In a fourth aspect the present application provides a method of controlling pests, comprising applying a pharmaceutical composition according to the third aspect of the present application to pine wood nematodes and/or pine wood nematode habitat, using a compound according to the first aspect of the present application or a salt thereof.
In some embodiments of the application, the concentration of the compound is from 10 to 10 5 μM。
The beneficial effects of the application are as follows:
the absence of cholesterol in the nematode will result in a decrease in the number of eggs of the next generation of nematodes. When cholesterol is added again, the fertility of the nematodes can be recovered. The nematode sperm plasma membrane has very rich cholesterol content, and the cholesterol and sphingomyelin constitute lipid which is favorable for the activation of sperm. Cholesterol can affect the composition of sperm lipids and thus hinder sperm activation. Cholesterol plays an important role in the formation and activity of the nematode sperm, as well as the number of offspring. Therefore, based on pine wood nematode sterol transporter (Sterol Carrier Protein, SCP 2) as a target, a friendly and effective inhibitor BxSCPI (SCP-2 inhibitor, SCPI) is designed and screened, and the accumulation and utilization of intracellular cholesterol are influenced by competing with cholesterol to bind with BxSCP-2, so that the growth and reproduction of nematodes are influenced. The BxSCPI compound is used for preventing and controlling the pine wood nematodes, and provides a new technology and a new thought for biological prevention and control of the pine wood nematodes.
The application adopts BxSCPI compound to prevent and treat pine wood nematodes; proved by experiments, the compound ID is 6574212, 7990798, 6433663 and 7655389, has a certain inhibition effect on nematode growth and good insecticidal activity; becomes a potential medicine for preventing and treating pine wood nematodes and provides a new control means for preventing and treating pine wood nematodes.

Detailed Description
The conception and the technical effects produced by the present application will be clearly and completely described in conjunction with the embodiments below to fully understand the objects, features and effects of the present application. It is apparent that the described embodiments are only some embodiments of the present application, but not all embodiments, and that other embodiments obtained by those skilled in the art without inventive effort are within the scope of the present application based on the embodiments of the present application.
Example 1
1.1.1 Experimental materials
1) Test pine wood nematodes and botrytis cinerea strains: is provided by plant protection of Shanxi national institute.
2) Test compound: purchased from MCE company compound library and cambridge small molecule compound library, usa.
1.1.2 research methods
1) Nematode culture collection
Reference Huang Xiaojuan et al (2014) when Botrytis cinerea is cultivated on PDA culture medium for 5-6 days and fully grows on 9cm dish, pine wood nematodes are inoculated, and the pine wood nematodes are placed in a constant temperature incubator at 27 ℃ for cultivation for 7 days, and when the nematodes all enter a tolerance state period. The nematode liquid is eluted by sterile water and collected in a 15ml centrifuge tube, and centrifuged at 4000r/min for 1min, and the supernatant is removed. Washing with sterile water for 2-3 times, and diluting to prepare the nematode suspension with the solubility of 2000 strips/mL.
2) Preparation of the Compounds
Referring to the study method of day et al (2019), 10 compounds were dissolved separately using DMSO. First, each drug concentration is prepared into mother liquor 3×10 4 Mu M, then diluted to 1X 10 sequentially with sterile water 4 μM、1×10 3 mu.M, 300. Mu.M, 100. Mu.M and 50. Mu.M.
The information about 10 compounds is shown in Table 1.
TABLE 1
3) Determination of influence and lethal effect of medicine on growth of pine wood nematodes
a. In an ultra clean bench, 20mL of the preparation was added to a sterile disposable culture dish with a diameter of 9cmThe PDA culture medium is cooled and solidified, a puncher with the diameter of 6mm is adopted to inoculate a cultured Botrytis cinerea colony at the center of each plate, and the culture is carried out for 5 days at the temperature of 27 ℃ until hyphae are fully distributed on the plate. The mycelium surface was uniformly sprayed with 100. Mu.M and 1X 10 concentrations, respectively, using a 1 mL-volume micro-spray bottle 3 μM、1×10 4 Mu M drug solution was sprayed uniformly with 0.5mL of each dish. After standing for 10min, 0.5mL of nematode solution (1000) is inoculated in each dish, an equal volume of DMSO solution with the concentration of the same dilution multiple is used as a control, 3 dishes are arranged in each treatment, and an equal volume of DMSO with the same concentration is used as a control. Sealing the culture dish with a Parafilm sealing film, placing the culture dish in a 26 ℃ incubator for dark culture, and observing and counting the death number and the growth condition of nematodes under an optical microscope every 24 hours. As a result, as shown in FIG. 1, the concentration of the inoculating compound was 1X 10 4 Mu M, compound IDs 6574212, 7990798, 6433663 and 7655389 have a certain inhibition effect on nematode growth. Compared with the control, the nematode is observed under a microscope after 120 hours, the nematodes treated by the 4 drugs are all dead, and the nematode carcasses are digested. And the growth vigor of the nematodes after the other drugs are treated is the same as that of the control DMSO, the nematodes are propagated in a large quantity, and the dish cover is filled with nematodes of different ages.
b. The 4 compounds screened were further validated using sterile 24-well cell culture plates. 100. Mu.L of the above prepared nematode suspension was added to each well, and then 100. Mu.L of each of the drug solutions at a concentration of 100. Mu.M and 50. Mu.M in this order was added, respectively, with the DMSO solution at the same dilution concentration as the control. Each concentration was set with 6 replicates of 1 well (about 200 nematodes). The culture plates are sealed by Parafilm sealing films and then placed in a 26 ℃ incubator for dark culture, the death number and the survival number of nematodes are observed and counted under an optical microscope every 12 hours, and the mortality rate is calculated. As shown in figures 2 and 3, the nematodes die in a large amount after 96 hours of drug treatment, and the final nematode survival rate is lower than 10%, so that the nematoda prevention and treatment effect is very good.
The present application has been described in detail in the above embodiments, but the present application is not limited to the above examples, and various changes can be made within the knowledge of those skilled in the art without departing from the spirit of the present application. Furthermore, embodiments of the application and features of the embodiments may be combined with each other without conflict.
