Abstract
The invention provides a urethra accurate drug delivery rod and a preparation method thereof. The drug delivery rod comprises a matrix rod, wherein the matrix rod comprises a handle and a fiber cotton rod body with pores on the surface, and the fiber cotton rod body is connected with the distal end of the handle; the distal end of the fiber cotton stick body is bent away from the axis to one side; the drug delivery stick also comprises a super-lubrication hydrogel coating coated on the fiber cotton stick body; the coating is prepared by the cross-linking reaction of raw materials including sodium alginate, hyaluronic acid, beta-sodium glycerophosphate and medicines for treating related diseases of urinary surgery in a cross-linking curing bath. The preparation method of the drug delivery rod is simple, the cost is low, the coating has ultra-high hydrophilicity and ultra-lubricity, and the uncomfortable feeling of a patient in the insertion process can be greatly reduced; the coating and the matrix rod have good anchoring effect, so that the medicine-carrying gel layer is prevented from falling off or sliding out in the inserting process; in addition, both the matrix rod and the coating can carry medicine and can stimulate the release of the medicine under the temperature and the pressure, thereby being beneficial to the treatment of diseases.

Technical Field
The invention relates to the field of medical instruments, in particular to a urethra accurate drug delivery rod and a preparation method thereof.
Background
Targeted or precise administration is the most effective route to improve drug utilization efficiency and reduce physical damage. At present, the administration through the urethra is a common administration mode in the treatment process of related diseases of urinary surgery, and particularly, the administration mode has a plurality of applications in the treatment of urethral stricture and postoperative rehabilitation of urethral stricture. At present, the common administration mode for treating the urethral stricture is to coat the drug ointment on the catheter, and then insert the catheter with the drug into the corresponding position through the urethra, thereby achieving the purpose of administration. However, the method has low administration efficiency, low drug utilization rate, unsatisfactory treatment effect, inconvenient use, easy secondary infection caused by improper operation, and the like. There is therefore a need for a more accurate way of urinary tract administration.

immersing the distal end of the fiber cotton stick body into sodium alginate solution, and fully absorbing and saturating;
immersing the distal end of the fiber cotton stick body into a composite solution of sodium alginate, hyaluronic acid, beta-sodium glycerophosphate and a medicament for treating related diseases of urinary surgery to enable the composite solution to be adhered to the fiber cotton stick body;
and thirdly, immersing the distal end of the fiber cotton stick body into crosslinking curing liquid, and crosslinking for 1-60 min to obtain the urethra precise drug delivery stick loaded with the super-lubrication drug-carrying hydrogel.
Further, the concentration of the sodium alginate solution in the first step is 1-10%.
Compared with the prior art, the invention has the following technical effects:
the preparation method of the drug delivery rod has simple process and low cost, and the synthesized hydrogel coating has ultrahigh hydrophilicity and super lubricity, so that the discomfort of a patient in the process of insertion can be greatly reduced; secondly, creatively selecting a fiber cotton stick as a matrix stick, wherein the fiber cotton stick has a loose porous structure and certain hardness and toughness, is deformable, can adsorb moisture and medicines, and because of the porous structure on the surface of the cotton stick, hydrogel can permeate into pores on the surface of the cotton stick, and an antenna is formed after solidification, so that a good anchoring effect is realized between a coating and the matrix stick, and the falling or sliding-out of a medicine-carrying gel layer in the inserting process is avoided; in addition, both the matrix rod and the hydrogel coating may be drug loaded and may stimulate drug release at temperature and pressure.

Detailed Description
The invention provides a urethra accurate drug delivery rod, which comprises a basal body rod 1, wherein the basal body rod 1 comprises a handle and a fiber cotton rod body, wherein the surface of the fiber cotton rod body is connected with the distal end of the handle, and the surface of the fiber cotton rod body is provided with pores; the distal end of the fiber cotton stick body is bent away from the axial line to one side; the dosing rod also comprises a super-lubricated hydrogel coating 2 (shown in figure 1) coated on the fiber cotton rod body;
wherein the super-lubricating hydrogel coating 2 is obtained by the cross-linking reaction of raw materials comprising sodium alginate, hyaluronic acid, beta-sodium glycerophosphate and medicines for treating related diseases of urinary surgery in a cross-linking curing bath. Because the porosity of the fiber cotton swab body (figure 2) can enable the hydrogel to enter the pores, an anchoring effect is formed between the super-lubrication hydrogel coating and the swab body, and the hydrogel coating can be firmly attached to the surface of the porous cotton swab.

In a preferred embodiment of the present invention, the mass ratio of the sodium alginate, the hyaluronic acid, the sodium beta-glycerophosphate and the drug for treating the urology related diseases included in the super-lubricated hydrogel coating 2 is 1-10: 0.1 to 5:1 to 60: 0.001-10.
In a preferred embodiment of the present invention, the medicament for treating a urologic related disease is a medicament for treating urethral stricture, and preferably includes one or more of diphenhydramine, tacrolimus, clobetasol propionate, dexamethasone, and the like.

In a preferred embodiment of the present invention, the angle α between the curved line formed by the distal end of the fiber cotton swab body and the axis is 10 to 30 °.
In a preferred embodiment of the present invention, the crosslinking curing bath is a soluble calcium salt solution including one or more of calcium chloride, calcium nitrate, calcium sulfate, and calcium silicate; the concentration of the soluble calcium salt is 0.1-10%.
The second aspect of the present invention provides a method for preparing the urethral precision drug delivery stick, comprising the steps of:
immersing the distal end of the fiber cotton stick body into sodium alginate solution, and fully absorbing and saturating;
immersing the distal end of the fiber cotton stick body into a composite solution of sodium alginate, hyaluronic acid, beta-sodium glycerophosphate and a medicament for treating related diseases of urinary surgery to enable the composite solution to be adhered to the fiber cotton stick body; wherein the concentration of the sodium alginate is 1-10wt%, the concentration of the hyaluronic acid is 0.1-5wt%, the concentration of the beta-sodium glycerophosphate is 1-60wt%, and the concentration of the medicament is 0.001-10wt%.
Immersing the distal end of the fiber cotton stick body into a crosslinking curing liquid, wherein the concentration of the crosslinking curing liquid is 0.1-10wt%, and crosslinking for 1-60 min to obtain the urethra precise drug delivery stick loaded with the super-lubrication drug-carrying hydrogel.
The drug delivery rod provided by the invention adopts nontoxic and biodegradable absorbed sodium alginate as a gel skeleton, hyaluronic acid as a super-lubrication buffer, beta-sodium glycerophosphate as a drug carrier and a temperature-sensitive control agent, and the drug delivery rod is crosslinked in a calcium ion solution to form a super-lubrication hydrogel coating; the hydrogel can be infiltrated into the porous fiber cotton stick body to form an antenna anchoring effect with the fiber cotton stick body, so that the hydrogel coating can be firmly attached to the surface of the porous cotton stick; the fiber cotton stick body has a certain hardness and toughness, the changeable porous fiber cotton stick body with a certain radian of the far end is convenient for entering the urethra and can alleviate the discomfort of patients.

Example 1
The embodiment provides a urethra accurate drug delivery rod loaded with super-lubrication drug-loaded hydrogel, and the preparation method comprises the following steps:
immersing the distal end of the fiber cotton stick body into a sodium alginate solution with the weight percent of 2%, and fully absorbing and saturating;
immersing the distal end of the fiber cotton stick body into a composite solution of 2wt% of sodium alginate, 1wt% of hyaluronic acid, 10wt% of beta-sodium glycerophosphate and 0.5wt% of bifeitin to enable the mixed solution with certain viscosity to adhere to the fiber cotton stick body;
and thirdly, immersing the distal end of the fiber cotton stick body into 3wt% of crosslinking curing liquid, and crosslinking for 30min to obtain the urethra precise drug delivery stick loaded with the super-lubrication drug-carrying hydrogel, wherein the structure of the urethra precise drug delivery stick is shown in figure 1.

Fig. 3 shows a photograph of the contact angle test of the urethral precision dosing rod, illustrating that the synthesized hydrogel coating has ultra-high hydrophilicity.
Fig. 4 shows the friction coefficient curve of the urethral precision drug delivery stick, and it can be seen that the friction coefficient of the surface of the hydrogel layer is as low as 0.098, indicating that the drug delivery stick has good lubrication performance.
Fig. 5 shows the drug release profile of the urethral precision drug delivery stick, and it can be seen that the drug content in the solution gradually increases with time, and the drug release reaches the maximum value when 60min is reached, that is, the drug delivery stick can rapidly release the drug within 60 min.
Example 2
The embodiment provides a urethra accurate drug delivery rod loaded with super-lubrication drug-loaded hydrogel, and the preparation method comprises the following steps:
immersing the distal end of the fiber cotton stick body into 3wt% sodium alginate solution, and fully absorbing and saturating;
immersing the distal end of the fiber cotton stick body into a composite solution of 3wt% of sodium alginate, 2wt% of hyaluronic acid, 30wt% of beta-sodium glycerophosphate and 1wt% of tacrolimus to enable the mixed solution with certain viscosity to be adhered on the fiber cotton stick body;
and thirdly, immersing the distal end of the fiber cotton stick body into 5wt% of crosslinking curing liquid, and crosslinking for 10min to obtain the urethra precise drug delivery stick loaded with the super-lubrication drug-carrying hydrogel.
Example 3
The embodiment provides a urethra accurate drug delivery rod loaded with super-lubrication drug-loaded hydrogel, and the preparation method comprises the following steps:
immersing the distal end of the fiber cotton stick body into a sodium alginate solution with the concentration of 6wt%, and fully absorbing and saturating;
immersing the distal end of the fiber cotton stick body into a composite solution of 6wt% of sodium alginate, 3wt% of hyaluronic acid, 10wt% of beta-sodium glycerophosphate and 2wt% of compound clobetasol propionate to enable the mixed solution with certain viscosity to be adhered on the fiber cotton stick body;
and thirdly, immersing the distal end of the fiber cotton stick body into a crosslinking curing liquid with the weight percent of 1 percent, and crosslinking for 60 minutes to obtain the urethra precise drug delivery stick loaded with the super-lubrication drug-carrying hydrogel.

the super-lubricating hydrogel coating is prepared by the cross-linking reaction of raw materials comprising sodium alginate, hyaluronic acid, beta-sodium glycerophosphate and medicines for treating related diseases of urinary surgery in a cross-linking curing bath.
2. The precise urethral dosing rod according to claim 1, wherein the mass ratio of sodium alginate, hyaluronic acid, beta-sodium glycerophosphate and the drug for treating urologic related diseases contained in the super-lubricated hydrogel coating is 1-10: 0.1 to 5:1 to 60: 0.001-10.

immersing the distal end of the fiber cotton stick body into a composite solution of sodium alginate, hyaluronic acid, beta-sodium glycerophosphate and a medicament for treating related diseases of urinary surgery to enable the mixed solution to adhere to the fiber cotton stick body;
and thirdly, immersing the distal end of the fiber cotton stick body into crosslinking curing liquid, and crosslinking for 1-60 min to obtain the urethra precise drug delivery stick loaded with the super-lubrication drug-carrying hydrogel.
10. The method according to claim 9, wherein the concentration of the sodium alginate solution in the first step is 1 to 10%.
