Description

Acrylate polymer, medical adhesive composition and medical adhesive plaster
Technical Field
The invention belongs to the technical field of medical pressure-sensitive adhesives, and particularly relates to an acrylate polymer, a medical adhesive composition and a medical adhesive plaster.
Background
The pressure-sensitive adhesive can be adhered to an adherend only by applying external force, is convenient to use, has good stability and is widely applied to the aspect of medical treatment and health. The application range of the current medical pressure-sensitive adhesive comprises wound treatment, biomedical device adhesion, transdermal drug release patch, medical label, medical adhesive tape and other applications. Among the various types of medical pressure-sensitive adhesives, acrylic pressure-sensitive adhesives have two major advantages: firstly, the acrylate pressure-sensitive adhesive does not need to be added with other additives, is a single-component adhesive, avoids potential safety problems caused by the additives, is polymerized by emulsion or solution, can be directly used as an adhesive for use as a polymerization product, has great advantages in the production process due to convenient operation, and can be used for preparing the acrylate pressure-sensitive adhesive meeting different use requirements by adjusting the content and the components of the comonomer, thereby having great advantages in use convenience.
With the increasing and wider requirements for the performance of the medical pressure-sensitive adhesive, the medical pressure-sensitive adhesive not only needs to satisfy the adhesive performance (initial adhesion, peeling force, permanent adhesion, and the like), but also needs to have different additional characteristics according to different applications, such as the comfort of the pressure-sensitive adhesive adhered to the skin and the compatibility with medicines. However, most of the existing acrylate pressure-sensitive adhesives are generally adhered to muscles, joints and body parts needing medicament action in the process of adhering to the skin, and the movement modes of the parts are different from those of other parts, so that better comfort is often needed; in addition, many drugs have polarity or water solubility, and are not well compatible with polyacrylate compounds, and when the drugs are added into pressure-sensitive adhesives, phase separation occurs, which is not favorable for the drugs to fully exert the functions.
Compared with acrylate compounds in CN201010557593.3 and CN201010557610.3, most of the acrylamide monomers are easy to cause skin irritation, the interaction force between the adhesive and the skin can be increased, and the discomfort during tearing can be caused. Adding the sodium alginate hydrogel into medical acrylic ester in CN201410445351.3, fully stirring to obtain sodium alginate glue, and preparing the sodium alginate intramuscular effect patch by using the glue. The sodium alginate hydrogel is incompatible with the acrylate adhesive, and a relatively uniform mixture of the sodium alginate hydrogel and the acrylate adhesive is formed as a result of sufficient stirring, so that sodium alginate cannot be effectively diffused in the acrylate adhesive, only a small amount of sodium alginate hydrogel particles dispersed on the surface can play a role, and the using effect of sodium alginate is limited.
The existing medical pressure-sensitive adhesive is intended to be applied to the skin with low irritation; can be firmly adhered without falling off for some reasons, and some can be added with oil-soluble drugs, but the compatibility of water-soluble drugs is poor. When the medical adhesive patch is applied to some affected parts, the user may feel uncomfortable due to environmental changes, such as from a warm and humid indoor environment to a cold and dry outdoor environment.
Therefore, the development of the medical adhesive plaster which has high comfort and good compatibility with the medicine and is beneficial to the medicine to play the role has important significance.

Detailed Description
The following detailed description of the embodiments of the present invention, it should be noted that these embodiments are merely illustrative, and not restrictive of the invention in any way.
In one aspect of the invention, an acrylate polymer is provided. According to an embodiment of the present invention, the acrylate polymer is copolymerized from at least an alkyl acrylate monomer, a carboxyl group-containing monomer and a phase change monomer, the carboxyl group-containing monomer excluding the phase change monomer.
The inventor finds that the alkyl acrylate monomer in the acrylate polymer provides basic pressure-sensitive characteristics, the carboxyl-containing monomer can improve the permanent adhesive force of the pressure-sensitive adhesive layer, the adhesive time is long, the phenomenon of peripheral tilting is not easy to occur even if the pressure-sensitive adhesive layer is attached to skin for a long time, the carboxyl is a hydrophilic structure, the water can be absorbed, the compatibility of the pressure-sensitive adhesive with the oil-soluble medicine and the water-soluble medicine is increased, and the phase-change monomer can improve the compatibility of the pressure-sensitive adhesive with the oil-soluble medicine and the water-soluble medicine and keep the temperature and humidity change of the attached part. The alkyl acrylate monomer, the carboxyl-containing monomer and the phase-change monomer all contain unsaturated double bonds, and the alkyl acrylate monomer, the carboxyl-containing monomer and the phase-change monomer can be copolymerized in a free radical copolymerization mode to form a copolymer. From this, the acrylic ester polymer of this application has and holds the adhesion well, and with oil-soluble medicine and water-soluble medicine compatibility good, thereby be used for medical adhesive plaster with the pressure-sensitive adhesive layer that this acrylic ester polymer and curing agent mix preparation and make medical adhesive plaster have fine moisturizing effect, be favorable to skin moisture's absorption, keep and the conduction, and this medical adhesive plaster can keep pasting a temperature, has fine heat preservation effect, so this medical adhesive plaster has fine comfort, this medical adhesive plaster pressure-sensitive adhesive layer adhesion is good simultaneously, be difficult for coming off from skin or substrate, and good with the compatibility of medicine, it plays a role to have the medicine of utilizing.
According to an embodiment of the present invention, the alkyl acrylate monomer is 50 to 80 parts by mass, the carboxyl group-containing monomer is 2 to 10 parts by mass, and the phase change monomer is 1 to 9 parts by mass based on 100 parts by mass of the total amount of the monomers. The inventors found that, if the amount of the alkyl acrylate monomer is less than 50 parts by mass, the pressure-sensitive property of the resulting adhesive layer is insufficient; if the alkyl acrylate monomer is used in an amount of more than 80 parts by mass, the risk of residual gum increases. If the using amount of the carboxyl-containing monomer is less than 2 parts by mass, the stripping force of the adhesive layer is too low, and firm adhesion is not facilitated; if the amount of the carboxyl group-containing monomer is more than 10 parts by mass, the adhesive layer peeling force is too large. If the amount of the phase change monomer is less than 1 part by mass, the phase change property cannot be exhibited; if the amount of the phase-change monomer is more than 9 parts by mass, the peeling force is significantly reduced, resulting in too low adhesion of the adhesive layer to the adherend. Further, the molecular weight of the acrylate polymer is 100000 to 800000. The inventors have found that if the molecular weight of the acrylate polymer is less than 100000, resulting in a high polymer molecular content, the adhesive risks residue; if the molecular weight of the acrylate polymer is larger than 800000, the viscosity of the adhesive is relatively high, and the processability is not ideal. Therefore, the acrylic ester polymer with the molecular weight of 100000-800000 can reduce adhesive residue and improve processability of the acrylic ester polymer.
Specifically, the polymerization method of the acrylate polymer in the present invention includes, but is not limited to, bulk polymerization, solution polymerization, emulsion polymerization and suspension polymerization, and solution polymerization is preferred. In the free radical copolymerization process, a large amount of heat is generated, and the invention prefers to add a monomer to make the reaction smoothly, namely, adding a part of monomer, initiator and solvent as substrates, and then slowly adding a mixed solution of other monomers and initiator. According to one embodiment of the invention, the acrylate polymer is obtained by: mixing part of alkyl acrylate monomer, part of carboxyl-containing monomer, phase change monomer and initiator to obtain first mixed solution; mixing the rest part of alkyl acrylate monomer, carboxyl-containing monomer, phase change monomer and rest initiator with solvent to obtain a second mixed solution; and dropwise adding the first mixed solution into the second mixed solution, stirring the second mixed solution while dropwise adding, and then preserving heat to prepare the acrylate polymer. Thereby, the polymerization reaction is sufficiently caused to take place, and a large amount of heat generation is avoided. It should be noted that, a person skilled in the art may select the solvent and the initiator used in the process of preparing the acrylate polymer according to actual needs, and may select the addition amount of each material in the first mixed solution and the second mixed solution according to actual needs, which is not described herein again.
According to the embodiment of the invention, the reaction time of the second mixed solution is 0.5-2 hours, the reaction temperature is 70-85 ℃, and the first mixed solution is added after the second mixed solution is reacted; the dropping time of the first mixed solution is 2-5 hours, and the temperature of the first mixed solution is 70-85 ℃ during dropping; after the dripping is finished, the temperature is kept at 70-85 ℃ for 3-8 hours. Thereby, the polymerization reaction is sufficiently performed, and a large amount of heat generation is avoided.
Further, the phase change monomer is a compound capable of performing phase change within a certain temperature range, for example, the phase change monomer capable of performing phase change within a temperature range of 20 to 50 ℃ is adopted in the present application, according to one embodiment of the present invention, the side group of the phase change monomer is long-chain polyether, and the long-chain polyether is connected with the acrylate main chain through a covalent bond, so that the compatibility of the long-chain polyether and the acrylate polymer is good, the phase change monomer can be uniformly dispersed in the acrylate polymer system, and the improvement of the compatibility of the acrylate polymer is facilitated. Meanwhile, the polyether chain segment is of a hydrophilic structure, can absorb moisture, is beneficial to absorption, maintenance and conduction of skin moisture, and enables the pressure-sensitive adhesive to have certain compatibility with oily and aqueous medicines. Thus, compatibility and moisture retention of the acrylate polymer can be improved. According to an embodiment of the present invention, the phase change monomer includes maleic acid polyethylene glycol monomethyl ether monoester, wherein the molecular weight of polyethylene glycol part in the maleic acid polyethylene glycol monomethyl ether monoester is 200-2000, the pressure sensitive adhesive has certain compatibility with the water soluble compound due to the function of ether group, and the maleic acid polyethylene glycol monomethyl ether monoester is a phase change compound and has phase change within the range of 20-50 ℃, so that the temperature change is not rapid. Thus, compatibility and moisture retention of the acrylate polymer can be improved.
According to an embodiment of the present invention, the alkyl acrylate monomer is an acrylate monomer having an alkyl group with 4 to 12 carbon atoms, and includes, for example, at least one of butyl methacrylate, hexyl methacrylate, 2-ethylhexyl methacrylate, n-octyl methacrylate, isooctyl methacrylate, n-nonyl methacrylate, isononyl methacrylate, n-decyl methacrylate, isodecyl methacrylate, butyl acrylate, hexyl acrylate, 2-ethylhexyl acrylate, n-octyl acrylate, isooctyl acrylate, n-nonyl acrylate, isononyl acrylate, n-decyl acrylate, and isodecyl acrylate.
According to an embodiment of the present invention, the above carboxyl group-containing monomer includes at least one of acrylic acid, methacrylic acid, carboxyethylacrylic acid, carboxyethylmethacrylic acid, carboxypentylglycolic acid, carboxypentylmethylacrylic acid, 2-acryloxyethylphthalic acid, 2-methacryloxyethylphthalic acid, 2-acryloxyethylsuccinic acid, 2-methacryloxyethylsuccinic acid, 2-acryloxyethylmaleic acid, 2-methacryloxyethylmaleic acid, carboxypolycaprolactone monoacrylate, carboxypolycaprolactone monomethacrylate, 2-acryloxyethyltetrahydrophthalic acid, and 2-methacryloxyethyltetrahydrophthalic acid. The carboxyl-containing monomer plays a role of a physical crosslinking point in a polymer system, so that the permanent adhesion of the pressure-sensitive adhesive can be improved, the adhesion time is long, and the phenomenon of warping around the pressure-sensitive adhesive is not easy to occur even if the pressure-sensitive adhesive is attached to the skin for a long time.
In yet another aspect of the invention, a medical adhesive composition is provided. According to an embodiment of the invention, the medical adhesive composition comprises the acrylate polymer and the curing agent. The inventor finds that the acrylic ester polymer with good permanent adhesion and good compatibility with oil-soluble medicines and water-soluble medicines is mixed with the curing agent, and the acrylic ester polymer forms a cross-linking network after curing, so that the mechanical property of the pressure-sensitive adhesive layer is improved, the pressure-sensitive adhesive layer can be attached to a base material and skin and is not easy to fall off from the skin or the base material, meanwhile, the obtained pressure-sensitive adhesive layer has certain compatibility with the oil-soluble medicines and the water-soluble medicines, and has a phase change phenomenon, so that the temperature change of the attached surface is small, and the using effect of the pressure-sensitive adhesive layer is improved.
Further, the mass ratio of the acrylate polymer to the curing agent is (50-100): (0.1-5). The inventors have found that if the content of the acrylate polymer is too high, the structure formed by curing becomes too dense, the adhesion to the substrate is poor, and solvent and monomer residues tend to occur; if the content of the acrylate polymer is too low, the adhesive layer is too thin, which is not favorable for the adhesive layer to be effectively adhered to an adherend. If the content of the curing agent is too low, the structure formed by curing is easy to be too loose, the structure is easy to adhere to the skin, residues are left after tearing, and if the content of the curing agent is too high, the reaction groups on the curing agent cannot be completely reacted. It should be noted that, a person skilled in the art may select a specific type of the curing agent according to actual needs as long as the above functions are achieved, and according to an embodiment of the present invention, the curing agent of the present application is a metal chelate, for example, the metal chelate includes one or more of aluminum chelate, iron chelate, and titanium chelate.
In order to further increase the permanent adhesion and initial adhesion of the pressure-sensitive adhesive layer and facilitate the long-term adhesion of the pressure-sensitive adhesive layer on the skin, tackifying resin is further added into the medical adhesive composition, and the addition amount of the tackifying resin is (50-100) by mass ratio of the acrylate polymer to the tackifying resin: (1-20) is the standard. The pressure-sensitive adhesive prepared according to the ratio has a better bonding effect, the residual monomers and solvent molecules of the pressure-sensitive adhesive layer are less in residue, and the using effect of the pressure-sensitive adhesive is improved. It should be noted that the specific type of the tackifying resin can be selected by those skilled in the art according to actual needs, and for example, the tackifying resin includes at least one of terpene resin, terpene resin derivative, rosin resin, and rosin resin derivative.
According to an embodiment of the present invention, the medical adhesive composition further comprises: the auxiliary agent comprises at least one of a cross-linking agent, a leveling agent, a stabilizing agent, a defoaming agent, a filler, a compatibilization auxiliary agent, a catalyst, a catalytic accelerator, a surfactant, a diluent, a plasticizer, a cross-linking inhibitor, an antioxidant and an anti-aging agent. It should be noted that the above solvents and the auxiliaries are all materials conventionally used in the art, and those skilled in the art can select specific types and addition amounts thereof according to actual needs, and the features and advantages described above for the acrylate polymer are also applicable to the medical adhesive composition, and are not described herein again.
In yet another aspect of the present invention, a medical adhesive patch is provided. According to an embodiment of the invention, the medical adhesive plaster comprises a base material, a pressure-sensitive adhesive layer and a release layer which are sequentially stacked, wherein the pressure-sensitive adhesive layer is prepared from the medical adhesive composition. The inventor finds that the medical adhesive plaster has a good moisturizing effect, is beneficial to absorption, maintenance and conduction of skin moisture, can keep the temperature of an adhered part, and has a good heat preservation effect, so that the medical adhesive plaster has good comfort, and meanwhile, the pressure-sensitive adhesive layer of the medical adhesive plaster has good adhesiveness, is not easy to fall off from skin or a base material, has good compatibility with medicines, and plays a role in utilizing the medicines.
Specifically, the material of the base material is not particularly limited, and examples thereof include materials excellent in transparency, mechanical strength, thermal stability, water-blocking property, isotropy, flexibility, dimensional stability, and the like, polyesters such as polyethylene terephthalate, polyethylene naphthalate, polybutylene terephthalate, and the like, cellulose polymers such as diacetylcellulose, triacetylcellulose, and the like, polycarbonate polymers, and the like. The release layer is a conventional material in the art, and can be selected by a person skilled in the art according to actual needs, which is not described herein again.
It should be noted that the features and advantages described above for the medical adhesive composition are also applicable to the medical adhesive patch, and are not described herein again.
The invention will now be described with reference to specific examples, which are intended to be illustrative only and not to be limiting in any way.
