背景技术
含有酶的口腔护理组合物描述于EP3192496、EP3192495、EP 3192565和EP3192564中。然而,仍然需要配制具有增强的酶活性的组合物。
本申请涉及用于增强口腔护理产品中的酶的活性的组合物和方法。此外,本发明克服了一些牙膏配制物可能发生的脱水收缩问题。
发明内容
本发明涉及牙膏组合物,该牙膏组合物包含酶、包含脂肪醇的聚乙二醇醚的表面活性剂、二氧化硅研磨剂、角叉菜聚糖和天然树胶(natural gum),该天然树胶选自刺梧桐树胶、瓜尔胶、黄原胶、阿拉伯树胶、黄蓍胶以及它们的混合物,其中黄原胶与角叉菜聚糖的重量比为1∶3至1∶1。

发明详述
根据本发明的组合物包含角叉菜聚糖。如果组合物包含大于角叉菜聚糖总含量的50重量%的λ-角叉菜聚糖,则是优选的。优选地,κ-角叉菜聚糖与λ-角叉菜聚糖的重量比为1∶3至2∶3,优选地,ι-角叉菜聚糖与λ-角叉菜聚糖的重量比为1∶6至1∶9。如果角叉菜聚糖包含λ-角叉菜聚糖、κ-角叉菜聚糖的混合物,则是优选的。还可以存在i-角叉菜聚糖。
优选地,角叉菜聚糖的含量为总组合物的0.1重量%至1.5重量%、更优选0.2重量%至1.0重量%。
本发明的组合物包含选自刺梧桐树胶、瓜尔胶、黄原胶、阿拉伯树胶和黄蓍胶的天然树胶,优选地本发明的组合物包含黄原胶。优选地,天然树胶、优选黄原胶的含量为总组合物的0.05重量%至1重量%,更优选为0.3重量%至0.7重量%。
本申请涉及包含酶的组合物,优选地所述组合物为个人护理组合物,更优选所述组合物为口腔护理组合物。本发明的组合物的优选产品形式是适于刷和/或冲洗口腔表面的那些。

在本发明的上下文中,单独的葡萄糖氧化酶或酶混合物的葡萄糖氧化酶部分是特别优选的。
优选地,酶的总含量为总组合物的0.01重量%至2重量%。更优选地为总组合物的0.05重量%至1重量%。
优选地,组合物中糖苷水解酶的总含量为总组合物的0.05至1重量%。
优选地,作用于供体的OH基团的氧化还原酶的总含量为总组合物的0.05至1重量%。
优选地,血红素过氧化物酶的总含量为总组合物的0.0005重量%至0.01重量%。
优选地,组合物中糖苷水解酶与其它酶的重量比大于1∶1,更优选大于3∶2。
本发明的组合物还可以包含其它蛋白质,诸如溶菌酶、乳铁蛋白和初乳。优选地,这些其它蛋白质以总组合物的0.001至0.5重量%、优选总组合物的0.005至0.1重量%的总含量存在。
根据本发明的组合物包含二氧化硅。用于本发明中的优选的二氧化硅是具有低折射率的二氧化硅。它可用作唯一的研磨性二氧化硅使用,或与低含量的其它研磨性二氧化硅例如根据EP 236070的那些结合使用。在本发明中用作研磨剂的低折射率二氧化硅优选为这样的二氧化硅,其具有在1.41至1.47、更优选1.43至1.45的范围内的表观折射率(R.I.),优选具有5至15μm的重均粒度,10至150m2/g的BET(氮)表面积,更优选约70-150cm3/100g的油吸收,但也可使用具有较低表观折射率的研磨性二氧化硅。合适的低折射率研磨性二氧化硅(例如具有在1.435和1.445之间的R.I.)的典型实例是购自Rhone Poulenc的Tixosil 63和73;购自Degussa的Sident10;购自Zeofinn的Zeodent 113;购自Huber的Zeodent 124、购自Crosfield Chemicals的Sorbosil AC 77(具有约1.440的R.I.)。这些二氧化硅在组合物中的量通常为总组合物的5至60重量%,更优选5至20重量%。
在一些实施方案中,如果将酶吸附/吸收到增稠二氧化硅上,则是优选的。优选地,所述二氧化硅具有小于150cm3/100g、更优选小于1cm3/100g的油吸收水平。特别优选的二氧化硅是Zeodent 165和Tixosil 43,因为这种类型的二氧化硅由于它们增强酶活性而优选。
所述口腔护理组合物将包含表面活性剂,所述表面活性剂包含按所述组合物的重量计0.2%至7%的量的脂肪醇的聚乙二醇醚。特别优选的是每单元具有20至40个环氧乙烷基团的聚乙二醇醚。这种合适的表面活性剂的实例包括被称为硬脂醇聚醚表面活性剂的表面活性剂组,如硬脂醇聚醚30。优选地,非离子表面活性剂的含量为总组合物的0.5至7重量%,更优选为总组合物的1至5重量%。
尽管可以存在其它表面活性剂,但如果将它们的含量限制在低于总组合物的0.5重量%、优选低于总组合物的0.1重量%,则是优选的。
在优选的产品形式中,基于组合物的总重量,按水和/或多元醇的总重量计,水和/或多元醇的量通常为至少10重量%、优选至少30重量%、更优选至少50重量%。优选地,水和/或多元醇的含量可以小于总组合物的90重量%、更优选小于85重量%。
根据本发明的洁齿剂组合物通常可包含各种相对量的水和多元醇的混合物作为含水连续相,其中水的量通常为10至50重量%(基于洁齿剂的总重量),多元醇的量通常为20至60重量%(基于洁齿剂的总重量)。
用于根据本发明的口腔护理组合物中,特别是洁齿剂组合物中的典型多元醇包括湿润剂,诸如甘油、山梨醇、聚乙二醇、聚丙二醇、丙二醇、木糖醇(和其它可食用多元醇)、氢化的部分水解的多糖以及它们的混合物。甘油和山梨醇是优选的,山梨醇是特别优选的。
所述口腔护理组合物,特别是洁齿剂组合物包含研磨材料。基于洁齿剂的总重量,研磨材料通常以0.5至75重量%、优选以3至60重量%的量存在。合适的研磨性清洁剂包括二氧化硅干凝胶、水凝胶和气凝胶以及沉淀颗粒状二氧化硅;碳酸钙、磷酸二钙、磷酸三钙、煅烧氧化铝、偏磷酸钠和偏磷酸钾、焦磷酸钠和焦磷酸钾、三偏磷酸钠、六偏磷酸钠、颗粒状羟基磷灰石以及它们的混合物。二氧化硅是特别优选的,因为它们可以发挥研磨剂和增强酶活性的手段的双重作用。优选地,组合物,特别是牙膏包含如上所述的二氧化硅基研磨剂。
依赖于所选择的材料,该组合物可以包含总组合物的约0.10至约15重量%比例的附加的无机或天然或合成增稠剂或胶凝剂。可用于本发明实践的合适的增稠剂或胶凝剂包括细碎分散的二氧化硅、锂蒙脱石、碳酸钙、胶态硅酸镁铝以及它们的混合物。优选的是无机增稠二氧化硅,诸如以商品名称Zeodent 165获自Evonik的无定形二氧化硅。
本发明的组合物可包含金属离子诸如锡或锌。优选地,锌离子源是氯化锌、乙酸锌、葡萄糖酸锌、硫酸锌、氟化锌、柠檬酸锌、乳酸锌、氧化锌、单甘油酸锌、酒石酸锌、焦磷酸锌、马来酸锌以及它们的混合物。
本发明的组合物可包含氟化物源,诸如氟化钠、氟化亚锡、单氟磷酸钠、氟化锌铵、氟化锡铵、氟化钙、氟化钴铵以及它们的混合物。
本发明的组合物还可以含有本领域常规的其它任选成分,诸如抗微生物剂,例如氯己定、血根碱提取物、甲硝唑、季铵化合物诸如氯化十六烷基吡啶;双胍类,诸如氯己定二葡糖酸盐、海克替啶、奥替尼啶、阿来西定;和卤代双酚化合物,诸如2,2'-亚甲基双-(4-氯-6-溴苯酚);
抗炎剂,诸如布洛芬、氟比洛芬、阿司匹林、吲哚美辛等;
抗龋齿剂,诸如氟化钠和氟化亚锡、氟化胺(aminefluoride)、单氟磷酸钠、三偏磷酸钠和酪蛋白;
牙斑缓冲剂,诸如尿素、乳酸钙、甘油磷酸钙和聚丙烯酸锶;
维生素类,诸如维生素A、维生素C和维生素E;
植物提取物;
植物可衍生的抗氧化剂,诸如类黄酮、儿茶素、多酚和单宁化合物以及它们的混合物;
脱敏剂,例如柠檬酸钾、氯化钾、酒石酸钾、碳酸氢钾、草酸钾、硝酸钾和锶盐;

**增稠二氧化硅
聚合物凝胶的脱水收缩归因于聚合物凝胶的物理和化学性质随时间的变化。例如,聚合物凝胶的物理变化与聚合物的渗透压和弹性调节有关,这是由于聚合物链的重排而发生的。化学变化可与例如氢键或静电相互作用有关。所有这些物理-化学变化通过形成凝胶网络的聚合物链的聚集而导致聚合物基质的强化,这转而排斥出(expel)连续相。因此,控制聚合物链随时间的聚集过程限制了脱水收缩。测量脱水收缩和监测聚合物链的聚集过程的方法可以通过检查在短时间(即2至3小时)内经历热变化的糊状物的行为来进行。对样品进行热处理增加了物理和化学相互作用随时间变化的速率,因此可减少实验时间,这进而又增加实验产出。温度循环改变(即,破坏,形成或增强)聚合物链的相互作用和键合(即,疏水相互作用,氢键合等)。这些不同的相互作用导致结构和网络的变化,潜在地使糊状物不稳定。通过温度循环测量弹性模量(G')和粘性模量(G”)以及tanδ(=G”/G')使得能够理解这些物理-化学变化。例如,G’随温度升高而降低突出了氢键的可逆断裂,导致更粘稠和液体状的样品。当温度冷却回去时,将发生新的分子间相互作用(离子和氢键合),从而导致结构改变。结构的破坏程度使得能够估计更容易随时间而经历脱水收缩的样品。

Claims (14)
1.一种牙膏组合物,其包含酶、包含脂肪醇的聚乙二醇醚的表面活性剂、二氧化硅研磨剂、角叉菜聚糖和天然树胶,所述天然树胶选自刺梧桐树胶、瓜尔胶、黄原胶、阿拉伯树胶、黄蓍胶以及它们的混合物,其中所述天然树胶与角叉菜聚糖的重量比为1∶3至1∶1。
2.根据权利要求1所述的组合物,其中所述天然树胶与角叉菜聚糖的重量比为1∶2至1∶1。
3.根据前述权利要求中任一项所述的组合物,其中所述天然树胶为黄原胶。
4.根据前述权利要求中任一项所述的组合物,其中所述酶选自糖苷水解酶、作用于供体的OH基团的氧化还原酶、血红素过氧化物酶或它们的混合物。

12.根据前述权利要求中任一项所述的组合物,其中所述角叉菜聚糖包含9∶4∶1至6∶3∶1重量比的λ-角叉菜聚糖、κ-角叉菜聚糖和i-角叉菜聚糖。
13.根据前述权利要求中任一项所述的组合物,其中所述脂肪醇的聚乙二醇醚每单元包含20至40个环氧乙烷基团。
14.以1∶3至1∶1的重量比的天然树胶和角叉菜聚糖在牙膏中用以减轻脱水收缩和增强酶活性的用途,其中所述天然树胶选自刺梧桐树胶、瓜尔胶、黄原胶、阿拉伯树胶、黄蓍胶以及它们的混合物。
