Abstract
本发明涉及一种用于防治畜禽肾损伤的中草药组合物及其制备方法。所述中草药组合物是通过将中草药原料进行挥发油提取、煎煮浓缩药液、混合三大步骤制备得到的;所述中草药原料由以下重量份的中草药组成:槲叶10~30份、姜皮10~30份、榆白皮5~15份、桂皮10~30份、陈皮10~40份、车前草1~10份、红毛五加皮10~20份、牡丹皮5~10份、姜黄5~15份。本发明在中兽医理论的基础上,将上述各中草药配伍组合,协同调理畜禽体质,可以最大限度地发挥各中草药的功能,从而有效地保护畜禽肾脏功能,减少肾水肿、尿酸盐沉积、关节肿大等问题的发生,而对畜禽其它脏器无任何副作用和损伤。
Description
技术领域
本发明涉及一种畜禽用中草药及其制备方法,特别涉及的是一种预防治疗畜禽肾功能损伤的中草药;还涉及该药物组合物的制备方法。
背景技术
近几年,由于畜禽疾病越来越严重,饲料原料品质的下降、饲料霉变,不合理使用药物等原因造成畜禽肾脏功能受到损伤。在家禽上表现为花斑肾、肾肿、肾出血、腹水等,在家畜上表现为肾肿大、血尿、蛋白尿、高尿酸症等,给畜禽养殖带来巨大损失,直接影响养殖效益。
由于其起因的复杂性,其针对性的治疗药物比较少,通常采用丙磺舒、碳酸氢钠等通肾药物,但会造成肾脏滤过功能的损伤,出现肾阳虚表现,容易导致腹泻脱水,在治疗期间畜禽出现大量死亡现象,从而影响了这类药物在临床上的推广使用。中草药在解决肾脏功能问题上具有数千年的历史,是解决畜禽肾损伤的一种重要方法。
发明内容
本发明所要解决的技术问题是提供一种副作用小、效果明显的防治畜禽肾损伤的中草药组合物及其制备方法。
本发明一方面提供了一种用于防治畜禽肾损伤的中草药组合物,所述中草药组合物的制备方法包括如下步骤:
(1)挥发油提取步骤:将姜皮、桂皮、牡丹皮、陈皮按重量配比混合,加水浸泡,煮沸提取挥发油,分别得到挥发油、药液和药渣;将挥发油和药液分别过滤后备用;
(2)煎煮浓缩药液步骤:将步骤(1)得到的药渣与槲叶、榆白皮、车前草、红毛五加皮、姜黄混合,加水煎煮2次,过滤,得到提取药液;将提取药液与步骤(1)得到的药液混合后浓缩,得到浓缩药液;
(3)混合步骤:在步骤(1)得到的挥发油中加入助溶剂,然后与步骤(2)得到的浓缩药液混合均匀,即制得所述中草药组合物。
所述制备方法中姜皮、桂皮、牡丹皮、陈皮、槲叶、榆白皮、车前草、红毛五加皮、姜黄的重量份数分别为:姜皮10~30份、桂皮10~30份、牡丹皮5~10份、陈皮10~40份、槲叶10~30份、榆白皮5~15份、车前草1~10份、红毛五加皮10~20份、姜黄5~15份。
作为一种优选的技术方案,所述制备方法中姜皮、桂皮、牡丹皮、陈皮、槲叶、榆白皮、车前草、红毛五加皮、姜黄的重量份数分别为:姜皮10份、桂皮10份、牡丹皮5份、陈皮10份、槲叶10份、榆白皮5份、车前草1份、红毛五加皮10份、姜黄5份。
作为一种优选的技术方案,所述制备方法中姜皮、桂皮、牡丹皮、陈皮、槲叶、榆白皮、车前草、红毛五加皮、姜黄的重量份数分别为:姜皮30份、桂皮30份、牡丹皮10份、陈皮40份、槲叶30份、榆白皮15份、车前草10份、红毛五加皮20份、姜黄15份。
作为一种优选的技术方案,所述制备方法中姜皮、桂皮、牡丹皮、陈皮、槲叶、榆白皮、车前草、红毛五加皮、姜黄的重量份数分别为: 姜皮20份、桂皮20份、牡丹皮7.5份、陈皮25份、槲叶20份、榆白皮8份、车前草5份、红毛五加皮15份、姜黄10份。
作为一种改进的技术方案,所述步骤(1)包括:将姜皮、桂皮、牡丹皮、陈皮按重量配比混合,按药材重量的8-10倍加水浸泡4h以上,多功能提取罐煮沸提取3h以上,分别收集挥发油和药液,过滤后备用;
作为一种改进的技术方案,所述步骤(2)包括:提取罐中加入槲叶、榆白皮、车前草、红毛五加皮、姜黄,与步骤(1)得到的药渣一起加水煮沸2次,每次加水量是药材重量的10倍,每次煮沸1.5h以上,过滤得到提取药液,与步骤(1)得到的药液混合后浓缩,得到按照原生药计1g/ml的浓缩药液。
作为一种改进的技术方案,所述步骤(2)包括:在步骤(1)得到的挥发油中,加入乙醇、吐温-80、OP-10、月桂酸聚氧乙烯酯,加入量分别不低于挥发油体积的0.5倍、0.2倍、0.1倍、0.2倍,搅拌均匀后,混入到步骤(2)得到的浓缩药液中,即得到中草药组合物。
本发明采用行气利水、健脾、活血的治疗法则,科学组方,调理脾肾功能,以达到防治肾功能损伤的目的。本发明中槲叶、姜皮作为君药,姜皮性辛凉,能行水消肿,槲叶性甘、味苦,利尿,促进水液代谢,共同配合起到行气利水的目的;姜黄作为臣药,性辛温,具有破血,行气,通经的作用,助姜皮行气,引药入肾;榆白皮、车前草作为臣药,助槲叶利水,通淋;桂皮、陈皮作为佐药,桂皮味辛甘、性热,有补元阳,暖脾胃,温阳化气的功效,陈皮性温,味辛,调中焦、理气健脾,以防君药行水利尿太过,造成脱水,牡丹皮活血化瘀,助君药利水;红毛五加皮性温,味辛,入肝、肾经,可祛湿,通关节,引诸药入肾;诸药相互配合调整气血精液运行正常,利尿排毒,将有毒有害物质排出体外,减轻肾脏负担,温阳化气,水液上输于脾、胃、肺等脏腑,保证机体对水的利用吸收,防止出现脱水,全面解决肾功能损伤问题。
由于采用了上述技术方案,本发明的有益效果是:将上述各中草药组有机结合,协同调理畜禽体质,可以最大限度地发挥各中草药的功能,从而有效地防治畜禽肾损伤,而对畜禽其它器官无任何副作用和损伤。且本发明在中草药加工过程中采用挥发油提取工艺,挥发油会影响本发明中草药组合物的治疗及预防效果。
实施方式
在下面的详细描述中,只通过说明的方式描述了本发明的某些示范性实施例。毋庸置疑,本领域的普通技术人员可以认识到,在不偏离本发明的精神和范围的情况下,可以用各种不同的方式对所描述的实施例进行修正。
实施例
本实施例所述中草药组合物,其制备方法如下:
取姜皮10kg、桂皮10kg、牡丹皮5kg、陈皮10kg置于多功能提取罐中,加8倍水浸泡4h,多功能提取罐煮沸提取3h,收集得到挥发油405毫升、药液203千克;将槲叶10kg、榆白皮5kg、车前草1kg、红毛五加皮10kg、姜黄5kg投入带药渣的多功能提取罐中,加入660kg水,煎煮1次,煮沸时间为1.5h,煎煮提取完成后,过滤得到提取药液,药渣加入660kg水再次煮沸1.5h,过滤得到提取药液,两次煎煮得到的提取药液与挥发油提取步骤得到的药液进行合并浓缩,得到65千克浓缩药液;在挥发油提取步骤得到的挥发油中,加入乙醇203ml、吐温-80 81 ml、OP-10 41ml、月桂酸聚氧乙烯酯81ml,搅拌均匀后,混入到煎煮浓缩步骤所得到的浓缩药液65kg中,加入苯甲酸钠0.3kg作为防腐剂,得到中草药组合物。
实施例
本实施例所述中草药组合物,其制备方法如下:
取姜皮30kg、桂皮30kg、牡丹皮10kg、陈皮40kg置于多功能提取罐中,加10倍水浸泡6h,多功能提取罐煮沸提取5h,收集得到挥发油1120毫升、药液580kg;将槲叶30kg、榆白皮15kg、车前草10kg、红毛五加皮20kg、姜黄15kg投入带药渣的多功能提取罐中,加入2000kg水,煎煮1次,煮沸时间为1.5h,煎煮提取完成后,过滤得到提取药液,药渣加入2000kg水再次煮沸1.5h,过滤得到提取药液,两次煎煮得到的提取药液与挥发油提取步骤得到的药液进行合并浓缩,得到200kg浓缩药液;在挥发油提取步骤得到的挥发油中,加入乙醇1120ml、吐温-80 560ml、OP-10 224ml、月桂酸聚氧乙烯酯560ml,搅拌均匀后,混入到煎煮浓缩步骤所得到的浓缩药液200kg中,加入苯甲酸钠1kg作为防腐剂,得到中草药组合物。
实施例
本实施例所述中草药组合物,其制备方法如下:
取姜皮20kg、桂皮20kg、牡丹皮7.5kg、陈皮25kg置于多功能提取罐中,加10倍水浸泡5h,多功能提取罐煮沸提取4h,收集得到挥发油700毫升、药液433kg;将槲叶20kg、榆白皮8kg、车前草5kg、红毛五加皮15kg、姜黄10kg投入带药渣的多功能提取罐中,加入1150kg水,煎煮1次,煮沸时间为1.5h,煎煮提取完成后,过滤得到提取药液,药渣加入1000kg水再次煮沸1.5h,过滤得到提取药液,两次煎煮得到的提取药液与挥发油提取步骤得到的药液进行合并浓缩,得到112kg浓缩药液;在挥发油提取步骤得到的挥发油中,加入乙醇700ml、吐温-80 350ml、OP-10 210ml、月桂酸聚氧乙烯酯210ml,搅拌均匀后,混入到煎煮浓缩步骤所得到的浓缩药液112kg中,加入苯甲酸钠0.5kg作为防腐剂,得到中草药组合物。
对比例:
一种中草药组合物,其制备方法如下:
取姜皮20kg、桂皮20kg、牡丹皮7.5kg、陈皮25kg置于多功能提取罐中,加10倍水浸泡5h,多功能提取罐煮沸提取4h,收集得到药液432kg;将槲叶20kg、榆白皮8kg、车前草5kg、红毛五加皮15kg、姜黄10kg投入带药渣的多功能提取罐中,加入1150kg水,煎煮1次,煮沸时间为1.5h,煎煮提取完成后,过滤得到提取药液,药渣加入1000kg水再次煮沸1.5h,过滤得到提取药液,两次煎煮得到的提取药液与挥发油提取步骤得到的药液进行合并浓缩,得到112kg浓缩药液;将乙醇700ml、吐温-80 350ml、OP-10 210ml、月桂酸聚氧乙烯酯210ml,搅拌均匀后,混入到煎煮浓缩步骤所得到的浓缩药液112kg中,加入苯甲酸钠0.5kg作为防腐剂,得到对比例的中草药组合物。
实施例三与对比例的主要区别是,对比例没有进行挥发油的提取保存,所制得的中草药组合物中没有挥发油。
应用试验及效果:
1、山东省昌乐县某养殖有限公司,2020年养殖的肉鸡大批发生肾功能损伤,表现为肾肿、花斑肾,腹水严重,部分肉鸡瘫痪、关节肿、采食量大幅下降,经诊断疑似霉菌毒素造成的肾功能损伤。按照每吨饮水中添加2L实施例一的中草药组合物,增强肾脏功能,利水排尿,促进毒素排出,连续使用3天后,肉鸡采食量恢复,瘫痪肉鸡恢复常态,养殖场内鸡群恢复正常。
2、实验室评价:选择42日龄海兰褐蛋鸡240只,随机分为6组(A-F组),每组40只,每组设8个重复,每个重复5只(5只为1笼),脚环标识。其中A组为健康对照组,始终饲喂正常饲料。B-F组饲喂高蛋白高钙饲料,饲喂至77日龄(共35天),剔除不符合痛风纳入标准鸡只,开始给药治疗;其中B组为模型对照组,不给药治疗;C组为实施例三中草药组合物组,按照每公斤组合物兑水500kg饮水添加; D组为对比例中草药组合物组,每公斤组合物兑水500kg饮水添加;E组为五皮散对照组,按照每公斤饲料添加5克拌料,F组为木通海金沙散对照组,按照每只鸡1g添加于饲料中。
至造模第35天,造模组病症积分均大于4,共180只;尿酸大于480μmol/L共170只,每组剖检1只,剩余165只纳入试验。
结果显示,连续给药治疗5天,C组实施例三中草药组合物组可显著改善痛风模型鸡的粪便、精神、采食等临床症状,可有效对抗高蛋白高钙饲料诱发的血清尿酸和血钙的升高,有效改善模型鸡的增重和料肉比。D组对比例中草药组合物组能改善痛风模型鸡的粪便、精神、采食等临床症状,但不能明显的降低高蛋白高钙饲料诱发的血清尿酸和血钙的升高,可改善模型鸡的增重和料肉比,相对实施例三中草药组合物效果较差;实施例三中草药组合物的治疗效果明显优于五皮散和木通海金沙散。
3、安徽省砀山县某养殖家庭农场,有母猪200头,2021年10月发现哺乳仔猪腹泻严重,粪便带有石灰渣样腹泻,母猪表现霉菌毒素中毒症状,出现皮肤黄疸,四肢无力,直肠出血,排出干燥的粪便,尿液色泽较深;病死仔猪解剖发现肾肿大、腹水、心包积液严重,部分皮下积水。
在母猪饮水中添加实施例一所述中草药组合物,按照每头50毫升添加,并结合脱霉剂、氟苯尼考粉连续使用10天,母猪霉菌毒素中毒现象减轻,仔猪腹泻率降低,死淘数量减少,粪便逐步恢复正常,猪群恢复健康。

Claims (10)
1.一种用于防治畜禽肾损伤的中草药组合物,其特征在于,所述中草药组合物的制备方法包括以下步骤:
(1)挥发油提取步骤:将姜皮、桂皮、牡丹皮、陈皮按重量配比混合,加水浸泡,煮沸提取挥发油,分别得到挥发油、药液和药渣;将挥发油和药液分别过滤后备用;
(2)煎煮浓缩药液步骤:将步骤(1)得到的药渣与槲叶、榆白皮、车前草、红毛五加皮、姜黄混合,加水煎煮2次,过滤,得到提取药液;将提取药液与步骤(1)得到的药液混合后浓缩,得到浓缩药液;
(3)混合步骤:在步骤(1)得到的挥发油中加入助溶剂,然后与步骤(2)得到的浓缩药液混合均匀,即制得所述中草药组合物。
2.如权利要求1所述的中草药组合物,其特征在于,所述制备方法中姜皮、桂皮、牡丹皮、陈皮、槲叶、榆白皮、车前草、红毛五加皮、姜黄的重量份数分别为:姜皮10~30份、桂皮10~30份、牡丹皮5~10份、陈皮10~40份、槲叶10~30份、榆白皮5~15份、车前草1~10份、红毛五加皮10~20份、姜黄5~15份。
3.如权利要求2所述的中草药组合物,其特征在于,所述制备方法中姜皮、桂皮、牡丹皮、陈皮、槲叶、榆白皮、车前草、红毛五加皮、姜黄的重量份数分别为:姜皮10份、桂皮10份、牡丹皮5份、陈皮10份、槲叶10份、榆白皮5份、车前草1份、红毛五加皮10份、姜黄5份。
4.如权利要求2所述的中草药组合物,其特征在于,所述制备方法中姜皮、桂皮、牡丹皮、陈皮、槲叶、榆白皮、车前草、红毛五加皮、姜黄的重量份数分别为:姜皮30份、桂皮30份、牡丹皮10份、陈皮40份、槲叶30份、榆白皮15份、车前草10份、红毛五加皮20份、姜黄15份。
5.如权利要求2所述的中草药组合物,其特征在于,所述制备方法中姜皮、桂皮、牡丹皮、陈皮、槲叶、榆白皮、车前草、红毛五加皮、姜黄的重量份数分别为: 姜皮20份、桂皮20份、牡丹皮7.5份、陈皮25份、槲叶20份、榆白皮8份、车前草5份、红毛五加皮15份、姜黄10份。
6.如权利要求1-5任一所述的中草药组合物,其特征在于,所述步骤(3)中的助溶剂由乙醇、吐温-80、OP-10和月桂酸聚氧乙烯酯组成。
7.如权利要求6所述的中草药组合物,其特征在于,所述助溶剂中乙醇、吐温-80、OP-10、月桂酸聚氧乙烯酯的体积分别不低于步骤(1)得到的挥发油体积的0.5倍、0.2倍、0.1倍、0.2倍。
8.如权利要求7所述的中草药组合物,其特征在于,所述中草药组合物的制备方法包括以下步骤:
(1)取姜皮20kg、桂皮20kg、牡丹皮7.5kg、陈皮25kg置于多功能提取罐中,加10倍水浸泡5h,煮沸提取4h,收集得到挥发油700毫升和药液433kg,药渣留在提取罐中,将挥发油和药液过滤后备用;
(2)将槲叶20kg、榆白皮8kg、车前草5kg、红毛五加皮15kg、姜黄10kg投入带药渣的多功能提取罐中,加入1150kg水,煎煮1次,煮沸时间为1.5h,过滤得到提取药液;向药渣中加入1000kg水,再次煮沸1.5h,过滤得到提取药液;将两次煎煮得到的提取药液与步骤(1)得到的药液进行合并浓缩,得到112kg浓缩药液;
(3)在步骤(1)得到的挥发油中加入乙醇700ml、吐温-80 350ml、OP-10 210ml、月桂酸聚氧乙烯酯210ml,搅拌均匀后,混入步骤(2)得到的浓缩药液中,即制得所述中草药组合物。
9.权利要求1-8任一所述中草药组合物在畜禽饲料生产中的应用。
10.权利要求1-8任一所述中草药组合物在制备具有治疗畜禽肾功能损伤功能的药品中的应用。
