Abstract
本发明属于医用/化妆品配制品领域,具体涉及一种含有胶原蛋白的皮肤修护组合物;组分包括胶原蛋白、水解血清蛋白、异甘草素、神经酰胺、润肤剂、植物油、复合植物发酵粉、乳化剂。本发明通过多种原材料的复合提供一种组合物,能够补充细胞间脂质基质,进而辅助角质层增厚,促进皮肤屏障自我修复;同时还能刺激细胞的新陈代谢,为细胞的再生提供稳定的环境;相比于现有的皮肤修复组合物效果更为显著。

技术领域
本发明属于医用/化妆品配制品领域,具体涉及含有胶原蛋白的皮肤修护组合物。
背景技术
人体皮肤是人体最大的器官,位于身体表面,由多层组织构成。它具有多种重要的功能,包括保护内部器官免受外界环境的伤害、调节体温、感知外界刺激和参与水分调节等。
皮肤保护屏障受损原因有多种,如物理因素刺激(摩擦、划伤、烫伤、医美创伤)、化学因素(强酸强碱的化学物,或者不当的刷酸或者过度使用刺激类的护肤品)、生物因素刺激(细菌、真菌、寄生虫等感染)、环境因素(干燥多风、过度曝光)、情绪因素(压力过大、产后焦虑);皮肤受损后引起炎症、出血、水肿、红肿、疼痛等症状,严重者还可能留下瘢痕或感染;皮肤受损后无法起到正常的屏障作用,需要修复。

优选的,所述极性润肤剂中包括以下中的至少一种:
碳酸二辛酯、肉豆蔻酸异丙酯、甘油。
优选的,所述植物油中包括以下中的至少一种:
亚麻籽油、玫瑰果油、霍霍巴油、甜杏仁油。
优选的,所述复合植物发酵粉成分包括水麻、远志、西洋接骨木;水麻、远志、西洋接骨木的质量比为20:4-6:1-1.5。
所述复合植物发酵粉的制备方法为:
(1)取水麻的地上部分、西洋接骨木的花、远志的根;分别洗净后,粉碎、灭菌;加入总质量1倍的水;
(2)灭菌后利用酵母菌发酵,发酵时间为48-72h;发酵温度为25-28℃;
(3)发酵后烘干至恒重,得复合植物发酵粉。
所述酵母菌为巴斯德酵母;接种的酵母菌的数量为≥105CFU/mL。。
优选的,以质量份计算还包括C相:
挥发性硅油20-40。
本发明中:
胶原蛋白与神经酰胺为皮肤自身含有的成分;能够帮助皮肤重构。其配比与人体皮肤中的含量类似;具体的,I型胶原蛋白含量最多,质地较硬,负责支撑皮肤硬度和坚固性。III型胶原蛋白具有较好的柔韧性和光泽度,与I结合型胶原蛋白增强皮肤稳定性和弹性,还能促进皮肤愈合。V型胶原蛋白主要负责促进愈合。
神经酰胺和胶原蛋白可以相互影响和促进。神经酰胺作用于真皮层可刺激成纤维细胞的活性,刺激产生更多胶原蛋白,并且抑制胶原酶活性,让胶原合成与降解处于动态平衡。胶原蛋白也可以通过增加皮肤的厚度和密度,改善角质层的水分保持能力,从而增加神经酰胺的含量。
由于本发明用于皮肤已经受损情况下的修护,所以大分子的胶原蛋白能通过已经损伤的皮肤屏障到达真皮层。
水解血清蛋白作用与胶原蛋白类似;异甘草素在本发明中主要起到抗炎和减少敏感性的效果。
本发明的极性润肤剂与植物油主要起到保湿、促进皮肤愈合的效果。
本发明的复合植物发酵粉富含多酚、黄酮、甾体等活性物质,除了保湿外还能起到抗氧化、抗菌的作用;且本发明发现,在发酵后,活性物质的含量明显增高,能够进一步促进皮肤的愈合。
本发明的挥发性硅油一方面作用为调节肤感,同时由于其挥发性使得在本发明涂抹后其余成分能快速的附着在皮肤上形成较为致密的膜;隔绝细菌、灰尘等外界刺激为皮肤修复提供稳定的环境。
本发明所述的乳化剂可以是任意能够实现乳化且符合相应法规所要求的物质,包括但不限于:聚甘油-10月桂酸酯、丙二醇月桂酸酯、PEG-5辛酸//癸酸甘油酯类、甘油辛酸酯、椰油酰胺MEA、丙烯酸(酯)类共聚物钠等。
本发明的有益效果:
本发明通过多种原材料的复合提供一种组合物,能够补充细胞间脂质基质,进而辅助角质层增厚,促进皮肤屏障自我修复;同时还能刺激细胞的新陈代谢,为细胞的再生提供稳定的环境;相比于现有的皮肤修复组合物效果更为显著。
具体实施方式
下面结合具体实施例,对本发明作进一步详细的阐述,下述实施例不用于限制本发明,仅用于说明本发明。以下实施例中所使用的实验方法如无特殊说明,实施例中未注明具体条件的实验方法,通常按照常规条件,下述实施例中所使用的材料、试剂等,如无特殊说明,均可从商业途径得到。
本发明中,水麻为荨麻科水麻属Debregeasia orientalis.。
远志为远志科植物远志Polygala tenuifoliaWilld.或卵叶远志Polygala
sibirica L.。
西洋接骨木为忍冬科接骨木属Sambucus Nigra.。
巴斯德酵母为巴斯德毕赤氏酵母Pichiapastoris。
水解血清蛋白为Hydrolyzed Serum Protein。
复合植物发酵粉的制备,包括以下步骤:
(1)将水麻的地上部分、西洋接骨木的花、远志的根分别洗净,洗净后切碎利用臭氧灭菌;灭菌后混合得混合原料;
(2)混合原料中加入其一倍体积量的无菌水,然后加入活化后的啤酒酵母菌厌氧发酵;啤酒酵母菌加入量为105CFU/mL。
(3)发酵后95℃下灭菌30min,灭菌后在至少50℃的烘干箱中烘干至恒重。
原料比例如下表1(单位质量份)所示。

含有胶原蛋白的皮肤修护组合物的制备,包括以下步骤:
(1)按表2-1、2-2所示称量各原料;
(2)将B相的成分加热至30-35℃,搅拌混合均匀,然后降温,得B相;
(3)将A相、B相以及其他组分混合,均质后的稳定混合物,得含有胶原蛋白的皮肤修护组合物。

获得的5组实施例、对比例1、2、3、5为具有一定流动性的不透明非固体。对比例4为固态粉末。
安全性测试
将获得的10组成品参照《2022化妆品安全技术规范》中的人体皮肤斑贴试验;实验前用去离子水稀释至50wt%的浓度;其中对比例4中额外加入3wt%的丙二醇月桂酸酯用于溶解。
皮肤封闭型斑贴试验的方法为:选择年龄为18-60周岁的人员50名,选用面积不超过50mm2、深度约1mm的合格斑试器材,取用0.020mL上述实施例和对比例制备的成品放入斑试器小室内,对照孔为空白对照(不置任何物质),将加有成品的斑试器用低致敏胶带贴敷于受试者的前臂曲侧,用手掌轻压使之均匀地贴敷于皮肤上,持续24h。分别于去除成品后30min(待压痕消失后)、24h和48h按表3标准观察皮肤反应,并记录观察结果,见表4。

油脂氧化稳定性测试
将上述各实施例和对比例制备而成含有胶原蛋白的皮肤修护组合物,进行油脂氧化稳定性测试,测试方法参照《GB/T21121-2007动植物油脂氧化稳定性的测定(加速氧化测试)》
同时为了更直观的观察油脂应用中的抗氧化能力,常见的化学合成抗氧化剂BHT,BHA作对比,添加量与实施例以及对比例中的山茶抗氧化组合物的添加量一致,选用空白植物油(无抗氧化剂添加)做对照,采用892型油脂氧化稳定性测试仪进行测试。

测试结果如表5,氧化诱导时间越长,则油脂的氧化稳定性越好。
抗氧化测试
测试原理:DPPH是一种稳定自由基,当有自由基清除剂存在时,自由基清除剂与DPPH单电子配对,使DPPH光吸收减弱;本发明通过DPPH自由基清除实验,测定溶液吸光度,以评定自由基清除剂的抗氧化能力。
测试方法:在10ml的比色管中依次加入4.0ml的DPPH无水乙醇溶液、1ml的试样(即稀释十倍);再加入无水乙醇至刻度值,混匀后在517nm波长处测吸光值,记为Ai,在室温避光30min后再次测吸光值,记为Aj,以同体积只加DPPH的无水乙醇溶液做空白组,测定吸光度,记为Ac;每组做3个重复,求其平均值,试验结果见表6,按下式

测试结果如表6所示;根据对比例1的结果显示,神经酰胺与血清蛋白没有直接的清除自由基的效果;根据对比例2、3、5的结果显示,复合植物发酵粉和植物油都具有较为显著的清除自由基的效果;但是单独使用时,复合植物发酵粉效果比植物油好,且复配后起到的效果更佳;对比例4中清除自由基的效果较弱,是由少量异甘草素带来的效果。
修复促进率测试
测试原理:斑马鱼的伤口愈合原理与哺乳动物一致。利用斑马鱼胚胎尾鳍修复实验,计算促进修复率以评估产品的促进修复功效。
测试方法:
挑选健康的斑马鱼胚胎,并以200μL鱼胚胎培养液中不超过1粒鱼胚胎的密度于28℃培养至受精后3天。
随机挑选42粒切除尾鳍的鱼胚胎,随机分成14组;其中一组为损伤模型对照组,剩余十三组为实验组。
转移至96孔板中,每孔含一粒鱼胚胎和0.2mL各组培养液,放置于28±1℃恒温箱中48±1h,放置于立体显微镜下对鱼胚胎按统一的拍照参数进行拍照分析,量度每粒鱼胚胎的再生尾鳍长度。
损伤模型对照组为鱼胚胎培养液;
实验组为将测试组按比例溶解到鱼胚胎培养液中。
实验结果见表7,按下式计算斑马鱼胚胎尾鳍修复促进率:
斑马鱼胚胎尾鳍修复促进率=[(样品处理组损伤斑马鱼胚胎尾鳍修复水平–损伤模型组斑马鱼胚胎尾鳍修复水平)*100/损伤模型组斑马鱼胚胎尾鳍修复水平]%。
根据样品对斑马鱼胚胎尾鳍修复促进率和促进率的统计学差异,对测试样本进行促进修复功效评估。

根据表7显示,对比例1只有一种胶原蛋白,且不含神经酰胺和血清蛋白;对比例2没有添加复合植物发酵粉;对比例3不含植物油;对比例4没有植物油和复合植物发酵粉;对比例5不含胶原蛋白、神经酰胺和血清蛋白;可见,胶原蛋白和神经酰胺的配合使用能够加强修复效果;而独立的植物油和复合植物发酵粉由于提供了一个较为稳定的条件,对修复具有微弱的促进作用,如果想要达到良好的修复需要配合A相的成分使用;而B相也能促进A相的修复效果。
炎症细胞抑制率测试
测试原理:斑马鱼的中性粒细胞在形态,生物化学和功能上,都与哺乳类动物相似。通过测试鱼胚胎中性粒细胞数目变化以评估产品的舒缓功效。
测试方法:挑选野生型AB品系斑马鱼胚胎,以200μL鱼胚胎培养液中不超过1粒鱼胚胎的密度于28℃培养至受精后3天。
随机挑选42粒鱼胚胎,随机分成14组;其中一组为损伤模型对照组,剩余十三组为实验组。
先用炎症诱导剂溶液(称取159.6mg硫酸铜(CuSO4·5H2O)溶于1000mL水中后,用鱼胚胎培养液稀释10倍)处理后,暴露于0.2ml各组的培养液中。
炎症模型对照组为鱼胚胎培养液;实验组为将测试组按比例溶解到鱼胚胎培养液中。
放置于28±1℃恒温箱中培养40-45min。转移至2mL离心管中,加入4%PFA(多聚甲醛溶液)溶液于室温固定、染色、漂色,放置于立体显微镜下对鱼胚胎按统一的拍照参数进行拍照、分析计算鱼胚胎细胞中性粒细胞的数目。
实验结果见表8,按下式计算斑马鱼胚胎炎症细胞(中性粒细胞)抑制率:
斑马鱼胚胎炎症细胞(中性粒细胞)抑制率=[(斑马鱼胚胎炎症模型对照组炎症细胞数目-斑马鱼胚胎炎症模型样品测试组炎症细胞数目)*100/斑马鱼胚胎炎症模型对照组炎症细胞数目]%。
根据样品对斑马鱼胚胎炎症细胞抑制率以及抑制率的统计学差异,对样品的舒缓功效进行评估。

根据表8显示,对比例1由于没有复配神经酰胺和血清蛋白,导致抑制效果比实施例弱;但与没有添加B相的对比例4效果好,本发明认为原因在于B相中含有多种的清除自由基的成分能够提升抑制效果。而对比例3与实施例相比利用甘油填补植物油,使得抑制效果稍弱,本发明认为是植物油中含有的不饱和脂肪酸导致的结果。对比例5中由于不含A相中的神经酰胺与蛋白;但由于能够清除自由基,使得具有一定的抑制效果。对比例2没有添加复合植物发酵粉,与实施例相比抑制效果较差,本发明认为复合植物发酵粉对减少炎症具有一定的促进作用。

1.含有胶原蛋白的皮肤修护组合物,其特征在于,以质量份计包括以下组分:
A相:
I型胶原蛋白 100

水解血清蛋白 4-8

极性润肤剂 300-400

所述复合植物发酵粉成分包括水麻、远志、西洋接骨木;
所述水麻、远志、西洋接骨木的质量比为20:4-6:1-1.5;
所述复合植物发酵粉的制备方法为:
(1)取水麻的地上部分、西洋接骨木的花、远志的根;分别洗净后,粉碎、灭菌;加入总质量至少1倍的水;
(2)灭菌后利用酵母菌发酵,发酵时间为48-72h;发酵温度为25-28℃;
