技术领域
本发明属于疾病动物模型构建方法技术领域,具体涉及一种系统性红斑狼疮的动物模型的构建。
背景技术
系统性红斑狼疮(systemic lupus erythematosus,SLE)是一种复杂的自身免疫性疾病,临床表现多样,以I型干扰素(Interferon,IFN)和自身抗体的过度产生为特征。SLE的发病机制尚不明确,遗传易感性、环境诱因和激素水平三者相互作用,共同促进了疾病的发生发展。近年来世界范围内SLE的发病率呈上升趋势,全球患病率为43.7/10万人,严重危害人类健康甚至威胁生命。开发能模拟临床症状、又兼具成本优势的动物模型,对研究SLE发病机制及安全有效的新型疗法具有重要意义。
目前SLE模型主要分为自发型和诱导型模型两种。自发型SLE小鼠模型具有良好的遗传背景及遗传稳定性,包括新西兰黑(NZB)和新西兰白(NZW)小鼠杂交的F1代NZBWF1小鼠,BXSB/MpJ小鼠,和MRL/lpr小鼠;人工诱导型小鼠包括淋巴细胞活性染色质诱导系统性红斑狼疮小鼠模型,空肠弯曲菌诱导的SLE小鼠模型,以及降植烷诱导的SLE小鼠模型等。虽然自发型小鼠模型是SLE的经典模型,但此类小鼠价格偏高,发病晚,易受环境因素影响,小鼠异质性大,且小鼠基因表型和发病机制与SLE患者有较大差别。人工诱导型小鼠模型价格相对较低,周期短,但其不具备遗传背景,与患者发病过程和临床表现差异较大,建模对研究者的实验操作要求较高。现有SLE动物模型大多缺少典型的SLE皮损改变,而与SLE患者不符的基因改变及致病过程限制了它们在发病机制、病理改变、疾病治疗等方面的应用,迫切需要基于患者遗传背景的自发型SLE小鼠模型,来推动SLE领域的研究,加深对疾病的认识。
角质形成细胞、免疫细胞及免疫分子共同构成皮肤局部免疫微环境,维持组织稳态。SLE患者常见皮肤受累,并且患者角质形成细胞中的炎症反应被放大。尽管尚无研究证明皮肤功能异常与SLE发病存在直接联系,但皮肤型红斑狼疮可能发展成更广泛或全身性的病变,且有一定几率进展为SLE;以及长期的I型IFN刺激会导致免疫细胞过度活化并诱导自身抗体的产生,这些临床现象和既往研究提示皮肤局部免疫功能异常可能促进系统性自身免疫反应并驱动SLE的发生发展。
PPARγ(过氧化物酶体增殖物激活受体γ)是PPAR通路的关键分子,能调控脂肪细胞分化和功能,调控免疫细胞的成熟和功能,影响组织和器官的细胞增殖,其信号失调还与肿瘤生成有关。目前尚未见到利用角质形成细胞条件性PPARγ敲除小鼠构建自发型SLE动物模型的报道。
发明内容
发明目的:本发明所要解决的技术问题是针对现有技术的不足,为SLE的发病机制、病理特征、治疗方法研究和开发提供一种新的自发型SLE动物模型。

(1)构建角质形成细胞条件性PPARγ敲除小鼠,所述角质形成细胞条件性PPARγ敲除小鼠包括PPARγflox/flox+/-Krt5-CreERT2+/-和PPARγflox/flox-/-Krt5-CreERT2+/-的C57BL/6小鼠;
(2)使用基因敲除激活剂敲除角质形成细胞中的PPARγ基因以介导疾病表型发生。
其中,所述基因敲除激活剂为代谢后可与雌激素受体突变体ERT结合,使CreERT2发挥Cre重组酶活性进行基因敲除的化合物。
在一种实施方式中,所述基因敲除激活剂为他莫昔芬或4-羟基他莫昔芬。
其中,他莫昔芬的使用方式为有针或无针化的腹腔注射;4-羟基他莫昔芬的使用方式为外用涂抹、皮肤贴敷、有针或无针化的皮下注射或皮内注射中的任意一种。
具体地,使用他莫昔芬作为基因敲除激活剂时,对角质形成细胞条件性PPARγ敲除小鼠连续腹腔注射他莫昔芬溶液1-7天,每天一次;使用4-羟基他莫昔芬作为基因敲除激活剂时,对角质形成细胞条件性PPARγ敲除小鼠身体部位无毛或剃毛后的皮肤连续涂抹4-羟基他莫昔芬溶液1-7天,每天一次,为了便于操作,可以选择双耳的腹侧及背侧的皮肤进行涂抹。
其中,所述他莫昔芬溶液按照100mg他莫昔芬、0.5ml乙醇、9.5ml玉米油的比例配置;所述4-羟基他莫昔芬溶液按照50mg 4-羟基他莫昔芬、1mlDMSO、9ml玉米油的比例配置;每次给小鼠注射的他莫昔芬溶液的剂量为75mg/kg(约100μl),每次给小鼠外用涂抹的4-羟基他莫昔芬溶液的剂量为10-80μL。
其中,小鼠在初次腹腔注射他莫昔芬7天后或外用涂抹4-羟基他莫昔芬7天后出现疾病表型。
具体地,所述角质形成细胞条件性PPARγ敲除小鼠通过如下方法构建得到:
将PPARγflox/flox和Krt5-CreERT2小鼠交配,得到杂交生成子一代,保留基因型为PPARγflox/flox+/-Krt5-CreERT2+/-的杂合子小鼠,将杂合子小鼠相互交配繁育得到基因型为PPARγflox/flox-/-Krt5-CreERT2+/-的小鼠。
本发明进一步提出了通过上述构建方法构建得到的自发型系统性红斑狼疮动物模型在研究SLE疾病机理上的应用。
本发明还提出了通过上述构建方法构建得到的自发型系统性红斑狼疮动物模型在筛选靶向治疗狼疮皮损的药物中的应用。
有益效果:本发明通过敲除小鼠角质形成细胞中的PPARγ,构建了一种自发型SLE动物模型,其不需要人工诱导,能条件性出现系统性自身免疫性炎症,包括局部皮肤脱毛、持续性皮损、持续性蛋白尿、血清自身抗体;组织学检查有真皮免疫细胞浸润,基底膜带IgG沉积,肾小球IgG沉积等系统性红斑狼疮样改变,模拟了系统性红斑狼疮的临床症状,为系统性红斑狼疮的相关研究提供了重要工具,为SLE的发病机制研究及药物开发提供了重要工具。该模型相较于其他狼疮鼠模型具有遗传背景而非药物诱导,发病迅速、高效稳定。在诱导角质形成细胞PPARγ敲除后1-2周,构建模型的所有小鼠均会出现一定程度的表型,建模成功率为100%,组内异质性小,可重复性强。此外,Krt5-CreERT2+/-PPARγflox/flox+/-及Krt5-CreERT2+/-PPARγflox/flox-/-小鼠可通过配种繁殖持续获得,经济方便。

下面结合实施例对本发明作进一步说明,但不应该理解为本发明上述主体范围仅限于下述实施例。在不脱离本发明上述技术思想的情况下,根据本领域普遍技术知识和惯用方法,做出各种替换和变更,均应包括在本发明的保护范围。
实施例1他莫昔芬腹腔注射激活的角质形成细胞PPARγ敲除诱导SLE模型。
1.实验材料
1.实验用药物及试剂:DMSO(Sigma-Aldrich),他莫昔芬(Sigma-Aldrich),玉米油(Beyotime),蛋白尿检测试纸(优利特),尿蛋白定量试剂盒(Nanjing JianchengBioengineering Institute),抗核抗体及抗双链DNA抗体ELISA试剂盒(Cusaibio),4%多聚甲醛(Biosharp),OCT包埋剂(Sakura),山羊抗鼠IgG-FITC
(abcam)。
1.2实验用动物
本实验采用的PPARγflox/flox及Krt5-CreERT2小鼠,周龄约6-12周,体重20-30g,由上海南方模式生物科技股份有限公司提供。Krt5-CreERT2小鼠通过krt5基因座的内源性启动子/增强子元件,能在角质形成细胞内定向表达CreERT2融合蛋白。CreERT2融合蛋白由雌激素受体(estrogen receptor,ER)的配体结合区突变体(ERT)与Cre重组酶蛋白组成。在没有他莫昔芬诱导的情况下,CreERT2蛋白在细胞质内处于无活性状态;而在他莫昔芬诱导后,他莫昔芬的代谢产物4-羟基他莫昔芬与ERT结合,可使CreERT2进核发挥Cre的重组酶活性。PPARγflox/flox小鼠即PPARγ条件性敲除小鼠,该小鼠PPARγ基因特定外显子的两端被插入了loxP位点,而loxP位点可以被活性的Cre重组酶切割。用Krt5-CreERT2小鼠与含有loxP侧翼序列的小鼠交配,生成基因型为Krt5-CreERT2+/-PPARγflox/flox+/-及Krt5-CreERT2+/-PPARγflox/flox-/-的后代小鼠,这些小鼠表达Krt5的细胞中的PPARγ基因可以被他莫昔芬诱导的Cre介导的基因重组所敲除。我们构建的Krt5-CreERT2+/-PPARγflox/flox+/-及Krt5-CreERT2+/-PPARγflox/flox-/-小鼠可以精准的在角质形成细胞中定向敲除PPARγ基因。并且相比一般的条件敲除小鼠,可以精准调控基因敲除时间,在同一个体上对比基因敲除前后的小鼠表型,能最大程度地模拟疾病发生的过程,是理想的SLE疾病模型小鼠。
C57BL/6小鼠,周龄约6-12周,体重20-30g,由江苏集萃药康有限公司提供。杂交生成子一代,保留PPARγflox/flox+/-Krt5-CreERT2+/-小鼠,继续繁育得到PPARγflox/flox-/-Krt5-CreERT2+/-小鼠。实验所用小鼠为PPARγflox/flox+/-Krt5-CreERT2+/-及PPARγflox/flox-/-Krt5-CreERT2+/-小鼠。饲养条件:室温18-20℃,湿度50-60%,明暗交替(12h),光度适度,通风洁净良好。所有实验均获得批准,并按照中国医学科学院皮肤病医院(中国医学科学院皮肤病研究所)伦理委员会的指引进行。
2.实验方法
2.1小鼠角质形成细胞敲除PPARγ
根据需求对PPARγflox/flox+/-Krt5-CreERT2+/-及PPARγflox/flox-/-Krt5-CreERT2+/-小鼠进行分组:对照组、PPARγ半敲除组、PPARγ全敲除组。将100mg他莫昔芬混于0.5ml乙醇中,加入9.5ml玉米油,使用涡旋器彻底混合悬浮液,并在37℃的超声波浴中超声处理20分钟,配成待用的他莫昔芬溶液。对照组腹腔注射100μl含有5%乙醇的玉米油;PPARγ半敲除组为PPARγflox/flox+/-Krt5-CreERT2+/-小鼠,腹腔注射100μl的他莫昔芬溶液(相当于1mg他莫昔芬);PPARγ全敲除组为PPARγflox/flox-/-Krt5-CreERT2+/-小鼠,腹腔注射100μl的他莫昔芬溶液(相当于1mg他莫昔芬)。三组小鼠连续腹腔注射对应溶液5天。
2.2PCR鉴定角质形成细胞PPARγ条件性敲除小鼠基因型
利用鼠尾基因型快速鉴定试剂盒(Beyotime),根据说明书,提取小鼠尾巴基因组DNA。取新鲜的小鼠尾巴:实验前用70%的乙醇冲洗剪刀和镊子。剪取0.2-1cm的小鼠尾尖制备模板DNA,并配制PCR反应体系:

PCR反应结束后进行琼脂糖凝胶电泳。检测小鼠基因型。引物序列如下:
PPARγflox/flox引物1:5’-CTTCCCCTTCCCCAAAATGAGTC-3’;
PPARγflox/flox引物2:5’-TCTGTGGCTGGACTACAGGA-3’;
Krt5-e(2A-CreERT2)引物1:5’-GTGGCTTACATTCTGCAACATTTT-3’;
Krt5-e(2A-CreERT2)引物2:5’-GGCCCACGCTTCACCAG-3’;
Krt5-e(2A-CreERT2)引物3:5’-GGATCCGCCGCATAACCAGT-3’。
其中,对于PPARγflox:变体(Mutant)大小为572bp,杂合子(Heterozygote)大小为572bp,野生型(Wild type)为445bp;
对于Krt5-Cre:变体(Mutant)大小为610bp,杂合子(Heterozygote)大小为610bp,野生型(Wild type)为453bp。
2.3小鼠皮损、尿蛋白及自身抗体监测
每周观察小鼠皮损,相机拍照留存。定时取小鼠清洁中段尿,使用优利特蛋白尿检测试纸检测尿蛋白。使用CBB法定量小鼠尿蛋白含量。使用ELISA试剂盒(Cusabio)检测外周血血清抗核抗体及抗双链DNA抗体含量。
2.4组织病理
第11天,小鼠麻醉后颈椎脱臼处死。用锋利手术刀和眼科剪取上述三组小鼠同一时间同一部位的前胸皮肤,约0.5厘米×0.5厘米,摊平于锡箔纸上,液氮速冻或固定于4%多聚甲醛。固定后进行脱水、浸蜡、包埋。将组织块切片后,进行摊片、烘片、脱蜡、苏木精和伊红染色,封片后用显微镜选取视野拍照,分析小鼠皮损中炎症细胞浸润。
2.5免疫荧光染色
将液氮速冻后的皮肤组织或肾脏用OCT包埋后,置于冰冻切片模具上,用冰冻切片机切片后,洗去OCT,免疫荧光封闭液(Beyotime)37℃封闭1小时后加入山羊抗鼠IgG-FITC(1:200)4℃过夜孵育,PBS清洗后用DAPI工作液室温染细胞核10分钟,PBS清洗后封片,用荧光显微镜成像观察皮肤及肾小球IgG沉积。
3.实验结果
图1A为角质形成细胞PPARγ条件性敲除小鼠基因型鉴定。目的小鼠为PPARγ纯合敲除(基因型为Krt5-CreERT2+/-PPARγflox/flox-/-,如58号),或PPARγ杂合敲除(基因型为Krt5-CreERT2+/-PPARγflox/flox+/-,如51、61号)小鼠。
图1B角质形成细胞PPARγ敲除第11天小鼠照片,可见脱毛及炎性皮损,对照组小鼠无皮损。说明角质形成细胞内PPARγ蛋白含量下降的小鼠会自发出现炎性皮损。
图1C小鼠角质形成细胞PPARγ敲除第11天皮肤组织学检查表明存在角化过度、棘层增厚、基底细胞液化变性,真皮层免疫细胞浸润。对照组小鼠同部位皮肤无明显炎性改变。以上结果再次证明角质形成细胞内PPARγ蛋白含量下降的小鼠会自发出现炎性皮损。
图1D角质形成细胞PPARγ敲除第11天小鼠尿蛋白及外周血抗双链DNA抗体含量。相较于对照组,PPARγ半敲除组及PPARγ全敲除组小鼠尿蛋白及外周血抗双链DNA抗体(外周血自身抗体抗双链DNA抗体,对照组vs PPARγ半敲除组vs PPARγ全敲除组:41.63ng/mlvs 306.81ng/ml vs 397.33ng/ml)明显上升。以上结果表明角质形成细胞中PPARγ降低会使小鼠出现尿蛋白及抗双链DNA抗体升高。
图1E角质形成细胞PPARγ敲除第11天小鼠肾小球IgG沉积。相较于对照组,PPARγ半敲除组及PPARγ全敲除组小鼠肾小球IgG沉积明显增加。以上结果表明角质形成细胞中PPARγ降低会使小鼠出现肾小球IgG沉积。
我们共使用14只PPARγ杂合敲除及PPARγ纯合敲除小鼠进行造模,14只小鼠均造模成功,造模成功率100%。以上结果表明,该动物模型表型与SLE临床症状类似。实施例24-羟基他莫昔芬双耳涂抹激活的局部角质形成细胞PPARγ敲除诱导SLE模型。
1.实验材料

1.2实验用动物
本实验所用小鼠为Krt5-CreERT2+/-PPARγflox/flox+/-及Krt5-CreERT2+/-PPARγflox/flox-/-小鼠。小鼠来源及饲养条件与实施例1 1.2相同。
2.实验方法
2.1小鼠局部角质形成细胞敲除PPARγ
根据需求对Krt5-CreERT2+/-PPARγflox/flox+/-及Krt5-CreERT2+/-PPARγflox/flox-/-小鼠进行分组:对照组、局部PPARγ半敲除组、局部PPARγ全敲除组。将50mg4-羟基他莫昔芬混于1mlDMSO中,加入9ml玉米油,使用涡旋器彻底混合悬浮液,并在37℃的超声波浴中超声处理20分钟,配成待用的4-羟基他莫昔芬溶液(5mg/ml)。对照组双耳涂抹含有10%DMSO的玉米油;局部PPARγ半敲除组为PPARγflox/flox+/-Krt5-CreERT2+/-小鼠,双耳涂抹50ul 5mg/ml 4-羟基他莫昔芬的溶液;局部PPARγ全敲除组为PPARγflox/flox-/-Krt5-CreERT2+/-小鼠,双耳涂抹50ul5mg/ml 4-羟基他莫昔芬的溶液。三组小鼠连续双耳涂抹对应溶液5天。
2.2PCR鉴定角质形成细胞PPARγ条件性敲除小鼠基因型
实验方法同实施例1 2.2。
2.3小鼠皮损、尿蛋白及自身抗体监测
每周观察小鼠皮损,相机拍照留存。定时取小鼠清洁中段尿,使用优利特蛋白尿检测试纸检测尿蛋白。使用CBB法定量小鼠尿蛋白含量。使用ELISA试剂盒(Cusabio)检测外周血血清抗核抗体及抗双链DNA抗体含量。
2.4组织病理

3.实验结果
图2A局部角质形成细胞PPARγ敲除第14天小鼠照片,可见脱毛及炎性皮损,对照组小鼠无皮损。说明角质形成细胞内PPARγ蛋白含量下降的小鼠会自发出现炎性皮损。
图2B局部小鼠角质形成细胞PPARγ敲除第17天皮肤组织学检查表明存在角化过度、毛囊角栓、棘层增厚、基底细胞液化变性,真皮层免疫细胞浸润。对照组小鼠同部位皮肤无明显炎性改变。以上结果再次证明角质形成细胞内PPARγ蛋白含量下降的小鼠会自发出现SLE样皮损病理改变。
图2C局部小鼠角质形成细胞PPARγ敲除第17天皮肤组织冰冻切片表明的基底膜带IgG沉积。对照组小鼠同部位皮肤无基底膜带IgG沉积。说明角质形成细胞内PPARγ蛋白含量下降的小鼠会自发出现皮肤基底膜带IgG沉积。
图2D局部角质形成细胞PPARγ敲除第14天小鼠尿蛋白。局部PPARγ半敲除组、局部PPARγ全敲除组小鼠尿蛋白明显上升,对照组小鼠尿蛋白无明显变化。以上结果表明角质形成细胞中PPARγ降低会使小鼠出现自发性蛋白尿。
图2E局部角质形成细胞PPARγ敲除第17天小鼠肾小球IgG沉积。相较于对照组,局部PPARγ半敲除组、局部PPARγ全敲除组小鼠肾小球有明显IgG沉积。以上结果表明角质形成细胞中PPARγ降低会使小鼠出现肾小球IgG沉积。
图2F局部角质形成细胞PPARγ敲除第17天小鼠外周血抗核抗体及抗双链DNA抗体含量。局部PPARγ半敲除组、局部PPARγ全敲除组小鼠外周血抗核抗体及抗双链DNA抗体含量明显高于对照组小鼠。以上结果表明角质形成细胞中PPARγ降低会使小鼠出现自身抗体。
我们共使用15只PPARγ杂合敲除及PPARγ纯合敲除小鼠进行造模,15只小鼠均造模成功,造模成功率100%。以上结果表明,该动物模型表型与SLE临床症状类似。
综上所述,我们建立了角质形成细胞PPARγ敲除诱导自发型SLE动物模型的构建方法,以期解决当前SLE模型与临床差异大、价格高、不稳定的难点。
本发明提供了一种通过敲除角质形成细胞PPARγ制备自发型SLE动物模型的思路及方法,具体实现该技术方案的方法和途径很多,以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。本实施例中未明确的各组成部分均可用现有技术加以实现。
Claims (10)
1.一种自发型系统性红斑狼疮动物模型的构建方法,其特征在于,包括如下步骤:
(1)构建角质形成细胞条件性PPARγ敲除小鼠,所述角质形成细胞条件性PPARγ敲除小鼠包括PPARγ flox/flox+/- Krt5-CreERT2+/-和PPARγ flox/flox-/- Krt5-CreERT2+/-的C57BL/6小鼠;
(2)使用基因敲除激活剂敲除角质形成细胞中的PPARγ基因以介导疾病表型发生。
2.根据权利要求1所述的构建方法,其特征在于,所述基因敲除激活剂为代谢后可与雌激素受体突变体ERT结合,使CreERT2发挥Cre重组酶活性进行基因敲除的化合物。

5.根据权利要求3所述的构建方法,其特征在于,使用他莫昔芬作为基因敲除激活剂时,对角质形成细胞条件性PPARγ敲除小鼠连续腹腔注射他莫昔芬溶液1-7天,每天一次;使用4-羟基他莫昔芬作为基因敲除激活剂时,对角质形成细胞条件性PPARγ敲除小鼠身体部位无毛或剃毛后的皮肤双耳的腹侧及背侧的皮肤连续涂抹4-羟基他莫昔芬溶液1-7天,每天一次。
6. 根据权利要求5所述的构建方法,其特征在于,所述他莫昔芬溶液按照100mg他莫昔芬、0.5ml乙醇、9.5ml玉米油的比例配置;所述4-羟基他莫昔芬溶液按照50mg 4-羟基他莫昔芬、1mlDMSO、9ml玉米油的比例配置;每次给小鼠注射的他莫昔芬溶液的剂量为75mg/kg,每次给小鼠外用涂抹的4-羟基他莫昔芬溶液的剂量为10-80μL。
