and finally, freezing the concentrated solution at-28 ℃ for 35h to obtain the solid medicament.
The traditional Chinese medicine preparation prepared by the invention is subjected to the following animal experiments:
(1) Grouping animals
80 detergent mice 8 weeks old and 20-25g in weight were selected and divided into 8 groups (10 mice per group) on average, wherein 1 group of mice was a blank group and only distilled water was drunk. In addition, 7 groups of mice freely drunk 3% of DSS solution, and 5 days later, distilled water was changed to 3 weeks, and mice as chronic models of polar colitis were established. The solid preparation prepared in the example is injected into 6 groups of model mice respectively, the injection dosage is 0.25g/Kg, and the solid preparation needs to be dissolved by normal saline before injection. The other model group was a control group and was not injected with any drug. Specifically, the results are shown in Table 1.
TABLE 1
Figure BDA0003930025580000041
Figure BDA0003930025580000051
(2) Animal feeding and treatment
After the treated mice are normally fed for 6 days, blood is collected, the mice are sacrificed, the colons of the mice are taken out, excrement is taken out for standby, the length of the colons of each group of mice is measured, the content of the colons is washed out by normal saline, one part of colons is used for preparing colons homogenate, one part of colons is soaked in 10 percent formaldehyde solution for fixation, and dehydration, transparence, paraffin embedding, slicing and HE dyeing are carried out.
(3) Analysis and conclusions
1) Colon length in mice
As can be seen from Table 2, the colon length of the control group mice is much shorter than that of the drug group mice, and the drug group mice can prevent the colon shortening phenomenon of the mice.

2) Colon section
FIG. 1 is a section of the colon of each of the mice in the drug groups 1-3 from left to right, and it can be seen from the section that the mucosal layer, submucosal layer, muscular layer and serosal layer of the colon of each of the mice in the drug groups are complete and clear in tissue structure, and the tight connection between cells is normal; the glands are regularly arranged, the goblet cells are rich and no inflammatory cell infiltration exists.
FIG. 2 is a colon section of a control mouse, and it can be seen from FIG. 2 that the colonic muscle layer of the control mouse is seriously damaged; the intercellular space of the lamina propria is increased, inflammatory cell infiltration exists, glands are reduced, arrangement is disordered, goblet cells are increased, and the morphology is abnormal.
The result shows that the traditional Chinese medicine preparation can reduce the colonic inflammation of mice and has a certain repairing effect.
3) Biochemical index of colon
As can be seen from Table 3, the feces, colon homogenates, and serum botulinum toxin LPS were higher in the control mice than in the blank. The feces, colon homogenate and serum botulinum toxin LPS of the mice in the medicament group are lower than those of the control group, which shows that the medicament can reduce the feces, colon homogenate and serum botulinum toxin of the mice and is probably one of the action mechanisms of the medicament for protecting the colon repair of depressed mice.
