒疫苗刺激,阳性对照孔加入1μg蛋白刺激,另外设置2个孔铺细胞不加蛋白刺激作为阴性对照,另2个孔加入CTL-Test无血清培养基作为空白对照;培养板置于37℃、5%CO2培养箱培养36h;倾倒孔内液体,加 PBS 200μl/孔洗涤5次,每次停留1min,最后一次,在吸水纸上扣干;用含有0.5%胎牛血清的PBS稀释检测抗体至1μg/ml(1:1000稀释),将稀释好的抗体工作液加入各实验孔,100μl//孔。室温孵育2小时;倾倒孔内液体,加PBS 200μl/孔,洗涤5次,每次停留1min,最后一次,在吸水纸上扣干;用含有0.5%胎牛血清的PBS以1:1000稀释 Streptavidin-ALP,加入各实验孔,100μl//孔。室温孵育1小时;倾倒孔内液体,加PBS 200μl/孔,洗涤5次,每次停留1min,最后一次,在吸水纸上扣干;用0.45μm的过滤器过滤待用底物溶液 (BCIP/NBT-plus),100μl/孔,室温避光静置5-30分钟,直至出现明显的斑点后停止显色;倾倒孔内液体,用去离子水/自来水洗涤终止显色,如果需要揭开板底座,用去离子水/自来水洗涤正反面及底座。将板放置在室温阴凉处,待其自然晾干后合上底座。ELISPOT板斑点计数,并记录斑点的各种参数,做统计分析。
结果示于图4。
由图4可以看出,将抗原蛋白(RBD蛋白或S蛋白)偶联于PLGA上,用所述抗原偶联的PLGA进行免疫,相对于未经偶联的抗原而言,可以显著提高T细胞免疫应答。例如,偶联于PLGA的同等量(如2-肌注-RBD (30μg+PLGA))、甚至更低量的免疫原蛋白(如1-肌注-RBD(10μg+PLGA))的IFN-γ量,显著高于未偶联的免疫原RBD蛋白(3- 肌注-RBD(5μg))的IFN-γ量。
由实验结果可以看出,将免疫原偶联于纳米颗粒上,可以显著提高疫苗的免疫效果。
尽管不希望受理论的束缚,但本发明人推测:因纳米颗粒上可以偶联多个免疫原(抗原),成为多价抗原,引起的免疫应答(包括体液应答和细胞应答)相对于未经偶联的单价抗原(免疫原)而言显著更强。
以冠状病毒为例,首先制备PLGA或其他聚合物纳米颗粒(任选地将新型冠状病毒S蛋白或RBD(receptor binding domain)蛋白或者编码所述蛋白的mRNA包裹进去),将冠状病毒的S蛋白或RBD蛋白偶联在纳米颗粒上,制成纳米颗粒疫苗,注射到体内,可以持续产生大量抗体,有效针对不同的冠状病毒突变株,从而开发出“泛冠状病毒疫苗”。这项技术不仅可以运用于开发冠状病毒疫苗,也可以运用于其他病毒疫苗的开发,例如HIV,HPV,HCV,EB,如寨卡病毒、狂犬病、流感病毒、巨细胞病毒、埃博拉病毒等,也可以运用于其他细菌、寄生虫疫苗的开发。
根据本申请公开内容,不需要过度实验就可以制备和使用本文中所公开和要求保护的所有疫苗、免疫原性组合物和方法等。尽管已经就优选实施方案描述了疫苗和方法等,但本领域技术人员应显而易见,可在不背离本发明的理念、精神和范围的情况下对本文中所描述的疫苗、免疫原性组合物和方法以及方法中的步骤或步骤顺序施加变化。更具体地说,可以用化学上和生理学上相关的某些试剂来替代本文中所描述的试剂,同时可实现相同或类似的结果。认为对本领域技术人员显而易见的所有此类相似的取代和修改都在如由所附权利要求所限定的本发明精神、范围和理念之内。
Claims (10)
1.一种新型冠状病毒疫苗,其包含免疫原性组合物,所述免疫原性组合物包含纳米颗粒和结合于纳米颗粒表面上的一种或更多种抗原,其中所述纳米颗粒选自以下的生物可降解聚合物纳米颗粒:聚乳酸-羟基乙酸、聚乳酸、聚己内酯、聚丁二醇丁二酸酯、聚苯胺、聚碳酸酯、乙丙交酯共聚物和乙交酯己内酯共聚物,优选选自聚乳酸-羟基乙酸、聚乳酸和聚己内酯,更优选为聚乳酸-羟基乙酸,所述抗原为新型冠状病毒特异性的抗原。
2.如权利要求1所述的疫苗,其中所述抗原包括选自新型冠状病毒的刺突蛋白和其受体结合域的一种或多种蛋白或多肽。
3.如权利要求2所述的疫苗,其中所述抗原包含一个或多个所述受体结合域。
4.如权利要求2所述的疫苗,其中所述抗原包含刺突蛋白。
5.如权利要求2所述的疫苗,其中所述抗原为所述受体结合域。
6.如权利要求2所述的疫苗,其中所述抗原为所述刺突蛋白或其包含受体结合域的片段。
7.如权利要求1-6中任一项所述的疫苗,其中所述抗原通过静电作用、疏水作用、氢键在内的物理吸附、化学共价偶联或特异性结合而结合到所述纳米颗粒上。
8.如权利要求7所述的疫苗,其中所述抗原通过化学偶联结合于所述纳米颗粒上。
9.如权利要求1-8中任一项所述的疫苗,其中所述纳米颗粒内部还包裹有所述抗原或其他抗原或者包含编码所述抗原或其他抗原的核苷酸序列的mRNA。
10.如权利要求1-9中任一项中所述的免疫原性组合物在制备用于预防或治疗新型冠状病毒感染或新型冠状病毒肺炎的疫苗中的用途。
