Abstract
本申请属于细胞治疗技术领域,尤其涉及仿生纳米酶纤维的制备方法及其在细胞治疗中的应用。本申请提供了一种仿生纳米酶纤维的制备方法,包括:将抗氧化金属纳米酶和生物可降解高分子聚合物,在溶剂条件下进行复合和纤维化,制得仿生纳米酶纤维;其中,所述抗氧化金属纳米酶负载在所述仿生纳米酶纤维上。本申请的仿生纳米酶纤维,可原位移植在损伤实体器官以调控其ROS稳态,还能与负载功能细胞药物起到协同增效增强细胞治疗效果的作用,有效解决现有细胞治疗药物容易在静脉注射后被肺内阻塞,或在移植后被巨噬细胞/吞噬细胞清除的问题。

技术领域
本申请属于细胞治疗技术领域,尤其涉及仿生纳米酶纤维的制备方法及其在细胞治疗中的应用。
背景技术
急性肝衰竭(ALF)是一种具有广泛肝细胞坏死和肝功能急性恶化的危及生命的疾病。然而,目前除肝移植外,尚无令人满意的治疗方法。据报道,活性氧(reactive oxygenspecies,ROS)的过度生成和过度积累是ALF的早期病理特征,并与疾病的进展密切相关,原因包括诱导的氧化应激、炎症风暴和大量肝细胞坏死。因此,早期有效消除ROS,避免肝细胞不可逆损伤,补充功能性肝细胞对ALF的治疗至关重要。
虽然有效清除ROS可以缓解局部恶劣的微环境,但ALF中大量肝细胞坏死和肝功能丧失仍需要功能性肝细胞进行必要的代偿。然而,供体有限、细胞增殖不良、体外维持肝细胞功能困难等阻碍了其治疗应用。间充质干细胞(mesenchymal stem cells,MSCs)具有多谱系分化潜能、自我更新能力以及抗氧化和抗炎功能,可以方便地在体外分离和扩增。此外,MSCs在临床应用中也表现出较低的免疫反应,且不存在伦理问题。研究证明,MSCs可以通过多种细胞因子阶段诱导有效地分化为功能性肝细胞样细胞(hepatocyte-like cells,HLCs)。MSCs来源的HLCs不仅具有抗炎活性,而且具有肝细胞特异性功能。因此,MSCs来源的HLCs显示出作为肝细胞治疗替代细胞来源的巨大潜力。然而,有研究表明,大多数植入细胞在静脉注射后被肺内阻塞,或在脾内移植后被巨噬细胞/吞噬细胞清除。此外,ALF肝脏的微环境是细胞治疗的一个关键考虑因素,细胞植入的炎症环境可引起移植细胞坏死和凋亡。
因此,亟需一种可有效清除ROS、副作用低、可原位移植功能性细胞的载体或者药物。
发明内容
本申请提供了一种仿生纳米酶纤维的制备方法和仿生纳米酶纤维,可原位移植在损伤实体器官以调控其ROS稳态,还能与负载功能细胞起到协同增效增强细胞治疗效果的作用,有效解决现有细胞治疗容易在静脉注射后被肺内阻塞,或在移植后被巨噬细胞/吞噬细胞清除的问题。
本申请第一方面提供了一种仿生纳米酶纤维的制备方法,包括:
将抗氧化金属纳米酶和生物可降解高分子聚合物,在溶剂条件下进行复合和纤维化,制得仿生纳米酶纤维;其中,所述抗氧化金属纳米酶负载在所述仿生纳米酶纤维上。
在一些实施例中,所述抗氧化金属纳米酶选自铜基纳米酶、金纳米酶、银纳米酶和铂纳米酶中的一种或多种。
作为优选,本申请所用的抗氧化金属纳米酶为铜基纳米酶Cu NZs。根据文献《Ultrasmall copper-based nanoparticles for reactive oxygen species scavengingand alleviation of inflammation related diseases》制备。
虽然铜基纳米酶Cu NZs具有抗氧化能力,但是常规静脉注射铜基纳米酶往往导致器官分布不理想、治疗效果差以及引起副作用的缺陷,因此,本申请发现将铜基纳米酶CuNZs与生物可降解高分子聚合物进行复合和纤维化,制备可原位移植的具有调控ROS稳态的仿生抗氧化纳米纤维,有效解决铜基纳米酶器官分布不理想的缺点。
在一些实施例中,所述生物可降解高分子聚合物选自聚乳酸聚乙醇酸共聚物PLGA、聚己内酯PCL、聚乳酸PLA和明胶中的一种或多种;
所述溶剂选自六氟异丙醇HFIP、二氯甲烷和四氢呋喃中的一种或多种。
作为优选,所述生物可降解高分子聚合物为聚乳酸聚乙醇酸共聚物PLGA;所述溶剂为六氟异丙醇HFIP。
更具体的,所述聚乳酸聚乙醇酸共聚物PLGA中,LA/GA=50/50,PLGA最大Mw为100000。所述聚乳酸聚乙醇酸共聚物PLGA在体系中为10wt/v%。
在一些实施例中,所述纤维化选自静电纺丝、3D打印或旋涂。
作为优选,所述纤维化为静电纺丝;静电纺丝的电压参数:13kV,静电纺丝的给料速率0.6mL h-1。
具体的,本申请的仿生纳米酶纤维制备方法包括:将Cu NZs和聚乳酸聚乙醇酸共聚物PLGA分散在六氟异丙醇HFIP中,进行静电纺丝成纳米纤维。收集静电纺丝纳米纤维,并将其置于真空以去除残留溶剂。
本申请第二方面提供了一种仿生纳米酶纤维,包括所述制备方法制得的仿生纳米酶纤维。
本申请第三方面公开了所述制备方法制得的仿生纳米酶纤维或所述仿生纳米酶纤维在制备原位细胞移植药物中的应用。
本申请第四方面提供了一种原位细胞移植药物,包括:
仿生纳米酶纤维和负载有功能性细胞的载药凝胶;
其中,所述仿生纳米酶纤维为所述制备方法制得的仿生纳米酶纤维。
在一些实施例中,所述功能性细胞选自功能性肝细胞样细胞、治疗性干细胞、原代肝细胞和肝细胞系AML-12中的一种或多种;所述功能性细胞还包括内皮细胞;
所述载药凝胶选自脱细胞基质dECM水凝胶、PLGA-PEG-PLGA、天然高分子水凝胶和胶原蛋白水凝胶中的一种或多种。
在一些实施例中,所述功能性细胞还包括内皮细胞,即功能性肝细胞样细胞、治疗性干细胞、原代肝细胞和肝细胞系AML-12中的一种或多种与内皮细胞组合得到的功能性细胞。
在一些实施例中,所述天然高分子水凝胶为透明质酸等水凝胶。
作为优选,所述功能性细胞为功能性肝细胞样细胞,功能性肝细胞样细胞(hepatocyte-like cells,HLCs)由MSCs通过多种细胞因子阶段诱导有效地分化得到。
具体的,HLCs具有与原代肝细胞相似的肝脏功能,并具有抗炎和促进再生作用。研究表明,肝细胞分化后,MSCs来源的HLCs获得了肝细胞功能,包括ALB形成、细胞色素P450酶活性、尿素分泌等。HLCs可以恢复肝脏氨和嘌呤代谢,降低转氨酶和氨水平,提高血清ALB水平,在肝病治疗中比未分化MSCs更有效。HLCs还减少了炎症因子IL-1β、IL-6和TNF-α的产生,以缓解CCl4诱导的ALF小鼠的炎症。因此,通过细胞因子诱导hADMSCs向肝系分化,获得用于治疗ALF的HLCs。
作为优选,所述载药凝胶为脱细胞基质(dECM)水凝胶,dECM水凝胶提供了理想的3D环境,有利于提高功能性细胞的治疗作用。
更为优选,所述脱细胞基质水凝胶为肝脏脱细胞外基质(dECM)水凝胶,肝脏脱细胞外基质(dECM)水凝胶可为负载的功能性细胞提供类似天然肝脏的微环境,且无不良免疫反应。然而,该dECM水凝胶的机械强度较弱,且损伤部位的恶劣微环境使得植入的功能性细胞难以发挥作用。本申请发现将具有调控ROS稳态能力的仿生纳米酶纤维与载药凝胶组合可有效提高载药凝胶中功能性细胞的治疗作用。
本申请第五方面公开了所述原位细胞移植药物在制备治疗肝脏疾病药物中的应用。
在一些实施例中,所述实体器官疾病包括肝脏疾病、皮肤创伤或心肌损伤修复。
作为优选,所述肝脏疾病包括肝纤维化或急性肝衰竭。
本申请开发了一种抗氧化金属纳米酶负载生物可降解高分子聚合物的仿生纳米酶纤维,该仿生纳米酶纤维复合了氧化金属纳米酶后,具有明显的ROS消除能力、良好的生物相容性和促血管生成作用。此外,本申请的仿生纳米酶纤维还可与载药凝胶组成的协同治疗平台,用于在实体器官中原位递送功能性细胞,以有效治疗实体器官疾病。本申请发现仿生纳米酶纤维可协同载药凝胶在实体器官中原位递送功能性细胞,一方面仿生纳米酶纤维对载药凝胶提供机械支撑,使后者在实体器官中提供3D环境以便功能性细胞发挥效果,另一方面仿生纳米酶纤维中的抗氧化金属纳米酶能有效清除ROS可以缓解局部恶劣的微环境,避免功能性细胞受到损伤器官的氧化微环境的干扰,促进功能性细胞及时发挥功能,并在早期缓解氧化应激和炎症。本申请提供的原位细胞移植药物对实体器官损伤的治疗效果显著、高效,表现为减少细胞凋亡和组织坏死,增加细胞增殖和血管化。

具体实施方式
本申请公开了仿生纳米酶纤维的制备方法及其在细胞治疗中的应用,用于解决现有技术中细胞治疗药物容易在静脉注射后被肺内阻塞,或在移植后被巨噬细胞/吞噬细胞清除的技术缺陷。
其中,以下实施例所用试剂或药物均为市售或自制。
以下实施例使用的引物如表1。

本申请实施例的试验流程如图1,①合成铜基纳米酶Cu NZs、②制备仿生纳米酶纤维(简称Cu NZs@PLGA纳米纤维)、③Cu NZs@PLGA纳米纤维与负载HLCs的dECM水凝胶组合形成原位细胞移植药物(简称HLCs/Cu NZs@fiber/dECM),该原位细胞移植药物用于递送hADMSCs诱导分化的HLCs,用于治疗CCl4诱导的ALF;图1(A,B)Cu NZs@PLGA纳米纤维、dECM水凝胶和复合材料HLCs/Cu NZs@fiber/dECM的合成;图1(C)HLCs/Cu NZs@fiber/dECM通过消除ROS、促进血管生成、抗炎、肝细胞相关功能和促进肝脏再生的作用对CCl4诱导的ALF治疗的示意图。
实施例1
本实施例提供了铜基纳米酶Cu NZs的合成与表征,具体包括:
将CuCl2(7447-39-4,Aladdin,China)分散在50mL的去离子水中,终浓度为10mM。在80℃搅拌10分钟后,将50mL 100mM抗坏血酸(50-81-7,AlfaAesar,USA)水溶液缓慢加入到CuCl2溶液中。使用1M NaOH(1310-73-2,Macklin,China)将混合溶液pH调整为8.5±0.5,并在80℃下搅拌12小时。反应结束后,离心去除沉淀,上清液使用10kDa透析管(TX132,Viskase,China)对去离子水进一步透析48小时。通过离心分离得到最终的Cu NZs。在120kV电压下,用透射电镜(TEM,FEI Tecnai G2 Spirit)对Cu NZs的形貌和尺寸进行了表征。采用x射线衍射仪(XRD,Empyrean)在45kV电压、40ma电流下对Cu NZs的晶体结构和氧化态进行了表征。
结果如图2和图3所示,Cu NZs首先通过透射电镜进行了表征,表明其分布均匀,平均粒径约为4.0nm(图2A,图3A)。Cu NZs的zeta电位约为-14.4mV(图2B)。根据标准XRD谱图,Cu NZs为Cu2O和Cu的化合物,说明Cu NZs的成功合成(图3B)。
实施例2
本实施例提供了采用静电纺丝制备Cu NZs@PLGA纳米纤维的方法:
1、将Cu NZs(600μg mL-1)和10wt/v%poly(乳酸-共乙醇酸)(PLGA,LA/GA=50/50,Mw~100,000,Shenzhen Polymtek Biomaterial)均匀分散在六氟异丙醇(HFIP,99.8%,Aladdin,China)中,在13kV电压下,给料速率0.6mL h-1静电纺丝成纳米纤维。收集静电纺丝纳米纤维,并将其置于真空中室温过夜以去除残留溶剂。采用相同的工艺制备了不添加CuNZs的普通PLGA纳米纤维。在20kV电压下,利用扫描电子显微镜(SEM,Quanta 400F)对纳米纤维进行形貌表征和能量色散谱(EDS)分析。
2、对Cu NZs、Cu NZs@PLGA纳米纤维的表征,结果如图3和图4所示,SEM成像表明,Cu NZs未明显改变PLGA纳米纤维的光滑表面和长纤维形态,平均直径为382.5±74.5nm(图3C,图4)。EDS元素映射成像发现C、O和Cu均匀分布在PLGA纳米纤维中(图3D),进一步证实了Cu NZs在纳米纤维中的成功负载。
实施例3
本实施例提供了Cu NZs和Cu NZs@PLGA纳米纤维对H2O2、·OH和O2·-的清除能力,具体包括:
1、检测清除H2O2的能力:
将不同浓度的Cu NZs置于磷酸盐缓冲溶液中,从0到200ng mL-1,分别与1mM H2O2(CS7730,G-Clone,China)在37℃下孵育2小时。剩余的H2O2由Amplex Red(Meilunbio,China)和HRP(Meilunbio,China)进行检测。Amplex Red与H2O2反应生成吸收峰在571nm的试卤灵。采用UV-2600紫外可见(UV-Vis)分光光度计(SHIMADZU,China)定量。不含Cu NZs的磷酸盐缓冲溶液作为对照。
含有不同量Cu NZs(0-200ng mL-1)的Cu NZs@PLGA纳米纤维浸泡在磷酸盐缓冲溶液中,用1mM H2O2在37℃下孵育0-10h。在2、4、6、8、10h检测各组剩余H2O2。以不含Cu NZs的PLGA纳米纤维作为对照。H2O2清除率计算公式:清除率(%)=(Ac-Ai)/Ac×100%。Ac是对照组的吸光度,Ai是样品的吸光度。
2、Cu NZs@PLGA纳米纤维的清除·OH能力检测:
采用典型的3,3',5,5'-四甲基联苯胺(TMB)(54827-17-7,Aladdin,China)显色法监测不同时间点的·OH清除率。Cu NZs@PLGA纳米纤维在醋酸/醋酸钠缓冲液(HAc/NaAc,0.5M,pH 4.5)(Aladdin,China)中预孵育,最终Cu NZs浓度为150ng mL-1,分别孵育0、2、6和12h。收集上清液,与含有1mM FeSO4(7782-63-0,Sangon,China)、1mM H2O2和250μM TMB的工作溶液在黑暗中孵育7分钟。无纳米纤维预处理的HAc/NaAc缓冲液作为对照。·OH氧化后的TMB(oxTMB)呈蓝色,用紫外-可见分光光度计测定在652nm处的吸光度。据此,采用与H2O2清除率相同的公式计算·OH清除率。
3、Cu NZs@PLGA纳米纤维清除O2·-的能力检测:
采用硝基四氮唑蓝(NBT)显色法研究Cu NZs@PLGA纳米纤维的O2·-清除活性。CuNZs@PLGA纳米纤维浸泡在Tris-HCl缓冲液(10mM,pH 7.4)(Biosharp,China)中,最终CuNZs浓度为150ng mL-1。预孵育0、2、6和12h后,收集上清液,分别与含有6.5mM L-甲硫氨酸(C11385354,Macklin,China)、37.5μM NBT(C11167116,Macklin,China)和10μM核黄素(C11507808,Macklin,China)的工作溶液均匀混合,并使用SCIENTZ03-II型紫外辐射仪(Scientz,China)进行紫外照射,波长254nm(60W,150mm),紫外照射导致核黄素和L-甲硫氨酸生成O2·-,进一步将NBT转化为甲瓒。光照后,用紫外-可见分光光度计测定混合溶液在560nm处的吸光度。O2·-的清除率按照H2O2清除率公式计算。
Cu NZs和Cu NZs@PLGA纳米纤维对H2O2、·OH和O2·-的清除效率检测,结果如图3及图5-7所示。随着Cu NZs浓度的增加,会逐渐引起571nm处的紫外吸收强度衰减和比色褪色,说明Cu NZs对H2O2的清除效率与浓度有关(图5)。值得注意的是,加入超低浓度(200ngmL-1)的Cu NZs可以消除高达83.4%的H2O2(图5C)。当Cu NZs被包裹在PLGA纳米纤维中时,H2O2的清除效率不受影响,并表现出时间和浓度依赖性(图6)。含有150ng mL-1和200ng mL-1等效Cu NZs浓度的Cu NZs@PLGA纳米纤维在10h时,对H2O2的清除率分别为68.1%和75.0%(图3G-I,图6)。此外,在去除H2O2的过程中,管壁上出现气泡,表明H2O2被Cu NZs转化为O2(图7)。此外,以Cu NZs当量浓度为150ng mL-1的Cu NZs@PLGA纳米纤维为例,测定Cu NZs@PLGA纳米纤维对·OH和O2·-的清除能力。Cu NZs@PLGA纳米纤维能够消除Fenton反应产生的·OH。随着孵育时间的延长,oxTMB的紫外吸收峰逐渐降低,说明Cu NZs@PLGA纳米纤维中的CuNZs能有效消除Fe2+和H2O2产生的·OH。特别是Cu NZs@PLGA纳米纤维在孵育12h后对·OH的清除效率接近100%(图3J-L)。此外,转化的NBT在560nm处的紫外吸光度下降也表明CuNZs@PLGA纳米纤维成功消除了O2·-,在12h时O2·-清除率约为48.5%(图3M-O)。综上所述,这些结果有力地证明了Cu NZs@PLGA纳米纤维能够有效地消除H2O2、·OH和O2·-。
实施例4
本实施例提供了Cu NZs@PLGA纳米纤维清除细胞内ROS的能力以及对HUVECs迁移和增殖影响的试验,包括:
1、细胞培养:
hADMSCs购自中国赛莱拉干细胞科技有限公司。hADMSCs在含5%血清替代品的无血清培养基中培养(G03010,Saliai,China)。第5和第9代之间的hADMSCs用于实验。AML12细胞购自国家认证细胞培养库(中国),用含有10%胎牛血清(FBS)(Gibco,美国)、1×胰岛素-转铁蛋白-硒补充剂(ITS)(ITSS-10201,Cyagen,中国)和40ng mL-1地塞米松(D4902,Sigma-Aldrich,美国)的DMEM/F-12培养基(11330-032,Gibco,美国)培养。人脐静脉内皮细胞(HUVECs)在含10%FBS、1%双抗(Gibco,USA)的DMEM/F-12中培养。用RPMI 1640培养基(Gibco,USA)、10%FBS、1%双抗和50μMβ-巯基乙醇(Sigma-Aldrich,USA)培养人单核细胞THP-1。RAW264.7细胞用DMEM培养基(Gibco,USA)、10%热灭活FBS、1%双抗培养。所有细胞在37℃、5%CO2的培养箱中培养。
2、测定Cu NZs@PLGA纳米纤维在AML12细胞中清除ROS的能力:
将AML12细胞以每孔2×104个细胞的密度接种在48孔板上,用不同含量的Cu NZs@PLGA纳米纤维(等效Cu NZs浓度从0ng mL-1到200ng mL-1)预孵育过夜,再用含有500μM H2O2的细胞培养液刺激细胞24小时,之后使用CCK-8评估细胞活力,并按照制造商的说明使用2',7'-二氯荧光素二乙酸(DCFH-DA,S0033S,Beyotime,China)标记细胞内ROS。10μM DCFH-DA染色液与AML12细胞在37℃黑暗环境下孵育30分钟,去除染色液后加入4%多聚甲醛(PFA,Biosharp,China)固定细胞,用0.2μg mL-14,6-二脒基-2-苯基吲哚(DAPI)在室温下染色15min,用倒置荧光显微镜(Nikon,日本)进行荧光成像,用流式细胞仪(Beckman,CytoFLEX,美国)进行平均荧光强度定量。
3、测定Cu NZs@PLGA纳米纤维的生物相容性:
将AML12细胞、hADMSCs和HLCs分别以每孔1×104细胞的密度接种于96孔板中。将Cu NZs@PLGA纳米纤维(等效Cu NZs浓度从0ng mL-1到200ng mL-1)与细胞孵育24小时,使用CCK-8评估细胞活力。通过使用酶标仪(BioTek)检测450nm处吸光度定量细胞活力。
4、测定Cu NZs@PLGA纳米纤维对HUVECs迁移和增殖的影响试验:
通过划痕试验研究了Cu NZs@PLGA纳米纤维对HUVECs迁移的影响。HUVECs以每孔5×104的密度接种于24孔板上,培养过夜。将细胞在无血清培养基中培养24小时,然后用1mL无菌吸管尖端在孔板底部划出一条直线。用磷酸缓冲液轻轻清洗细胞两次以清除细胞碎片,然后用含有1%FBS和Cu NZs@PLGA纳米纤维(等效Cu NZs浓度从0ng mL-1到200ng mL-1)的完全培养液培养细胞。分别在24和48小时对划痕区域进行拍照成像,使用Image J软件(NIH,USA)测量和统计。细胞迁移统计公式:细胞迁移(%)=((S0-St)/S0)×100%,其中S0为初始划痕区域面积,St为24或48小时的划痕区域面积。
EdU试剂盒(K1076,APExBIO,USA)用于评估Cu NZs@PLGA纳米纤维对HUVECs增殖的影响。细胞处理与划痕试验中相同,只是每个孔的细胞接种密度较低,为1×104。在Cu NZs@PLGA纳米纤维处理48h后使用EdU试剂盒对HUVECs进行染色,并用倒置荧光显微镜拍照成像,用Image J测量EdU阳性信号的增殖细胞,HUVECs增殖率(%)=EdU阳性细胞/总细胞×100%。在每个孔中随机选择的三个区域对EdU阳性细胞进行定量。
结果如图8-图10所示,将细胞与Cu NZs@PLGA纳米纤维(含有等效Cu NZs浓度0ngmL-1至200ng mL-1)孵育24小时后,CCK-8实验显示,与未处理的空白组相比,细胞存活率在95.0至107.9%之间变化,表明Cu NZs@PLGA纳米纤维具有良好的生物相容性(图9)。随后,受Cu NZs@PLGA纳米纤维优异的ROS清除能力的启发,本实施例检测Cu NZs@PLGA纳米纤维对细胞内ROS的清除能力及细胞保护能力。通过流式细胞术分析DCFH-DA染色后细胞内ROS生成情况,当Cu NZs@PLGA纳米纤维中Cu NZs当量浓度达到150ng mL-1时,Cu NZs@PLGA纳米纤维处理后DCF荧光强度较对照组下降1.8倍。结果表明,细胞内ROS可以明显消除(图8B-C),与DCFH-DA染色图像一致(图8E)。同样,CCK-8试验(图8D)也证实,与对照组(细胞活力53.0%)相比,Cu NZs@PLGA纳米纤维在等效Cu NZs浓度为100ng mL-1和150ng mL-1时,明显提高了H2O2处理后的细胞存活率,分别达到82.2%和88.3%。因此,Cu NZs@PLGA纳米纤维能够明显缓解氧化应激引起的细胞损伤,从而促进高ROS条件下肝细胞的存活。后续实验中,将等效150ng mL-1Cu NZs的Cu NZs@PLGA纳米纤维作为最适浓度。
本实施例分别采用划痕实验和EdU染色成像进一步测定了Cu NZs@PLGA纳米纤维对HUVECs迁移和增殖的影响。如图8F-G所示,与未处理的对照组相比,Cu NZs@PLGA纳米纤维孵育后,更多的细胞向划痕中央迁移,且呈浓度依赖模式。特别是,在加入Cu NZs当量浓度为150ng mL-1的Cu NZs@PLGA纳米纤维处理24h后,划痕区域基本被细胞覆盖。Cu NZs@PLGA纳米纤维处理48h后,各浓度组划痕区域完全恢复(图8F-G)。EdU染色成像显示,CuNZs@PLGA纳米纤维处理48h后,EdU-阳性细胞增多(图8H-I)。与未处理的对照组相比,150ngmL-1Cu NZs@PLGA纳米纤维浓度组中HUVECs增殖率增加了21.1%(图10)。Cu NZs@PLGA纳米纤维的促血管生成作用可能加速移植HLCs与宿主肝脏的整合及功能化,从而促进肝脏再生。上述结果说明,Cu NZs@PLGA纳米纤维具有促血管生成从而促进肝脏再生的作用。
实施例5
本实施例提供了体外hADMSCs诱导分化的HLCs及其表征,具体包括:
1、利用细胞因子诱导hADMSCs向肝细胞谱系分化。首先在含有20ng mL-1表皮生长因子(EGF,PeproTech,美国)和10ng mL-1碱性成纤维细胞生长因子(bFGF,PeproTech,美国)的低糖DMEM(Gibco,美国)无血清培养液中培养两天。随后,细胞在含有20ngmL-1肝细胞生长因子(HGF,PeproTech,美国)、10ng mL-1bFGF和0.61mg mL-1烟酰胺(Sigma-Aldrich,美国)的低糖DMEM培养液中诱导分化一周。之后,在含有20ng mL-1oncostatin M(OSM,PeproTech,美国)、1mM地塞米松和1×ITS的低糖DMEM培养液中继续培养两周。培养基每周更换两次。
如图11A为hADMSCs诱导肝分化时间表示意图。用ALB测定试剂盒(CH0101002,Maccura,中国)和尿素测定试剂盒(CH0101051,Maccura,中国)分别于第7天和第21天测定培养液上清中产生的白蛋白(ALB)和尿素。细胞在4%PFA中固定15分钟,按照制造商说明分别用Oil Red O(Sigma-Aldrich,USA)和periodic acid-Schiff(PAS,Sigma-Aldrich,USA)对累积的脂质和糖原进行染色。
2、实时定量PCR和免疫荧光染色检测hADMSCs是否成功诱导分化为HLCs,以及检测HLCs的特征,具体包括:
实时定量PCR:总RNA通过TRIzon试剂(CW0580S,Cwbiotech,中国)从细胞中提取,使用HiFiScript cDNA合成试剂盒(CW2569M,Cwbiotech,中国)进行反转录。实时荧光定量PCR使用UltraSYBR混合物(CW2601,Cwbiotech,中国),采用QuantStudioTM5系统(AppliedBiosystems)进行检测。所有数据以ACTB或GAPDH的CT值进行归一化处理,使用delta-deltaCt法计算相对表达量。
免疫荧光染色:细胞在4%PFA常温下固定15min,用5%山羊血清孵育1h,4℃一抗孵育过夜。兔抗人一抗稀释如下:ALB(16475-1-AP,Proteintech,美国)(1:500),E-cadherin(710161,Invitrogen,美国)(1:200),DPP4(PAB32046,中国)(1:200),HNF4α(61189,Active Motif,美国)(1:200),ABCC2(PAB35870,中国)(1:200)。用AlexaFluor 488山羊抗兔二抗(1:500)常温下避光孵育2h,用磷酸缓冲液洗涤2次。用0.2μg mL-1DAPI染色细胞核,常温下孵育15min。最后,使用倒置荧光显微镜成像。
3、测定HLCs的抗炎能力,具体包括:
将THP-1细胞以每孔8×105的密度接种于6孔板上,用含100ngmL-112-myristate13-acetate(PMA)(Sigma-Aldrich,USA)的培养基处理48h诱导THP-1细胞(M0)活化。然后去除含PMA的培养基。用新鲜培养基和HLCs上清液(1:1)培养THP-1细胞(M0)24或48h。
用脂多糖(LPS,100ng mL-1)(Sigma-Aldrich,USA)刺激THP-1(M0)和Raw264.7细胞24h诱导M1极化。然后,除去含脂多糖的培养基。用新鲜培养基和HLCs上清液(1:1)处理极化的THP-1和Raw264.7细胞24h。收集细胞,通过qPCR检测THP-1和Raw264.7细胞中IL 1β、TNFA、NOS2、TGFβ1(促炎因子)、IL 10(抗炎因子)、MRC 1(M2型巨噬细胞标志物)的表达,评估巨噬细胞的极化状态。
4、hADMSCs诱导分化后的HLCs特征结果如图11-13。本实施例应用细胞因子的两步分化方案诱导干细胞肝分化(图11A)。分化后,纺锤状的hADMSCs变为多边形形状(图11B,图12)。与hADMSCs相比,诱导的HLCs分别通过PAS染色和Oil Red O染色显示出明显的糖原产生和脂质积累(图11B)。此外,肝细胞相关蛋白,如ALB、HNF4α、ABCC2、E-cadherin和DPP4也在HLCs中高表达(图11C-E,图13)。从细胞分化开始到结束,追踪肝细胞特异性基因的mRNA表达。HLCs中ALB和CYP3A4基因在第21天的mRNA表达量是第0天的10.0倍以上(图11F)。相反,与第7天相比,未成熟肝细胞标志物AFP水平在第21天明显降低(图11F)。此外,分化21天后HLCs培养基上清液中ALB和尿素的分泌水平分别比分化第7天高5.3倍和2.1倍(图11G)。综上所述,这些结果表明,hADMSCs被成功诱导成具有典型肝细胞形态和相关功能的HLCs,有利于改善ALF的肝脏生物功能。
5、HLCs的炎症调节作用结果如图14-15。本实施例测定了HLCs上清液对巨噬细胞极化的影响。首先,将HLCs上清液与THP-1(M0)孵育24或48h,通过qPCR分析巨噬细胞的基因表达,评估巨噬细胞的极化情况。与THP-1(M0)相比,随着处理时间的延长,HLCs上清液处理的THP-1中促炎基因IL 1β、NOS2和TGFβ1的表达明显降低,而抗炎基因IL 10和MRC 1在48h时分别升高3.3倍和33.9倍(图14A-B)。这表明HLCs上清可以使巨噬细胞向M2极化(抗炎表型)。接下来,本实施例进一步探究HLCs上清是否可逆转炎症状态下巨噬细胞极化。结果显示,LPS处理后THP-1的IL 1β、TNFA、TGFβ1表达上调,表明THP-1(M0)成功诱导为M1表型(促炎表型)。值得注意的是,与LPS处理组相比,HLCs上清处理的THP-1中IL 1β、TNFA和TGFβ1的表达明显下降,接近THP-1的水平(M0)。相比之下,经HLCs上清处理后,IL 10和MRC 1水平显著升高至23.6倍和21.1倍(图14C-D)。同时,本实施例在RAW264.7细胞中也得到了类似的结果(图15)。这些结果表明,HLCs上清可有效促进炎症状态下巨噬细胞表型的转化。
此外,本实施例还研究了HLCs是否对高氧化应激下的肝细胞具有保护作用。H2O2处理后,单独培养的AML12细胞(对照组)观察到大量漂浮的死亡细胞和碎片,但与HLCs或hADMSCs共培养的细胞则没有(图16)。与单独培养的AML12细胞以及与hADMSCs共培养的AML12细胞相比,与HLCs共培养的AML12细胞具有更好的细胞活力(图24)。通过对AML12细胞中抗氧化基因表达的表征,本实施例发现当AML12细胞与HLCs共培养时,抗氧化基因SOD2、NQO1和KEAP1的mRNA表达显著上调2.2-3.8倍(图16C)。因此,在ALF治疗中,HLCs可能通过提高宿主肝细胞的抗氧化能力和细胞活力来促进肝脏再生。
实施例6
本实施例提供了dECM水凝胶的制备、生物相容性及其对肝功能的影响试验,具体包括:
1、dECM水凝胶的制备:
约200克猪肝,在去除连接的组织后轻轻切碎。肝组织需经过-80℃冷冻、常温解冻的3次循环,期间用去离子水冲洗。肝组织在含1%TritonX-100、0.1%NH3·H2O、1%青霉素/链霉素、0.01mM苯甲基磺酰氟(PMSF)的去离子水中脱细胞3天,每天换液3次。在冷冻干燥前,脱细胞肝组织在无菌水中洗涤1小时。所有步骤都在150转/分钟的搅拌下进行。为了制备dECM原液,用10%胃蛋白酶(Sigma-Aldrich,美国)对脱细胞肝组织进一步消化,以dECM:胃蛋白酶(10:1)的重量比加入到0.1M盐酸中,并在常温下搅拌48小时。用扫描电镜(SEM)表征了dECM水凝胶的形态。
2、检测dECM水凝胶的生物相容性及其对肝功能的影响,包括:
将dECM原液用碱性介质稀释至终浓度为5mg mL-1,并用1M NaOH将pH调至7.4。将HLCs或hADMSCs以1×106cells mL-1的密度均匀分散dECM溶液中,加入96孔板,37℃孵育1h诱导凝胶化。形成稳定的dECM水凝胶后,加入细胞培养液。24小时内,用Calcein-AM/PI染色测定细胞活力。第3天收集培养基上清,检测HLCs产生的ALB和尿素,同时制备负载HLCs的dECM水凝胶冷冻切片,并进行肝细胞相关标志物蛋白的免疫荧光染色,包括ALB、HNF4α、ABCC2和E-cadherin。以dECM水凝胶培养的hADMSCs为对照。
3、测定Cu NZs@PLGA纳米纤维增强dECM水凝胶对ROS的清除能力:
本试验评估在氧化应激状态下,Cu NZs@PLGA纳米纤维对dECM水凝胶培养的HLCs的保护作用。以1×106mL-1的细胞密度将HLCs包载于dECM中,加入96孔板中,在37℃下形成凝胶,与Cu NZs@PLGA纳米纤维(等效Cu NZs浓度为150ng mL-1)预孵育过夜。用500μM H2O2刺激细胞24h,Calcein-AM/PI染色检测细胞活力;DCFH-DA染色检测细胞内ROS,酶标仪测定平均荧光强度用于ROS定量。
4、Cu NZs@PLGA纳米纤维增强dECM水凝胶的表征结果如图17-18。
本实施例使用了肝脏dECM水凝胶(图17A)原位递送HLCs,肝脏dECM水凝胶保留了肝脏的生物活性成分,并且,3D培养环境促进了HLCs的成熟。dECM在37℃孵育1h后,形成的dECM水凝胶,具有相对较大的存储模量(G')大于损失模量(G")(图17B,图18A)。SEM图像显示dECM水凝胶呈多孔结构,脱细胞后的无残留细胞(图17C)。然而,弹性模量小于100.0Pa的凝胶可能不便于移植手术。因此,我们使用Cu NZs@PLGA纳米纤维作为dECM水凝胶的机械支撑(图18B)。细胞嵌入后,HLCs均匀分布在Cu NZs@PLGA纳米纤维结合的dECM水凝胶中(图17D)。
5、肝脏dECM水凝胶与生物相容性结果如图17-18。
在dECM水凝胶中培养3天后(3D培养),HLCs显示出良好的细胞活力(图18C),肝细胞相关标记物仍维持高表达,包括ALB、HNF4α、E-cadherin和ABCC2(图17E-H)。在dECM水凝胶中培养的HLCs,其ALB和CK18基因的mRNA表达量比在2D平板中的高8.0倍以上。同时,药物代谢相关基因CYP3A4也被显著上调(图17I)。此外,HLCs培养基上清液中ALB和尿素的分泌也进一步增加(图17J),特别是3D培养的HLCs的尿素含量比2D培养的增加了2.3倍。因此,dECM水凝胶具有良好的生物相容性,dECM水凝胶有利于维持HLCs表型,增强肝细胞相关功能。
6、测定Cu NZs@PLGA纳米纤维对dECM水凝胶中的HLCs的细胞保护作用,结果如图19。
本试验在Cu NZs@PLGA纳米纤维(等效Cu NZs浓度为150ng mL-1)预处理后,与对照组相比,Cu NZs@PLGA纳米纤维明显减少了H2O2引起的HLCs死亡(图19A),这是因为Cu NZs明显缓解了细胞内高ROS水平(图19B)。
实施例7
本实施例提供了采用原位细胞移植药物(HLCs/Cu NZs@fiber/dECM)治疗ALF小鼠试验,具体包括:
1、所有的动物实验都是按照中山大学动物保护与使用委员会批准的方案进行的(SYSU-IACUC-2022-001799)。腹腔注射30(v/v)%CCl4的玉米油溶液(5μL g-1)处理小鼠24h,以建立ALF小鼠模型。雄性C57BL/6小鼠(6~8周龄,18~22g)随机分为6组,其中(1)空白(n=5):健康小鼠;(2)对照组(n=5):假手术的ALF小鼠;(3)fiber/dECM(n=5):ALF小鼠植入PLGA纳米纤维结合dECM水凝胶;(4)Cu NZs@fiber/dECM(n=5):ALF小鼠植入Cu NZs@PLGA纳米纤维结合的dECM水凝胶;(5)hADMSCs/Cu NZs@fiber/dECM(n=5):ALF小鼠植入hADMSCs/Cu NZs@fiber/dECM;(6)HLCs/Cu NZs@fiber/dECM(n=5):ALF小鼠植入HLCs/CuNZs@fiber/dECM。移植1天或3天后,腹腔注射0.6%戊巴比妥钠溶液(阿拉丁,中国),剂量为10μL g-1,采集血液和器官。使用AST检测试剂盒(CH0101202,Maccura,中国)、ALT检测试剂盒(CH0101201,Maccura,中国)和ALB测定试剂盒(CH0101002,Maccura,中国)按照制造商的方案分别检测血清中的谷草转氨酶(AST)、谷丙转氨酶(ALT)和白蛋白(ALB)水平。采集的心脏、肝脏、脾脏、肺和肾脏在4%PFA中固定,石蜡切片后进行苏木精和伊红染色(G1005,Servicebio,China)。采用TUNEL凋亡检测试剂盒(G1501,Servicebio,China)检测肝脏细胞凋亡。Ki67染色(GB11141,Servicebio,China)检测肝脏细胞增殖。此外,在肝脏切片中进行TNF-α(GB11188,Servicebio,China)和IL-6(GB11141,Servicebio,China)免疫荧光染色,以评估炎症程度。应用二氢乙二胺(DHE,GDP1018,Servicebio,China)染色检测冷冻肝脏切片的ROS水平。用Image J进行平均荧光强度定量。对植入HLCs/Cu NZs@fiber/dECM周围的肝组织进行CD31/HNF4α免疫荧光染色(抗CD31抗体,GB11063-1,Servicebio,China),以说明植入物的血管生成和血管化。
2、Cu NZs@PLGA纳米纤维增强dECM水凝胶递送HLCs进行ALF治疗的结果如图20-26。
上述实施例证明了Cu NZs@PLGA纳米纤维具有强大的ROS清除效率和促血管生成作用,dECM水凝胶可增强HLCs的肝功能。因此,本实施例测定了HLCs/Cu NZs@fiber/dECM协同治疗CCl4诱导的ALF的治疗潜力(图20A)。CCl4处理后,肝脏颜色变浅,质地变硬,靠近脉管区有大量细胞坏死(图21)。根据HE染色图像,Cu NZs@PLGA纳米纤维的植入在第1天显著减少了17.9%-29.9%的坏死区域(图20B,E)。同样,TUNEL和Ki67染色也显示,与“对照组”和“fiber/dECM”组相比,“Cu NZs@fiber/dECM”组的ALF小鼠肝脏中凋亡细胞更少,增殖细胞更多。提示Cu NZs在ALF氧化应激初始阶段的细胞保护作用。值得注意的是,与hADMSCs/CuNZs@fiber/dECM相比,植入HLCs/Cu NZs@fiber/dECM的ALF小鼠表现出更少的组织坏死和更明显的细胞增殖,这表明植入的HLCs具有肝功能代偿和对宿主肝细胞的额外保护作用(图20C-D,F-G)。这些结果与体外实验结果一致。用HLCs/Cu NZs@fiber/dECM治疗的小鼠血清AST和ALT水平降低,血清ALB水平恢复,也证明了HLCs优越的治疗效果(图20H-J)。在炎症和氧化应激方面,与未进行治疗的对照组相比,Cu NZs@PLGA纳米纤维的植入在第1天明显减少了炎症因子如TNF-α、IL-6以及细胞内ROS的积累(图21)。此外,未经处理的对照组小鼠肝细胞中TNFA、IL-6和COX2基因的mRNA表达量比正常小鼠高10.0倍以上,经Cu NZs@PLGA纳米纤维处理后,这些基因的mRNA表达量大幅下降(图22G-I)。特别是IL1和INOS基因的mRNA表达恢复到正常范围内(图22J-K),而“Cu NZs@fiber”组抗炎基因IL10水平较未处理的对照组增加了5.0倍以上(图22L)。此外,植入HLCs进一步降低了ALF小鼠肝组织中促炎基因TNFA和IL1的mRNA表达,并上调了抗炎基因IL10的表达。与对照组相比,HLCs/Cu NZs@fiber/dECM处理后,TNFA和IL1基因表达量分别显著降低2.6倍和2.0倍,IL10基因表达量显著上调了9.3倍,表明HLCs/Cu NZs@fiber/dECM对局部恶劣微环境的调节具有优势。
HLCs/Cu NZs@fiber/dECM对CCl4诱导的ALF治疗3天疗效结果如图23,肝脏坏死明显减少,以炎症细胞浸润为主(图23A-B)。Tunel阳性细胞较少,Ki67阳性细胞在各组肝组织中广泛分布(图23C-D)。HLCs/Cu NZs@fiber/dECM组血清AST和ALT水平与正常值相近,明显低于其他治疗组(图23E-F)。与此同时,经HLCs/Cu NZs@fiber/dECM处理的小鼠血清ALB完全恢复(图23G)。HLCs/Cu NZs@fiber/dECM还减缓了ALF小鼠在初始阶段的体重减轻,并从第3天开始加速体重恢复(图23H)。此外,Cu NZs@PLGA纳米纤维对炎症风暴和细胞内过量ROS的缓解在第3天TNF-α、IL-6和DHE染色中得到证实(图24A-C)。与hADMSCs/Cu NZs@fiber/dECM组相比,HLCs/Cu NZs@fiber/dECM组小鼠肝细胞中TNFA、COX2、NLRP3基因mRNA表达明显降低(图24D-F)。因此,HLCs/Cu NZs@fiber/dECM可以持续有效地缓解炎症风暴,抑制氧化应激,为宿主肝细胞建立理想的微环境,从而促进肝脏再生。此外,促血管生成的Cu NZs还促进了宿主CD31阳性内皮细胞向负载HLCs的dECM水凝胶(HNF4α阳性细胞)迁移,并在整个植入物中形成血管,这表明HLCs/Cu NZs@fiber/dECM的治疗效果可能部分归因于植入物的充分血管化(图25)。主要脏器HE染色用于评价HLCs/Cu NZs@fiber/dECM的全身毒性。治疗3天后,小鼠的心、脾、肺、肾均无明显变化,说明HLCs/Cu NZs@fiber/dECM在体内具有良好的生物相容性(图26)。综上所示,这些基于血液和肝脏组织的抗炎、肝功能、增殖和凋亡分析结果证实了HLCs/Cu NZs@fiber/dECM通过结合Cu NZs的抗氧化和促血管生成活性,以及HLCs的抗炎和肝细胞功能,对ALF具有良好的协同治疗作用。
实施例8
本申请实施例提供了上述实施例中治疗1天后的治疗组(HLCs/Cu NZs@fiber/dECM:HLC-1和HLC-2)与对照组(control:Ctrl-1和Ctrl-2)贴附区域肝组织转录组分析,探究HLCs/Cu NZs@fiber/dECM对ALF小鼠的协同治疗作用的治疗机制
1、RNA测序:
分离对照组和HLCs/Cu NZs@fiber/dECM组的肝组织(n=2),在TRIzon试剂中研磨以提取mRNA。利用高质量的RNA样本构建测序文库。使用Illumina NovaseqTM6000(Illumina,USA)按照供应商推荐的方案进行2×150bp对端测序(PE150)。所有数据均采用Lc-Bio Technology(杭州,中国)提供的R-package进行处理。
2、统计分析:
使用统计软件GraphPad Prism 9(GraphPad software,USA)进行所有统计分析。数据以均数±标准差表示,采用Student's t-test或单因素方差分析进行比较。*P<0.05,**P<0.01,***P<0.001,****P<0.0001,或#P<0.05,##P<0.01,###P<0.001,####P<0.0001表示统计有显著性差异。ns表示没有显著性差异。
结果如图27-30所示。火山图显示,HLCs/Cu NZs@fiber/decm处理组肝脏中有833个上调基因和1184个下调基因差异表达(图27A)。此外,KEGG(the Kyoto EncyclopediaofGenes and Genomes)通路分析显示,差异基因富集的前20个通路涉及细胞周期、肝脏生物合成、药物和物质代谢、凋亡和肝脏再生(图27B)。此外,HLCs/Cu NZs@fiber/decm处理组中高表达的药物代谢相关基因及其基因集富集分析(GSEA)表明,细胞色素P450参与的药物代谢在ALF治疗中的重要作用(图27C,图28)。除了参与肝脏再生的重要通路Hippo信号通路外,与凋亡相关的p53信号通路也在治疗中有重要意义。GSEA结果显示,植入HLCs/Cu NZs@fiber/dECM后,p53信号通路中88%的关键基因被差异下调(图27C,图29)。值得注意的是,介导肝再生的关键通路Wnt信号通路中部分基因显著上调(图30),其中作为肝细胞增殖/自我更新标志的AXIN2基因在HLCs/Cu NZs@fiber/dECM组中高表达。这些结果与TUNEL和Ki67染色结果一致(图20C和20D)。此外,转录组分析显示,在植入HLCs/Cu NZs@fiber/dECM的小鼠肝脏中,与炎症或氧化应激相关的基因如IL33和HIF1A显著下调,而抗氧化基因如SOD1、SOD2、GPX1和CAT以及与血管生成相关的VEGFA和ANG基因则高表达(图8D)。更重要的是,在HLCs/Cu NZs@fiber/dECM组小鼠肝组织中,肝脏再生及肝细胞相关基因如ALB、HNF4A、DPP4、ABCC2、TTR、UGT1A1、GSTM1、SULT1A1的表达明显升高(图27D)。
综上所述,ROS过多、严重炎症风暴、大面积肝坏死是ALF的主要病理特征,因此,清除ROS减少肝细胞坏死、补充肝细胞实现肝功能代偿,从而促进肝脏再生是ALF治疗的关键。因此,本申请实施例开发了一种由负载抗氧化金属纳米酶的纳米纤维和负载功能性细胞的凝胶(HLCs/Cu NZs@fiber/dECM)组成的协同治疗平台,用于治疗CCl4诱导的ALF,并证明了HLCs/Cu NZs@fiber/dECM良好的治疗效果。
本实施例成功合成了具有良好生物相容性和高ROS清除效率的Cu NZs并进行了表征。即使负载于PLGA纳米纤维,Cu NZs的抗氧化效果也不受影响。Cu NZs@PLGA纳米纤维在等效Cu NZs浓度低至150ng mg-1时即可有效清除ROS,提高了肝细胞在体外和体内的存活率。此外,Cu NZs@PLGA纳米纤维还能显著促进HUVECs的迁移和增殖,有利于植入物在体内的快速血管化。
由于大多数细胞在脾内移植后可被巨噬细胞或吞噬细胞清除,而肝脏的炎症环境也会损害移植细胞的活力,因此本实施例将HLCs负载在载药凝胶(如dECM水凝胶),并与CuNZs@PLGA纳米纤维组合原位递送HLCs。结果表明,dECM作为HLCs的载体,其多孔结构和天然成分提供了良好的3D培养环境,维持/增强了HLCs的肝功能,有利于HLCs在损伤肝脏中实现肝功能代偿。而且,本实施例证明了Cu NZs@PLGA纳米纤维在dECM水凝胶中对H2O2具有清除作用,并对包载的HLCs具有细胞保护作用。此外,dECM水凝胶的机械强度较低,原位移植难以保持其结构不被破坏,Cu NZs@PLGA纳米纤维增强的dECM水凝胶提高了体内移植的可操作性。Cu NZs@PLGA纳米纤维还可以减少植入物粘附在邻近器官或组织上,促进HLCs与宿主肝脏的整合。
此外,本实施例在体内实验中评估了HLCs/Cu NZs@fiber/dECM的协同治疗效果。一方面,抗氧化纳米酶Cu NZs能高效清除过量的ROS,防止肝组织坏死恶化。如图6和图23所示,细胞凋亡减少导致炎症因子的积累减少,从而避免肝脏发生严重的炎症风暴(图22、24),改善的微环境也有利于HLCs的存活。如图25所示,Cu NZs还能促进植入物的血管生成和血管化。另一方面,HLCs具有肝细胞相关的功能,并在dECM水凝胶中进一步维持或增强。图20和图23显示,HLCs显著提高ALF小鼠ALB水平。此外,HLCs具有固有的抗炎活性,从而协同保护宿主肝细胞免受氧化应激,加速肝脏再生。正如预期的那样,HLCs/Cu NZs@fiber/dECM对CCl4诱导的ALF表现出优异的治疗效果。综上所述,HLCs/Cu NZs@fiber/dECM的协同作用有助于ALF小鼠的有效肝脏再生,并促进移植细胞在体内的长期肝功能整合。
本实施例进一步进行转录组分析,以探究潜在的治疗机制。从差异基因表达热图来看,如图27D所示,氧化应激和炎症相关的基因差异下调,这与HLCs/Cu NZs@fiber/dECM的ROS消除和抗炎能力相对应。高表达的血管生成相关基因表明Cu NZs的促血管生成作用。通过KEGG和GSEA分析,进一步探讨HLCs/Cu NZs@fiber/dECM影响细胞凋亡的途径。研究表明,急性肝损伤中大量的肝细胞衰老可能会打破肝细胞再生与衰竭之间的平衡。我们的结果显示,经过HLCs/Cu NZs@fiber/dECM处理后,p53信号通路中富集的基因差异下调,AXIN2显著上调。肝细胞中AXIN2的上调被认为是肝细胞自我更新、细胞周期再进入和再生的表面标志物。这些结果表明,HLCs/Cu NZs@fiber/dECM可能通过影响Wnt信号通路来阻止肝细胞衰老,促进肝再生。此外,在肝脏生物合成、药物物质代谢等途径中也存在丰富的差异基因。值得注意的是,细胞色素p450药物代谢通路的相关基因在HLCs/Cu NZs@fiber/dECM组肝脏中差异上调。此外,肝细胞相关基因也高表达,提示HLCs肝功能代偿的潜在机制可能与物质生物合成和药物代谢有关。总的来说,转录组分析也证实了HLCs/Cu NZs@fiber/dECM在ALF治疗中抗氧化、促进血管生成、抗炎症和肝细胞相关功能的协同作用。
Claims (10)
1.一种仿生纳米酶纤维的制备方法,其特征在于,包括:
将抗氧化金属纳米酶和生物可降解高分子聚合物,在溶剂条件下进行复合和纤维化,制得仿生纳米酶纤维;其中,所述抗氧化金属纳米酶负载在所述仿生纳米酶纤维上。
2.根据权利要求1所述的制备方法,其特征在于,所述抗氧化金属纳米酶选自铜基纳米酶、金纳米酶、银纳米酶和铂纳米酶中的一种或多种。
3.根据权利要求1所述的制备方法,其特征在于,所述生物可降解高分子聚合物选自聚乳酸聚乙醇酸共聚物PLGA、聚己内酯PCL、聚乳酸PLA和明胶中的一种或多种;
所述溶剂选自六氟异丙醇HFIP、二氯甲烷和四氢呋喃中的一种或多种。
4.根据权利要求1所述的制备方法,其特征在于,所述纤维化选自静电纺丝、3D打印或旋涂。
5.一种仿生纳米酶纤维,其特征在于,包括如权利要求1~4任意一项所述的制备方法制得的仿生纳米酶纤维。
6.权利要求1~4任意一项所述的制备方法制得的仿生纳米酶纤维或权利要求5所述的仿生纳米酶纤维在制备原位细胞移植药物中的应用。
7.一种原位细胞移植药物,其特征在于,包括:
仿生纳米酶纤维和负载有功能性细胞的载药凝胶;
其中,所述仿生纳米酶纤维为权利要求1~4任意一项所述的制备方法制得的仿生纳米酶纤维。
8.根据权利要求7所述的原位细胞移植药物,其特征在于,所述功能性细胞选自功能性肝细胞样细胞、治疗性干细胞、原代肝细胞和肝细胞系AML-12中的一种或多种;所述功能性细胞还包括内皮细胞;
所述载药凝胶选自脱细胞基质dECM水凝胶、PLGA-PEG-PLGA、天然高分子水凝胶和胶原蛋白水凝胶中的一种或多种。
9.权利要求7或8所述的原位细胞移植药物在制备治疗实体器官疾病药物中的应用。
10.根据权利要求9所述的应用,其特征在于,所述实体器官疾病包括肝脏疾病、皮肤创伤或心肌损伤修复。
