胶质母细胞瘤(GBM)侵袭性强,致死率极高,五年生存率低于5%。目前,胶质母细胞瘤的主要治疗方法是手术切除、放疗和化疗。以替莫唑胺(一种烷化剂)为代表的传统化疗药物可以有效地穿过血脑屏障(BBB),以增强胶质母细胞瘤的治疗效果,在替莫唑酰胺化疗过程中加入低强度交变电场,可以显著提高胶质母细胞瘤患者的生存率。尽管这些改善令人鼓舞,但胶质母细胞瘤患者的长期生存率仍然极低。另外,化疗和放疗的全身毒性副作用也限制了其临床应用。胶质母细胞瘤的难治性主要是由于遗传异质性、侵袭性和浸润性肿瘤生长以及免疫抑制肿瘤微环境(TME)等。血脑屏障和胶质瘤干细胞的存在也增加了胶质母细胞瘤对化疗和放疗的抵抗力。为了提高治疗效果,迫切需要开发新的治疗方法来对抗胶质母细胞瘤。
近年来,基于肿瘤组织局部热损伤的光热疗法(PTT)逐渐发展成为临床肿瘤治疗的最佳选择之一。光热疗法具有高选择性和非侵入性的优点,它通过光热转换剂(PCAs)将吸收的光能转换为热量,并导致肿瘤组织局部过热,从而引发肿瘤细胞死亡。然而,光热疗法在治疗胶质母细胞瘤中的临床应用需要解决以下问题:现有技术中的光热转换剂虽然具有良好的光热转换效率,但是其生物相容性欠佳,易对生物体产生毒性作用;光热转换剂需要穿过血脑屏障,并在胶质母细胞瘤组织中积聚,才能增强光热疗法的治疗效果,但是现有技术中的大部分光热转换剂难以穿过血脑屏障充分实现治疗效果;采用光热疗法治疗肿瘤后,容易发生肿瘤复发和转移。因此,亟需研发一种能够穿过血脑屏障的、可在胶质母细胞瘤组织中大量积聚的、生物相容性好的、可以减少肿瘤复发和转移的几率的、用于光热疗法的纳米药物传递系统,以提高治疗效果,进而提高胶质母细胞瘤患者的生存率。
发明内容
本发明意在提供一种异黑素类仿生纳米粒,以解决现有技术中的缺少针对胶质母细胞瘤的可穿越血脑屏障的、生物相容性好的、可以减少肿瘤复发和转移的几率的、基于光热疗法的纳米粒的技术问题。
为达到上述目的,本发明采用如下技术方案:
一种异黑素类仿生纳米粒,包括异丙黑素纳米粒和癌细胞膜,所述异丙黑素纳米粒上负载有PD-L1小分子短肽抑制剂。
本方案还提供了一种异黑素类仿生纳米粒的制备方法,包括以下依次进行的步骤:
S1:获取胶质母细胞瘤细胞膜;
S2:将异丙黑素纳米粒和CLP002混合搅拌,获得负载有CLP002的异丙黑素纳米粒;
S3:将负载有CLP002的异丙黑素纳米粒分散在含有胶质母细胞瘤细胞膜的PBS溶液中,获得分散液;再经超声处理获得异黑素类仿生纳米粒。
本方案还提供了一种异黑素类仿生纳米粒在制备治疗胶质母细胞瘤的药物中的应用。
本技术方案的原理以及有益效果在于:
免疫检查点阻断疗法在各种肿瘤的治疗中显示出巨大的潜力,但其对胶质母细胞瘤(GBM)的治疗效果却不尽如人意。由于免疫原性低、T细胞浸润不佳以及血脑屏障(BBB)的存在,使免疫检查点阻断疗法相关试剂或药物难以到达胶质母细胞瘤组织。针对上述问题,本技术方案开发了一种仿生纳米治疗平台(AMNP@CLP@CCM)用于胶质母细胞瘤靶向的光热疗法(PTT)和免疫检查点阻断疗法(Immune checkpoint blockade therapy,ICB)的协同治疗。
本技术方案将免疫检查点抑制剂(PD-L1小分子短肽抑制剂,CLP002)加载到异丙黑素纳米粒(AMNPs)中,然后在异丙黑素纳米粒外涂覆癌细胞膜(CCM)。由于癌细胞膜的归巢效应,AMNP@CLP@CCM可成功穿过血脑屏障并将CLP002输送至胶质母细胞瘤组织。作为一种天然光热转换剂,异丙黑素纳米粒可用于肿瘤光热疗法。光热疗法增加的局部温度不仅增强了血脑屏障穿透,而且上调了胶质母细胞瘤细胞上的PD-L1水平。重要的是,光热疗法可以有效刺激免疫原性细胞死亡,以诱导肿瘤相关抗原暴露并促进T淋巴细胞浸润,这可以进一步放大胶质母细胞瘤细胞对CLP002介导的免疫检查点阻断疗法的抗肿瘤免疫反应,使得原位胶质母细胞瘤出现了显著生长抑制现象。因此,AMNP@CLP@CCM在光热疗法和免疫检查点阻断疗法协同治疗原位胶质母细胞瘤中显示出了巨大的潜力。
另一方面,肿瘤复发和转移是光热疗法疗效不佳的主要原因。肿瘤免疫疗法可以通过激活宿主的免疫系统来有效杀死残留和转移的肿瘤细胞,进而克服上述肿瘤复发和转移的问题。在本技术方案中,将CLP002多肽整合到异黑色素上,CLP002是一种PD-L1的抑制剂,可以阻止PD-L1和PD-1的结合,进而提高T细胞活性以及数量,通过激活宿主的免疫系统来有效避免肿瘤转移。因此,光热疗法和免疫检查点阻断疗法协同治疗可以有效治疗肿瘤,并避免肿瘤复发和转移。
综上所述,本技术方案通过1,8-二羟基萘(1,8-DHN)的氧化聚合合成了黑色素类似物AMNP,并用作光热转换剂,同时负载CLP002,构建获得纳米载体。CLP002肽是一种低分子量抗PD-L1抑制剂,通过阻断PD-1和PD-L1之间的相互作用,进而用于免疫检查点阻断疗法治疗,将GL261(小鼠胶质母细胞瘤细胞)细胞膜涂覆在纳米平台的表面,以模拟源细胞的高度复杂功能。数据表明AMNP@CLP@CCM仿生纳米治疗平台具有很强的血脑屏障穿透和归巢能力,通过光热疗法和免疫检查点阻断疗法协同治疗可以有效抑制原位胶质母细胞瘤的生长,从而显示出原位胶质母细胞瘤治疗的前景。
进一步,所述癌细胞膜来自胶质母细胞瘤细胞。
进一步,聚所述PD-L1小分子短肽抑制剂为CLP002。
进一步,所述异丙黑素纳米粒由如下方法制备:1,8-二羟基萘溶于乙腈,然后加入水搅拌,再加入高碘酸钠溶液,再经搅拌、离心和洗涤,获得异黑素核心。
进一步,1,8-二羟基萘、乙腈、水和高碘酸钠溶液的用量比为150mg:7.5mL:138.5mL:4mL;高碘酸钠溶液的浓度为0.25M。
进一步,异丙黑素纳米粒和CLP002的质量比为10:5。
进一步,在S3中,所述分散液中的异丙黑素纳米粒的浓度为1mg/mL。
进一步,在S1中,胶质母细胞瘤细胞膜溶于PBS缓冲液中,每1.5×107个细胞中提取的胶质母细胞瘤细胞膜使用1mLPBS缓冲液溶解。
综上所述,发明人通过将CLP002加载到AMNPs中,然后在其表面涂覆GL261细胞膜以形成AMNP@CLP@CCM纳米粒。该纳米粒可以穿过血脑屏障,靶向原位胶质母细胞瘤,并实现光热治疗和免疫检查点阻断疗法的协同作用。在该纳米平台中,AMNPs既用作光热治疗的生物衍生光热转换剂,也用作CLP002递送的纳米载体,而CLP002肽通过阻断PD-1和PD-L1之间的相互作用而用于免疫检查点阻断疗法治疗。AMNP@CLP@CCM显著提高了纳米粒的血脑屏障穿透能力和肿瘤靶向能力,这一点通过体外transwell和体内靶向实验得到证实。体外和体内实验均表明,AMNPs具有强烈的光热消融能力来诱导肿瘤细胞凋亡和免疫性细胞死亡(ICD),CLP002多肽在阻断PD-L1方面具有良好的生物活性。重要的是光热治疗和免疫检查点阻断疗法的结合显著增强了T细胞的免疫应答,改善了胶质母细胞瘤的免疫抑制微环境,从而提高原位胶质母细胞瘤抑制作用。

具体实施方式
下面结合实施例对本发明做进一步详细的说明,但本发明的实施方式不限于此。若未特别指明,下述实施例以及实验例所用的技术手段为本领域技术人员所熟知的常规手段,且所用的材料、试剂等,均可从商业途径得到。
实施例1:纳米粒的合成以及性能表征
(1)细胞膜的提取
对GL261/HepG2/4T1/CT26细胞进行常规培养,然后用0.25%胰酶消化,然后900g离心5min。使用预冷的PBS缓冲液(pH=7.4)重悬细胞,再600g离心6min。收集细胞沉淀,使用低渗裂解缓冲液重悬细胞,低渗裂解缓冲液中含有蛋白质提取试剂和苯甲磺酰氟(PMSF)(Beyotine Institute of Biotechnology)。然后,在冰浴条件下孵育10-15min。接下来使用反复冻融的方法,在上述缓冲液中将细胞破碎。然后,4℃下700g离心10min,取上清液。上清液14000g离心30min,收集细胞膜。然后将收集的细胞膜溶于含有PMSF的PBS缓冲液中,储存于-20℃备用,1.5×107个细胞对应1mLPBS缓冲液。
GL261-Luc(表达荧光素酶的小鼠胶质母细胞瘤细胞),GL261细胞,Raw264.7小鼠巨噬细胞,bEnd.3小鼠脑微血管内皮细胞,CT26小鼠直肠癌细胞,HepG2人肝细胞癌细胞均使用高糖DEME培养基培养,其中,含有10%FBS,1%双抗和2mM L-谷氨酰胺,培养环境为37℃、5%CO2。4T1小鼠乳腺癌细胞使用1064培养,培养基中含有10%FBS和1%双抗,培养环境为37℃、5%CO2。
(2)纳米粒的合成
AMNP和AMNP@CCM按照如下方法合成:
将1,8-二羟基萘(1,8-Dihydroxynaphthalene,1,8-DHN,150mg)分散在乙腈(CH3CN,7.5mL)中,直至完全溶解,然后加入138.5mL超纯水并持续搅拌5min。然后快速加入0.25MNaIO4(4mL),持续搅拌20h,接下来12000g离心30min,并用超纯水洗三次,获得AMNPs。
将AMNPs分散在100μL的细胞膜的PBS溶液中,AMNPs的终浓度为1mg/mL。上述混合物在冰上进行超声处理30min,然后在4℃条件下洗涤并离心,获得AMNP@CCM。
AMNP@CLP按照如下方法合成:
将10mgAMNPs和5mgCLP002(WHRSYYTWNLNT,Xi'an ruixi)在4mL系统中混合并持续搅拌24h。通过离心收集上清液,检测和计算CLP002的载药量。负载到AMNPs上的CLP002通过280nm紫外光谱仪测量。载药量的计算方法为:
载药量(DL%)=纳米粒中的药物质量(Weight of drug in micelles)/材料和药物的重量(Weight of materials and drugs)×100%。
AMNP@Cy5.5的制备方法同上,只是使用2mg Cy5.5(WHRSYYTWNLNT,Xi'an ruixi)代替CLP002。
AMNP@CLP@CCM和AMNP@Cy5.5@CCM的制备方法同AMNP@CCM的制备方法,只是使用AMNP@CLP或者AMNP@Cy5.5代替AMNP。
(3)纳米粒的表征
AMNPs的透射电镜(TEM)图像详见图1a,AMNP@CCM的TEM图像详见图1b,两种纳米粒均为球形结构,并可以看到AMNP@CCM外层的癌细胞膜。AMNPs(168nm)和AMNP@CCM(216nm)的粒径分布图详见图1c,由AMNPs的35nm粒径变为了AMNP@CCM的45nm粒径,说明细胞膜的负载成功。图1d展示了AMNPs(-32.5±0.71mV),CLP((22.13±0.7mV),CCM(-12.31±1.76mV),AMNP@CLP(13.53±0.2mV),AMNP@CLP@CCM(-19.46±0.81mV)的Zeta电位。图1e展示了AMNP,AMNP@CLP和CLP的红外光谱。AMNPs在3429cm-1有明显的O-H键的吸收峰,在1630cm-1有明显的C=C键的吸收峰。在负载CLP002后,AMNP@CLP展示了CLP002在1513cm-1和670cm-1的吸收峰,分别对应-COO-和N-H。结果显示CLP002已被成功负载。
为了确认光热效应以及药物释放特性,进行了如下实验。图1f展示了AMNPs溶液的在不同浓度下的升温曲线(808nm激光辐射,1W/cm2,10min)。图1g展示了100μg/mL AMNPs溶液在不同激光强度下的温度变化情况。图1h展示了100μg/mL AMNPs溶液在0.5W/cm2加热和自然冷却条件下的五个循环的温度变化情况。上述实验证明了AMNPs的光热转化效力以及稳定性。
实施例2:体外免疫逃逸、靶向性以及穿透血脑屏障效果的实验研究
GL261-Luc细胞和Raw264.7细胞分别以5×104个/皿的密度接种到培养皿中,常规培养24h。然后,加入AMNP@Cy5.5和AMNP@Cy5.5@CCM(GL261细胞膜)孵育2h,再用PBS清洗细胞三次,使用4%多聚甲醛固定,DAPI染色展示细胞核,激光共聚焦观测细胞对纳米粒的摄入量,并使用流式细胞术观测细胞对纳米粒的摄入情况。实验流程参见图2a,激光共聚焦实验结果参见图2b(红色:Cy5.5;蓝色:DAPI;比例尺:25μm),流式细胞术实验结果参见图2c。Cy5.5用于标记和追踪AMNP@CCM NPs,实验结果说明GL261细胞膜的使用可以逃逸免疫细胞的识别,但是可以被GL261-Luc细胞大量摄入。
制备由不同细胞膜包裹AMNP的纳米粒,细胞膜的来源包括GL261细胞、CT26细胞、4T1细胞、HepG2细胞(AMNP@HepG2,AMNP@4T1,AMNP@CT26,AMNP@GL261)。大致实验过程为:将GL261细胞按照1×105个细胞每孔接种在6孔板中,常规培养24h,然后将培养基换成新鲜的含有AMNP@Cy5.5@CCM的培养基,再共培养4h,培养完成后用PBS洗细胞,使用4%多聚甲醛固定,DAPI染色展示细胞核,进行激光共聚焦观察。实验结果参见图2d(比例尺:25μm)。实验结果说明AMNP@GL261显示出特异性靶向能力。
为了研究纳米粒穿透血脑屏障的能力,将bEnd.3细胞(6.7×104个/孔)接种在transwell实验上方腔室中,培养7天形成单层细胞。监测单层细胞的完整性,直至bEnd.3单层细胞的TEER值(transendothelial electrical resistance)超过150Ω·cm2。将GL261细胞(6.7×104个/孔)接种在transwell实验下方腔室中,培养24h。将AMNP@Cy5.5@CCM(GL261细胞膜、CT26细胞膜、4T1细胞膜、HepG2细胞膜)放入上方腔室中,共培养8h后,取下方腔室的细胞进行流式细胞术检测。实验流程参见图2e,实验结果(流式细胞术分析被GL261细胞摄入的纳米粒情况)参见图2f(Mean±s.d.,n=3)。AMNP@Cy5.5@CCM(GL261细胞膜)展示出了最高的摄入量,是其他组别的3.88到4.27倍,说明在AMNPs外包裹GL261细胞膜,有助于纳米粒跨越血脑屏障,并靶向目的癌细胞。
实施例3:体外光毒性实验、光热效应引起的凋亡和ICD现象研究以及纳米粒与PD-L1的结合能力的研究
(1)体外光毒性
将bEnd.3细胞和Raw264.7细胞(1×104个细胞每100μL培养基每孔)接种在96孔板中,培养24h后,加入不同浓度的AMNP@CLP@CCM,其中AMNPs的浓度为50μg/mL-800μg/mL。完成孵育之后,使用CCK-8法检测细胞存活率。每组实验细胞存活率均在90%以上,说明细胞毒性小,生物相容性好(实验结果未展示)。
GL261细胞(1×104个细胞每100μL培养基每孔)接种在96孔板中,培养24h后,加入不同浓度的AMNPs和AMNP@CCM孵育4h,其中AMNPs的浓度为50μg/mL-800μg/mL。然后,使用无药的培养基洗细胞,再使用(或不使用)808nm激光处理(0.75W/cm2,5min),再培养24h,使用CCK-8法检测细胞存活率,实验结果参见图3a。实验结果说明,癌细胞膜的包覆可以提升GL261细胞杀伤效果。
将GL261细胞接种在共聚焦培养皿中(5×104个细胞每皿),培养24h,用PBS洗涤,然后加入AMNPs和AMNP@CCM。其中,AMNPs的浓度为200μg/mL,孵育4h。然后使用808nm激光处理(0.75W/cm2,5min),再培养4h,calcein AM/PI染色后,进行荧光共聚焦显微观察,实验结果显示癌细胞膜的包覆可以提升GL261细胞杀伤效果,且激光处理可以提升治疗效果(图像未展示)。
(2)光热效应引起的凋亡和ICD现象(immunogenic cell death)
使用流式细胞术评价光热效应带来的细胞凋亡情况。GL261细胞(1×105个细胞每孔)接种在6孔板中,培养24h。然后加入AMNPs和AMNP@CCM(AMNP浓度200μg/mL),孵育4h,接下来更换新鲜培养基,进行(或者不进行)808nm激光处理(0.75W/cm2,5min),接着孵育4h。然后收集细胞,加入Annexin V、FITC和PI,进行流式细胞术检测。实验结果参见图3b。实验结果显示,AMNP@CCM的细胞凋亡百分比为81.4%,远高于AMNP组(65.9%)和对照组(1.27%),表明AMNP@CCM可以有效地引起肿瘤组织中的癌细胞凋亡。
GL261细胞(1×105个细胞每孔)接种在6孔板中,培养24h。然后加入AMNPs和AMNP@CCM(AMNP浓度200μg/mL),孵育4h,接下来更换新鲜培养基进行(或者不进行)808nm激光处理(0.75W/cm2,5min),接着孵育6h。接下来收集细胞,使用PE anti mouse CRT抗体(Biolegend,USA)处理,在4℃条件下孵育20min,再进行流式细胞术和激光共聚焦分析。GL261细胞(5×104个细胞每孔)接种在激光共聚焦培养皿中,培养24h。然后加入AMNPs和AMNP@CCM。接下来更换新鲜培养基进行(或者不进行)808nm激光处理(0.75W/cm2,5min),接着孵育12h。固定细胞,使用Alexa647anti-HMGB1抗体(Biolegend,USA)染色,并进行DAPI染色,染色后进行激光共聚焦观察。使用激光共聚焦观察结果如图3c所示(比例尺:25μm),GL261细胞与AMNP和AMNP@CCM共孵育后,在808nm激光照射下,细胞膜上呈现出强烈的绿色荧光(CRT,calreticulin),细胞质中呈现出丰富的红色荧光(HMGB1,high-mobilitygroup box 1)。通过流式细胞术进一步验证CRT表达水平,GL261细胞用AMNP@CCM处理,并用808nm激光照射,释放出最显著水平的CRT,详见图3e。使用现有技术的试剂盒测量ATP的释放情况,采用上述培养和处理方式,进一步测量了各种处理情况下,细胞ATP的释放情况,实验数据详见图3d(mean±s.d.,n=3)。AMNP和AMNP@CCM处理后,在808nm激光照射下,GL261细胞ATP分泌明显增强,其中以AMNP@CCM组的作用效果最强。
结果表明,AMNP可以在808nm激光照射下有效地触发GL261细胞的ICD效应。AMNP@CCM具有针对GL261细胞的优越靶向能力,从而产生更好的肿瘤消融能力,并改善对“冷”肿瘤的免疫应答。此外,在光热处理后,孵育24小时,使用免疫荧光检测癌细胞上PD-L1的表达,结果如图3f所示(比例尺:100μm),PD-L1的表达通过“光热效应”上调,表明光热效应可能提升肿瘤细胞对免疫检查点阻断疗法的敏感性。此外,当CLP002与肿瘤细胞共培养时,在光热效应作用下,几乎没有发现红色荧光,这表明CLP002可以有效阻断PD-L1。
(3)纳米粒与PD-L1的结合能力
将GL261细胞接种在6孔培养板(1×105个细胞每孔)中过夜培养。然后加入干扰素-γ(IFN-γ,20nM)以刺激PD-L1在肿瘤细胞表面的过表达。在24小时后,使用FBS收集细胞并阻断。将细胞悬液分为四组PBS1(未染色,阴性对照),PBS2(用PE标记的PD-L1染色,用作阳性对照表明肿瘤细胞膜上PD-L1表达的总量),AMNP@CLP@CCM(pH6.0)和AMNP@CLP@CCM(pH6.0,激光功率0.75W/cm2,5min)。然后,所有细胞用PBS洗涤两次,使用PE标记的PD-L1(CD274)抗体染色20min。最后细胞经过洗涤收集之后,使用流式细胞术进行检测,抗体的结合能力与荧光强度呈负相关。由于CLP002通过阻断PD-L1免疫检查点发挥作用,药物本身对肿瘤细胞没有任何毒性作用。接下来,发明人测量了从AMNP@CLP@CCM上释放的CLP002对GL261细胞膜上的PD-L1的亲和性,实验结果如图3g和图3h(mean±s.d.,n=3)所示。γ干扰素刺激后,PD-L1在GL261细胞膜上表达率为93.63%。使用AMNP@CLP@CCM处理后,PD-L1在细胞膜上的表达率降低至80.86%。808nm激光照射进一步增强了阻断效应(21.74%),表明AMNP@CLP@CCM能够抑制PD-1/PD-L1通路。
为了进一步研究光热疗法对CLP002活性的影响,使用不同强度的808nm激光照射AMNP@CLP@CCM持续5min(0.5W/cm2,0.75W/cm2,1.5W/cm2)。纳米粒与肿瘤细胞孵育后,清洗所有样品,并用PE标记的PD-L1抗体染色20min,使用流式细胞术检测荧光强度。为了研究激光照射对PD-L1与CLP002结合能力的影响,通过流式细胞术测量不同照射功率处理后PD-L1在细胞表面的结合力。结果表明,激光照射强度对CLP002小分子阻断剂的生物活性没有显著影响,即使功率增加到1.5W/cm2。
实施例4:关于纳米粒穿透血脑屏障和在肿瘤组织中聚集的能力的体内实验研究
(1)动物模型
雄性C57小鼠(6-8周)购自Enswell公司,并按照SPF条件饲养。使用处于对数生长期的GL261-Luc细胞配置细胞悬液,浓度为1×105个/mL。将GL261 Luc细胞(5μL)通过微型注射器缓慢接种到C57小鼠的右侧纹状体中(横向:2.0mm,纵向:1.0mm,深度:3.5mm)。使用IVIS图像观测肿瘤生长情况。
(2)纳米粒在小鼠体内分布情况
尾静脉注射AMNP@Cy5.5和AMNP@Cy5.5@CCM(AMNP浓度2mg/mL,200μL)到荷瘤小鼠体内,拍摄实时荧光图像。在评估靶向能力之前,使用生物发光(Luc)和MRI(T1)检查肿瘤大小和位置的一致性。实验结果参见图4a:荧光信号随时间逐渐增加,并在12h达到最大值,AMNP@Cy5.5@CCM组比AMNP@Cy5.5组的靶向性更为优异。
此外,大脑和主要器官的离体荧光图像如图4b所示(注射后12h),其中AMNP@Cy5.5@CCM组在大脑中的荧光强度是对照组AMNP@Cy5.5组的2.9倍(图4c)。使用荧光共聚焦检测两组的冷冻肿瘤组织切片,以进一步研究纳米粒在肿瘤部位的靶向能力和积聚情况。图4d展示了AMNP@CCM的在荷瘤小鼠体内PTT正位图像。AMNP@Cy5.5@CCM具有优异的靶向神经胶质母细胞瘤能力,很少渗入正常脑组织(图4f,细胞核:DAPI;两组纳米粒均显示Cy5.5荧光;比例尺:250μm,虚线表示胶质瘤和正常脑组织的界限)。这些结果表明AMNP@Cy5.5@CCM可以穿过血脑屏障并选择性地聚集在肿瘤部位。图4g为AMNP@Cy5.5和AMNP@Cy5.5@CCM的扩散深度量化分析(从正常组织到深层肿瘤组织,Image J软件),证明癌细胞膜的添加可以增加纳米粒入侵肿瘤组织的深度。图4h为AMNP@Cy5.5@CCM浸入肿瘤组织的2.5D图像(Image J软件)。
(3)体内光热效应
为了研究体内光热效应对胶质母细胞瘤原位植入肿瘤模型的影响,将荷瘤小鼠分为三组:PBS,AMNPs和AMNP@CCM(AMNP浓度2mg/mL,200μL),静脉注后12小时,用808nm激光(1W/cm2)从一个小的颅骨窗口对肿瘤组织进行处理,监测局部肿瘤温度,实验结果参见4d和4e。在AMNP@CCM组,激光照射5min后,肿瘤部位温度达到48.2℃;对于AMNPs组,肿瘤部位温度轻度升高至43.7℃。实验结果再次表明癌细胞膜赋予AMNP@CCM具有优异的胶质母细胞瘤靶向性。
实施例5:AMNP@CLP@CCM的PTT和ICB的协同作用的体内实验研究
参照实施例4建立动物模型,在第7天进行体内成像以确认肿瘤形成,然后将荷瘤小鼠随机分为5组(n=5):(1)对照组(PBS处理);(2)AMNP@CCM;(3)AMNP@CLP@CCM;(4)AMNP@CCM+L;(5)AMNP@CLP@CCM+L。在上述各组中,AMNP剂量均为2mg/mL、200μL。在第1、3和5天,给小鼠静脉注射治疗。注射12小时后,使用808nm激光照射AMNP@CCM+L组和AMNP@CLP@CCM+L组在1W/cm2下持续5分钟,激光束的光斑根据小鼠的头骨窗口进行调整。在第5、12、19和26天,对小鼠生长情况进行检测,操作流程参见图5a。试验结束后,处死小鼠,取主要器官(心脏、肝脏、脾脏、肺和肾脏)和脑胶质瘤组织进行H&E染色。
图5b为不同治疗后不同时间点荷瘤小鼠的生物发光图像,图5c为不同治疗后荷瘤小鼠脑部的H&E染色图像。图5d为不同处理下生物发光图像图5b中的量化的荧光强度。图5e为不同处理下荷瘤小鼠的体重变化曲线,不同处理并没有造成各组的小鼠体重差异。图5f为通过荧光共聚焦检测的不同处理下胶质瘤组织的细胞凋亡情况(蓝色:细胞核;红色:凋亡细胞;比例尺:100μm)。
在治疗过程中,PBS组和AMNP@CCM组中,肿瘤体积增加明显;而在AMNP@CLP@CCM组和AMNP@CCM+L组中,肿瘤体积增长被轻微抑制;AMNP@CLP@CCM+L组中,肿瘤体积增长被显著抑制(图5b和图5d)。通过TUNEL的免疫荧光染色进一步检测肿瘤细胞的凋亡,AMNP@CLP@CCM+L组的细胞凋亡水平最高。
实施例6:光热免疫疗法介导的免疫应答
为了探索抗肿瘤免疫反应,将荷瘤小被随机分为如实施例5的五组(n=3),并给予不同的治疗。治疗结束后的两天处死所有接受治疗的荷瘤小鼠,收集其颈部淋巴结和脾脏,制备单细胞悬浮液。用APC抗小鼠CD3e、FITC抗小鼠CD4和PE抗小鼠CD8a处理上述细胞,以此来鉴定细胞毒性T细胞。用FITC抗小鼠CD11、APC抗小鼠CD80和PE抗小鼠CD86处理上述细胞,用以分析DC细胞的成熟情况。并通过流式细胞术测定上述样品。收集脑肿瘤组织,制备冷冻切片,通过免疫荧光进一步检测CD8+T细胞、钙网蛋白(CRT)和PD-L1在肿瘤中的表达。
实验结果表明,在激光处理组中,肿瘤组织中CRT的绿色荧光明显更高,AMNP@CCM或AMNP@CLP@CCM可以通过将CRT暴露于凋亡肿瘤细胞表面,进而激活ICD以及抗肿瘤免疫应答(图6a;蓝色:细胞核;绿色:CRT;比例尺:100μm)。如图6b和图6c(mean±s.d.,n=3)所示,通过流式细胞术检测颈部淋巴结中DC细胞(CD11c+、CD86+和CD80+)的成熟情况。其中,AMNP@CLP@CCM+L组和AMNP@CCM+L组中,mDCs的比例较高,分别为50.9%和41.1%,比对照组高2.4倍和1.9倍。
PTT可激活全身免疫应答,以增加T细胞浸润,并上调肿瘤细胞表面PD-L1的表达,这有使得肿瘤对ICB治疗更加敏感,并扩大治疗效益。发明人通过流式细胞术分析脾脏中的T淋巴细胞(CD8+T细胞),如图6d和图6e(mean±s.d.,n=3)所示。在AMNP@CLP@CCM+L组中,CD8+T细胞的含量超过41.2%,几乎是对照组的1.6倍。与AMNP@CLP@CCM组和AMNP@CCM+L组相比,AMNP@CLP@CCM+L组的CD8+T细胞的数量分别增加了1.3倍和1.2倍。结果表明,PTT联合ICB增强了胶质母细胞瘤中T细胞的免疫反应,改善了胶质母细胞瘤的免疫抑制微环境。如图6f所示,通过免疫荧光进一步检测肿瘤中的CD8+T细胞,这与上述结果一致。相对其他组,在AMNP@CLP@CCM+L组中,肿瘤组织中红色荧光的CD8+T细胞的数量显著增加,进一步证明了PTT和ICB治疗的协同效应。
CLP002通过阻断PD-L1发挥作用,发明人通过免疫荧光检测了肿瘤组织中PD-L1的表达。如图6g所示AMNP@CLP@CCM组和AMNP@CLP@CCM+L组与对照组相比,几乎没有观察到绿色荧光,这意味着PD-1/PD-L1轴之间的有效阻断。值得注意的是,在光热消融下,肿瘤细胞表面PD-L1表达增强(AMNP@CCM+L组),这与其他研究的结果一致。因此,PTT将冷肿瘤转换为热肿瘤,提升抗PD-L1在肿瘤组织中的表达,并同时通过CLP002阻断PD-1/PD-L1轴,提升基于ICB的癌症免疫治疗的疗效,从而实现协同抗癌治疗。

Claims (7)
1.一种异黑素类仿生纳米粒,其特征在于:包括异丙黑素纳米粒和癌细胞膜,所述异丙黑素纳米粒上负载有PD-L1小分子短肽抑制剂;
所述癌细胞膜来自胶质母细胞瘤细胞;
所述PD-L1小分子短肽抑制剂为CLP002;
所述异丙黑素纳米粒由如下方法制备:1,8-二羟基萘溶于乙腈,然后加入水搅拌,再加入高碘酸钠溶液,再经搅拌、离心和洗涤,获得异丙黑素纳米粒。
2.根据权利要求1所述的一种异黑素类仿生纳米粒,其特征在于:1,8-二羟基萘、乙腈、水和高碘酸钠溶液的用量比为150mg:7.5mL:138.5mL:4mL;高碘酸钠溶液的浓度为0.25M。
3.根据权利要求2所述的一种异黑素类仿生纳米粒,其特征在于:异丙黑素纳米粒和CLP002的质量比为10:5。
4.根据权利要求1-3中任一项所述的一种异黑素类仿生纳米粒的制备方法,其特征在于:包括以下依次进行的步骤:
S1:获取胶质母细胞瘤细胞膜;
S2:将异丙黑素纳米粒和CLP002混合搅拌,获得负载有CLP002的异丙黑素纳米粒;
S3:将负载有CLP002的异丙黑素纳米粒分散在含有胶质母细胞瘤细胞膜的PBS溶液中,获得分散液;再经超声处理获得异黑素类仿生纳米粒。
5.根据权利要求4所述的一种异黑素类仿生纳米粒的制备方法,其特征在于:在S3中,所述分散液中的异丙黑素纳米粒的浓度为1mg/mL。
6.根据权利要求5所述的一种异黑素类仿生纳米粒的制备方法,其特征在于:在S1中,胶质母细胞瘤细胞膜溶于PBS缓冲液中,每1.5×107个细胞中提取的胶质母细胞瘤细胞膜使用1mLPBS缓冲液溶解。
7.根据权利要求1-3中任一项所述的一种异黑素类仿生纳米粒在制备治疗胶质母细胞瘤的药物中的应用。
