Abstract
本发明属于化妆品技术领域,具体涉及一种敏感肌适用的美白组合物及其制备方法和应用。本发明通过将特定用量配比的麦角硫因、十一碳烯酰基苯丙氨酸、天女木兰提取物组合物、二裂酵母发酵产物滤液组合物、二甲基甲氧基苯并二氢吡喃醇组合,使得本发明化妆品组合物具有协同增效的全效美白、抗衰和修护等功效,将其应用到化妆品中,能够促进皮肤细胞新陈代谢,抑制并消除色素沉着,抵御紫外线、高能可见光和红外线,清除自由基,能够有效缓解并全面改善肤色粗糙、暗黄以及晒斑等多种皮肤问题,使皮肤皱纹减少以及更加紧致,并且具有出色的使用感受。另外,本发明美白组合物无刺激性,特别适用于敏感肌。

技术领域
本发明属于化妆品技术领域,具体涉及一种敏感肌适用的美白组合物及其制备方法和应用。
背景技术
随着生活水平的不断提高,人们对美容护肤的要求也越来越高,日常生活中的环境污染、紫外线的损伤、化学品的刺激、面部清洁不彻底导致的油污、黑色素量的增多、油性痤疮肌肤、工作压力、生活压力、辐射衰老等问题都会造成肌肤成为敏感肌;但是爱美之心人皆有之,敏感肌肤的人群也有保持皮肤美白的需求。
目前传统的美白方法常常通过调控黑色素,即抑制黑色素生成、运输及排泄,促进皮肤的代谢循环,从而达到美白功效,但在此过程中常常会消耗皮肤大量的水分,容易造成皮肤干燥,同时如果大量美白剂残留在皮肤表层,容易引起皮肤刺激、过敏等副作用,造成角质层蛋白流失,继而导致皮肤免疫屏障功能受损;而且目前的美白产品大多都是通过较单一的路径抑制黑色素的形成来达到美白的效果,美白效果有待进一步提升,同时产品的稳定性差,或者刺激性大,长时间使用易造成皮肤敏感。因此,目前市场上销售的大部分功效确切的美白产品实质上并不适于敏感肌肤人群使用。
特别针对敏感皮肤人群研制的,改善皮肤暗沉和泛黄的护肤产品非常少,且美白产品的效果、刺激性与配方成分的配伍性息息相关,因此温和或低刺激性的美白护肤品,一直是人们不断的追求目标,所以非常有必要开发一款敏感肌适用的美白产品。
CN111407680A公开了一种美白护肤品及其制备方法,含有烟酰胺、乙酰壳糖胺、3-o-乙基抗坏血酸、羟癸基泛醌、苯乙基间苯二酚和白藜芦醇这几种主要的美白成分,可从抑制黑色素产生、抑制黑色素转移、还原已形成的黑色素多重路径联合发挥美白作用。CN113768818A公开了一种美白抗衰组合物、化妆品及其制备方法,由以下组分组成:熊果苷0.05-3份、抗坏血酸葡糖苷0.05-3份、谷胱甘肽0.001-0.1份、烟酰胺0.5-5份、凝血酸0.1-2份、二肽二氨基丁酰苄基酰胺二乙酸盐0.0005-0.05份、乙酰基四肽-2 0.00005-0.005份和寡肽-4 0.0001-0.005份,选用熊果苷、抗坏血酸葡糖苷、谷胱甘肽、烟酰胺和凝血酸作为美白组分;选用二肽二氨基丁酰苄基酰胺二乙酸盐、乙酰基四肽-2和寡肽-4作为抗衰组分,美白组分与抗衰组分之间具有协同增效作用,共同作用能显著提高组合物的美白抗衰功效。但是,这些美白组合物在配方过程中主要考虑的是美白功效而没有针对敏感肌肤进行特别设计,导致这些美白组合物存在作用机理比较单一,美白效果也不够好的问题,同时这些美白组合物产品的稳定性,以及长期使用后的皮肤敏感性也不得而知。
本发明人针对现有技术中存在的上述缺陷,开发了一种包含麦角硫因、十一碳烯酰基苯丙氨酸、天女木兰提取物、二裂酵母发酵产物滤液、二甲基甲氧基苯并二氢吡喃醇等多种活性成分的敏感肌适用的美白组合物。本发明美白组合物更适用于敏感肌,且全效美白、抗衰、修护功效更强。可以将本发明美白组合物方便地用于各种化妆品中,以赋予目标化妆品上述功效。
发明内容
本发明的目的是克服现有技术的不足,提供一种敏感肌适用的美白组合物及其制备方法和应用。本发明通过将特定用量配比的麦角硫因、十一碳烯酰基苯丙氨酸、天女木兰提取物组合物、二裂酵母发酵产物滤液组合物、二甲基甲氧基苯并二氢吡喃醇组合,使得本发明化妆品组合物具有协同增效的全效美白、抗衰和修护等功效。另外,本发明美白组合物无刺激性,特别适用于敏感肌。
具体地,通过以下几个方面的技术方案实现了本发明:
在第一个方面中,本发明提供了一种敏感肌适用的美白组合物,所述美白组合物包含按照重量百分比计的下述原料:麦角硫因0.001%-30%、十一碳烯酰基苯丙氨酸0.01%-30%、天女木兰提取物组合物0.1%-10%、二裂酵母发酵产物滤液组合物10%-95%和二甲基甲氧基苯并二氢吡喃醇0.001%-30%。
作为可选的方式,在上述美白组合物中,所述美白组合物包含按照重量百分比计的下述原料:麦角硫因0.01%-20%、十一碳烯酰基苯丙氨酸0.05%-20%、天女木兰提取物组合物0.1%-5%、二裂酵母发酵产物滤液组合物10%-50%和二甲基甲氧基苯并二氢吡喃醇0.01%-10%。
作为可选的方式,在上述美白组合物中,所述美白组合物包含按照重量百分比计的下述原料:麦角硫因0.01%-2%、十一碳烯酰基苯丙氨酸0.05%-5%、天女木兰提取物组合物0.1%-5%、二裂酵母发酵产物滤液组合物10%-30%和二甲基甲氧基苯并二氢吡喃醇0.01%-5%。
作为可选的方式,在上述美白组合物中,所述美白组合物包含按照重量百分比计的下述原料:麦角硫因0.02%、十一碳烯酰基苯丙氨酸1%、天女木兰提取物组合物3%、二裂酵母发酵产物滤液组合物20%和二甲基甲氧基苯并二氢吡喃醇0.05%。
作为可选的方式,在上述美白组合物中,所述美白组合物的活性成分由麦角硫因、十一碳烯酰基苯丙氨酸、天女木兰提取物组合物、二裂酵母发酵产物滤液组合物和二甲基甲氧基苯并二氢吡喃醇组成。
作为可选的方式,在上述美白组合物中,所述天女木兰提取物组合物由按照重量百分比计的下述原料制成:天女木兰(MAGNOLIA SIEBOLDII)提取物0.1%、丁二醇60.0%和水39.9%。
作为可选的方式,在上述美白组合物中,所述二裂酵母发酵产物滤液组合物由按照重量百分比计的下述原料制成:二裂酵母发酵产物滤液99.1%、1,2-己二醇0.5%和对羟基苯乙酮0.4%。
在第二个方面中,本发明提供了上述第一个方面所述的美白组合物的制备方法,所述制备方法包括以下步骤:
按重量百分比称量各组分原料,将各组分依次加入反应釜混合搅拌均匀,即获得一种敏感肌适用的美白组合物。
在第三个方面中,本发明提供了上述第一个方面所述的美白组合物或者采用上述第二个方面所述制备方法制备得到的美白组合物在制备敏感肌适用的美白化妆品中的应用。
在第四个方面中,本发明提供了上述第一个方面所述的美白组合物或者采用上述第二个方面所述制备方法制备得到的美白组合物在制备预防或改善无规律色素过度沉着的敏感肌适用的美白化妆品中的应用。
作为可选的方式,在上述应用中,所述无规律色素过度沉着是老年斑、雀斑、黑斑、黄褐斑、晒斑或蝴蝶斑中的一种或多种。
作为可选的方式,在上述应用中,所述美白化妆品为爽肤水、乳液、精华、霜、膏、面膜或冻干粉。
与现有技术相比,本发明的有益效果是:

具体实施方式
为了便于本领域技术人员的理解,下面对本发明中涉及的各种主要活性成分进行描述。
如本文所用,在本发明美白组合物中使用的“麦角硫因”是一种人体必需的稀有氨基酸,具有超强、广谱、稳定、持久的靶向抗氧化作用,能清除体内尤其是线粒体内部的过量自由基,同时具有防止紫外线辐射损伤、美白和抗衰老等多种功能。麦角硫因是一种极其稳定的水溶性抗氧化剂,可添加在各类配方中,显著减缓皮肤老化现象、淡化色斑、亮白肌肤、增加皮肤弹性。
如本文所用,在本发明美白组合物中使用的“十一碳烯酰基苯丙氨酸”是促黑色素细胞激素α-MSH在黑色素合成过程中的对抗剂,可作用于由α-MSH引发的黑色素生成反应的各个阶段,并且可从上游拦截α-MSH信号,从而从多个环节更快、更全面、更有效地抑制黑色素的生成,效果更加明显、持久。体内实验中可7天快速提亮肤色,14天淡化色斑,对于各种肤色人群均具有美白提亮或淡化色斑的效果,并可增效抗坏血酸葡糖苷的美白效果,安全不刺激。可用于各种使皮肤皙透的配方中。
如本文所用,在本发明美白组合物中使用的“天女木兰提取物”通过清除自由基,抑制多巴的自氧化,抑制黑素小体在黑素细胞树突中的运输及转移至角质形成细胞,多靶点抑制黑色素形成和转移,从而达到有效可见的美白功效。人体实验表明能够抑制炎症或UV照射后引起的色素沉着,并明显改善黄褐斑。
如本文所用,在本发明美白组合物中使用的“二裂酵母发酵产物滤液”通过促进外皮蛋白和神经酰胺合酶-3的表达增强皮肤屏障,通过抑制黑色素的形成美白亮肤,同时还具有控油,抗炎舒缓等益肤功效。
如本文所用,在本发明美白组合物中使用的“二甲基甲氧基苯并二氢吡喃醇”是强效稳定的抗氧化剂,对氧自由基(ROS)、氮自由基(RNS)和碳酰自由基(RCS)具有三重捕捉功能;低浓度时即可抑制脂质过氧化反应;防止过氧亚硝基阴离子和一氧化氮对皮肤细胞的损伤,通过抑制UVA产生的氧自由基,达到内在光保护作用,同时具有抗污染和皮肤防护&解毒功效。适用于抗自由基、抗污染、抗氧化、抗衰老和防护产品。
本发明中使用的各种化妆品原料均为市售产品。
化妆品领域的技术人员能够根据实际需要在不同的化妆品(例如,但不限于爽肤水、乳液、精华、霜、膏或面膜)中添加本发明化妆品组合物。

实验方法参见本申请人的在先申请CN114712258A。简言之,通过构建酪氨酸酶催化反应体系,来评价实施例1-3和对比例1-5制备得到的面霜对酪氨酸酶的抑制效果。在pH=6.8的磷酸缓冲液体系中,酪氨酸酶可催化L-酪氨酸产生黑色素,在475nm处具有最大吸收峰。通过在反应体系中添加实施例1-3和对比例1-5制备得到的面霜作为酪氨酸酶抑制剂,可以减少黑色素的生成,通过利用黑色素生成量的减少程度来评价实施例1-3和对比例1-5制备得到的面霜美白效果。
DPPH是一种以氮为中心的稳定自由基,测定DPPH清除率是目前评价天然活性物质抗氧化活性中的简便、快速,灵敏的方法。通过构建DPPH自由基清除反应体系,评估含实施例1-3和对比例1-5制备得到的面霜的抗氧化活性。
实验结果如表1所示。

由表1的实验结果可以看出,本发明实施例1-3制备得到的面霜在美白和清除自由基方面的作用显著优于对比例1-4制备得到的面霜。如对比例6所示,单独的用于制备面霜的常规化妆品基础成分基本上没有美白和清除自由基方面的作用。
对于对比例1-4而言,如上表1所示,对比例1(用玻色因替换麦角硫因)、对比例2(用山茶花提取物替换天女木兰提取物)对酪氨酸酶的抑制率和DPPH自由基清除率是最差的。提示尽管麦角硫因和天女木兰提取物在本发明美白组合物中不是含量最高的活性成分,但是麦角硫因和天女木兰提取物均是本发明美白组合物中发挥美白和清除自由基效果的关键且不可替代的成分,因为将其换成化妆品领域中经常与麦角硫因或天女木兰提取物组合或互换使用的常用化妆品成分(如玻色因或山茶花提取物)后组合物的美白和清除自由基的效果显著下降。对比例3(不含十一碳烯酰基苯丙氨酸)和对比例4(不含二甲基甲氧基苯并二氢吡喃醇)对酪氨酸酶的抑制率和DPPH自由基清除率也显著不及实施例1-3,提示上述两种成分也是本发明美白组合物必不可少的重要活性成分。由此可见,本发明美白组合物的各种活性成分之间产生了显著的协同增效作用,任一替换或删除一种或多种活性成分之后,其美白和清除自由基的作用就会显著降低。
效果实施例2:本发明美白组合物的斑马鱼美白功效试验
委托广州环特智鱼优检生物科技有限公司检测,项目编号:5611-1。以下内容为报告节选,如有需要可提供完整检测报告。
1.实验室试验简述
1.1方法来源:《斑马鱼美白功效评价实验标准操作规程》
1.2试验起止日期:2022年12月06日-2022年12月08日1.3体系及样本量
用鱼品系:野生型AB品系斑马鱼。
斑马鱼鱼龄:受精后6小时(6hpf)。
每组实验样本量:15尾(N=10)。
成鱼饲养及繁殖方法:按照本公司实验室标准饲养和繁殖方法,符合国际AAALAC认证(认证编号:001458)的要求。
1.4实验步骤
(1)随机选取斑马鱼于6孔板中,每孔15尾。
(2)水溶给予样品(制备实施例1),同时设置正常对照组,每孔容量为3mL。
(3)28℃条件下避光孵育45h。
(4)每个实验组随机选取10尾斑马鱼置于解剖显微镜下拍照,用高级图像处理软件分析并采集数据,分析斑马鱼头部黑色素信号强度(S),根据公式计算样品的美白功效,判断其是否具有美白功效。
1.5适用性及局限性
适用于化妆品及其原料的美白功效测试,要求样品能溶于水或制备成能在水中均匀分散的悬浮液。
1.6判定依据
统计学分析p<0.05,功效值≥20%,判定为有效。
2.检测结果
检测结果如表2和图1所示。实验结果表明,测试样品舒敏美白霜(制备实施例1)的头部黑色素信号强度与正常对照组相比,明显降低,揭示了该样品具有美白功效。

效果实施例3:本发明美白组合物的美白功效人体试验
委托广州金评检测研究院有限公司检测,报告编号:JPTI-GX23004。以下内容为报告节选,如有需要可提供完整检测报告。
1.材料和方法
1.1受试物
受试物:制备实施例1制备得到的面霜
阴性对照:空白对照
阳性对照:7%抗坏血酸制品
1.2受试者
共30人,男性4人,女性26人,年龄18至59岁,平均年龄39.07±12.11岁,符合受试志愿者入选标准。
1.3仪器设备
日光模拟仪、皮肤色度仪、皮肤黑素检测仪、皮肤肤色色卡,各项性能指标符合测定规范要求。
1.4环境要求
试验过程中视觉评估和仪器测试环节在温度为21±1℃,相对湿度为50±10%RH的环境下进行,且视觉评估在恒定光照(色温5500-6500K的日光灯管或LED光照)条件下进行。受试者需在温度为21±1℃,相对湿度为50±10%RH的环境下至少30分钟后方可进行评估和测试。
1.5试验方法
按照《化妆品安全技术规范》化妆品祛斑美白功效测试方法的具体要求进行检验。选取受试者背部作为试验部位,每个黑化测试区域面积不小于0.5cm2。检测前24小时预测受试者皮肤对紫外线照射的最小红斑量(MED),根据测试结果调整紫外线照射剂量。用日光模拟仪在相同照射点按0.75倍MED剂量每天照射1次,连续照射4天,照射结束后的4天为皮肤黑化期,不做任何处理。在照射结束后的第5天,对各测试区域进行视觉评估和仪器检测,剔除ITA°值与全部测试区域均值相差大于5的区域。当天开始在各测试区域按照产品随机表涂抹相应受试物,各区域之间至少间隔1cm,涂抹面积3cm*3cm,按涂样量2.00±0.05mg/cm2连续涂抹产品4周。分别在涂抹产品前(0周),涂抹后1周、2周、3周、4周对受试部位进行视觉评估和仪器检测,并记录。
1.6统计方法
应用SPSS软件进行数据的统计分析。计量资料表示为均值±标准差,并进行正态分布检验,符合正态分布要求,自身前后的比较采用配对T检验,否则采用两个相关样本秩和检验;等级资料使用前后的比较,采用两个相关样本秩和检验;测试区和对照区之间比较采用独立样本t检验或秩和检验;同时,计算各参数随时间变化的回归系数(斜率k值),显著性水平均为<0.05。
1.7结果判定
试验产品涂抹前后任一时间点肤色视觉评分差值或ITA°值与阴性对照相比有显著性改善(P<0.05),或经回归系数分析整体判断试验产品与阴性对照相比皮肤黑化显著改善时(P<0.05),则认定产品具有祛斑美白功效,否则认为试验试验产品无祛斑美白功效。
2.检验结果
计量资料检验结果和等级资料检验结果见原始报告,如有需要可提供完整检测报告。
3.试验结论
使用试验样品后,试验产品组的视觉肤色等级改善程度在第3周时显著性优于阴性对照组(P<0.05)。综上所述,在本试验条件下,试验样品具有祛斑美白的功效。
效果实施例4:本发明美白组合物敏感肌适用温和无刺激1.乳酸刺痛评价
1.1测试原理
研究认为,皮肤敏感的发生是一种涉及皮肤屏障-皮肤神经系统-皮肤免疫细胞的复杂过程。在内在和外在因素的共同作用下,皮肤屏障功能受损,引起感觉神经传入信号增加,导致皮肤对外界刺激的反应增强,引发皮肤免疫炎症反应。使用产品前后乳酸刺痛试验的评分,可直接反映产品对皮肤屏障功能的修护作用。
本发明通过对使用产品前后乳酸刺痛评价来测试组合物(实施例1-3和对比例1-5)的皮肤屏障修护功效。
1.2测试方法
测试环境:测试环境温度为22±1℃,湿度为50±5%,并且进行实时动态检测;
测试志愿者:年龄在16-60岁之间(妊娠或哺乳期妇女除外),共40人,每组5人;无严重系统疾病,无免疫缺陷或自身免疫性疾病者;既往对护肤类化妆品无严重过敏史者;近一个月内未曾使用激素类药物及免疫抑制剂者;未参加其他临床试验者;按规定使用受试药物且资料齐全;测试前所有自愿者应填写知情同意书。
测试步骤:
测试前准备:受试部位前2-3天不能使用任何产品(化妆品或外用药品)。
测试过程:受试者在恒温恒湿室内静坐30分钟,然后在受试者左、右侧鼻唇沟分别使用等量的10%乳酸水溶液和蒸馏水,并在0.5、2、5min的时候询问受试者的自我肤感症状,按四分法进行评分:没有刺痛感0分,轻度刺痛感为1分,中度刺痛感为2分,重度刺痛感为3分。同一个志愿者的测试由同一个测量人员完成。
1.3测试结果
乳酸刺痛改善率计算公式:R=(R1-R2)/R1×100%
R:乳酸刺痛改善率;
R1:使用样品前乳酸刺痛评分;
R2:使用样品28天后乳酸刺痛评分;
乳酸刺痛评分=0.5min+2min+5min刺痛评分;
实验结果如表3所示。

表3中的实验结果体现出显著的差异性,说明本发明美白组合物除了美白功效以外还能够提供修护皮肤屏障的功效。如对比例6所示,单独的用于制备面霜的常规化妆品基础成分基本上没有修护皮肤屏障方面的作用。
通过比较对比例1-4与实施例1-3的改善率,可以看出本发明美白组合物在修护皮肤屏障方面也具有协同增效的作用,当有部分组分替换或缺失时,组合物的功效作用明显下降。
乳酸刺痛试验结果说明本发明敏感肌适用的美白组合物具有修护皮肤屏障的功效,且通过本发明美白组合物的使用,可以明显的降低皮肤的敏感性,缓解外界刺激。
2.对本发明组合物(制备实施例1)的安全性评价
2.1皮肤科医生评估
使用测试样品期间皮肤科医生未发现与产品(制备实施例1)有关的不良反应。如下表4所示。

2.2受试者自评
使用测试样品28天后,制备实施例1组受试者未发现产品有刺激现象,100.00%认为产品温和无刺激。
2.3产品满意度评估
使用测试样品28天后,制备实施例1组受试者对测试产品的整体满意度为100.00%。
2.4测试结论
在皮肤科医生的控制下,通过观察35名年龄在25-55岁的敏感肌成年受试者,在连续28天使用本发明组合物,评价其引起人体皮肤不良反应潜在的可能性。皮肤科医生评估结果为35名受试者在连续使用测试样品28天后临床均未出现不良反应。说明本发明组合物产品适用于敏感肌。
在连续28天使用本发明组合物期间,35名敏感肌受试者未发现产品有刺激现象,100.00%认为产品温和无刺激。
综上所述,以上测试结果提示本发明组合物产品适用于敏感肌且温和无刺激。
显然,本领域的技术人员可以对本发明进行各种改动和变型而不脱离本发明的精神和范围。这样,倘若本发明的这些修改和变型属于本发明权利要求及其等同技术的范围之内,则本发明也意图包含这些改动和变型在内。
Claims (2)
1.一种敏感肌适用的美白面霜,其特征在于:所述美白面霜由按照重量百分比计的下述原料制成:麦角硫因0.02%、十一碳烯酰基苯丙氨酸1%、天女木兰提取物组合物3%、二裂酵母发酵产物滤液组合物20%、二甲基甲氧基苯并二氢吡喃醇0.05%和化妆品基础成分75.93%,所述天女木兰提取物组合物由按照重量百分比计的下述原料制成:天女木兰(MAGNOLIA SIEBOLDII)提取物0.1%、丁二醇60.0%和水39.9%,所述二裂酵母发酵产物滤液组合物由按照重量百分比计的下述原料制成:二裂酵母发酵产物滤液99.1%、1,2-己二醇0.5%和对羟基苯乙酮0.4%。
2.根据权利要求1所述的美白面霜的制备方法,其特征在于:所述制备方法包括以下步骤:
按重量百分比称量各组分原料,将各组分依次加入反应釜混合搅拌均匀,即获得一种敏感肌适用的美白面霜。
