技术领域
本发明涉及动物疾病模型构建技术领域,具体涉及一种食蟹猴NASH模型的构建方法。
背景技术
非酒精性脂肪肝病(nonalcoholic fatty liver disease,简称NAFLD)是全球第一大慢性肝病,其中从单纯的脂肪肝进展为非酒精性脂肪性肝炎(non-alcoholicsteatohepatitis,简称NASH)是疾病进展、发展为肝细胞癌的关键步骤。当前对NASH疾病进展的特征认识还不够,针对NASH的药物研发除了经常遇到的靶点雷同的问题外,还面临着更多方面的挑战。
最大的挑战来源于药效模型种类很多,使用不同模型进行药效学实验造成在项目之间很难直观的相互比较,对项目质量评价带来很多模糊的空间,且各模型仅能在一定程度上模拟NASH的症状,但NASH模型背后的机理可能与人类特征相差十万八千里。
目前临床前药效的评价动物模型大多采用大鼠和小鼠的模型,从种属及基因等维度上,鼠和人的相近程度远远低于非人灵长类,进而鼠NASH模型的进展与人类特征相差甚远,导致研究鼠NASH的病理特诊与临床上人类NASH的病变特征相差较大,还有部分通过低剂量的酒精在非人灵长类上进行NASH造模,成模周期长,通常需三年之久。
发明内容
本发明的目的在于提供一种食蟹猴NASH模型的构建方法,以解决上述背景技术中提到的技术问题。
为了实现上述目的,本发明公开了一种食蟹猴NASH模型的构建方法,该构建方法包括:根据饲养标准对食蟹猴通过诱导饲料进行饲养,其中,诱导饲料为动物油和高胆固醇的混合物,对食用诱导饲料后的食蟹猴进行NASH症状检测,确认食蟹猴NASH模型构建成功。
优选地,诱导饲料通过烤制而成。
可选地,诱导饲料的能量为4600-4900Kcal/kg。
优选地,诱导饲料的能量为4789.5Kcal/kg。
进一步地,诱导饲料的营养成分包括蛋白质、脂肪和碳水化合物。
可选地,饲养标准包括每只食蟹猴食用的诱导饲料的量为190-210g/天。
可选地,将190-210g的诱导饲料分两顿。
可选地,该方法中还通过辅食和水对食蟹猴进行饲养,其中,辅食包括蔬菜和水果。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,下面通过结合附图和具体实施例对本发明方案进行详细阐述。应当理解,下述说明仅是为了解释本发明,并不对其内容进行限定。
1、动物信息
如下表1所示:

2、诱导饲料营养成分
诱导饲料为动物油和高胆固醇的混合物,其动物油如猪油,混合物经烤制再形成诱导饲料,其中,诱导饲料的能量为4789.5Kcal/kg,诱导饲料的营养成分包括蛋白质、脂肪和碳水化合物,如下表2所示:

3、建模动物饲养管理
上述实验用15只食蟹猴编号为01、02、…、15,根据饲养标准对15只食蟹猴通过上述诱导饲料进行饲养。
环境:为食蟹猴提供动物房,动物房的室温控制在18-26℃、湿度控制在40-70%,每只食蟹猴独用一个不锈钢笼具,笼具规格的长×宽×高为90×90×90cm,且动物房和笼具每天清洗1次,笼具每两周更换一次,动物房的地面和墙面每2周消毒1次。
饲养标准:除试验需要禁食外,食蟹猴每日自由饮水,每只食蟹猴食用的诱导饲料的量为200g,并将200g诱导饲料分为上午和下午两顿投喂,在下午还可通过辅食和水对食蟹猴进行饲养,其辅食可以是适量的新鲜蔬菜或水果,共计诱导饲料的食用周期为50周/只,另外,还可定时对食蟹猴摄入的诱导饲料的量进行监控,如每24小时称量诱导饲料的剩余量。
4、NASH症状检测
对食用诱导饲料后的食蟹猴进行NASH症状检测,NASH症状检测的方法使用体重、血液、磁共振成像、肝脏活检和计算机断层扫描,以监控建模过程。
检测方法1、体重
需对每只食蟹猴每周检测一次。
检测方法2、血液
需对每只食蟹猴每周检测一次,利用奥林巴斯AU640设备对食蟹猴血液生化中的肝功能指标进行检测,即对谷丙转氨酶(ALT)、谷草转氨酶(AST)、胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-c)和低密度脂蛋白(LDL-c)进行检测,以判断NASH进展。
检查方法3、磁共振成像
需对每只食蟹猴每三个月检测一次,利用磁共振进行肝脏脂肪密度(PDFF)检测,该方法是一种无创检测技术,可以随时对食蟹猴进行检测确认NASH症状的进展,初步了解整个肝脏的脂肪含量和分布情况。
检测方法4、肝脏活检
需对每只食蟹猴在超声波引导下每四周检测一次,与检测方法3不同,肝脏活检是一种有创的检测技术,根据检测方法1反应的肝脏情况,再通过肝脏活检可以更加深入的了解肝脏组织的形态学的改变。
检测方法5、计算机断层扫描
需对每只食蟹猴每三个月检测一次,计算机断层扫描结果可以直观的反应肝脏的致密度,NASH早期扫描的致密度较低,随着NASH的进展,如肝脏组织中纤维化的产生,NASH后期扫描的致密度升高。
5、建模检测数据
参与试验的15只食蟹猴中,结合血液、磁共振成像、计算机断层扫描和肝脏活检的检测结果,可观察在不同饲养周期时的NASH进展,最终确认通过本发明方法构建的食蟹猴NASH模型均构建成功。
另外,为了与建模用15只食蟹猴形成对比,每次检测还对照了10只健康的食蟹猴作为正常组。
1)血液检测:对通过本发明方法饲养的15只食蟹猴作为模型组,进行肝功能指标检测,且将未进行NASH模型构建的10只食蟹猴作为正常组,对比在第0、10、16、30、50周时模型组和正常组肝功能指标变化情况,检测得到的模型组(图中标记为:model)和正常组(图中标记为:sham)的肝功能指标中ALT平均值、AST平均值、TG平均值、TC平均值、HDL-c平均值、LDL-c平均值如图1所示。
结论:由图1中各项平均值(平均值已加减标准误)可见,在第10周开始,模型组的肝功能的各项指标均显著高于正常组,到第50周时,通过本发明方法构建的食蟹猴NASH模型构建成功。
2)磁共振成像检测:对通过本发明方法饲养的15只食蟹猴作为模型组,进行PDFF检测,且将未进行NASH模型构建的10只食蟹猴作为正常组,对比在第24周时模型组和正常组PDFF变化情况,检测得到的正常组(图中标记为:naive)和模型组(图中标记为:week 24)成像参照图2所示,相应的根据图2中脂肪的密度计算得出的PDFF平均值如图3所示。
结论:由图3中PDFF平均值(平均值已加减标准误)可见在24周时模型组与正常组相比PDFF显著性增高,且相比正常组,模型组在24周时PDFF增加了23%,而正常组并无明显增加,此时模型组处于NASH进展期,通过本发明方法构建的食蟹猴NASH模型构建成功。
3)计算机断层扫描检测:对通过本发明方法饲养的15只食蟹猴作为模型组,进行CT值检测,且将未进行NASH模型构建的10只食蟹猴作为正常组,对比在第24周时模型组和正常组CT变化情况,检测得到的正常组(图中标记为:naive)和模型组(图中标记为:model-24wks)成像参照图4所示,相应的根据图4中CT扫描图计算得到的CT平均值如图5所示。
结论:由图4中CT平均值(平均值已加减标准误)可见,在第24周时,与正常组相比模型组肝脏硬度降低,此时模型组处于NASH进展期,通过本发明方法构建的食蟹猴NASH模型构建成功。
4)肝脏活检检测:对通过本发明方法饲养的15只食蟹猴作为模型组,进行肝脏活检检测,对比第10、16、30、50周(图中标记为week-10、week-16、week-30、week-50)的肝脏活检变化情况,通过HE染色和天狼星红染色成像分别参照附图6和7所示。
结论:通过图6看到,各时间段均可见大量的脂肪沉积在肝组织中,在第10周时已出现肝细胞气球样变的发生,随着饲养时间越久,可见到有少量炎细胞浸润产生,通过图7看到,在第10周时,肝细胞索之间已有极少量的纤维化产生,随着饲养时间越久,纤维化的生成变多,且在第50周时已经可以明显看到玫红色阳性染色的较远纤维沉积,通过本发明方法构建的食蟹猴NASH模型构建成功。
通过本发明食蟹猴NASH模型的构建方法,相比使用兔子、小鼠、仓鼠等,不仅模型更与人类特征最接近,而且饲养周期适宜,方便在不同饲养周期随时检测,从而了解NASH相关的各项数值变化情况,更容易对NASH的进展进行研究。

1.一种食蟹猴NASH模型的构建方法,其特征在于,所述构建方法包括:
根据饲养标准对所述食蟹猴通过诱导饲料进行饲养,其中,所述诱导饲料为动物油和高胆固醇的混合物;
对食用所述诱导饲料后的所述食蟹猴进行NASH症状检测,确认食蟹猴NASH模型构建成功。
2.如权利要求1所述的构建方法,其特征在于,所述诱导饲料的能量为4600-4900Kcal/kg。
3.如权利要求2所述的构建方法,其特征在于,所述诱导饲料的营养成分包括蛋白质、脂肪和碳水化合物。
4.如权利要求1所述的构建方法,其特征在于,所述饲养标准包括每只所述食蟹猴食用的所述诱导饲料的量为190-210g/天。
5.如权利要求4所述的构建方法,其特征在于,将190-210g的所述诱导饲料分两顿。
6.如权利要求1所述的构建方法,其特征在于,还通过辅食和水对所述食蟹猴进行饲养,其中,所述辅食包括蔬菜和水果。
7.如权利要求1所述的构建方法,其特征在于,所述食蟹猴的体重为2-2.5kg。
