Description

Uterus warming patch and manufacturing process thereof
Technical Field
The application relates to the technical field of medicines, in particular to a uterus warming patch and a manufacturing process thereof.
Background
Dysmenorrhea is one of the most common gynecological diseases of women. Modern medicine believes that dysmenorrhea occurs in association with vegetative nerve functional disturbance and prostaglandin release. The traditional Chinese medicine considers that the disease location of dysmenorrhea is in uterus and Chong and ren meridians, and the main pathogenesis is 'obstruction leading to pain' or 'failing to nourish leading to pain'. Clinically, abdominal pain during menstruation is mostly due to cold syndrome and blood stasis, and this disease is treated by warming meridians to dispel cold, activating blood circulation, removing blood stasis and alleviating pain. Modern pharmacological studies prove that the lower abdomen stasis removing soup can obviously relieve the uterine swelling of a rat caused by a plastic tube, and has a relatively obvious inhibiting effect on subcutaneous granuloma of the rat caused by a cotton ball, which indicates that the soup has an anti-inflammatory effect. In clinical application, aiming at different symptoms of dysmenorrheal, the medicines are increased and decreased according to symptoms, are flexibly tailored, and are combined to play the effects of warming channels and expelling cold, activating blood and dissolving stasis, and promoting qi circulation and relieving pain, so that cold people can be warmed, stasis people can be dissolved, the pathogeny is removed, channels and collaterals and qi and blood are smooth, thoroughfare and conception vessels and uterus blood are harmonized, and the circulation is not painful, and the symptoms are not repeated periodically along with menstruation.
At present, the lower abdomen stasis removing decoction evolves except traditional Chinese medicine decoction, granules, pills, capsules and plaster preparations are available on the market. The decoction can be added or subtracted according to symptoms, can fully exert the comprehensive curative effect and characteristics of various components of the prescription medicine, has quick effect, but needs to be prepared and taken at the same time, is easy to mildew after long-term storage, has poor taste, is complicated to decoct and is inconvenient to carry. The granule maintains the traditional decoction basis, overcomes the difficulty in decocting the traditional Chinese medicines, is more convenient to carry, store and take compared with a liquid preparation, but has the advantages of high price, easy deliquescence, poor mixing performance during the dissolution, easy layering, frequent taking and poor taste. The pill has the advantages of slow dissolution and drug release, prolonged drug effect, reduced toxicity and irritation, reduced adverse reaction, portability, inconvenient administration, easy dissolution and slow disintegration due to improper production and operation, poor taste, and difficulty in swallowing. The capsule can cover up the special smell of the medicine and improve the stability of the medicine, has relatively low price, quick response and portability, but has the phenomena of difficult swallowing and more times of taking.

Transdermal administration of Chinese herbs is a common administration way, which allows the effective ingredients of Chinese herbs to enter the systemic circulation through skin absorption, resulting in systemic or local therapeutic effects. The transdermal drug delivery preparation can avoid the first pass effect of the liver and the damage of the gastrointestinal tract, reduce the drug delivery times, prolong the drug delivery time, maintain constant effective blood concentration, reduce the toxic and side effects of the drug and improve the curative effect. However, because the transdermal administration permeation rate of traditional Chinese medicine is generally low, improving the transdermal efficiency of the effective components of traditional Chinese medicine is particularly important for carrying out the transdermal administration of traditional Chinese medicine. Modern pharmacy can improve the absorption of the effective ingredients of the traditional Chinese medicine on the skin by preparing the effective ingredients of the traditional Chinese medicine into a nano preparation, and provides a preparation process of the following formula for enhancing the efficacy of the formula.
A manufacturing process of a uterus warming patch comprises the following steps:
pulverizing the raw materials with ultramicro-nanometer powder, and filtering with ultrafiltration membrane with pore diameter of 0.001-0.01 μm to obtain nanopowder and residue; heating Vaseline into liquid, stirring and mixing the Vaseline liquid and the nano powder, wherein the ratio of the Vaseline liquid to the nano powder is 1 mL/g, cooling to obtain an ointment, adding the ointment onto a non-woven fabric blank patch, wherein the mass of the ointment is 5-10g, covering the non-woven fabric blank patch added with the ointment with paper, and refrigerating and storing at the temperature of 2-10 ℃.
The raw materials are crushed into nano powder to prepare the patch which has good absorption performance and high bioavailability, can be used for treating or assisting in treating chronic and easily-recurrent dysmenorrhea for a long time, makes up the defects of bitter taste, high price, difficult decoction and the like of traditional Chinese medicine decoction, exerts the advantages of the traditional Chinese medicine acupoint patch, can fully utilize the medicine, exerts the medicine effect to the greatest extent and ensures the treatment effectiveness. The solid lipid nanoparticles can form a film on the surface of skin because a high-concentration surfactant is not used in the preparation process, promote the absorption of the medicament through the hydration of epidermis after partial sealing, and increase the contact time and the area of the medicament and the horny layer, thereby being beneficial to the percutaneous absorption of the medicament.
In the embodiment, the method also comprises the steps of adding ethanol into the filter residue, carrying out reflux extraction at 60-80 ℃ for 30-50min, then filtering, centrifuging at 800-1000r/min for 5-10min, concentrating until the water content of the thick paste is 20-30% to obtain thick paste, and then stirring and mixing the thick paste with vaseline liquid and nano powder.
The filter residue has larger grain diameter, so that the filter residue is directly prepared into ointment which is not beneficial to percutaneous absorption, and the effective components in the filter residue are extracted and then mixed with the nano powder, thereby not only enhancing the content of the effective components of the medicament, but also enhancing the medicament absorption, improving the bioavailability of the medicament and leading the patch to have better treatment effect on dysmenorrheal and cold womb.
