本发明涉及酶解技术领域,尤其涉及一种农作物秸秆的预处理方法。
背景技术
木质纤维素生物精炼厂利用生物质生产可再生燃料和化学品,为应对化石能源枯竭和环境恶化提供了一个重要的、变革性的、有利的机会。木质纤维素生物质主要由三种生物聚合物组成:纤维素、半纤维素和木质素,形成一种耐(生物)化学转化的复合材料。分馏木质纤维素生物质的一个根本挑战是在不损失碳水化合物的情况下获得更多可加工木质素部分,并尽量减少不良产品的产生。因此,迫切需要高效的预处理技术来分离和保存纤维素、半纤维素和木质素。在典型的生物炼制中,预处理被认为是打破木质纤维素生物质的天然顽固结构和改善酶水解的主要步骤。作为生物质综合转化过程的一个组成部分,预处理不仅在技术上可行,而且要在经济上也可行。在过去的几十年中,纤维素乙醇工业中已开发出许多从木质纤维素生物质中分离木质素的预处理策略。在这些预处理技术中,乙二胺(EDA)在提高生物乙醇生产中木质纤维素多糖的利用率、提高木质素的水溶性和减少其缩合方面表现出优异的性能。EDA预处理面临的主要挑战是EDA的成本和与EDA回收相关的系统复杂性。
苛刻的预处理条件会导致木质素发生不可逆的降解和缩合反应,并形成抑制剂。这些不可逆的变化会导致低质量的木质素流股,不适合后续的升级放大过程。热化学预处理方法几乎完全集中于最大化单体碳水化合物,可能会导致许多缩合的木质素结构。
发明内容
有鉴于此,本发明提供一种农作物秸秆的预处理方法,该方法不仅可以获得高碳水化合物(木糖和葡萄糖)和可加工的木质素产量,而且可以提高EDA试剂的利用效率。
为了实现上述发明目的,本发明提供以下技术方案:
一种农作物秸秆的预处理方法,包括以下步骤:
将农作物秸秆干燥、粉碎后与乙二胺混匀,100-130℃反应45-60min;
将反应后的农作物秸秆冷却后置于烘箱中,105℃~120℃回收乙二胺1~2.5h,回收乙二胺,获得预处理的农作物秸秆。
本发明中,所述农作物秸秆包括玉米秸秆、小麦秸秆、水稻秸秆和高粱秸秆。
一些具体实施例中,所述反应的具体为120℃反应60min。
本发明中,所述农作物秸秆与乙二胺的质量体积比为0.8~1g:1mLmL,具体可为。
本发明中,所述冷却为:将所述反应的反应釜在自来水流动条件下降温1h。
一些实施方案中,所述回收优选为:105℃回收1~2.5h。一些具体实施例中,所述回收具体为:105℃回收2.5h、105℃回收1h或105℃回收2h。
本发明提供一种利用秸秆制备木质素、葡萄糖和木糖的方法,包括:
利用本发明所述的预处理方法处理农作物秸秆,将预处理的农作物秸秆用水洗涤后进行固液分离,分别获得预处理固体和水洗液;水洗液依次经乙醇沉淀、冷冻干燥,获得含木质素的组分1;
将预处理固体干燥至含水量≤10%,酶解,获得含木糖和葡萄糖的组分2。
本发明中,所述酶解的条件为:50℃、200rpm,酶解发酵96h,pH为4.8。一些具体实施例中,所述pH用0.05M的柠檬酸盐缓冲溶液维持pH为4.8。
本发明中,预处理固体按照1%(w/v)的葡聚糖负荷,添加纤维素酶和半纤维素酶。具体的,本发明中,预处理的玉米秸秆的酶解在100mL锥形瓶中以1%(w/v)葡聚糖负荷进行酶解,其反应体系总体积为20mL。在本发明中按照1.8FPU酶/g葡聚糖的比例添加所使用的酶,其中Cellic CTec2(Novozymes,120FPU/mL)与Cellic HTec2(Novozymes,108FPU/mL)比例为9:1。使用柠檬酸钠缓冲液(pH4.8,50mM)来维持混合物的pH。酶解过程在50℃和200rpm摇床中进行。反应96h后,终止酶解。在一定酶解时间间隔取样并12000rpm离心取上清,经0.22微米滤膜过滤后通过HPLC测定糖浓度。基于预处理玉米秸秆中的葡聚糖和木聚糖含量分别计算葡萄糖和木糖产量。
本发明提供的利用秸秆制备葡萄糖和木糖的方法,所述酶解后还包括:将酶解液离心,取上清经滤膜过滤的步骤。
一些实施方案中,所述滤膜的孔径为0.22μm。
本发明提供的农作物秸秆的预处理方法,包括以下步骤:将玉米秸秆干燥、粉碎后与乙二胺混匀,100-130℃反应45-60min;将反应体系冷却后置于烘箱中,60℃干燥2.5~96h,回收乙二胺,获得预处理的农作物秸秆。该方法在乙二胺处理农作物秸秆后于特定的温和条件下回收乙二胺,本发明乙二胺处理农作物秸秆后在特定的较为温和的条件下回收乙二胺,大大提高了可加工木质素以及碳水化合物(木糖和葡萄糖)的产量,而且还提高了EDA试剂的利用效率。

本发明提供了一种农作物秸秆的预处理方法。本领域技术人员可以借鉴本文内容,适当改进工艺参数实现。特别需要指出的是,所有类似的替换和改动对本领域技术人员来说是显而易见的,它们都被视为包括在本发明。本发明的方法及应用已经通过较佳实施例进行了描述,相关人员明显能在不脱离本发明内容、精神和范围内对本文的方法和应用进行改动或适当变更与组合,来实现和应用本发明技术。
本发明采用的试材皆为普通市售品,皆可于市场购得。
下面结合实施例,进一步阐述本发明:
实施例1
1)将玉米秸秆(购自天津)用蒸馏水清洗,在60℃下干燥。将干燥的玉米秸秆进行粉碎,收集20至80目之间的部分并在室温下储存。
2)将10g粉碎后的CS(干重,dw)装入250ml螺旋瓶,负载为1ml EDA/g CS,预浸泡12小时,然后转移到有聚苯(PPL)内衬的不锈钢反应器中。EDA预处理在120℃(在油浴中1小时)的温和条件下进行(step1)。之后,不锈钢反应容器用流动的自来水快速冷却。预处理CS通过烘箱加热进行EDA回收(设置温度分别为60℃、105℃、120℃和150℃)(step2)。EDA回收后,用蒸馏水按10ml水对1g固体的比例洗涤三次预处理固体。预处理的玉米秸秆(CS)用蒸馏水以10毫升水每克固体的比例洗涤三次,通过过滤进行固液分离,分别获得水洗液和预处理固体物,计算预处理固体物的回收率,结果见表1。
3)从洗涤液中分离木质素,以1:2(v/v)洗涤液:乙醇进行乙醇沉淀。对不溶性底物进行离心,然后对洗涤液馏分进行旋转蒸发除去乙醇,收集浓缩液。经冷冻干燥得到EDA木质素样品。
本研究中,EL样品代表EDA预处理(step1)得到的木质素,EL-60℃、EL-105℃、EL-120℃和EL-150℃样品分别代表EDA预处理和回收过程(第2步)通过60℃至150℃加热得到的木质素。
4)预处理的玉米秸秆的酶解在100mL锥形瓶中以1%(w/v)葡聚糖负荷进行,其反应体系总体积为20mL。所使用的酶用量为1.8FPU酶/g葡聚糖,其中Cellic CTec2(Novozymes,120FPU/mL)与Cellic HTec2(Novozymes,108FPU/mL)的体积为9:1。使用柠檬酸钠缓冲液(pH4.8,50mM)来维持混合物的pH。酶解过程在50℃和200rpm摇床中进行。反应96h后,终止酶解。在一定酶解时间间隔取样并12000rpm离心取上清,经0.22微米滤膜过滤后通过HPLC测定糖浓度。基于预处理玉米秸秆中的葡聚糖和木聚糖含量分别计算木聚糖转化率和木聚糖转化率。结果见表2和表3。
葡聚糖转化率(%)=酶解产物中葡萄糖质量/(预处理样品中葡聚糖质量×1.11)。
木聚糖转化率(%)=酶解产物中木糖质量/(预处理样品中木聚糖质量×1.13)
5)使用Smartlab衍射仪(日本Rigaku)测量了玉米秸秆原料(粒径小于125μm)的预处理固体的结晶度。在2θ=10°到40°范围内扫描衍射强度,步长为0.02°,速率为4°/分钟。
6)木质素表征
本研究中,木质素样品“EL-60℃”表示“酶解+60℃-2.5h回收乙二胺(即EL-60℃-2.5h)”、EL-105℃(EL-105℃-2.5h)、EL-120℃
(EL-120℃-2.5h)、EL-150℃(EL-150℃-4h)。
对获得到木质素样品采用凝胶渗透色谱法(GPC)测定了分子量分布:将分离得到的木质素样品溶于浓度为1mg/ml的0.1MNaOH中,并注入到含有2个300×7.8mm(L.×I.D.)的GPC体系(Waters e2695)中。TSKgel GMPWxl柱(TOSOH,东京)系列。流动相为0.1MNaOH,流速1.0ml/min,烘箱温度30℃。设置紫外检测器在280nm和254nm波长处测量吸光度。采用聚苯乙烯(SPS)标准品(210-17000Da)和愈创木酰基甘油-β-愈创木酰基醚计算表观分子量与停留时间的三级校正拟合型标准曲线,通过凝胶渗透色谱法(GPC)测定木质素的分子量,结果见图1。
采用Netzsch STA449F3同步热分析仪(DSC/DTA-TG)在氮气气氛下,升温速率为10℃/min,对木质素样品进行了热重分析,结果见图2。
采用傅里叶变换红外光谱(FTIR)和二维核磁(HSQC)方法测定了官能团和结构,方法如下:
将提前干燥脱水的样品与光谱级溴化钾以1:100的质量比研磨后压片。FTIR光谱通过红外光谱分析仪(BrukerTENSOR 27)扫描得到,扫描波长为4000-400cm-1,结果见图3。
将100mg木质素样品溶于600μL 99.9%的DMSO-d6(含有TMS内标)进行二维核磁表征。二维核磁图谱用BrukerAVANCE III(600MHz)光谱仪测定脉冲序列为hsqcedetgpsisp2.2,扫描次数为64,时间增量为256。木质素中β-O-4,β-β连接键和氨基相关结构的半定量结果参照先前的文献计算得到,结果见图4和表4。
木质素样品的C,H,N以及S的含量通过ElementarVario EL CUBE元素分析仪测定分析,结果见表5。

表5木质素样品的元素分析
表4和表5中,CEL样品为纤维素酶直接酶解未处理秸秆得到的木质素样品,EL样品表示EDA预处理后不回收乙二胺制备获得的木质素,EL-60℃表示采用本发明预处理方法(60℃加热2.5h回收乙二胺)处理玉米秸秆之后制备得到的木质素。EL-105℃表示采用105℃加热2.5h回收乙二胺处理玉米秸秆之后制备得到的木质素,EL-120℃表示采用120℃加热2.5h下回收乙二胺处理玉米秸秆之后制备得到的木质素。EL-150℃表示采用150℃加热4h下回收乙二胺处理玉米秸秆之后制备得到的木质素。
由表4可知,本发明特定的条件下回收EDA的农作物秸秆的预处理方法成功把氮元素引入了木质素结构中,这种含氮结构有助于木质素的高值化利用,同时回收的EDA可重复使用,降低了生产成本。
由图1和2可知,本发明在105℃~120℃加热1~2.5h条件下进行乙二胺回收的预处理方法能够促进木质素解聚为低分子量的单体和寡聚物。
由表5可知,本发明成功把氮元素引入到木质素结构中。
由图3和4,表4可知,EDA回收预处理能有效地裂解生物质中乙酰酯键,促进氨解生成酰胺类化合物和席夫碱结构化合物;这些氨基化木质素可促进木质素的高值化,可作为有前途的增值产品前体。结合表2~3木聚糖和葡聚糖转化率结果,105℃乙二胺回收2.5小时的预处理方法效果最优,不仅能够有效的提高了酰胺、氨基这些氨化结构,促进木质素的高值化,同时还保证了较高的木聚糖和葡聚糖转化率,应用价值更高。
以上结果显示,本发明提供的EDA的回收预处理方法提高了碳水化合物的可及性和酶解性能,赋予木质素下游高值化,提高了工艺可行性,实现了生物质利用的可持续性。
以上仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (10)
1.一种农作物秸秆的预处理方法,其特征在于,包括以下步骤:
将反应秸秆干燥、粉碎后与乙二胺混匀,100-130℃反应45-60min;
将反应后的农作物秸秆冷却后置于烘箱中,105℃~120℃回收乙二胺1~2.5h,获得预处理的农作物秸秆。
2.根据权利要求1所述的预处理方法,其特征在于,所述农作物秸秆包括玉米秸秆、小麦秸秆、水稻秸秆和高粱秸秆。
3.根据权利要求1所述的预处理方法,其特征在于,所述农作物秸秆与乙二胺的质量体积比为1g:0.8~1mL。
4.根据权利要求1所述的预处理方法,其特征在于,所述冷却为:将所述反应的反应釜在自来水流动条件下降温1h。
5.根据权利要求1所述的预处理方法,其特征在于,所述回收的温度为105℃,时间为1~2.5h。
6.一种利用农作物秸秆制备木质素、葡萄糖和木糖的方法,其特征在于,包括:
利用权利要求1~5任一项所述的预处理方法处理农作物秸秆,将预处理的农作物秸秆用水洗涤后进行固液分离,分别获得预处理固体和水洗液;
水洗液依次经乙醇沉淀、冷冻干燥,获得含木质素组分1;
将预处理固体干燥至含水量≤10%,添加纤维素酶和半纤维素酶进行酶解,获得含木糖和葡萄糖的组分2。
7.根据权利要求7所述的方法,其特征在于,所述酶解的条件为:50℃、200rpm,酶解发酵96h,pH为4.8。
8.根据权利要求6所述的方法,其特征在于,所述纤维素酶和半纤维素酶的体积比为9:1。
9.根据权利要求所述的方法,其特征在于,所述酶解后还包括:将酶解液离心,取上清经滤膜过滤的步骤。
10.根据权利要求所述的方法,其特征在于,所述滤膜的孔径为0.22μm。
