Abstract
本发明提供原位选择性监测铅有效态的薄膜梯度扩散方法,包括非晶型硅酸锰(MnSi)的制备及其通过相转化技术制备MnSi负载的PVDF(PMn)膜,并应用于薄膜梯度扩散技术(DGT)装置结合相。步骤1.采用简易的共沉淀法80℃水浴制备MnSi;步骤2.通过相转化技术将MnSi负载于PVDF膜孔中,制备PMn膜;步骤3.将PMn膜代替DGT结合相,按照过滤膜在上,扩散相在中间,结合相在底部的方式组装;步骤4.探究PMn‑DGT的性能。本发明制备出了一种对铅具有高选择性和高吸附性能的MnSi的环境功能材料,并且将其转化为PMn膜,并利用该膜进行铅的原位监测。

技术领域
本发明属于环保技术领域,尤其涉及一种原位选择性监测铅有效态的薄膜梯度扩散方法。
背景技术
土壤作为陆地生态系统中最为重要的组成部分之一,是人类赖以生存和发展的最基础的自然资源。然而科技和经济快速发展遗留的历史环境问题正在影响人类的生活质量,其中土壤重金属污染因其隐蔽性、不可逆性和长期性,正成为影响全球人类健康和环境的问题。土壤重金属污染关系生态系统平衡和农产品和食品的质量安全,及时发现并解决问题是阻止污染扩散的关键。然而土壤基质成分复杂,重金属在土壤中的存在形态多种多样,并且pH、有机质含量、氧化还原电位以及环境的变化等因素均会影响重金属的迁移转化,改变土壤中重金属的生物有效态的含量。因此,如何有效监测土壤中重金属的生物有效态含量是目前研究人员关注的焦点。为了促进我国的可持续发展,原位监测土壤重金属生物有效态含量的变化情况以及及时采取有效的修复措施对保护我国的耕地面积和提高人类的生活品质具有重要的意义。
众所周知,重金属的毒性和生物积累主要与它们的生物可利用部分有关,即实际进入受体并影响生物体的元素部分,而不是总浓度。基于此,重金属生物有效态的监测不仅对环境危害风险评估至关重要,而且对污染修复也不可或缺。目前,关于土壤中重金属的存在形态,暂时还没有一个公认的分析步骤和标准。但是关于土壤中重金属的生物有效态的界定,不同研究领域的科学家,鉴定略有不同。环境学科的研究学者认为:土壤中能够被生物吸收、累积或者能产生毒害效应的形态,被定义为重金属有效态或生物有效态。原因在于土壤中这部分重金属活性强、毒性大,能随着土壤孔隙水的迁移而迁移转化,并且较为容易被植物的根系吸收进入食物链。除此之外,土壤环境氧化还原电位、微生物的行为以及pH的改变均能影响生物有效态的含量。
目前,在环境学科领域原位监测土壤重金属有效态的方法主要包括传统化学浸提法(Tessier,BCR等)、薄膜梯度扩散技术等。其中DGT技术提供了一种新型的原位被动监测策略,为准确、有效地评估环境中不稳定、生物可利用性组分中的目标物质浓度提供了应用潜力。与其他几种被动采样方法相比,DGT技术因其装置构造简单、现场部署方便、对环境的适用性高,低检测限和高元素能力、评估分析物的生物可利用性分数以及测量目标的多功能性而受到研究人员的广泛关注。
目前,自DGT技术问世以来,研究人员在结合相的优化和研发方面做出了巨大的努力,研发了各种各样的结合相材料。早期使用的结合相为一种离子交换树脂的聚丙婦醜胺凝胶(Chelex-100),它对Cu、Cd、Mn、Ni等部分金属离子均有较好的吸附和富集,能适应复杂的环境干扰和变化。但无论DGT技术用于何种污染物的监测,其核心部分在于结合相对待测物质的高吸附和解吸性能。然而,基于环境金属的多元化以及部分地区污染物的典型性,原位选择性分析典型重金属污染物的有效态含量对于评估其毒性以及监测变化情况尤为重要。但是,如果监测地区污染严重,有效态含量高,则需要将更多的结合剂浸渍到一个单一的结合相中,反过来看这又增加了制备结合凝胶的难度,因为在凝胶铸造过程中所需的固体物质的数量也增加了。此外,使用更多类型的结合剂可能会导致在动力学和能力方面的DGT性能显著降低。虽然目前使用的DGT结合相可高效吸附待测物质并在酸溶剂中解离,但解离时间长,一方面增加了DGT的时间成本,另一方面易造成监测结果偏差。除此之外,一方面增加了DGT的时间成本,另一方面易造成监测结果偏差。除此之外,结合相材料制备复杂、成本高、特征选择性差等问题是DGT装置大面积应用的瓶颈之一。
鉴于土壤重金属污染监测和修复的计数难题,特别是硅酸盐环境功能材料的开发及应用方面的局限性,有必要提出一种高选择性和高吸附性能的功能材料作为结合相吸附剂,解决DGT技术在土壤监测方面选择性差等问题。
发明内容
本发明的目的在于解决上述现有技术存在的缺陷,提供原位选择性监测铅有效态的薄膜梯度扩散方法,其中方法采用一种合成简单的高选择性和高吸附性能的环境功能材料作为结合相吸附剂。
本发明采用如下技术方案:
高选择性和高吸附性能的环境功能材料制备方法包括:
步骤1.在搅拌器的作用以及80℃水浴条件下,将Na2SiO3溶液滴加到MnCl2前体溶液中(按照Mn:Si摩尔比为1:1),然后待絮状物产生并稳定后持续反应2h得到MnSi颗粒物;
步骤2.产物离心后收集,用超纯水多次洗涤以去除其他杂质,将收集的材料冷冻干燥48h并储存在样品袋中贴标签,备用。
高选择性和高吸附性能的环境功能材料,由高选择性和高吸附性能的环境功能材料的制备方法制得。
一种高选择性高吸附容量的PMn膜,包括:
采用简单的相转化技术制备PVDF支持的MnSi结合相膜(简称PMn膜)。
具体是,将合成的0.5gMnSi与1.25gPVP和2.5gPVDF粉末均匀混合后(同时按照相同方法制备了MnSi/PVDF质量比为0、0.05、0.1的PMn膜),完全溶解在25mL氮,氮二甲基甲酰胺(DMF)溶液中,在60℃水浴条件下,密封剧烈搅拌12h,使聚合物完全溶解形成均匀的铸膜液。待反应结束后,倒入少量铸膜液于刮膜专用玻璃上,采用刮膜器(膜的厚度500μm)刮膜,然后迅速放入水中固化,待其在水中固化,自然脱落后取出PMn膜。最后,将生成的PMn膜用去离子水冲洗数次,采用打孔器切割得到大小一致、直径为2.5cm的圆片,并在使用前保存在去离子水中。

步骤1.将制备得到的PMn膜替换DGT装置的结合相,按照过滤膜在上,扩散相在中间,结合相在底部的方式组装;
步骤2.为了测试PMn-DGT的原位监测性能,需要对待测土壤进行前期培养
称取对应的土壤样品100g于塑料盒中,加入40mL水以达到土壤最大持水能力。搅拌均匀,采用保鲜膜封口,并盖上塑料盖,防止水分的快速流失。将预培养的土壤在25℃下培养24h,作为DGT装置待监测土壤样品(实验设置三个平行)。将准备好的DGT装置按照说明组装和部署,轻轻地敲击DGT几次,以确保采样器的过滤器表面与土壤之间的完全接触。其次,将所有DGT装置平整放入含有少量水的塑料袋中,保持水分的平衡,在25℃下培养24h;
步骤3.部署和培养完成,取出DGT装置,纯水清洗DGT装置表面粘附的土壤样品并拆卸,用镊子小心取出DGT结合相,放入5mL离心管。整个实验过程中,设置8个处理组,PMn-DGT和市售DGT分别部署在4个土壤中(4个土壤Pb的有效态含量存在差异)每个处理设置3个重复。取出的DGT首先采用2mL1mol/L的HNO3溶液洗脱2h测定DGT富集Pb的含量。实验过程所获得的溶液通过0.45μm孔径的PES注射器过滤器,然后用ICP-MS上机分析样品中重金属含量。

本发明提出了通过简单的共沉淀法成功制备出MnSi作为一种Pb2+高选择性、高吸附材料,并用于DGT结合相选择性监测土壤Pb有效态含量。MnSi在有无有机物存在的体系中均表现出高吸附性能和选择性,同时抗干扰能力强,最大吸附容量高达569.2mg·g-1。
本发明研发的一种新型、高选择性的PMn-DGT,实现了污染土壤Pb有效态的原位选择性监测,为更好的评估土壤重金属Pb的活性和毒性提供新的理论基础和技术依据。

为使本发明的目的、技术方案和优点更加清楚,下面本发明中的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
本发明的PMn-DGT结合相中的吸附剂MnSi通过简易的共沉淀法制备,再将其负载于PVDF膜,即可作为PMn-DGT结合相。步骤简单、可实现固体废弃物的资源化利用
实施例
如图8所示,本发明的高选择性和高吸附性能的环境功能材料制备方法,包括:
在水浴搅拌器的作用以及80℃水浴条件下,将Na2SiO3溶液滴加到MnCl2前体溶液中(按照Mn:Si摩尔比为1:1),然后待絮状物产生并稳定后持续反应2h得到MnSi颗粒物。
步骤2.产物离心后收集,用超纯水多次洗涤以去除其他杂质,将收集的材料冷冻干燥48h并储存在样品袋中贴标签、备用。
一种高选择性高吸附容量的PMn膜,包括:
采用简单的相转化技术制备了PVDF支持的MnSi结合相膜(简称PMn膜)。具体是,首先,将合成的0.5gMnSi与1.25gPVP和2.5gPVDF粉末均匀混合后(同时按照相同方法制备了MnSi/PVDF质量比为0、0.05、0.1的PMn膜),完全溶解在25mL氮,氮二甲基甲酰胺(DMF)溶液中,在60℃水浴条件下,密封剧烈搅拌12h,使聚合物完全溶解形成均匀的铸膜液。待反应结束后,倒入少量铸膜液于专用玻璃上,采用刮膜器(膜的厚度500μm)刮膜,然后迅速放入水中固化,待其在水中固化,自然脱落后取出PMn膜。最后,将生成的PMn膜用去离子水冲数次,采用打孔器切割得到大小一致、直径为2.5cm的圆片,并在使用前保存在去离子水中。
原位选择性监测铅有效态的薄膜梯度扩散方法,包括:
步骤1.将上述制备的PMn膜替换DGT装置的结合相,按照过滤膜在上,扩散相在中间,结合相在底部的方式组装;
步骤2.为了测试PMn-DGT的原位监测性能,需要对待测土壤进行前期培养。称取对应的土壤样品100g于塑料盒中,加入40mL水以达到土壤最大持水能力。搅拌均匀,采用保鲜膜封口,并盖上塑料盖,防止水分的快速流失。将预培养的土壤在25℃下培养24h,作为DGT设备待监测土壤样品(n=3,表示设置3个平行试验)。将准备好的DGT装置按照说明组装和部署,轻轻地敲击DGT几次,以确保采样器的过滤器表面与土壤之间的完全接触。其次,将所有DGT平整放入含有少量水的塑料袋中,保持水分的平衡,在25℃下培养24h;
步骤3.部署和培养完成,取出DGT,纯水清洗DGT表面粘附的土壤样品并拆卸,用镊子小心取出DGT结合相,放入5mL离心管。整个实验过程中,设置8个处理组,PMn-DGT和市售DGT分别部署在4个土壤中(4个土壤Pb的有效态含量存在差异)每个处理设置3个重复。取出的DGT首先采用2mL1mol/L的HNO3溶液洗脱2h测定DGT富集Pb的含量。实验过程所获得的溶液通过0.45μm孔径的PES注射器过滤器,然后用ICP-MS上机分析样品中重金属含量。
实验测试:
实验1
采用X射线衍射仪(BrukerD8Advance)用于研究MnSi的主要成分和结晶度。使用傅里叶变换红外光谱(FT-IR:Nexus670)粉体常规压片模式进一步测试了材料表面官能团。高分辨率透射电子显微镜图像(HR-TEM:FEITecnaiF20)用于表征MnSi并观察表面形貌和高分辨图像。同时,结合选区电子衍射图谱(SAED)鉴定材料的非晶型结构。
通过图1中,(a)-(d)可知,MnSi的XRD衍射峰宽,表明MnSi的结晶性能较差(图1,(a))。然而,在PMn膜的XRD衍射峰中没有检测到MnSiO3的峰,这可能是由于MnSi-PVDF杂化中MnSi的非晶态结构。这种现象的发生可能是因为PVDF与MnSi的界面效应,导致MnSi均匀分散在PVDF膜表面和内部结构中,抑制了MnSi粒子的出峰。FTIR光谱结果验证了MnSi和PVDF表面的官能团(图1,(b)),这些官能团为吸附提供了丰富的位点。除此之外,MnSi高分辨透射电镜图像(HR-TEM)清晰的显示了MnSi在亚微米级的表面结构,可以看到MnSi没有明显的晶格条纹,并且是无序的非晶型结构(图1,(c))。同时,选定区域的电子衍射图SAED中没有明显的电子衍射环,(图1,(d)),这也进一步验证了MnSi的非晶结构,与XRD结果一致。
实验2
通过批量吸附实验探讨了MnSi对常见的几种重金属(Co2+、Ni2+、Cd2+、Zn2+、Cu2+和Pb2+)在单元和多元重金属体系下的吸附性能。
如图2所示,为了明确MnSi对不同阳离子的选择性,将体系分为单元金属体系和多元金属体系,开展了相应的吸附实验。首先评估了CO2+、Ni2+、Cd2+、Zn2+、Cu2+和Pb2+这6种常见的重金属阳离子。批量吸附实验结果表明无论在单元还是多元金属体系(图2中,(a)-(b)),MnSi均表现出对Pb2+的优异的选择性。单元金属体系中,MnSi对Pb2+的吸附最强,吸附量高达569.2mg/g,对Cu2+表现出的吸附性能次之,吸附量为314.16mg/g。从图中可以得到MnSi在单元体系下对六种常见的重金属离子的吸附顺序为:Pb2+>Cu2+>Zn2+>Cd2+>Ni2+>Co2+。多元金属体系中,MnSi对Pn2+的选择性同样明显,而MnSi对其他四种金属离子吸附量微乎其微。这可能是因为在多元体系中,MnSi吸附剂的量一定,其表面暴露的活性位点数量有限,多元金属共存时,金属离子之间的竞争性增强。而MnSi对Pb2+的亲和力更强,因此,在溶液中优先结合Pb2+,其他金属离子的吸附量相比于单元体系有较大的下降幅度也就不足为奇。
实验3
通过批量吸附实验探讨了在不同初始pH和常规阳离子、阴离子条件下,MnSi对Pb2+吸附性能的影响。
如图3所示,对于溶液中的离子和固体之间的相互作用,初始溶液pH作为重要因素可能会影响吸附剂的吸附行为,如吸附剂的表面性质、吸附质的化学形态等。因此,研究了不同初始pH条件下,MnSi在单金属体系和PHP存在体系对Pb的吸附能力。结果如图3中(a)、(b)所示,pH在5-8范围内对MnSi去除溶液中Pb2+的性能无显著影响。但是初始pH为3时,两个体系均表现出明显的抑制作用。这可能是因为溶液呈强酸性,溶液中存在了大量的H+占据了MnSi的吸附位点,吸附剂表面发生明显的质子化,抑制了MnSi对Pb2+的吸附。
基于无机离子在水体环境中广泛存在,无机离子的存在可能会占据吸附位点来干扰吸附剂性能,常见的阴离子:Cl-、NO3 -、HCO3 -、SO4 2-和H2PO4 -和同时存在上述阴离子的体系(All)。结果表明,与空白对照组相比,适量阴离子的加入对Pb2+的去除效率并没有显著影响(如图3,(c))。同样,图7中,(d)结果表明10mmol/LNa+、Mg2+和Ca2+离子的存在对该体系没有表现出明显的抑制作用。上述实验现象在一定的浓度范围内,MnSi对Pb2+的吸附可以适应水体中不同离子的干扰以及环境的变化。
实验4
除了将吸附剂替换为PMn-DGT,其他吸附实验步骤同实验2。
我们设计了PMn-DGT装置在多元金属同时存在且初始浓度(5mg·L-1)相同体系下,PMn-DGT对多种金属离子的富集量。图4结果反映了PMn-DGT在该体系下对金属离子的吸附性能大小为Pb2+>Cu2+>Zn2+>Cd2+>Co2+>Ni2+,仍然是表现出对Pb2+的选择性。基于此,通过该方法将MnSi转化为PMn-DGT结合相吸附材料并没有改变材料的自身特性。因此,PMn-DGT装置依旧保留了对Pb2+的选择富集。
实验5
将合成的0.25gMnSi与0.25gPVP和0.5gPVDF粉末均匀混合后(MnSi不同负载量按比例添加),完全溶解在25mL氮,氮二甲基甲酰胺(DMF)溶液中,在60℃水浴条件下,密封剧烈搅拌12h,使聚合物完全溶解形成均匀的铸膜液。待反应结束后,倒入少量铸膜液于刮膜专用玻璃上,采用刮膜器(厚度500μm)刮膜,然后迅速放入水中固化,待其在水中固化,自然脱落后取出PMn膜。
鉴于MnSi对Pb2+优异的吸附性能,我们将其作为DGT结合相吸附材料。同时,利用PVDF膜液遇水固化的特点,将PVDF作为MnSi的基底材料,采用相转化技术将MnSi均匀分散在膜液中备用。为了获得具有最大吸附能力、性能稳定的PMn膜,设置了反应混合物中MnSi与PVDF的质量比,并以相同的条件下进行吸附Pb2+的反应。如图5所示,当MnSi负载量为0时,吸附材料的用量为0.1g·L-1,PVDF膜自身对Pb2+有一定的吸附能力,去除率仅为4.97%。随着MnSi负载量的增加,该体系中Pb2+的去除率呈现增大的趋势,当MnSi/PVDF质量比为0.2时,Pb2+的去除率达到89.33%。然而,继续增加MnSi的量,PVDF在固化成膜过程中不成形,过量的MnSi可能会影响PVDF成膜性质,严重影响后续的使用,因此后续实验选择MnSi/PVDF质量比为0.2的PMn膜。
实验6吸附实验同实验2
如图6所示,我们设计了PMn-DGT装置在多元金属同时存在且初始浓度(5mg·L-1)相同体系下,PMn-DGT对多种金属离子的富集量。图6结果反映了PMn-DGT在该体系下对金属离子的吸附性能大小为Pb2+>Cu2+>Zn2+>Cd2+>Co2+>Ni2+,仍然是表现出对Pb2+的选择性。基于此,通过该方法将MnSi转化为PMn-DGT结合相吸附材料并没有改变材料的自身特性。因此,PMn-DGT装置依旧保留了对Pb2+的选择性富集。
实验7
PMn-DGT实际应用:称取对应的土壤样品100g于塑料盒中,加入40mL水以达到土壤最大持水能力。搅拌均匀,采用保鲜膜封口,并盖上塑料盖,防止水分的快速流失。将预培养的土壤在25℃下培养24h将准备好的DGT装置按照说明组装和部署,轻轻地敲击DGT几次,以确保采样器的过滤器表面与土壤之间的完全接触。其次,将所有DGT平整放入含有少量水的塑料袋中,保持水分的平衡,在25℃下培养24h。部署和培养完成,取出DGT,纯水清洗DGT表面粘附的土壤样品并拆卸,用镊子小心取出DGT结合相,放入5mL离心管,取出的DGT首先采用2mL1mol·L-1的HNO3溶液洗脱2h测定DGT富集Pb的含量,再用2mL0.5mol·L-1H2O2和0.5mol·L-1NaOH混合液洗脱2h测定DGT富集Sb的含量。
由于实际土壤的基质更为复杂,在验证PMn-DGT的实际应用价值中,我们采集了成都市某废弃铅酸蓄电池附近的4个点位土壤,因此,采用DGT技术培养24h测定了实际污染土壤中Pb和Sb的有效态含量。铅酸蓄电池厂附近土壤呈碱性,有机质含量较低。同时,采用纯水和DTPA两种溶液浸提以得到两种形式的土壤有效态含量,分别定义为溶解性有效态和DTPA有效态。分别将两种方式测定的有效态含量与PMn-DGT和市售DGT测定CDGT含量进行相关性分析,结果如图7所示。4个土壤样品中,PMn-DGT对土壤中Pb和Sb有效态的检测结果均有相同的趋势,相关性较好。CDGT(Pb)与两种形式有效态中,PMn-DGT相关性略优于市售DGT,PMn-DGT测定同一土壤有效态均显著高于市售DGT,这可能是归因于结合相中MnSi对Pb的高选择性和吸附性能。此外,DGT技术测定的CDGT(Sb)与两种形式有效态中,PMn-DGT相关性分别为R2分别为0.9060和0.9927,显著优于市售DGT。可能的原因是PMn-DGT部署时间与富集含量的线性关系较好,这与前面相关结论一致。因此,证实了PMn薄膜作为新型DGT结合相用于铅酸蓄电池附近污染土壤Pb和Sb有效态含量的可行性。
由于采集的实际污染土壤只有Pb和Sb超标,为了体现PMn-DGT在实际污染土壤堆Pb的选择性检测,我们对比了两种DGT对Pb和Sb检测的有效态含量。图7中,(e)-(f)结果表明,4个土壤的DGT检测结果中,PMn-DGT测定的Pb的有效态含量均显著高于市售DGT,而Sb的有效态含量显著低于市售DGT,结合上述MnSi在多金属体系中对Pb的高选择性,考虑这种现象归因于PMn结合相对Pb的亲和力更强,更容易优先吸附通过扩散相的Pb。
最后应说明的是:以上实施例仅用以说明本发明的技术方案,而非对其限制;尽管参照前述实施例对本发明进行了详细的说明,本领域的普通技术人员应当理解:其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的精神和范围。
Claims (1)
1.原位选择性监测铅有效态的薄膜梯度扩散方法,其特征在于,包括:
步骤1.将制备得到的PMn膜替换DGT装置的结合相,按照过滤膜在上,扩散相在中间,结合相在底部的方式组装;
步骤2.为了测试PMn-DGT的原位监测性能,需要对待测土壤进行前期培养;
称取对应的土壤样品100g于塑料盒中,加入40mL水以达到土壤最大持水能力,搅拌均匀,采用保鲜膜封口,并盖上塑料盖,防止水分的快速流失;将预培养的土壤在25℃下培养24h,作为DGT装置待监测土壤样品,将准备好的DGT装置按照说明组装和部署,轻轻地敲击DGT几次,以确保采样器的过滤器表面与土壤之间的完全接触;其次,将所有DGT装置平整放入含有少量水的塑料袋中,保持水分的平衡,在25℃下培养24h;

其中,PMn膜的制备方法包括:合成0.5gMnSi与1.25gPVP和2.5gPVDF粉末均匀混合后,完全溶解在25mL氮,氮二甲基甲酰胺溶液中,在60℃水浴条件下,密封剧烈搅拌12h,使聚合物完全溶解形成均匀的铸膜液,待反应结束后,倒入少量铸膜液于刮膜专用玻璃上,采用刮膜器刮膜,膜的厚度为500μm,然后迅速放入水中固化,待其在水中固化,自然脱落后取出PMn膜,最后,将生成的PMn膜用去离子水冲洗数次,采用打孔器切割得到大小一致、直径为2.5cm的圆片,并在使用前保存在去离子水中;
MnSi按照如下步骤制备:
步骤1.在搅拌器的作用、以及80℃水浴条件下,将Na2SiO3溶液滴加到MnCl2前体溶液中,按照Mn:Si摩尔比为1:1,然后待絮状物产生并稳定后持续反应2h得到MnSi颗粒物;
搅拌器为机械搅拌器,转速为500r/min;
步骤2.产物离心后收集,用超纯水多次洗涤以去除其他杂质,将收集的材料冷冻干燥48h并储存在样品袋中贴标签,备用。
