Abstract

Background
With the increasing working and living pressure and the deterioration of the external environment, the alopecia is not limited to occur in the elderly people any more, and the sick people are obviously young, which becomes a big problem disturbing the physical and mental health of people. Aiming at the problem, a large number of alopecia preventing and hair growing products appear on the market, but the symptoms of the alopecia symptoms are various and are linked with the external environment, genetic genes, immunoregulation and even living habits, and the existing products are difficult to treat the symptoms and the root causes.

Chinese patent CN201810317289.8 discloses a turmeric oil-control, anti-hair loss and hair-growing scalp essence and a preparation method thereof, wherein biota orientalis leaf extract, ginger root extract, dimethylol hydantoin, panthenol and other raw materials are adopted to act to achieve the effects of diminishing inflammation, resisting allergy, removing dandruff and controlling oil.
The prior art still cannot give consideration to the balance between the effects of hair loss prevention and hair growth, safety, no stimulation and cost of the hair washing and caring composition.
Disclosure of Invention
The invention provides the hair loss preventing and growing composition containing the plant extract, solves the problem that the safety, the hair loss preventing effect and the cost of hair washing and caring products in the prior art cannot be considered at the same time, and realizes the hair loss preventing and growing composition which is safe, mild and non-irritant.
In order to solve the above problems, the present invention provides a composition for preventing hair loss and promoting hair growth containing plant extracts, which comprises the following raw materials: plant extracts and polypeptides.
In some preferred embodiments, the plant extract is extracted from a combination of one or more of red clover, ginger, mulberry, camellia oleifera, cacumen biotae, polygonum multiflorum, soapberry, fructus viticis, and apple.
Further preferably, the plant extracts include apple (MALUS DOMESTICA) fruit cell culture extract and apple (MALUS DOMESTICA) fruit cell culture extract.
In some preferred embodiments, the polypeptide comprises a combination of one or more of copper peptide, acetyl hexapeptide-3, palmitoyl pentapeptide-3, acetyl tetrapeptide-5.
Further preferably, the polypeptide is acetyl tetrapeptide-5.
More preferably, the weight ratio of the apple fruit cell culture extract, the red clover extract and the acetyl tetrapeptide-3 is (50-80): (0.03-0.1): (0.03-0.1).
Under the influence of factors such as heredity, inflammation and environment, the problem of alopecia is difficult to eliminate completely, and the problem is a problem which puzzles consumers. In the research process, the invention discovers that the joint action of the plant extract and the polypeptide can activate Wnt signal conduction of scalp, activate the growth of dormant hair follicle and reduce seborrheic alopecia caused by excessive secretion of male hormone. In some preferred embodiments, the anti-alopecia composition is applied to scalp essence, and the raw materials of the scalp essence comprise, by weight:
phase A: 0.5-5% of lubricant, 0.1-1% of anti-inflammatory agent, 0.01-0.5% of EDTA disodium and the balance of water;
phase B: 1-10% of hair loss prevention and hair growth promoting composition, 0.1-4% of skin conditioner, 0.1-2% of hydroxylated saturated cyclane and 0.1-1% of preservative.
The sum of the weight ratios of the A phase and the B phase is 100 percent.
Further preferably, the raw materials of the scalp essence comprise:
phase A: 2% of lubricant, 0.4% of anti-inflammatory agent, 0.1% of EDTA disodium and the balance of water;

The preparation method of the scalp essence comprises the following steps:

Performance testing
In vitro hair follicle testing
1. Experimental materials:
1.1 mouse whisker Hair follicle
1.2 culture solution: williams' E serum-free medium (Gibco, lot: 2066552) containing 2mM L-glutamine, 10. Mu.g/mL bovine insulin, 10ng/mL hydrocortisone, 100U/mL penicillin, 100. Mu.g/mL streptomycin
1.3 control group: culture solution
1.4 Positive group: using culture solution to prepare minoxidil liniment with concentration of 10 μ M, sichuan Mei Da Kanghua pharmaceutical industry Limited
1.5 sample groups: samples (example 2) were prepared at concentrations of 70mg/mL,7mg/mL,0.7mg/mL using the culture broth.
2. The test steps are as follows:
2.1 isolating hair follicles: c57BL/6 mice were sacrificed humanely, bilateral whiskers were cut off, the upper lips were sterilized with 75% ethanol 3 times, bilateral whisker pads were cut off and placed in 1:1 (volume) of HBSS: PBS (100U/mL penicillin, 100. Mu.g/mL streptomycin in PBS), and mixing the mixture with 1:1 (volume) of HBSS: PBS was washed 3 times, connective tissue around the hair follicle was carefully removed under a dissecting microscope, the hair follicle was bluntly separated from the surrounding tissue with microsurgical forceps in the direction of the hair follicle, the distal end of the hair follicle was gently grasped, the hair was carefully pulled out of the subcutaneous tissue, and the separated hair follicles were placed in different wells of a 24-well plate.
2.2 Hair follicle culture: at 37 ℃ C, 5% CO 2 Incubate for 14 days under conditions, containing 0.5mL of medium per well.
2.3 sample adding: adding samples on the next day of culture, adding culture medium containing samples with different concentrations into corresponding holes, arranging 8 multiple holes in each group, placing 2-3 isolated hair follicles in each hole, and changing the liquid once every 3 days.
2.4 image analysis: whisker hair follicle length was photographed on days 1 and 14, and morphologically observed and measured.

Compared with a control group, the hair follicle relative growth rate of the positive group is statistically different (p is less than 0.05); compared with the control group, the sample group has statistical difference (p < 0.05) of the relative growth rate of hair follicles of 70mg/mL and 7mg/mL, and under the experimental condition, the sample of the example 2 has certain effect on promoting the hair growth at the concentration of 70mg/mL and 7mg/mL.

1. Experimental Material

2. Experimental procedure
2.1 preparation of type II 5 α -reductase: 10 male rats were taken, fasted overnight, cervical-free and sacrificed by removal of the cervical spine, prepared with pre-cooled buffer at 0 ℃ in a glass homogenizer to 1: homogenizing 5 (by mass), centrifuging 3000g for 10min, centrifuging 10000g of supernatant for 45min, and packaging the supernatant into EP tubes with 1mL per tube to obtain enzyme extract, and storing in refrigerator at-80 deg.C.
2.2 type II 5 α -reductase Activity assay: the reaction system contained 0.25mL of phosphate buffer, 0.70mL of enzyme extract, 0.1mL of testosterone solution (0.5 mg/mL), 0.35mL of NADPH solution (1 mg/mL), and 0.6mL of 10% ethanol (0.4. Mu.M finasteride in the positive control tube and solution in the sample tube). Replacing the enzyme extract with phosphate buffer solution in the control tube, and adding 2.0mL of dichloromethane to stop the reaction; the sample tube reacts at 37 ℃ for 30min, 2.0mL of dichloromethane is added after the reaction is finished to stop the reaction, and the sample tube is shaken for 1min and then centrifuged at 3000r/min for 10min. The upper aqueous phase was removed, 1.0mL of the organic phase was removed and evaporated to dryness, the residue was dissolved in 1.0mL of methanol and the area of the testosterone peak (T) was determined by high performance liquid chromatography. Each reaction was set up with 3 parallel channels.
2.3 analysis of results

2.5 conclusion of the experiment
The sample (example 2) was found to significantly inhibit type II 5. Alpha. -reductase activity at concentrations of 100%,50%,30%, 10%.
3. Stability test
The scalp essence provided in example 2 was placed at-10 ℃ for 30 days, then placed at 45 ℃ for 30 days, and taken out and returned to room temperature, and no difference between the physical and chemical conditions of the sample and the initial state, delamination, precipitation, and discoloration were observed.
4. Irritation test
The scalp essence obtained in example 1 was subjected to irritation test, and the test report is shown in fig. 6 to 9; the polypeptide scalp care essence for the plant cultivation of the test object is specifically the scalp essence provided in example 2.
Claims (10)
1. A hair loss preventing and hair growing composition containing plant extracts is characterized by comprising the following raw materials: plant extracts and polypeptides.
2. The composition for preventing hair loss and growing hair according to claim 1, wherein the plant extract is extracted from one or more of red clover, ginger, mulberry, camellia oleifera, cacumen biotae, fleece-flower root, soapberry, fructus viticis and apple.
3. The composition for preventing hair loss and promoting hair growth containing plant extracts as claimed in claim 2, wherein the plant extracts comprise apple extract and red clover extract.
4. The use of the anti-hair loss and hair growth composition containing the plant extract according to any one of claims 1 to 3, wherein the anti-hair loss and hair growth composition is applied to scalp essence, hair conditioner, hair care essential oil and hair mask.
5. The use of the composition for preventing alopecia and promoting hair growth containing plant extracts as claimed in claim 4, wherein the composition for preventing alopecia is applied to scalp essence, and the raw materials of the scalp essence comprise, by weight:
phase A: 0.5-5% of lubricant, 0.1-1% of anti-inflammatory agent, 0.01-0.5% of EDTA disodium and the balance of water;
phase B: 1-10% of hair loss prevention and hair growth promoting composition, 0.1-4% of skin conditioner, 0.1-2% of hydroxylated saturated cyclane and 0.1-1% of preservative.
The sum of the weight ratios of the A phase and the B phase is 100 percent.
6. The use of the composition for preventing hair loss and promoting hair growth containing plant extracts as claimed in claim 5, wherein the lubricant comprises one or more of glyceryl polyether, polyoxyethylene fatty acid ester, castor oil, and vaseline.
7. The use of the composition for preventing alopecia and promoting hair growth containing plant extracts as claimed in claim 5 or 6, wherein the anti-inflammatory agent comprises one or more of natural ingredients, dipotassium glycyrrhizinate, allantoin, and bisabolol.
8. The use of the anti-alopecia and hair-growing composition containing plant extracts as claimed in claim 7, wherein the natural ingredients comprise one or more of a combination of Chondrus crispus extract, aloe vera extract, hamamelis virginiana extract, chamomile extract, and calendula extract.
9. The use of the composition for preventing alopecia and promoting hair growth containing plant extracts as claimed in any one of claims 5-8, wherein the skin conditioner comprises one or more of quaternary ammonium salt, polysiloxane and its derivatives, hydrolyzed collagen, isopropyl isostearate, sclerotium rolfsii gum, hyssop extract, gentian extract, tetrandra root extract, and polyol ester.
10. The use of the composition for preventing alopecia and promoting hair growth containing plant extracts as claimed in any one of claims 5 to 9, wherein the number of hydroxyl groups in the hydroxylated saturated cycloalkane is > 4.
