Abstract
本发明涉及一种小鼠疾病造模方法,通过模拟经受类似车祸、地震等事故中的空间剧烈反转、震荡以及光线改变的过程,并对小鼠在上述环境下造模期间进行高脂饲料喂养,以此构建更加确切的小鼠模型,来模拟事故中人类经受眩晕、无助、空间剧烈震荡与反转过程后,导致精神状态改变的抑郁模型。本发明提供的构建事故后抑郁症小鼠疾病模型的方法,操作过程不复杂,利于操作,其中的变量易于控制,有较高的可操作性和可重复性,并且能够在较短时间内建成小鼠疾病模型,建模过程和得到的小鼠模型都具有良好的稳定性。
Description
技术领域
本发明涉及一种小鼠快速造模方法,通过模拟经受类似车祸、地震等事故中的剧烈空间反转、震荡、光线改变的过程,并对小鼠在造模期间进行高脂饲料喂养,以此来更加快速的构建抑郁症小鼠模型,并能够充分模拟事故中人类经受眩晕、无助、剧烈空间震荡与反转过程后,导致精神状态改变的抑郁模型。
背景技术
随着生活节奏加快,抑郁症日渐成为人类身心健康的严重威胁。抑郁症以情感低落、思维迟缓、意志活动减退为主要表现,是心境障碍的主要类型,重症抑郁有近15%的自杀率。在临床抑郁病例中,有较大一部分患者是由于经历了某种事故,比如交通意外后精神受到创伤导致的抑郁病症,因此,为了探索严重事故后抑郁症发病的机制,并针对性地开发治疗此种事故后抑郁症的药物,领域内迫切需要构建合适的动物模型。
动物模型能帮助人们更好的认识抑郁症,对于抑郁症的治疗和抗抑郁药的开发有重要的作用,主要的抑郁模型有以下几种:药物诱发抑郁模型、应激模型、获得性无助、慢性轻度不可预见性的应激等。近年来,通过改变周围环境使动物行为异常而建立的抑郁症模型,为抑郁症的研究拓宽了思路,常用的应激因子有:昼夜颠倒、禁食、禁水、垫料潮湿、行为束缚、强迫游泳、电击足底、高温环境、噪音干扰、陌生气味等等,将几种不同的应激刺激在实验中应用来完成抑郁症动物模型的构建。
现有的小鼠抑郁模型传统构建方法,主要是对小鼠施加持续的应激因子:电击、饥饿、束缚等严重刺激从而导致其抑郁,最常用的还有束缚法建模,而上述方法建模过程耗时长,效率低,极大的加长了科研周期,提高科研成本,且上述方法建模小鼠其发病机制与事故后抑郁明显不同,无法应用于探索严重事故后抑郁症发病的机制以及针对性地开发治疗此种事故后抑郁症的药物。
进一步地,在临床实践中我方研究者发现,较多的抑郁患者有高脂饮食的生活习惯,由此启发我们将高脂饲料和模拟事故发生现场的应激因子相结合来构造事故后抑郁症小鼠疾病模型。
因此,本发明是采取给予小鼠特定配方的高脂饲料结合将小鼠不定时地置于空间反转、震荡环境的方法,来构建事故后抑郁症小鼠疾病模型,实现小鼠模型的快速构建,并实现对严重事故后抑郁症发病机制的探索以及促进针对性地治疗此种事故后抑郁症的药物开发工作。
发明内容
本发明是采取给予小鼠特定配方的高脂饲料结合将小鼠不定时地置于空间反转和震荡环境中的方法,来构建事故后抑郁症小鼠疾病模型。
本发明提供一种快速构建事故后抑郁症小鼠疾病模型的方法,具体步骤包括:
(1)准备一定数量的雄性小鼠,筛选出快感缺失较明显的小鼠用于造模;
(2)对上述筛选后的小鼠以特定配比的高脂饲料进行饲养;
(3)将高脂饲料饲养后小鼠分组,分别置于统一规格的透明圆筒中;
(4)将放有小鼠的圆筒进行空间反转和震荡实验;
(5)验证小鼠模型的构建是否成功。
优选地,所述步骤(1)中的可以通过进行糖水偏好试验筛选出快感缺失较明显的小鼠;
优选地,所述步骤(2)中所述高脂饲料成分的质量配比为:基础饲料71.5%,猪油20%,蔗糖5%,胆固醇3%,胆盐0.5%;
优选地,所述步骤(2)中的高脂饲料饲养至少持续7天;
优选地,所述步骤(3)中,小鼠分配为1只/筒,保持透气,并将圆筒和小鼠处于强照射光源之下;

有益效果:
本发明提供的构建事故后抑郁症小鼠疾病模型的方法,操作过程不复杂,利于操作,其中的变量易于控制,有较高的可操作性和可重复性,并且能够在较短时间内建成小鼠抑郁症疾病模型,建模过程和得到的小鼠模型都具有良好的稳定性,极大地缩短了研究周期,提高了科研效率。
通过强光刺激、空间反转、震荡和闪烁光源处理,高度模仿了事故冲击中出现的应激因子,针对性地完成对人类事故后郁抑症疾病的小鼠模型建立,更加契合的模拟并提供了郁抑症疾病的外部成因,从而对于重大事故后抑郁症的发病机制、药物靶点探索和药物研发,具有重大的意义,在事故后抑郁症疾病研究领域,提供了有效的动物模型工具,为临床治疗铺设道路。

具体实施方式
下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1:
传统的小鼠抑郁症模型建立方法采用慢性束缚法构建小鼠抑郁模型,本申请采用上述方法作为对照组2。
实验原理:动物慢性应激能较真实的模拟出人类生活的应激情境以及事故中无法逃离的客观环境,慢性束缚应激是常见的慢性心理应激造模方法,常用于建立动物的抑郁样情绪类疾病模型。具体操作步骤:<1>小鼠的束缚装置由聚酯材料构成,透明可视,顶端和底部有多个直径1cm通气孔,有助于小鼠的呼吸畅通、排泄物的排出,避免对建模带来干扰;<2>试验开始后,在每天的10点至16点将小鼠放置于束缚器内(如附图1所示),用配套的塞托封闭开口并固定塞托,使固定后,动物在行为限制器内的空间长度小于其身长,并保证只有头部可以自由活动而四肢活动受限。如此限制小鼠活动6小时,期间禁食禁水;<3>连续束缚21天,分别在第7天、第14天和第21天进行强迫游泳试验,检验动物的抑郁程度。
一、实施例1实验步骤:
(1)选取30只SPF级雄性C57BL/6J小鼠,检疫结束后根据体重随机分为正常对照组1(6只)、束缚抑郁造模普通饲料组2(6只)、束缚抑郁造模高脂饲料对照组3(6只)、空间反转和震荡抑郁造模普通饲料组4(6只)、空间反转和震荡抑郁造模高脂饲料组5(6只);
(3)为了使建模效率更高,上述所述的30只小鼠,可通过进行糖水偏好试验筛选出快感缺失较明显得到,其实验原理实:抑郁症伴随多种行为表型,其中一个重要表现是快感缺失(Anhedonia),即个体无法从奖励或愉快的活动中体验到快乐。在动物模型实验中,通常用糖水偏好实验(Sucrose preference test,简称SPT)来测试快感缺失症状。啮齿类动物天生对甜食有强烈的欲望,在可自由选择分别含有蔗糖溶液和普通水的两个饮水装置时,会有选择地喝甜味的蔗糖溶液。然而,当啮齿类动物处于抑郁状态时,啮齿类动物不会倾向性去喝蔗糖溶液。因此检测动物对蔗糖溶液的偏好程度可作为评估动物快感缺失症状及抑郁程度的有用手段。具体操作步骤:<1>准备6-只生长状态一致的小鼠,试验开始前,小鼠单笼饲养,每个笼内放两瓶饮用水,让小鼠自由适应;<2>在试验第一天时,将其中一瓶饮用水换成1%的蔗糖水;<3>在试验第二天早上,将每笼的糖水和饮用水互换位置,并于当天晚上对小鼠进行禁食禁水(约8小时);<4>在试验第三天早上,每笼放置预先称量过的,已经确定体积的一瓶糖水和一瓶饮用水,让小鼠自由饮用约12h;<5>在试验第三天晚上,称取剩余糖水和饮用水的量,计算得到小鼠对糖水与饮用水的消耗量,按下列公式计算动物的糖水偏好指数:糖水偏好指数=蔗糖水/(饮用水+蔗糖水)×100%,对60只小鼠筛选得到糖水偏好指数较低的30只。
(3)对照组1不进行任何操作,将束缚抑郁造模高脂饲料对照组3、空间反转和震荡抑郁造模高脂饲料组5进行高脂饲料饲养7天,而后将束缚抑郁造模组2和3进行束缚实验,将空间反转和震荡实验组4和5组进行空间反转和震荡实验,高脂饲料成分的质量配比为:基础饲料71.5%,猪油20%,蔗糖5%,胆固醇3%,胆盐0.5%;
(4)步骤3中的空间反转和震荡实验步骤为:小鼠分配为1只/筒,保持透气,并将圆筒和小鼠处于强光照射之下;放有小鼠的圆筒在机械带动下剧烈震荡并反转,同时照射光源不断闪烁,持续时间为2分钟后停止震荡和反转,光线也不再闪烁,间隔t时间后圆筒再次剧烈震荡并反转伴随光源不断闪烁,30秒≤t≤30分钟,相邻两次的时间间隔保持不同(防止小鼠形成记忆),如此反复持续进行6小时,重复操作3天,如图3-4所示。
(5)验证实验:强迫游泳试验。
造模后7和14天时,进行验证。实验原理:“绝望”为抑郁症的明显特征,其行为表现是:在明显的外部不利环境中放弃挣扎。对于啮齿类动物,可利用其恐水的天性,将其放置于深度足以淹没动物的水中,正常小鼠在水中会表现出挣扎行为,而抑郁小鼠表现为静止不动的“绝望”行为,静止的时间长短可以反应小鼠的“绝望”程度。具体操作步骤:<1>试验开始前,将小鼠放置于将要开展试验的房间内适应1小时;<2>给强迫游泳装置的圆筒内加足水,水温控制在24±1℃;<3>设置并调试动物行为视频分析系统的摄像头及软件;<4>将小鼠放置于强迫游泳装置的圆筒内,采用动物行为视频分析系统检测小鼠在6min内的运动情况,记录后4min的不动时间。
二、实验结果(如图2所示):
(1)在造模后第7天,与正常对照组1相比,束缚抑郁造模普通饲料组2、束缚抑郁造模高脂饲料对照组3的小鼠不动时间无明显变化,而空间反转和震荡抑郁造模普通饲料组4、空间反转和震荡抑郁造模高脂饲料组5的小鼠不动时间均显著增加(P≤0.05),与空间反转和震荡抑郁造模普通饲料组4相比,空间反转和震荡抑郁造模高脂饲料组5的小鼠不动时间均显著增加(P≤0.05);
(2)在造模后第14天,与正常对照组1相比,束缚抑郁造模普通饲料组2的小鼠不动时间无明显变化,束缚抑郁造模高脂饲料对照组3的小鼠不动时间显著增加(P≤0.05),空间反转和震荡抑郁造模普通饲料组4、空间反转和震荡抑郁造模高脂饲料组5的小鼠不动时间均显著增加;与束缚抑郁造模普通饲料组2相比较,束缚抑郁造模高脂饲料对照组3的小鼠不动时间均显著增加(P≤0.05);与束缚抑郁造模普通饲料组2相比较,空间反转和震荡抑郁造模普通饲料组4的小鼠不动时间均显著增加(P≤0.05);与束缚抑郁造模高脂饲料组3相比较,空间反转和震荡抑郁造模高脂饲料组5的小鼠不动时间均显著增加(P≤0.05);与空间反转抑郁造模普通饲料组4相比,空间反转和震荡抑郁造模高脂饲料组5的小鼠不动时间均显著增加(P≤0.05);
三、实验结论:
(1)空间反转和震荡抑郁造模比束缚法造模的耗时少,效率高,能够快速建模;
(2)空间反转和震荡抑郁造模比束缚法造模得到的小鼠模型具有更为明显的抑郁症表现,是更好的疾病研究模型;
(3)高脂饲料喂养对抑郁小鼠的疾病模型制备具有促进作用;
(4)空间反转和震荡辅以高脂饲料喂养是更优的事故后小鼠抑郁症疾病模型建立方法。

1.本发明提供一种快速构建事故后抑郁症小鼠疾病模型的方法,具体步骤包括:
(1)准备一定数量的雄性小鼠,筛选出快感缺失较明显的小鼠用于造模;
(2)对上述筛选后的小鼠以特定配比的高脂饲料进行饲养;
(3)将高脂饲料饲养后小鼠分组,分别置于统一规格的透明圆筒中;
(4)将放有小鼠的圆筒进行空间反转和震荡实验;
(5)验证小鼠模型的构建是否成功。
