Abstract
本发明涉及一种化合物在制备肺纤维化粘度响应性荧光探针中的应用。本发明将化合物<Ⅰ>制备成肺纤维化粘度响应性近红外荧光探针,通过监测粘度的变化,可很好用于患者肺纤维化进程的检测。该近红外荧光探针对粘度具有高度的响应性,粘度敏感性范围为1cp~948cp,能够抗背景干扰,并在广泛pH范围内具有高度稳定性,对极性的变化不敏感。同时该荧光探针具有良好的生物相容性,在抗真菌药物诱导的细胞功能障碍和脂多糖诱导的细胞炎症模型中,具有监测活细胞微黏度波动的能力。近红外荧光探针在体内对肺纤维化早期的可视化足够敏感,且肺纤维化的严重程度与粘度的增加呈正相关,可作为临床准确预测肺纤维化进展的快速、可靠的工具。化合物<Ⅰ>结构式如下:

技术领域
本发明属于医疗检测技术领域,具体涉及一种化合物在制备肺纤维化粘度响应性荧光探针中的应用。
背景技术
特发性肺纤维化(Idiopathic pulmonary fibrosis,IPF)是一种常见的慢性、进行性的间质性肺疾病,主要特性为人体肺泡异常增殖,肺泡壁受损而渐渐演变成纤维化状态,其发病机制尚不明确。IPF早期症状表现仅仅是咳嗽,并不会引起患者的重视,但随着病情的持续发展,纤维化加重,给患者的身体及生活会造成极大困扰,通常患者确诊时肺纤维化已经发展到中晚期阶段,确诊后中位期一般只有3~4年。特发性肺纤维化因病因复杂,发病机制不明,使得其诊断和治疗面临巨大的困难,目前缺乏对IPF有针对性的治疗手段,对于IPF的早期诊断手段也极为缺乏。
到目前为止,仅有两种药物被推荐用于IPF的治疗——Nintedanib(尼达尼布)和Pirfenidone(吡非尼酮),这两种药物都有减缓IPF进展的作用,但仍不能改变或逆转纤维化病变。因此,对于IPF的早诊断和早治疗是提高患者生存率的关键手段。
IPF患者发病时会出现严重的咳嗽和呼吸困难,在肺部CT影像上可以观察到肺泡出现弥漫性异常增殖、肺实质和间质有不同程度的纤维化。常用的临床影像学方法如光诱导电子转移(PET)、磁共振成像(MRI)和计算机断层扫描(CT)等,只有在肺组织水平能观察到症状时,才能有效诊断IPF。另外,苏木精-伊红(H&E)和Masson染色等病理方法大多较为复杂,无法在体内成像,这些都增加了IPF早期诊断的难度。
文献《干细胞在特发性肺纤维化治疗中的研究与应用综述》(四川大学学报(医学版),2021,52(3):373-379)中记载:“目前,肺纤维化的发病机理尚不明确,无有效的治疗手段延缓病情进展。”“针对IPF的诊断,目前主要依靠胸部HRCT(高分辨率CT)及外科活检。”“尽管肺活检对于IPF的诊断意义重大,但多数患者在疾病早期往往未进行有效的病理相关检测,其发生发展的具体过程至今未明”。
文献《特发性肺纤维化的发病机制与临床治疗研究进展》(吉林医学,2021,42(12))中记载:“特发性肺纤维化是一种进展缓慢,但病死率极高的肺部疾病,肺纤维化发病率呈逐年上升的趋势,发病原因不明,病理机制复杂,多种信号通路相互交错关联。”“现代医学对于防治特发性肺纤维化,尚且缺乏有效的方法和药物,至今没有确切有效的治疗手段。”
文献《特发性肺纤维化的研究进展》(中国民间疗法,2020,28(03))中记载:“目前IPF的诊断主要根据其临床症状,并结合胸部高分辨率CT、血清学检测等进行综合诊断。”
上述文献记载表明:目前对于特发性肺纤维化的发病原因不明,病理机制复杂,早期症状不明显,无有效的诊断和治疗手段,主要根据临床症状和胸部HRCT和外科活检手段来进行病情的诊断。
据报道,生理微环境如pH、粘度、极性等在病理过程的早期就会发生异常变化,并随着疾病的发展而不断变化。当线粒体粘度水平异常时,会影响线粒体正常功能代谢,导致细胞凋亡,进而使生物体产生病症,如:阿尔兹海默症、帕金森症和胰岛素抵抗等。对于IPF而言,持续的慢性炎症过程和过量的细胞外基质沉积可能会增加活细胞和肺组织的粘度;然而,因IPF的发病机制无法明确,病因也极其复杂,粘度在IPF中的潜在生物学作用尚未被完全了解,另外,粘度的异常变化在活细胞中反应迅速,易受生物微环境的影响和随着病理周期的影响而发生较大变化,因此目前尚无针对于粘度检测来用于诊断IPF进程的方法。
现有对于粘度的检测手段较为精确的是荧光探针法,目前对粘度荧光探针的设计主要是关于分子转子的设计,受粘度环境的影响,荧光强度随着旋转体的分子内旋转而改变,从而达到检测和识别目的。天津理工大学硕士论文《检测粘度及生物活性物种荧光探针的合成及性能研究》中记载,可以用于粘度荧光探针设计的分子转子仅包括以下几种有限的荧光分子结构:推拉式荧光分子转子(如硫磺素连接苯并噻唑)、BODIPY型荧光分子转子(硼二吡咯亚甲基)、菁类荧光分子转子(可调节分子结构中共轭聚甲川链的长度)和罗丹明类荧光分子转子。
上述这些分子转子结构构建的荧光探针在用于粘度检测时仍然存在诸多问题,如发射波长较短、光稳定性较差、响应敏感性较低,对粘度的选择性较差,易受背景荧光的干扰,以及受其他环境因素如pH、温度和极性的影响较大。
大连理工大学硕士论文《苯并噻唑半菁染料荧光探针的设计合成及应用》中有如下记载:“菁染料普遍斯托克斯位移较小(通常约20nm),这也是许多染料(如BODIPY类、罗丹明B)都存在的问题。斯托克斯位移较小在一定程度上影响染料的光物理性能和测试结果,如造成荧光淬灭现象和造成仪器测试误差,这个缺陷在生物分析方面表现尤为突出。”“另外,菁染料存在水溶性差和稳定性差的缺点,而有机溶剂的存在有时候会影响染料与基质结合的过程,甚至改变基质的生物活性及两者结合之后产物的光谱性能。”“传统的分子转子局域于DCVJ及其衍生物,它们结构较为单一,在稳定情况下可准确测定环境粘度,然而却容易受环境极性的干扰。除此之外,生物体内环境复杂多变,容易导致染料各处浓度出现不同梯度的变化,从而给细胞内粘度检测带来极大的困难。”“BODIPY类染料由于具有较强的耐光性和较高的摩尔消光系数被用于环境粘度检测。除此之外,这类染料也可以用作各种各样的pH探针。事实上,许多担载有氮原子或羟基的BODIPY衍生物被报道作为荧光探针检测水相或水-有机相混合溶液的pH,但是它们之中很少能够检测细胞中的pH。”这些记载表明,现有的荧光染料在用于粘度检测时或多或少存在上述问题。
基于此,如何在四种分子转子主体结构基础上进一步设计荧光探针结构,来解决粘度荧光探针存在的上述问题,目前并没有定论。如何针对各类分子转子结构具有的特性来设计分子链的结构和进行不同的修饰,以期得到不同结构的粘度荧光探针化合物,赋予探针更多的功能性和更好的检测效果,以期能够实现将其用于特发性肺纤维化的检测和诊断,成为粘度荧光探针研究中的难点。
专利文献CN 112779001A报道了一种近红外粘度荧光探针,其是一种罗丹明类染料,该探针具有近红外激发和发射性质,能实现溶液中粘度的检测,并实现细胞内成像,但该探针仅涉及用于对恶性肿瘤、动脉粥样硬化、高血压、糖尿病和阿尔茨海默病等疾病的检测,因其存在抗干扰能力较弱、受溶剂极性的影响较大、稳定性较差,而无法用于IPF的诊断。
专利文献CN 114605397A报道了一种粘度近红外荧光探针,其也是一种罗丹明类染料,可用于活细胞中粘度的检测,该探针具有较好的抗干扰能力,但是由于该探针抗活性氮的干扰能力较弱,受溶剂极性变化的影响较大,同时其粘度敏感性范围较窄,无法用于IPF的精确诊断。
专利文献CN 114181204A公开了一种检测粘度的近红外荧光探针,该粘度荧光探针的结构中含有二甲氨基苯基和苯并噻唑基团,属于推拉式荧光分子转子,但该荧光探针的发射波长较短,量子产率较低,同时探针的耐极性变化稳定性仍有待提高,无法用于IPF的诊断。
现有开发出的可用于检测粘度的荧光探针,因其荧光强度不够、对粘度的响应性不够敏感、对pH的稳定范围不够宽泛以及耐背景干扰能力不够强,仅能用于几种有限的疾病检测,而均无法用于IPF进程的检测。其原因是这些粘度检测荧光探针用于监测不同肺纤维化状态以及临床肺纤维化患者样本的粘度变化时,存在检测精确度不高、探针稳定性较差、对粘度的检测范围较窄,同时易受背景干扰,对极性的变化较为敏感的缺陷。
因此,能否开发出一种高通量、多样本、高特异性和敏感性的近红外生物检测技术,用于在体内及临床患者标本中探索不同肺纤维化状态的变化,以期对IPF实现早期精准诊断,成为亟待解决的技术问题。
发明内容
本发明就是为了解决现有技术存在的问题,从而提供一种化合物在制备肺纤维化粘度响应性荧光探针中的应用。本发明的技术目的在于,提供一种对肺纤维化疾病的诊断具有高度特异性和敏感性的近红外生物检测技术,能够实现从肺纤维化早期到晚期进行精准检测,以用于临床对肺纤维化进程的精准诊断。
为了实现上述技术目的,本发明采用的技术方案如下:
本发明提供了一种化合物在制备肺纤维化粘度响应性荧光探针中的应用,该化合物的结构式如下式<Ⅰ>所示:
所述应用包括化合物<Ⅰ>在制备具有广泛pH范围内稳定性好的肺纤维化粘度响应性近红外荧光探针方面的应用,所述广泛pH范围为pH在4-9之间。
本发明是将式<Ⅰ>所示的化合物制备成近红外荧光探针,通过监测生物体内或体外粘度的波动变化,作为患者肺纤维化进程的检测工具。
本发明的上述化合物<Ⅰ>并未被现有报道所记载,其制备方法可以参见实施例1。与化合物<Ⅰ>结构相近的报道,仅见于1964年美国专利(US3384487A)及其同族公开的染料2。该美国专利中记载染料2作为一种吲哚丁二烯类染料,具有良好的吸光特性,是一种荧光化合物。
该美国专利公布于58年前,受限于当时的技术发展水平,人们仅发现其公布的染料2可作为照相显影剂或漂白剂使用,远未涉及当时尚未发展起来的生物检测领域。同时,在此专利之后,针对该专利中所涉及的相关化合物,人们并未发现其它用途,同时几乎停止了对这些化合物的进一步应用研究。
值得说明的是,如在背景技术中所述,在粘度响应性荧光探针方面的研究中,人们虽然对众多荧光性物质开展了大量的研究,但所得的荧光探针仍然存在检测精确度不高、探针稳定性较差、对粘度的检测范围较窄、易受背景干扰以及对极性的变化较为敏感的缺陷。
进一步而言,能适于肺纤维化检测的粘度响应性荧光探针则更为难得,同时还存在检测灵敏度低的问题。如文献《上海药物所研发用于诊断肺纤维化的小分子荧光探针》(化工新型材料,2020,48(02))中记载,该探针的荧光强度仅高于正常小鼠6倍,检测条件受到较大限制。也即是说,虽然本领域已开展了粘度响应性荧光探针的研究,但所得探针均难以用于特发性肺纤维化的检测。
经过大量的实验摸索,发明人意外地发现,将本发明化合物<Ⅰ>用于粘度高度响应性近红外荧光探针,发现其可以很好用于特发性肺纤维化进程的检测,能够从肺纤维化早期到肺纤维化晚期对细胞内粘度的变化实现精准灵敏的检测,有希望用于特发性肺纤维化的精准诊断。本发明的检测体系采用的是甘油和PBS的混合体系。
本发明的近红外荧光探针用于检测粘度时,具备粘度的检测范围宽、检测精度高、敏感度高的特点。而现有的粘度检测荧光探针,要么存在荧光强度低、检测精度低,要么存在粘度检测范围较窄,敏感度低的缺点,要么是容易受到各种背景因素的干扰和极性变化的干扰,难以精准用于特发性肺纤维化的检测。
本发明探索出了一种将化合物<Ⅰ>作为近红外荧光探针,用于检测肺纤维化在受测对象中不同状态下粘度变化,来精准监测肺纤维化从早期到纤维化(晚期)的过程。本发明的近红外荧光探针,对环境中粘度的变化有高度选择性和敏感性,其响应性极好,具有反应速度快、准确性高的特点,同时该近红外荧光探针能够因电荷的转移激发出不同的荧光颜色变化,上述特点决定了该近红外荧光探针化合物能够很好用于特发性肺纤维化的检测。
本发明的粘度响应性荧光探针中的C-C键在低粘度环境下可以自由旋转,从而导致荧光猝灭;在高粘度环境下,由于旋转抑制,荧光恢复。另外,该化合物结构中的磺酸基和吲哚盐可增强其水溶性,并能通过活细胞的质膜渗透。此外,该粘度响应性荧光探针的拉推结构和较大的共轭长度能够发射近红外波长,适用于活体成像。
此外,该近红外荧光探针化合物在抗真菌药物诱导的细胞功能障碍和脂多糖诱导的细胞炎症模型中均表现出良好的生物相容性,具有监测微粘度波动的能力。其在体内对肺纤维化早期的可视化足够敏感,且肺纤维化的严重程度与粘度的增加呈正相关,该检测技术有望成为临床准确预测肺纤维化进展的一种快速而可靠的工具。
本发明首次将化合物<Ⅰ>作为一种粘度响应性近红外荧光探针,通过粘度的变化来显示肺纤维化从早期到纤维化的过程。该近红外荧光探针在低粘度条件下几乎没有荧光,在高粘度条件下近红外发射(704nm)明亮。同时,近红外荧光探针具有良好的生物相容性,在抗真菌药物诱导的细胞功能障碍和脂多糖诱导的细胞炎症模型中,具有监测活细胞微黏度波动的能力。近红外荧光探针在体内对肺纤维化早期的可视化足够敏感,且肺纤维化的严重程度与粘度的增加呈正相关。因此,近红外荧光探针有望成为临床准确预测肺纤维化进展的快速、可靠的工具。
本发明将粘度响应性荧光探针化合物<Ⅰ>用于肺纤维化的检测,具有检测方法简单,具备高通量、多样本、高特异性和敏感性的检测特点,其粘度高效响应性范围在1cp~948cp之间,检测精度远远高于现有的粘度近红外荧光探针,可很好用于检测或辅助检测待测样本是否为放射性肺纤维化,为放射性肺纤维化的诊断提供了新的检测标准,为该疾病的机制及防治提供了新的线索。
进一步的是,所述应用包括化合物<Ⅰ>在制备具有抗背景因子干扰的肺纤维化粘度响应性近红外荧光探针方面的应用,所述背景因子包括活性氧、活性氮、活性硫和/或金属离子。
进一步的是,所述活性氧包括H2O2、HOCl、·OH和·O2 -中的至少一种,所述活性氮包括NO和ONOO-中的至少一种,所述活性硫包括S2 -、GSH和Cys中的至少一种,所述金属离子包括Na+、K+、Ca2+、Cu2+、Zn2+、Mg2+和Fe3+中的至少一种。
进一步的是,所述应用包括化合物<Ⅰ>在制备具有抗极性干扰的肺纤维化粘度响应性近红外荧光探针方面的应用,所述极性干扰为极性溶剂和非极性溶剂之间的变化。
进一步的是,所述应用包括化合物<Ⅰ>在制备具有抗极性干扰的肺纤维化粘度响应性近红外荧光探针方面的应用,所述极性干扰为极性溶剂和非极性溶剂之间的变化。
本发明的有益效果如下:
(1)本发明提供了一种化合物在制备肺纤维化粘度响应性荧光探针中的应用,本发明提供的近红外荧光探针对环境中粘度的变化具有高度选择性和敏感性,具有响应性好、反应速度快、准确性高的特点,同时该近红外荧光探针能够因电荷的转移激发出不同的荧光颜色变化,可以很好用于监测体内和体外粘度的变化;
(2)本发明提供的一种化合物在制备肺纤维化粘度响应性荧光探针中的应用,可以通过对粘度变化的高度响应性和敏感性,用于精准检测肺纤维化的进程,特别是能够用于对肺纤维化早期的诊断。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例对本发明进行具体描述,有必要指出的是,以下实施例仅用于对本发明进行解释和说明,并不用于限定本发明。本领域技术人员根据上述发明内容所做出的一些非本质的改进和调整,仍属于本发明的保护范围。
实施例1
本发明提供的近红外荧光探针化合物的结构式如下式<Ⅰ>所示:
该近红外荧光探针的制备方法如下:
(1)将2,3,3-三甲基吲哚(1.36g,10mmol)加入到1,2-二氯苯溶液(15mL)中,然后向其中缓慢加入1,3-丙烷磺酸内酯(1.59g,10mmol),将上述混合溶液在120℃下搅拌12h,真空脱除溶剂后,用乙醚洗涤紫色产物,真空干燥后备用;
(2)将步骤(1)所得产物(212mg,0.5mmol)和(Z)-3-(4-(二甲氨基)苯基)丙烯酰基醛(87.5mg,0.5mmol)溶于15mL无水甲醇中,然后加入1mL哌啶进行搅拌,混合溶液在N2气氛下回流12h,然后冷却至室温,经旋转蒸发,用洗脱液(DCM/MeOH=20/1,v/v)进行柱层析,得到近红外荧光探针,记为NIR-PF。
表征结果如下:
1H NMR(400MHz,Methanol-d4)δ(ppm):8.29(1H,dd,J1=3.2Hz,J2=15.2Hz),7.97(1H,d,J=8.8Hz),7.63(4H,m),7.53(1H,m),7.46(1H,m),7.28(1H,m),6.96(1H,d,J=14.4Hz),6.88(1H,d,J=9.2Hz),6.80(1H,d,J=9.2Hz),4.49(1H,m),4.41(1H,m),3.19(3H,s),3.11(3H,s),2.92(2H,m),2.09(2H,m),1.98(2H,m),1.77(6H,d,J=10.8Hz),1.29(4H,m).
HRMS m/z:calcd 453.2206,found 453.2207[M]+。
实验例1
(1)NIR-PF的表征
本发明制备的近红外荧光探针NIR-PF,其中二乙基胺(供电子)基团与半花青素(吸电子)基团通过缩合反应连接,C-C键在低粘度环境下可以自由旋转,从而导致荧光猝灭;NIR-PF在高粘度条件下,由于旋转抑制,荧光恢复(如图1)。
此外,磺酸基和吲哚盐可增强半花青素染料的水溶性,并倾向于通过活细胞的质膜渗透。NIR-PF的拉推结构和较大的共轭长度有望发射近红外信号,适用于活体成像。
(2)NIR-PF对粘度的光谱特性
为了考查NIR-PF是否能够高效用于检测粘度变化,对NIR-PF在PBS缓冲液和甘油中的光学性能进行了研究。
如图2中A分图所示,NIR-PF在PBS缓冲溶液中的主吸收峰位于551nm处,而在甘油中的最大吸收峰位于564nm处,红移约13nm,这是由于在高粘度条件下NIR-PF形成了更平面的构型,从而增加了共轭程度。
如图2中B分图所示,NIR-PF在704nm处从PBS缓冲液到甘油溶液的近红外荧光增强约54.4倍,NIR-PF的绝对荧光量子效率也从0.004(PBS缓冲溶液)提高到0.210(甘油),提升了50倍。荧光在低粘度下的猝灭是由于分子转子的自由旋转导致非辐射衰减;相反,高粘度会限制C-C键的旋转,减少非辐射途径,从而恢复荧光。
综上,上述结果表明,NIR-PF能够很好用于检测粘度的变化。
(3)对荧光变化的影响因素考查
首先考查了不同极性溶剂对荧光变化的影响,从图3中A分图中可以看出,吸收峰变化不大,NIR-PF在不同溶剂中荧光较弱,而在甘油中荧光较强(图3中B分图),表明NIR-PF对极性变化不敏感。
除极性变化外,pH、稳定性等其他反应条件对粘度的检测也至关重要。如图4所示,在不同比例的甘油-PBS混合物条件下,随着时间的增加,NIR-PF的荧光非常稳定,随着甘油-PBS混合物比例从0%增加到100%,荧光强度逐渐增加。此外,荧光光谱在30min内也保持不变(图5)。
进一步验证了pH对NIR-PF荧光变化的干扰。如图6所示,在甘油-PBS混合环境(0%、25%、50%、75%)下,NIR-PF的荧光在pH值4.0~9.0范围内保持稳定。此外,不同pH值下荧光强度的增加趋势与粘度的增加一致。
上述结果表明:NIR-PF探针具有很好的稳定性,可以在不受pH变化、极性变化和微环境背景干扰的情况下,精准检测粘度的变化。
(4)竞争分子对粘度选择性的考查
考虑到复杂生物条件下大量大分子的存在,测试了许多生物相关的大分子存在下NIR-PF对粘度的选择性的影响,包括活性氧(H2O2、HOCl、·OH、·O2 -)、活性氮(NO、ONOO-)、活性硫(S2 -、GSH、Cys)和金属离子(Na+、K+、Ca2+、Cu2+、Zn2+、Mg2+、Fe3+)。
结果发现,NIR-PF在加入分析物后显示出微弱的荧光,而在甘油引起的粘度条件下加入NIR-PF,可以观察到明亮的近红外荧光(图7),表明NIR-PF对粘度的响应性能优于其他竞争物。
(5)NIR-PF对粘度变化的荧光和荧光寿命测量
在排除干扰和优化响应条件后,进一步评估NIR-PF是否可以很好用于监测粘度的变化。
如图8中A分图所示,NIR-PF在704nm处的荧光强度随粘度的增加(从1cP增加到1000cP)而逐渐增大,并且粘度变化与NIR-PF荧光强度在1cP到1000cP范围内的线性拟合良好(图8中B分图)。
进一步测量了NIR-PF在不同粘度环境下的荧光寿命。根据时间分辨的发光衰减曲线,得到NIR-PF的荧光寿命随着粘度的增加从0.17ns(PBS)延长到1.20ns(甘油)(图8中C分图)。同时,NIR-PF在较大的动态范围内也显示出荧光寿命(logτ)和粘度(logη)之间良好的线性关系(图8中D分图)。这些结果证实了NIR-PF能够精准用于定量检测荧光粘度变化和荧光寿命模式。
(6)活细胞粘度变化的细胞毒性和荧光成像
通过CCK-8试验对RAW 264.7细胞和A549癌细胞进行细胞毒性实验。从图9中可以看出,NIR-PF(0~20μM)作用24h后,90%以上的RAW264.7细胞和A549癌细胞仍然存活,表明NIR-PF的细胞毒性很低。
进一步验证NIR-PF是否可以用于监测活细胞粘度的变化。将不同培养皿中的A549细胞分别用Nys或Mon处理1h后,再用NIR-PF孵育30min。如图10中A分图所示,仅用NIR-PF处理的细胞荧光较弱,荧光强度随Nystatin或Monensin引起的细胞内粘度的增加而大大增强,说明NIR-PF可用于检测活细胞粘度的变化。
进一步研究NIR-PF检测炎症细胞模型中粘度波动的可行性。RAW264.7细胞经脂多糖(LPS)预处理4h后,与NIR-PF共孵育。如图10中B分图和C分图所示,与仅用NIR-PF处理的对照组相比,经LPS处理后的细胞荧光明显增强;而LPS和强ROS清除剂n-乙酰半胱氨酸(NAC)预处理的细胞,也表现出与对照组一样的微弱荧光,说明RAW264.7细胞的粘度相对较低。表明NIR-PF在生物系统粘度波动监测方面具有很大的潜力。
实验例2
(一)不同时间博来霉素诱导的肺纤维化模型检测
选用特异性无病原体的8周龄雄性C57BL/6小鼠(购自北京维港河实验动物科技有限公司),小鼠体重为18-22g,所有小鼠均在四川大学动物实验中心饲养。
实验过程进行以下处理:将小鼠腹腔注射戊巴比妥钠(40mg/kg)进行麻醉,然后气管内注射70μL生理盐水(NS)或博莱霉素(BLM,5mg/kg)用于诱导肺纤维化。第0d、3d、7d、14d分别采用尾静脉注射NIR-PF(50μM、100μL),分别在640nm激发波长和700nm发射波长下拍摄全身荧光图像。处死小鼠后,采集肺荧光图像,将肺解剖标本固定于10%甲醛溶液中进行苏木精-伊红染色(H&E)和Masson染色,并将保存在液氮中的肺标本用于共聚焦成像。所有方案均经四川大学动物保护与使用委员会同意。
取肺纤维化患者的临床组织标本,将获得的组织用PBS洗涤后,用NIR-PF(10μM)探针孵育30min,PBS洗涤3次后用于成像。
(二)博来霉素诱导肺纤维化不同时间的活体显像
将小鼠按博来霉素不同作用时间(0d、3d、7d、14d)分为4组。小鼠尾静脉注射NIR-PF后进行成像。如图11所示,对照组小鼠肺区荧光不明显,第三天肺部荧光明显增强,说明早期粘度增加。此外,随着博来霉素刺激时间的延长,小鼠的平均荧光强度逐渐增加,说明肺粘度随肺纤维化严重程度的增加而增加。离体成像实验的荧光增强趋势与体内成像一致。这些结果表明,肺纤维化从早期开始,粘度水平逐渐升高。
为了证实NIR-PF的诊断效果,对肺切片进行测量。从离体肺组织中获得不同小鼠肺切片。如图12所示,肺的荧光强度随着博来霉素作用时间的增加而增强,结果与体内和体外成像实验一致。为进一步评估粘度与肺纤维化程度的相关性,采用H&E染色和Masson染色。如图13所示,与对照组相比,博来霉素组有明显的病理表现,如炎症细胞浸润、肺泡结构破坏、肺泡间隙增宽等。此外,肺纤维化的严重程度与博来霉素刺激时间呈正相关。
这些结果表明NIR-PF可以作为早期肺纤维化诊断的一种荧光探针,具有高度敏感性和检测精准度。
(三)临床患者组织肺纤维化的检测
将临床确诊为肺纤维化患者的组织用NIR-PF(10μM)孵育30min,PBS洗涤3次。从图14中A分图的3D图像中可以看出,患者组织的荧光较亮,而正常组织的荧光较弱,说明患者组织的粘度高于正常组织。H&E染色和Masson染色结果与影像学结果一致。此外,组织也直接在近红外成像系统和曝光下成像,患者组织的荧光强度也强于对照组(图14中B分图和C分图)。这些结果表明NIR-PF在临床应用中可作为一种快速、可靠的肺纤维化诊断工具。

2.根据权利要求1所述的应用,其特征在于,所述应用包括化合物<Ⅰ>在制备具有抗背景因子干扰的肺纤维化粘度响应性近红外荧光探针方面的应用,所述背景因子包括活性氧、活性氮、活性硫和/或金属离子。
3.根据权利要求2所述的应用,其特征在于,所述活性氧包括H2O2、HOCl、·OH和·O2 -中的至少一种,所述活性氮包括NO和ONOO-中的至少一种,所述活性硫包括S2 -、GSH和Cys中的至少一种,所述金属离子包括Na+、K+、Ca2+、Cu2+、Zn2+、Mg2+和Fe3+中的至少一种。
4.根据权利要求1所述的应用,其特征在于,所述应用包括化合物<Ⅰ>在制备具有抗极性干扰的肺纤维化粘度响应性近红外荧光探针方面的应用,所述极性干扰为极性溶剂和非极性溶剂之间的变化。
5.根据权利要求1所述的应用,其特征在于,所述应用包括化合物<Ⅰ>在制备具有粘度敏感性的肺纤维化粘度响应性近红外荧光探针方面的应用,所述荧光探针的粘度敏感性变化范围为1cp~948cp。
