技术领域
本发明涉及一种蛋白抑制剂、一种试剂组及其应用,属于细胞生物学、癌症生物学与药物研发技术领域。
背景技术
三阴乳腺癌的定义来自于肿瘤细胞表面分子的差异进行分类,三阴性乳腺癌是雌激素受体(ER)、孕激素受体(PR)和原癌基因Her-2均为阴性的乳腺癌。三阴乳腺癌通常是通过手术、化疗、放射治疗等方式来达到治疗目的的。三阴乳腺癌在乳腺癌当中是比较特殊的,因为这种乳腺癌对靶向治疗和内分泌治疗都不敏感,所以只能在发现的时候尽早的采取手术治疗,并且在疾病有转移的情况时,手术之后再采取一定的辅助治疗措施。总体来说,三阴乳腺癌易转移、易复发,死亡率高,除了化疗没有靶向治疗与免疫治疗药物。
小细胞肺癌虽仅占肺癌发病总量的15%,但恶性程度高,死亡率高;小细胞肺癌的癌细胞通常生长速度快,容易扩散和转移,对化疗和放疗较为敏感。
结肠癌在所有消化系统的恶性肿瘤中,排在第三位,发病率还是比较高,属于比较难治愈的肠癌疾病。结肠癌的治疗方法主要有:手术治疗:这也是最主要的治疗方式,适用于早期和中期的一些结肠癌;化学药物治疗:一般结肠癌根治后,有复发或者转移的可能,因此术前或者术后化疗可能提高根治术后五年的生存率。像结肠癌化疗常用的药物就是氟尿嘧啶类的药物,其他的还有阿霉素类的药物;放射治疗:术前的放疗可以缩小肿瘤,提高切除率;术后放疗可以杀死残留的肿瘤细胞。
由此可见,以上的癌症甚至更多均缺乏明确的治疗靶标,缺少被批准使用的靶向治疗药物,免疫治疗药物亦没有取得较好的治疗效果,因此目前临床上均以化疗为主要治疗手段。虽然在治疗初期部分病人对化疗药物的响应度良好,但随即许多病人出现化疗耐受,导致无药可用和最终的死亡。
发明内容
为了克服现有技术的不足,本发明的第一个目的在于提供一种蛋白抑制剂,该蛋白抑制剂对于CILK1蛋白进行抑制,有利于对癌细胞进行抑制,并且可以与化疗药物联用。
本发明的第二个目的在于提供一种包括上述蛋白抑制剂的试剂组,该试剂组提供了蛋白抑制剂与化疗药物联用的方案,具有更有效地杀死癌细胞的效果。
本发明的第三个目的在于提供一种上述蛋白抑制剂的应用,对于CILK1蛋白抑制机理的研究也有很好的启发和引导。
实现本发明的第一个目的可以通过采取如下技术方案达到:一种蛋白抑制剂,蛋白抑制剂为抑制CILK1蛋白的抑制剂。
进一步地,蛋白抑制剂为通过抑制苏氨酸-157和酪氨酸-159的磷酸化修饰,以抑制CILK1蛋白的蛋白抑制剂。
进一步地,蛋白抑制剂为CILK1-C28或CILK1-C30;CILK1-C28的化学结构式为Ⅰ;CILK1-C30的化学结构式为Ⅱ;
进一步地,蛋白抑制剂包括CILK1-C28和CILK1-C30,CILK1-C28和CILK1-C30的浓度均为1-20μM。

实现本发明的第二个目的可以通过采取如下技术方案达到:一种试剂组,试剂组包括蛋白抑制剂、顺铂和依托泊苷;蛋白抑制剂为CILK1-C30;CILK1-C30的化学结构式为Ⅱ;
进一步地,顺铂的浓度为0.2-1.8μg/mL。
进一步地,依托泊苷的浓度为0.2-100μg/mL。
实现本发明的第三个目的可以通过采取如下技术方案达到:一种蛋白抑制剂的应用,将蛋白抑制剂应用于CILK1蛋白抑制机理的研究。
本发明的设计原理如下:
CILK1(Ciliogenesis Associated Kinase 1,纤毛发生相关激酶1)是一种丝苏氨酸蛋白激酶,一般认为它在纤毛发生以及纤毛类疾病中起重要作用。目前,对于它在癌症中的作用一无所知。本方案中蛋白抑制剂的研究具有颠覆性的创新,为行业提供了全新的癌症治疗方案以及新思路。首先,对于杀伤癌细胞使用何种抑制剂,首先要确定具体癌细胞内的分子靶点(蛋白质),此步骤需要专业科研人员的大量的实验探索,难以凭空想到。第二,即使确定了某种蛋白质是治疗某类肿瘤的分子靶点,研发它的抑制剂是非常困难的,需要数年时间的专业探索,运用多种专业技术手段。
本方案是行业中首次确立了CILK1是癌症特别是三阴乳腺癌、小细胞肺癌和结肠癌的治疗分子靶点,具有理论上的原创性。然后提供了CILK1的特异性小分子抑制剂,首次证明CILK1抑制剂可以杀死三阴乳腺癌、小细胞肺癌和结肠癌细胞,具有极高的创新度和重大应用价值。
相比现有技术,本发明的有益效果在于:
1、本发明确立了CILK1是癌症特别是三阴乳腺癌、小细胞肺癌和结肠癌的治疗分子靶点,基于以上所开发的全新抑制剂,使用能有效杀死癌细胞;
2、本发明的试剂组提供了蛋白抑制剂与化疗药物联用的方案,具有更有效地杀死癌细胞的效果;
3、本发明蛋白抑制剂对于CILK1蛋白抑制机理的研究也有很好的启发和引导。

一种蛋白抑制剂,为通过抑制苏氨酸-157和酪氨酸-159的磷酸化修饰,以抑制CILK1蛋白的蛋白抑制剂。
蛋白抑制剂为CILK1-C28或CILK1-C30;CILK1-C28的化学结构式为Ⅰ;CILK1-C30的化学结构式为Ⅱ;
其中,蛋白抑制剂是CILK1-C28和CILK1-C30,CILK1-C28和CILK1-C30的浓度均为1-20μM。
蛋白抑制剂可以和化疗药物联合使用。

其中,紫杉醇的浓度为1-50nM。
一种试剂组,试剂组包括蛋白抑制剂、顺铂和依托泊苷;蛋白抑制剂为CILK1-C30。
其中,顺铂的浓度为0.2-1.8μg/mL。
其中,依托泊苷的浓度为0.2-100μg/mL。
蛋白抑制剂的应用,为将蛋白抑制剂通过抑制CILK1的激酶活性,以抑制癌细胞的活力。
蛋白抑制剂的应用,为将蛋白抑制剂用于CILK1蛋白抑制机理的研究:苏氨酸-157和酪氨酸-159的双磷酸化修饰是反映CILK1蛋白活性的重要指标。CILK1-C30和CILK1-C28能明显抑制这两个氨基酸基团的磷酸化修饰,而不影响CDK1(其蛋白序列、结构与CILK1类似)的磷酸化水平,说明它们能特异性抑制CILK1的活性。
人类细胞内的丝苏氨酸蛋白激酶总数达上百种,它们彼此间是独立、不相同的蛋白质,对于不同的癌症的作用机理不同。比如,丝苏氨酸蛋白激酶Akt与MAPKs被分别发现在癌症中的重要作用在当时是重要的理论突破与创新,但目前没有CILK1在癌症中作用的学术报道,更加没有针对CILK1蛋白抑制剂的记载,是因为行业中还未有意识到CILK1在癌症进展中的重要作用,也没有意识到研发CILK1抑制剂的重要性和科学与应用价值。
实施例1:
使用Western blot方法,使用三种CILK1:pTyr159-CILK1(酪氨酸-159磷酸化的CILK1蛋白)、pThr157-CILK1(苏氨酸-157磷酸化的CILK1蛋白)和CILK1,pTyr159-CILK1识别第159位酪氨酸上有磷酸基团修饰的CILK1蛋白,pThr157-CILK1识别第157位苏氨酸上有磷酸基团修饰的CILK1蛋白,CILK1自身抗体对于CILK1蛋白无论是否有修饰均能识别;两种CDK1:CDK1和pCDK1和Actin抗体进行实验。
Western blot杂交实验步骤:
1.用冰冷的PBS缓冲液洗涤培养皿中DMSO(二甲基亚砜,对照Ctr)、CILK1-C30(浓度1μM和10μM)、CILK1-C28(浓度1μM和10μM)分别处理的三阴乳腺癌细胞(MDA-MB-231和BT549)两次,在蛋白裂解缓冲液中破碎细胞收集裂解物。
2.高速离心提取蛋白质混合物,蛋白浓度由BCA试剂盒(KEGEN)测定。
3.每道60毫克蛋白质混合物装载于7-15%(W/V)的SDS-PAGE胶上进行电泳分离。
4.将SDS-PAGE胶上的分离蛋白质电转移到PVDF膜上。
5.PVDF膜在5%(W/V)脱脂牛奶中室温孵育1.5小时。
6.PVDF膜加入一抗(稀释浓度1:1000),于4度冰箱过夜孵育。具体使用的一抗列举如下:anti-CILK1(ab196964,Abcam),anti-pCILK1(Tyr-159)(#103269,Thermo FisherScientific),anti-pCILK1(Thr-157)(SPC-996,StressMarq),anti-CDK1(GTX108120,GeneTex),anti-pCDK1(Thr-161)(ZEN-BIOSCIENCE),anti-actin(Santa Cruz)。
7.TBST缓冲液洗涤PVDF膜3次,每次10分钟。
8.在室温下,使用HRP标记二抗(稀释浓度1:5000)孵育PVDF膜1-1.5小时。
9.TBST缓冲液洗涤PVDF膜3次,每次10分钟。
10.ECL反应曝光显影。
结果如图1和2所示,CILK1-C30和CILK1-C28特异性抑制CILK1蛋白的磷酸化,因此抑制它的激酶活性,证明CILK1-C30和CILK1-C28是CILK1的特异性抑制剂。
实施例2:
CCK8方法检测细胞活力,测定药物抑制细胞活力的IC50值。
CCK8细胞活力测定实验步骤:
1.每个96-孔板细胞培养孔铺入1500个三阴乳腺癌细胞,将培养板在培养箱预培养24小时(37℃,5%v/v CO2)。
2.分别加入不同浓度的CILK1-C30或CILK1-C28,孵育设定的时间。
3.每10μL Cell Counting Kit-8(CCK8)(Bimake,B34302)试剂与100μL细胞培养液一起加入培养孔孵育2小时。
4.使用酶标仪在450nm波长下测量培养孔的吸光度值。
5.依据不同浓度CILK1-C30或CILK1-C28孵育下测定的细胞活力值,绘制活力曲线,计算出CILK1-C30或CILK1-C28抑制癌细胞活力的IC50值。IC50值表示抑制50%癌细胞活力所需的药物浓度,因此它的数值越小代表药物杀伤力越强。
CILK1-C30对于BT549的抑制情况如图3所示,对于MDA-MB-231的抑制情况如图4所示。
CILK1-C28对于BT549的抑制情况如图5所示,对于MDA-MB-231的抑制情况如图6所示。
实施例3:
紫杉醇(PTX)是三阴乳腺癌新辅助化疗的基本药物。CILK1-C30或CILK1-C28与紫杉醇联合使用明显的降低了紫杉醇的杀伤IC50值,更有效的杀死三阴乳腺癌细胞。
1.每个96-孔板细胞培养孔铺入1500个三阴乳腺癌细胞,将培养板在培养箱预培养24小时(37℃,5%v/v CO2)。
2.加入不同浓度的CILK1-C30或CILK1-C28和紫杉醇,孵育设定的时间。
3.每10μL Cell Counting Kit-8(CCK8)(Bimake,B34302)试剂与100μL细胞培养液一起加入培养孔孵育2小时。
4.使用酶标仪在450nm波长下测量培养孔的吸光度值。
5.依据不同浓度的CILK1-C30或CILK1-C28和紫杉醇孵育下测定的细胞活力值,绘制活力曲线,计算出抑制癌细胞活力的IC50值。
用浓度为2μM的CILK1-C30处理细胞,将紫杉醇抑制MDA-MB-231癌细胞活力的IC50值由6.883纳摩降至2.882纳摩,如图7所示,IC50值降低表示紫杉醇杀伤力增强。
用浓度为1μM的CILK1-C30处理细胞,将紫杉醇抑制BT549癌细胞活力的IC50值由4.751纳摩降至2.886纳摩,如图8所示。
用浓度为2μM的CILK1-C28处理细胞,将紫杉醇抑制MDA-MB-231癌细胞活力的IC50值由7.075纳摩降至2.001纳摩,如图9所示。
用浓度为1μM的CILK1-C28处理细胞,将紫杉醇抑制BT549癌细胞活力的IC50值由6.391纳摩降至1.798纳摩,如图10所示。
紫杉醇单独使用对三阴乳腺癌细胞的杀伤效力有限,且临床上常出现癌细胞对紫杉醇耐药的情况,CILK1-C30或CILK1-C28与紫杉醇的联用明显提高了杀伤效力。
实施例4:
CILK1-C30与紫杉醇联用对结肠癌细胞也有杀伤效果:
1.每个96-孔板细胞培养孔铺入1500个结肠癌细胞(RKO和HCT116),将培养板在培养箱预培养24小时(37℃,5%v/v CO2)。
2.加入不同浓度的C30和紫杉醇,孵育设定的时间。
3.每10μL Cell Counting Kit-8(CCK8)(Bimake,B34302)试剂与100μL细胞培养液一起加入培养孔孵育2小时。
4.使用酶标仪在450nm波长下测量培养孔的吸光度值。
5.绘制活力曲线,计算出紫杉醇抑制癌细胞活力的IC50值。
如图11,使用浓度为2微摩的CILK1-C30处理细胞,将紫杉醇抑制RKO癌细胞活力的IC50值由5.600纳摩降至3.729纳摩。
如图12,使用2.5微摩浓度的CILK1-C30处理细胞,将紫杉醇抑制HCT116癌细胞活力的IC50值由14.06纳摩降至8.810纳摩。
实施例5:
顺铂(DDP)和依托泊苷(VP-16)是小细胞肺癌化疗的基本药物,二者联用是化疗中常见的EP方案。CILK1-C30与顺铂和依托泊苷联合使用显著的降低了EP方案的杀伤IC50值,相比单独化疗更有效的杀死小细胞肺癌细胞(H446是小细胞肺癌细胞株,H446R是H446对EP方案耐药的衍生细胞株)。
1.每个96-孔板细胞培养孔铺入1500个小细胞肺癌细胞,将培养板在培养箱预培养24小时(37℃,5%v/v CO2)。
2.加入不同浓度的CILK1-C30、顺铂(DDP)和依托泊苷(VP-16),孵育设定的时间。
3.每10μL Cell Counting Kit-8(CCK8)(Bimake,B34302)试剂与100μL细胞培养液一起加入培养孔孵育2小时。
4.使用酶标仪在450nm波长下测量培养孔的吸光度值。
5.绘制活力曲线,计算出依托泊苷抑制癌细胞活力的IC50值,如图13-14所示。
图13所示,单独使用VP-16抑制H446活力的IC50值为38.31微克/毫升。使用浓度为1微摩的CILK1-C30处理细胞,将VP-16抑制H446活力的IC50值由38.31微克/毫升降至12.00微克/毫升。使用0.5微克/毫升的DDP处理细胞,将VP-16抑制H446活力的IC50值由38.31微克/毫升降至4.774微克/毫升。使用CILK1-C30/DDP一起处理细胞,将VP-16抑制H446活力的IC50值由38.31微克/毫升降至1.210微克/毫升。
如图14所示,单独使用VP-16抑制H446R细胞活力的IC50值为159.9微克/毫升。使用浓度为1微摩的CILK1-C30处理细胞,将VP-16抑制H446R活力的IC50值由159.9微克/毫升降至54.39微克/毫升。使用1.5微克/毫升的DDP处理细胞,将VP-16抑制H446R活力的IC50值由159.9微克/毫升降至16.23微克/毫升。使用CILK1-C30/DDP处理细胞,将VP-16抑制H446R活力的IC50值由159.9微克/毫升降至7.038微克/毫升。
顺铂和依托泊苷(EP方案)对小细胞肺癌细胞的杀伤效力有限,且临床上常出现癌细胞对EP方案耐药的情况,CILK1-C30与EP方案的联用明显提高了杀伤效力。
对于本领域的技术人员来说,可根据以上描述的技术方案以及构思,做出其它各种相应的改变以及变形,而所有的这些改变以及变形都应该属于本发明权利要求的保护范围之内。
