技术领域
本发明大体涉及生物医疗技术领域,具体而言,涉及一种特异性抑制ATG16L1表达的siRNA或shRNA。
背景技术
细胞自噬(autophagy)是真核细胞内一种高度保守的分解代谢过程。细胞通过细胞自噬不仅可以回收利用各种基本的生命物质,同时也可以清除入侵病原体、受损线粒体等有害物质。因此,细胞自噬在众多的生理过程中扮演着重要角色。与此同时,众多的人类疾病均与细胞自噬的功能异常密切相关,比如癌症、神经退行性疾病等。在经典的巨自噬(macroautophagy)过程中,细胞通过吞噬泡(phagophore)的引发、延伸进而形成自噬体(autophagosome)来包裹细胞内待降解的自噬底物,比如糖原、蛋白聚集物、受损细胞器和入侵病原体等。在吞噬泡的引发和延伸阶段,WIPI2b蛋白招募并激活ATG12~ATG5-ATG16L1复合物,使其发挥类E3酶的活性,实现对ATG8家族蛋白的脂质化修饰,进而促进吞噬泡的延伸和自噬体的形成过程。然而,其中WIPI2b蛋白招募ATG16L1复合物——这一关键自噬步骤的具体分子机制仍不清楚。
自噬相关16样蛋白1(RecombinantAutophagy Related Protein 16Like Protein1,ATG16L1)在高等真核生物中被鉴定为与Atg16相似,Atg16是一种酵母蛋白,以前被表征为Atg12-Atg5/Atg16复合物的亚基。在酵母中,这种复合物催化Atg8在前自噬体结构上的脂化,因此是自噬体形成所必需的。在高等真核生物中,ATG16L1也几乎完全作为ATG12-ATG5/ATG16L1复合物的一部分存在,并且在自噬中具有相同的基本功能。
现有技术的研究表明,ATG16L1在经典自噬过程中的重要作用。而自噬过程与众多疾病具有广泛的关联性。但现有技术中缺乏有效阻断ATG16L1的相关工具。
RNA干扰(RNAi)是有效沉默或抑制目标基因表达的过程,该过程通过双链RNA(dsRNA)使得目标基因相应的mRNA选择性失活来实现的。RNA干扰由转运到细胞细胞质中的双链RNA激活。沉默机制可导致由小干扰RNA(siRNA)或短发夹RNA(shRNA)诱导实现靶mRNA的降解,或者通过小RNA(miRNA)诱导特定mRNA翻译的抑制。
发明内容
本发明的涉及特异性抑制ATG16L1表达的核酸,其为siRNA或shRNA,包含SEQ IDNO:1所示的正义链和SEQ ID NO:2所示的反义链,所述正义链和反义链互补共同形成RNA二聚体。
可选的,所述正义链中的至少一个核苷酸具备化学修饰。
可选的,所述正义链中至少一个核苷酸中的核糖2’-OH被甲氧基所取代。
可选的,所述正义链第6位核苷酸中的核糖2’-OH被甲氧基所取代。
本发明还涉及一种能够产生如上所述核酸的DNA分子。

本发明的涉及特异性抑制ATG16L1表达的核酸,其为siRNA或shRNA,包含SEQ IDNO:1所示的正义链和SEQ ID NO:2所示的反义链,所述正义链和反义链互补共同形成RNA二聚体。
在本发明中,所述siRNA,即小干扰RNA(small interfering RNA)是一种双链小RNA分子,其由完全互补的正义链和反义链组成,由Dicer(RNAaseⅢ家族中对双链RNA具有特异性的酶)加工而成。所述正义链和反义链互补共同形成RNA二聚体,并且所述反义链的序列为与ATG16L1mRNA序列在高等严紧条件下杂交的序列。所述双链RNA正义链和反义链的长度均为10~30个核苷酸;优选地,长度均为15~27个核苷酸;更优选19~23个核苷酸,也可以选择20、21或者22个核苷酸。siRNA是siRISC的主要成员,激发与之互补的目标mRNA的沉默。RNA干扰(RNAinterference,RNAi)是指内源性或外源性双链RNA(dsRNA)介导的细胞内mRNA发生特异性降解,从而导致靶基因的表达沉默,产生相应的功能表型缺失的现象。
在本发明中,所述shRNA,即小发卡或短发卡RNA(a small hairpin RNA orshorthairpin RNA,shRNA)是一段具有紧密发卡环(tight hairpin turn)的RNA序列,其包含正义链片段、反义链片段以及连接正义链片段和反义链片段的茎环结构,常被用于RNA干扰沉默靶基因的表达。其中正义链和反义链的序列互补,并且所述反义链片段的序列与ATG16L1 mRNA中10~30个连续的核苷酸序列相同,优选地,所述反义链片段与ATG16L1mRNA中15~27个连续的核苷酸序列相同;更优选地,所述反义链片段与ATG16L1mRNA中19~23个核苷酸相同,也可以选择19、20或者21个连续的核苷酸序列相同,或者为与上述各序列在高等严紧条件下杂交的序列。shRNA的发卡结构可被细胞机制切割成siRNA,然后siRNA结合到RNA诱导沉默复合物上(RNA-inducedsilencing complex,RISC),该复合物能够结合到目的mRNAs并将其降解。
进一步地,所述shRNA的茎环结构的序列可为本领域的常规选择,例如选自以下任一种序列:UUCAAGAGA、AUG、CCC、UUCG、CCACC、CUCGAG、AAGCUU和CCACACC。
在本发明中,“互补”是指序列能够在高等严紧条件下杂交。“杂交条件”根据测量杂交时所用条件的“严紧性”程度来分类。严紧性程度可以以例如核酸结合复合物或探针的解链温度(Tm)为依据。例如,“最大严紧性”典型地发生在约Tm-5℃(低于探针Tm 5℃);“高等严紧性”发生在Tm以下约5℃~10℃;“中等严紧性”发生在探针Tm以下约10℃~20℃;“低严紧性”发生在Tm以下约20℃~5℃。作为替代,或者进一步地,杂交条件可以以杂交的盐或离子强度条件和/或一或多次的严紧性洗涤为依据。例如,6×SSC=极低严紧性;3×SSC=低至中等严紧性;1×SSC=中等严紧性;0.5×SSC=高等严紧性。从功能上说,可以采用最大严紧性条件确定与杂交探针严紧同一或近严紧同一的核酸序列;而采用高等严紧性条件确定与该探针有约80%或更多序列同一性的核酸序列。
对于要求高选择性的应用,典型地期望采用相对严紧的条件来形成杂交体,例如,选择相对低的盐和/或高温度条件。Sambrook等(Sambrook,J.等(1989)分子克隆,实验室手册,Cold Spring HarborPress,Plainview,N.Y.)提供了包括中等严紧性和高等严紧性在内的杂交条件。
为便于说明,用于检测本发明的多核苷酸与其它多核苷酸杂交的合适的中度严紧条件包括:用5×SSC、0.5%SDS、1.0mM EDTA(pH8.0)溶液预洗;在50-65℃下在5×SSC中杂交过夜;随后用含0.1%SDS的2×、0.5×和0.2×SSC在65℃下各洗涤两次20分钟。本领域技术人员应当理解,能容易地操作杂交严紧性,如改变杂交溶液的含盐量和/或杂交温度。例如,在另一个实施方案中,合适的高度严紧杂交条件包括上述条件,不同之处在于杂交温度升高到例如60℃~65℃或65℃~70℃。
在一些实施方式中,所述正义链中的至少一个核苷酸具备化学修饰。
在一些实施方式中,所述正义链中至少一个核苷酸中的核糖2’-OH被甲氧基所取代。
在一些实施方式中,所述正义链第6位核苷酸中的核糖2’-OH被甲氧基所取代。
根据本发明的再一方面,还涉及能够产生如上所述核酸的DNA分子。
在一些实施方式中,所述DNA分子经密码子优化以在生物体中表达。
本发明的任何DNA分子、多核苷酸和/或载体等都可以进行密码子优化,以便在任何感兴趣的生物体中表达。密码子优化在本领域中是众所周知的,并且涉及使用物种特异性密码子使用表针对密码子使用偏好性对核苷酸序列进行修饰。密码子使用表是基于对感兴趣的生物体/物种的最高表达基因的序列分析而生成的。当核苷酸序列要在细胞核中表达时,密码子使用表是基于对感兴趣物种的高表达核基因的序列分析而生成的。通过将物种特异性密码子使用表与天然多核苷酸序列中存在的密码子进行比较来确定核苷酸序列的修饰。如本领域所理解的那样,核苷酸序列的密码子优化导致核苷酸序列与天然核苷酸序列(或称优化前的核苷酸序列)具有小于100%的同一性(例如,约70%、71%、72%、73%、74%、75%、76%、77%、78%、79%、80%、81%、82%、83%、84%、85%、86%、87%、88%、89%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%或99.9%,以及其中的任何范围或值),但其仍编码与原始天然核苷酸序列编码的多肽具有相同功能的多肽。因此,在本发明的一些实施方案中,本发明的DNA分子是经密码子优化的,用于在感兴趣的特定物种中表达,例如特定动物物种和人等。
根据本发明的再一方面,还涉及包含如上所述DNA分子的载体。
本文所用的术语“载体”是指包含多核苷酸或与多核苷酸缔合的大分子或生物大分子缔合物,其可用于介导多核苷酸向细胞的转移、递送或导入。用于转化宿主生物体的载体在本领域是众所周知的。通用载体类别的非限制性实例包括病毒载体、质粒载体、噬菌体载体、噬菌粒载体、粘粒载体、fosmid载体、噬细菌体、人工染色体、微环载体(minicircle)或者农杆菌属(Agrobacterium)双元载体,呈双链或单链线性或环状形式,它们可能是或可能不是自我传递或移动的。在一些实施方式中,所述的载体是病毒载体。在一些实施方案中,所述病毒载体可包括但不限于腺病毒载体、腺相关病毒(AAV)载体、慢病毒载体或逆转录病毒载体。
本发明还涉及递送系统,其包含i)如上所述的核酸,或如上所述的DNA分子,以及ii)递送媒介物。
在一些实施方式中,所述递送媒介物包括一种或多种脂质体、一种或多种外泌体、一种或多种微囊泡、一种或多种树状大分子、一种或多种无机纳米粒子、一种或多种细胞穿膜肽、基因枪、一种或多种质粒、一种或多种病毒载体(在一些实施方式中,所述病毒载体如上所定义),以及它们所组成的组。
脂质体可以为阳离子脂质体或中性脂质体,其可通过公知的方法进行制备或修饰,例如加入聚乙二醇(PEG)修饰的脂质体可以有效防止脂质体载体的聚集并增加其稳定性。脂质体或脂质转染配制品可以通过本领域技术人员已知的方法制备。这样的方法描述于例如WO 2016205764和美国专利号5,593,972、5,589,466、和5,580,859中,将各个文献通过引用以其全文并入本文。
树枝状大分子是一种具有明确的分子结构、可精确控制的化学结构和独特的多价性质的特殊聚合物家族,逐渐成为基因传递的非病毒载体。典型的树枝状大分子例如聚(酰氨基胺)(PAMAM)树枝状聚合物,其可做进一步修饰,例如在PAMAM表面修饰核碱基类似物2-氨基-6-氯嘌呤构建衍生物AP-PAMAM,或者通过硫酸软骨素(CS)与PAMAM偶联制备CS-PAMAM等等。
无机纳米粒子可选择金纳米粒子(AuNPs)、磁性纳米粒子、介孔二氧化硅纳米粒子(MSNs)等。
细胞穿膜肽(cell-penetratingpeptides,CPPs)是一类具有较强跨膜转运能力的小分子肽,可携带多肽、蛋白质和核酸等多种大分子物质进入细胞。其可以为阳离子型CPPs(如TAT,Penetratin,Polyarginine,P22N,DPV3和DPV6等)、两亲型CPPs(可以由疏水性肽序列和NLSs共价连接而成,或者从天然蛋白质中分离获得,如pVEC,ARF(1-22)和BPrPr(1-28))、疏水型CPPs(一般只含有非极性氨基酸残基,净电荷量约低于氨基酸序列总电荷量的20%)。
在一些实施方式中,所述递送经由质粒进行。所述剂量可以是足够数量的质粒以引发响应。在一些情况下,质粒组合物中质粒DNA的合适量可以是从约0.1至约2mg。质粒将通常包括(i)启动子;(ii)编码靶向核酸的CRISPR相关蛋白和/或辅助蛋白的序列,每个序列与启动子(例如,相同的启动子或不同的启动子)可操作地连接;(iii)可选择标志物;(iv)复制起点;以及(v)位于(ii)的下游并与其可操作地连接的转录终止子。质粒还可以编码CRISPR复合物的RNA组分,但这些组分中的一种或多种可以替代地在不同的载体上编码。施用频率在医学或兽医学从业者(例如,医师、兽医师)或本领域技术人员的范围内。

根据本发明的再一方面,还涉及药物组合物,其包含如上所述的递送系统以及药学上可接受的赋形剂。
术语“药学上可接受的”指当分子本体、分子片段或组合物适当地给予动物或人时,它们不会产生不利的、过敏的或其他不良反应。可作为药学上可接受的赋形剂或其组分的一些物质的具体示例包括磷酸,柠檬酸,和其它有机酸;抗氧化剂(例如,抗坏血酸和甲硫氨酸);抗菌剂(例如,十八烷基二甲基苯氯化铵,氯化六烃季铵,苯扎氯铵,酚,丁醇或苯甲醇,烷基尼泊金,邻苯二酚,间苯二酚,环己醇,3-戊醇,或间甲酚);低分子量(不到约10kDa)多肽;蛋白,例如,血清白蛋白,明胶,或免疫球蛋白;亲水性聚合物,例如,聚乙烯吡咯烷酮;氨基酸(例如,甘氨酸,谷氨酰胺,天冬酰胺,组氨酸,精氨酸,或赖氨酸);单糖,二糖和其它碳水化合物(包括例如,葡萄糖,甘露糖,或葡聚糖);螯合剂(例如,EDTA);糖(例如,蔗糖,甘露醇,海藻糖,或山梨醇);成盐反离子;金属复合物;和/或非离子型表面活性剂(例如,包括TWEENTM,PLURONICSTM,或聚乙二醇)。此外,根据配制方法,可以由本领域普通技术人员适当选择常用的填充剂,稀释剂,结合剂,增湿剂,崩解剂,和/或表面活性剂。
药物组合物中还可以包含核酸稳定剂。用于稳定化和保持核酸的稳定化药剂的例子包括阳离子化合物、去污剂、离液盐、核糖核酸酶抑制剂、螯合剂等及其混合物。稳定剂可以包括例如诸如多聚甲醛的交联固定剂或诸如乙醇的沉淀剂。稳定剂可以通过在细胞分子之间形成共价键或通过将一些细胞内分子沉淀或通过其它方法来起作用。在一些实施方式中,稳定剂包括细胞裂解缓冲液。细胞透化缓冲液也是本领域已知的,并且可以包含使细胞膜透化从而允许探针和染料穿过膜的去污剂。在细胞裂解缓冲液中使用的去污剂的例子包括但不限于TweeruTriton X-100、皂草苷、NP-40等。针对给定的最终用途调整细胞裂解和透化剂的浓度。当以过低的浓度存在时,细胞裂解和透化可能不能达到最佳。在过高的浓度下,可能出现不希望的细胞破坏。可以进行常规的根据经验的步骤来确定在每种情况下优选的路线。在一些实施方式中,稳定剂包括氯仿、苯酚、TRIZOL。但在更优选的实施方式中,稳定剂是易于被除去的或者对细胞毒性较低的成分,最优选是药学上可接受的成分。
本发明还涉及如上所述核酸,或如上所述的DNA分子,或如上所述的递送系统在制备用于促进伤口愈合的药物中的应用。优选为促进糖尿病患者相关的伤口愈合的药物。
下面将结合实施例对本发明的实施方案进行详细描述。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中未注明具体条件的实验方法,优先参考本发明中给出的指引,还可以按照本领域的实验手册或常规条件,还可以参考本领域已知的其它实验方法,或者按照制造厂商所建议的条件。
下述的具体实施例中,涉及原料组分的量度参数,如无特别说明,可能存在称量精度范围内的细微偏差。涉及温度和时间参数,允许仪器测试精度或操作精度导致的可接受的偏差。
实施例
一、实验方法
1.细胞培养和处理。
大鼠采用1%戊巴比妥钠腹腔过量注射麻醉处死。在超净工作台上取出大鼠眼球,立即用70%医用酒精溶液浸泡30秒。取出眼球并沿角膜边缘切开。移除晶状体和角膜并分离视网膜组织。去除视网膜中较大的主要血管,将剩余的组织在冰预冷的PBS缓冲液中冲洗两次,然后用剪刀剪成碎片。将剪下的视网膜组织通过200目筛网过滤,过滤组织的上层用冰预冷的PBS缓冲液重新悬浮。将重悬液于4℃2500RPM离心12分钟以收集离心沉淀物。在视网膜毛细血管组织中加入10倍体积的0.1%Ⅱ型胶原酶消化液,37℃水中震荡45分钟。然后将消化液反复泵入注射器并通过100目筛网过滤。4℃1500RPM离心10分钟,收集离心沉淀。离心沉淀细胞用10倍体积的h-DMEM培养基洗涤2次,用DMEM完全培养基(含20%FBS、200μg/mL肝素钠、400μg/mL ECGS、100单位/mL青霉素)重悬细胞钠,100μg/mL硫酸链霉素)。将细胞接种于0.5%明胶烧瓶中,置于37℃、5%CO2培养箱中培养。培养48小时后,换液去除未贴壁的细胞,待细胞汇合度达到70%时,用胰蛋白酶消化细胞,按1:3的比例传代。
在六孔板中培养至约70%的汇合度后,按照制造商的说明,使用Lipofectamine3000用化学合成的ATG16L1 siRNA和阴性对照(siRNA NC)转染小鼠肾皮层上皮细胞(RCEC)。转染24小时后,将细胞分别在5.5mM葡萄糖(NG)或25mM葡萄糖(HG)培养基中分别培养24小时和96小时。
细胞分为以下六组:i)NG;ii)HG;iii)NG+siRNANC;iv)HG+siRNA NC;v)NG+ATG16L1 siRNAAL06;vi)HG+ATG16L1 siRNA AL06。
2.Real-time PCR。
使用TRIzol试剂(Invitrogen,Carlsbad,Canada)提取总RNA,然后使用逆转录酶试剂盒(Takara,Shiga,Japan)逆转录mRNA。qRT-PCR反应由ABI Prism 7500序列检测系统(Applied Biosystems,Foster City,CA)监测,并对每个样品重复进行。
PCR反应混合物(20μl)由2μl cDNA模板,0.6μl正向和反向引物和10μl 2×SYbr-green PCR Mix(Takara)组成。靶向ATG16L1的引物(F:5'-GCAAGCCGAATCTGGACT-3',R:5'-CCTGAGACTATCCGTGCAT-3')用于Real-time RT-PCR扩增。通过融合曲线分析和凝胶电泳确定PCR特异性,并通过标准曲线法分析数据。以β-actin为内参,用2-ΔΔCt法计算目的基因水平。
3.Westernblotting。
细胞用PBS洗涤3次后,用RIPA Biotechnology buffer(BeyotimeBiotechnology,中国上海)提取细胞总蛋白。按照说明书的方案,使用BCA蛋白质测定试剂盒(Beyotime Biotechnology,中国上海)测量蛋白质浓度。通过10%SDS-PAGE凝胶分离蛋白质并转移至PVDF膜。将膜用5%脱脂牛奶在37℃下封闭2小时,然后与特异性一抗在4℃下孵育过夜。1×TBST洗膜3次后,辣根过氧化物酶偶联二抗37℃孵育2小时。使用增强的化学发光试剂使蛋白质条带可视化。
4.伤口愈合实验
将细胞接种在6孔板中并培养至约90%汇合。使用无菌移液管的尖端划伤细胞层以形成伤口。用PBS去除细胞碎片并更换新鲜培养基。然后将细胞在不同的处理组中孵育。在0和48小时在显微镜下拍摄相同视野的图像。每个实验重复三次。使用Image J软件评估伤口愈合的百分比。
5.Transwell侵袭试验。
将不同组处理的RCEC细胞分别稀释至1×105/mL。将200μL无血清DMEM细胞悬液加入到涂有Matrigel的上层transwell室中。将含有20%FBS的600μL DMEM装入下室。在37℃孵育24小时后,入侵细胞用4%多聚甲醛固定,用0.1%结晶紫染色。拍摄数码照片并使用Image J软件测量迁移细胞。
6.统计分析。
测量数据表示为平均值和标准偏差。独立学生t检验用于比较两组之间的正态分布连续变量。卡方检验或Fisher精确检验用于比较非连续变量。实验至少独立重复三次。使用SPSS 20.0(SPSS Inc.,Chicago,IL,USA)进行统计分析。p<0.05的值被认为具有统计学意义。
二、实验结果
1.单2’-O-Me修饰对siRNA沉默效率和稳定性的影响。
本发明所采用的siRNA的序列为:
Sense Sequence:5’-UCAUUCUUCUGAUGCUGCCAGGAGA-3’(SEQ ID NO:1)
Antisense Sequence:5’-UCUCCUGGCAGCAUCAGAAGAAUGA-3’
(SEQ ID NO:2)
沉默效率和稳定性的检测模型为检测RCECs中ATG16L1的动态表达及siRNA转染后的表达变化。将siRNA分子转染RCECs后,于高糖条件下培养24h后收集细胞并进行Real-time PCR。
针对正义链的不同位点分布进行2’-O-Me修饰,结果如下表所示:
其中,抑制率=(NC阴性对照组mRNA相对表达水平-siRNA组的mRNA相对表达水平)÷NC阴性对照组mRNA相对表达水平。
NC为转染的是相对于实验组乱序的siRNA。
选取其中抑制率最高的AL06、AL08、AL10、AL21再次进行Real-time PCR验证,此次与24h验证的区别在于,选取的时间点为高糖条件下培养96h。结果如下:

从上述结果可以看出,AL06在96h抑制率最高,这一方面可能与其24h的高活性有关,但更重要的是正义链第6位核苷酸的单2’-O-Me修饰使得其稳定性获得更多的提升,因而能够持续发挥抑制作用。
2.蛋白水平的变化
为进一步验证AL06的效果,我们进一步采用WB的方法检测了ATG16L1的变化水平。总体而言,相同浓度葡萄糖不同时间处理24h后各组细胞中ATG16L1的mRNA表达均低于处理96h后的细胞。与暴露于正常葡萄糖的细胞相比,暴露于高糖24h的细胞中ATG16L1蛋白表达增加,而ATG16L1 siRNAAL06组细胞中ATG16L1蛋白水平降低(图1中A,C)。与处理24小时的相应组相比,暴露于正常和高葡萄糖96小时的细胞中ATG16L1蛋白表达增加(图1中B、C)。
3.ATG16L1对RCECs迁移能力的影响
用ATG16L1 siRNA或对照siRNA转染的RCEC被机械划伤以产生伤口。在暴露于正常葡萄糖或高葡萄糖96小时后,我们测量了伤口愈合的程度。通过检查与初始伤口区域相比的伤口区域,我们发现高糖处理显着增强了RCECs细胞的迁移能力(图2)。此外,与对照组相比,转染ATG16L1 siRNAAL06后,正常组和高糖组RCECs的细胞迁移能力均受到显着抑制(p<0.01),表明ATG16L1在促进迁移中起关键作用RCEC的能力。
4.ATG16L1对RCECs侵袭能力的影响。
为了验证ATG16L1在细胞侵袭中的作用,我们进一步进行了transwell迁移试验。结果表明,与正常葡萄糖组相比,高糖组RCEC的侵袭率显着增加(图3)。与对照组相比,转染ATG16L1 siRNAAL06后高糖组和正常糖组RCECs的侵袭率均得到显着抑制(p<0.01),表明ATG16L1也增强了RCECs的侵袭能力。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准,说明书及附图可以用于解释权利要求的内容。
Claims (10)
1.特异性抑制ATG16L1表达的核酸,其为siRNA或shRNA,包含SEQ ID NO:1所示的正义链和SEQ ID NO:2所示的反义链,所述正义链和反义链互补共同形成RNA二聚体。
2.根据权利要求1所述的核酸,所述正义链中的至少一个核苷酸具备化学修饰。
3.根据权利要求2所述的核酸,所述正义链中至少一个核苷酸中的核糖2’-OH被甲氧基所取代。
4.根据权利要求3所述的核酸,所述正义链第6、8、10、21位核苷酸中的核糖2’-OH被甲氧基所取代;优选所述正义链第6位核苷酸中的核糖2’-OH被甲氧基所取代。
5.一种能够产生权利要求1~4任一项所述核酸的DNA分子。
6.包含权利要求5所述DNA分子的载体。
7.递送系统,其包含i)权利要求1~4任一项所述核酸,或权利要求5所述的DNA分子,以及ii)递送媒介物。
8.根据权利要求7所述的递送系统,所述递送媒介物包括一种或多种脂质体、一种或多种外泌体、一种或多种微囊泡、一种或多种树状大分子、一种或多种无机纳米粒子、一种或多种细胞穿膜肽、基因枪、一种或多种质粒、一种或多种病毒载体,以及它们所组成的组。
9.药物组合物,其包含权利要求8所述的递送系统以及药学上可接受的赋形剂;优选还包含核酸稳定剂。
10.权利要求1~4任一项所述核酸,或权利要求5所述的DNA分子,或权利要求7或8所述的递送系统在制备用于促进伤口愈合的药物中的应用。
