Abstract
本申请涉及一种茋苷化合物在制备抗UV‑B致皮肤光老化护肤品中的应用,属于化妆品技术领域,所述茋苷化合物具体包括曲札茋苷、藏黄苷或二苯乙烯苷中的至少一种,本发明效果显示曲札茋苷、藏黄苷或二苯乙烯苷在5、20、50及150μg/mL浓度下均能显著增加‑UVB照射(剂量为60mj/cm2)人永生角质形成细胞(HaCaT)的相对活力,对其损伤有显著保护作用,同时可抑制细胞活性氧的产生、炎症因子IL‑1、IL‑6及TNF‑α分泌及基质金属蛋白酶MMP‑1分泌。

技术领域
本申请涉及一种茋苷化合物在制备抗UV-B致皮肤光老化护肤品中的应用,属于化妆品技术领域。
背景技术
皮肤老化包括内源性老化和外源性老化,内源性老化是由遗传因素调控并随时间推移导致机体功能逐渐衰退的自然老化过程;外源性老化是由外界因素包括紫外线辐射、坏境污染、饮食、吸烟、坏境等引起的皮肤过早老化的过程,紫外线辐射是引起的皮肤老化的主要外界因素,长期紫外线照射引起的老化即光老化。皮肤光老化会严重影响美观,出现干燥、粗糙、毛细血管扩张、弹性降低、不规则色素沉着、脂溢性角化等表现。紫外线辐射促进和加速皮肤衰老发生和发展的过程,导致人的容貌过早老态而影响外在形象和面貌。
紫外线按照波长分类为长波紫外线(UVA),中波紫外线UVB和短波紫外线(UVC)。UVC波长短,穿透能力弱,在臭氧层已被吸收,极少到达地球表面;UVA穿透能力很强,可穿透臭氧层,到达地球表面的紫外线大约有98%为UVA;UVB穿透能力中等,能到达地球表面的只有约2%。UVB主要作用于皮肤表皮及真皮浅层。虽然到达地球表面的UVB只是太阳紫外线的一小部分,但急性暴露于高剂量的UVB会导致急性皮肤损伤如红斑、水疱等晒伤样病变;还可导致光毒反应、光变态反应;长期低剂量UVB辐射则会导致DNA损伤、尿苷酸光异构化等可致皮肤肿瘤发生。UVB光损伤作用是同剂量UVA的800~1000倍,除可直接导致DNA损伤,UVB照射皮肤后诱导皮肤细胞产生活性氧自由基(ROS),过量的ROS会打破细胞自身抗氧化系统的平衡,进而引起氧化应激。细胞在接受紫外线辐射后同样会产生大量的炎症因子,能导致皮肤局部的炎性反应,是引起皮肤光老化的主要诱发因素。
人表皮中的主体细胞成分是角质形成细胞,此类细胞经过增殖和分化,最终形成包括基底层、棘层、粒层及角质层的表皮复层结构。人角质形成细胞是表皮中重要的细胞,也是紫外线敏感的靶细胞。近年来研究表明,人角质形成细胞在光老化的发生和发展中起重要作用,在加重炎症反应方面也尤为突出。目前,人永生化角质形成细胞HaCaT细胞株已被广泛应用于化妆品的抗衰老及透皮性的研究中。
茋苷类成分属于茋类(stilbenoids)化合物的糖苷,具有二苯乙烯母体结构,广泛分布于植物中,具有抗肿瘤、抗炎、抗氧化、保护心脑血管、抑菌、抗病毒等多种作用。目前,尚未见茋苷成分用于UVB所致皮肤光老化方面的报道。
发明内容
本申请提供了一种茋苷成分及其在对抗UVB所致光老化损伤护肤品中的用途,
本申请提供的茋苷成分中的茋(stilbenoids)也称为“二苯乙烯类化合物”,具有如下化学结构通式:
式I中,R选自-H、-OH、-OCH3、-O-葡萄糖基、-O-取代的葡萄糖基中的至少一种。
茋苷类成分为R基至少有一个-O-葡萄糖基。
所述茋苷类成分为藏黄苷、曲札茋苷或二苯乙烯苷。
具体地,曲札茋苷化学名为(E)-1-(3,5-二羟苯基)-2-(3-羟基-4-O-D-吡喃葡萄糖苯基)乙烯或3,5,3′,4′-四羟基茋-3′-O-β-葡萄糖苷,结构式如式II:
具体地,藏黄苷化学式化学名为(E)-1-(3,5-二羟苯基)-2-(4-羟基-3-O-D-吡喃葡萄糖苯基)乙烯或3,5,3′,4′-四羟基茋-4′-O-β-葡萄糖苷,结构式如式III:
具体地,二苯乙烯苷化学名为stilbeneglycoside,2,3,5,4’-四羟基二苯乙烯-2-O- -D-葡萄糖苷。结构式如式IV:
曲扎茋苷、藏黄苷及二苯乙烯苷在蓼科植物(Polygonaceae)中含量丰富,如曲札茋苷在拉萨大黄(Rheum lhasaense A.J.Liet P.K.Hsiao)干燥根茎中含量可达4%,藏黄苷在藏边大黄(Rheum emodi D.Don)干燥根茎中含量可达7%,二苯乙烯苷在何首乌(Fallopia multiflora(Thunb.)Harald)干燥根茎中含量不少于0.70%。因此这些茋苷化合物可通过简单的分离纯化技术获得化合物单体,也可通过化学方法进行合成。
本发明的另一用途是提供上述茋苷成分在对抗UV-B所致光老化损伤护肤品中的用途。
优选地,所述用途包括提高UV-B照射角质形成细胞的活力。
优选地,所述应用包括降低UV-B照射角质形成细胞中活性氧自由基(ROS)的含量。
优选地,所述用途包括抑制UV-B照射角质形成细胞的分泌IL-1α、IL-6及TNF-α炎症因子。
优选地,所述用途包括抑制UV-B照射角质形成细胞中分泌基质金属蛋白酶MMP-1分泌。
本发明提供茋苷化合物可按现有方法将其制成常规皮肤护理产品,包括但不限于:面霜、护手霜、眼霜、身体乳、保湿水、精华液、纯露、沐浴乳、洗发水、护发素、发油、按摩油、精华油、身体油、面膜、眼膜、防晒霜、防晒喷雾等。

本申请中首次将曲札茋苷、藏黄苷及二苯乙烯苷用于对抗UV-B所致皮肤光老化护肤品中的用途。
与现有技术相比,本发明有益效果如下:
1.如前所述,UVB是皮肤光老化的主要紫外线,人角质形成细胞是表皮中重要的细胞,也是UVB引起皮肤光老化症状的主要紫外线敏感靶细胞。本发明显示曲札茋苷、藏黄苷及二苯乙烯苷在5、20、50及150μg/mL浓度下均能显著增加能使UVB照射(剂量为60mj/cm2)人永生角质形成细胞(HaCaT)的相对活力,对其损伤有显著保护作用。
2.HaCaT在UVB刺激后,ROS含量显著增加,产生氧化应激,显示细胞内氧化作用与抗氧化作用平衡被打破,从而产生大量具有高度活性的活性氧自由基(ROS)。ROS包括超氧阴离子(·O2-)、过氧化氢(H2O2)、羟自由基(·OH)等,参与了脂质、蛋白的氧化及DNA损伤,进而引起组织氧化损伤。研究证明,高水平的ROS可导致细胞凋亡或坏死。本发明显示曲札茋苷、藏黄苷及二苯乙烯苷在5、20、50及150μg/mL浓度下均能显著降低损伤细胞的活性氧(ROS)含量或水平,对损伤细胞的氧化应激具有显著逆转作用,从而达到保护细胞、避免凋亡或坏死的作用。
3.UVB辐射后,不仅直接产生ROS损伤细胞,而且可激活IL-1、IL-6、及TNF-α等促进炎症细胞因子表达,同时抑制抗炎症细胞因子的表达,使细胞因子调控网络失衡,从而引起细胞炎症反应。这些细胞因子还能诱导MMPs的表达,导致细胞外基质降解、抑制胶原合成,进而加速光老化的过程。本研究实施例中显示,经过UVB辐射后,光老化模型HaCaT细胞中的炎症因子IL-1、IL-6和TNF-α水平均升高,同时细胞中的基质金属蛋白酶MMP-1水平也显著升高,提示UVB辐射加重HaCaT细胞光老化。相比UVB照射组,本发明显示曲札茋苷、藏黄苷及二苯乙烯苷在5、20、50及150μg/mL浓度下降低光老化模型HaCaT细胞IL-1、IL-6、TNF-α和MMP-1的表达水平,能够减少UV辐照后胶原蛋白和弹性蛋白的降解。
4.综上,本发明提供3个茋苷成分能够显著降低UVB光老化HaCaT细胞的ROS含量、炎症因子IL-1、IL-6、TNF-α和基质金属蛋白酶MMP-1的表达水平,近而抑制UVB所致损伤细胞的氧化应激反应和炎症反应,降低UVB所致HaCaT细胞的损伤作用和凋亡(死亡),提高细胞的活力,达到对抗UVB所致光老化的功效,在皮肤保护方面具有良好的应用价值。
另外,本发明所提供的有效物质单体,成分明确可控,天然资源含量丰富,活性高(5μg/mL浓度即起效),化合物颜色为白色,在护肤品制备中具有较强的实用性。
具体实施方式
下面结合实施例详述本申请,但本申请并不局限于这些实施例。
实施例1:制备茋苷面霜
(1)配料表如表1所示

60℃恒温水浴加热融化油相(1-7,总量200克),同时加热水相(8-20,总量800克),保持温度边搅拌边把水相慢慢加入到油相中,加完后保温持续搅拌30分钟,待油相和水相充分乳化,冷却至35℃以下加入添加剂(21-23,总量10克)搅拌30分钟,灌装入50g面霜瓶,冷却即得。
实施例2:制备茋苷面膜胶

(2)制法
将2项加入到8项中,加热到80~90℃,待PVA完全溶胀后加入3项,降温到50~60℃时加其余项的混合物,恒温水浴持续搅拌30分钟全部溶解,灌装,冷却至室温得可剥离膏状面膜胶。
实施例3茋苷类化合物对UV-B损伤角质形成细胞的保护作用
1实验材料及主要仪器
1.1材料
1.1.1实验细胞人永生化角质形成细胞株(HaCaT)购于武汉大学中国典型培养物保藏中心。
1.1.2主要试剂DMEM培养基、胎牛血清、胰蛋白酶-EDTA消化液均购于美国Gibco公司;DMSO溶液购于美国Sigma-Aldrich公司;PBS缓冲液、青链霉素混合溶液(100X)购于索莱宝中国公司;CCK-8试剂盒购于索莱宝中国公司;活性氧检测试剂盒(DHE荧光探针法)购于北京百奥莱博科技有限公司;人白细胞介素-1β(IL-1)ELISA试剂盒;人白细胞介素-6(IL-6)ELISA试剂盒及人肿瘤坏死因子(TNF-α)ELISA试剂盒购于美国MERK公司;MMP-1ELISA试剂盒购于上海碧云天生物科技有限公司。
1.1.3主要仪器超净工作台(苏州净化公司);CO2细胞培养箱、-80℃低温冰箱、酶标仪(美国Thermo公司);UVB辐照灯(荷兰皇家飞利浦电子公司)、高速离心机(湖南湘仪公司)。
2实验方法
2.1细胞培养
将HaCaT细胞复苏,在超净工作台上将细胞悬液移入25T培养瓶中,加入完全培养基(含有体积分数10%的胎牛血清,终浓度为1μmol/L的青霉素和链霉素的DMEM培养基)至5mL,使细胞分布均匀后置于37℃、5%CO2细胞培养箱中培养至贴壁。用磷酸缓冲盐溶液(PBS,浓度为0.01mol/L,pH=7.4)清洗细胞。
2.2不同茋苷对UVB诱导光老化HaCaT细胞影响
2.2.1建立UVB诱导光老化HaCaT细胞模型
将接种于96孔板,培养至密度为80%左右,设置空白组、模型组和实验组,每组设置6个复孔取均值。空白组不经UVB照射,加入10μL培养液处理2小时,然后于37℃培养24h后用于实验。模型组HaCaT细胞加入10μL培养液处理2小时,然后采用60mJ/cm2的UVB照射处理,于37℃培养24h后用于实验。实验组分为5μg/mL组、20μg/mL组、50μg/mL组、150μg/mL组共4个实验组,分别加入10μL不同茋苷培养液,使其茋苷终浓度为5、20、50、150μg/mL,然后采用60mJ/cm2的UVB照射处理,于37℃培养24h后用于实验。
2.2.2不同浓度的茋苷对UVB诱导光老化HaCaT细胞活力的影响实验
按2.2.1步骤处理各组细胞,培养24h后根据CCK-8试剂盒说明书,向每孔加入10μL的CCK溶液,置于培养箱内孵育30min后,用酶标仪测定在450nm处的吸收值(A),计算细胞的相对活力。
细胞相对活力计算公式:
细胞相对活力=(As/Ab)×100%
其中:As为模型组及实验组样品在450nm处的吸收值;
Ab为空白组样品在450nm处的吸收值。
2.2.3不同浓度的茋苷对UVB诱导光老化HaCaT细胞ROS含量影响实验按2.2.1步骤处理各组细胞,培养24h后根据活性氧检测试剂盒(DHE荧光探针法试剂)盒说明书,向每孔加入10μL稀释500倍的DHE溶液,置于培养箱内避光孵育30min后,用新鲜溶液清洗细胞3次,在激发波长535nm,发射波长610nm用荧光酶标仪测定细胞内活性氧水平,计算活性氧的含量。
2.2.4不同浓度的茋苷对UVB诱导光老化HaCaT细胞炎症因子水平影响实验
按2.2.1步骤处理各组细胞,培养24h后细胞培养液在4℃、8000rpm离心20min,收集各组细胞上清液,按照各个试剂盒说明应用ELISA法进行检测并计算出各组细胞内炎症因子IL-1α、IL-6、TNF-α的含量。
2.2.5不同浓度的茋苷对UVB诱导光老化HaCaT细胞MMP-1蛋白水平影响实验
按2.2.1步骤处理各组细胞,培养24h后细胞培养液在4℃、12000r/min条件下离心10min,收集上清液。用BCA蛋白定量试剂盒对上清液中的蛋白进行定量,确定终浓度为10μg/mL。根据酶联免疫吸附测定(ELISA)试剂盒说明书测定分泌蛋白MMP-1的表达水平。
2.3统计学方法用SPSS22.0软件对实验数据进行统计学分析,计量资料用均数±标准差表示,多组间数据采用单因素方差分析,组间两两比较采用LSD-t检验,两组间数据比较采用t检验。以P<0.05为差异有统计学意义。
3实验结果
3.1不同浓度的茋苷对UVB诱导光老化HaCaT细胞活力的影响
实验结果见表3。与空白组比较,模型组细胞相对活力显著降低,差异有统计学意义(P<0.05);与模型组比较,曲札茋苷及二苯乙烯苷各剂量组非常显著地提高HaCaT细胞活力,差异均有统计学意义(P<0.01),藏黄苷各剂量组显著提高HaCaT细胞活力,差异有统计学意义(P<0.05)。各茋苷对细胞活力的影响有剂量依赖关系。

实验结果见表4。与空白组比较,模型组细胞ROS含量显著升高,差异有统计学意义(P<0.05);与模型组比较,曲札茋苷及二苯乙烯苷各剂量组非常显著地降低HaCaT细胞ROS含量,差异均有统计学意义(P<0.01);藏黄苷5、20及50μg/mL剂量组显著降低HaCaT细胞ROS含量(P<0.05),50g/mL剂量组效果更加显著(P<0.01)。各茋苷对细胞活力的影响有剂量依赖关系。

3.3不同浓度的茋苷对UVB诱导光老化HaCaT细胞炎症因子水平影响
实验结果见表5。与空白组比较,模型组细胞炎症因子IL-1、IL-6及TNF-α表达水平均显著升高,差异均有统计学意义(P<0.01);与模型组比较,试验茋苷各剂量组均显著地降低HaCaT细胞细胞炎症因子IL-1、IL-6及TNF-α表达水平,差异均有统计学意义(P<0.05或P<0.01)。各茋苷对细胞炎症因子的影响剂量依赖关系不明显,低剂量显示出更好活性的趋势。

实验结果见表4。与空白组比较,模型组细胞MMP-1蛋白表达水平显著升高,差异均、有统计学意义(P<0.01);与模型组比较,试验茋苷各剂量组均显著地降低HaCaT细胞MMP-1蛋白表达水平,差异均有统计学意义(P<0.05或P<0.01)。各茋苷对细胞炎症因子的影响剂量依赖关系不明显。

Claims (9)
1.一种茋苷化合物的应用,其特征在于,所述茋苷化合物具体包括曲札茋苷、藏黄苷或二苯乙烯苷中的至少一种,所述应用包括在制备抗UV-B所致皮肤光老化护肤品中的应用。
2.根据权利要求1所述的应用,其特征在于,所述应用为制备抗UV-B所致人皮肤光老化护肤品中的应用。
3.根据权利要求2所述的应用,其特征在于,所述护肤品包括:面霜、护手霜、眼霜、身体乳、保湿水、精华液、纯露、沐浴乳、洗发水、护发素、发油、按摩油、精华油、身体油、面膜、眼膜、防晒霜或防晒喷雾。
4.根据权利要求1所述的应用,其特征在于,所述应用包括提高UV-B照射角质形成细胞的活力,所述细胞为HaCaT。
5.根据权利要求1所述的应用,其特征在于,所述应用包括降低UV-B照射角质形成细胞中活性氧自由基(ROS)的含量。
6.根据权利要求1所述的应用,其特征在于,所述应用包括抑制UV-B照射角质形成细胞分泌炎症因子IL-1α、IL-6或TNF-α。
7.根据权利要求1所述的应用,其特征在于,所述应用包括抑制UV-B照射角质形成细胞分泌基质金属蛋白酶MMP-1。
8.根据权利要求4-7任一所述的应用,其特征在于,使用时的浓度大于5μg/mL。
9.根据权利要求4-7任一所述的应用,其特征在于,使用时的浓度为5~150μg/mL。
