Abstract
本发明公开了一种靶向PBX1启动子区G‑四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用,所述治疗黑色素瘤药物具有如下任意一种或多种用途:抑制黑色素瘤细胞增殖;抑制黑色素瘤细胞迁移;抑制黑色素瘤细胞侵袭;抑制黑色素瘤生长;抑制黑色素瘤肺转移。本发明对靶向PBX1启动子区G‑四链体的反义寡核苷酸的鉴定为新一代抑制黑色素瘤药物的开发提供了一个全新的药物靶点以及新的治疗手段和思路。

技术领域
本发明涉及生物医药领域,尤其涉及靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用。
背景技术
黑色素瘤(Melanoma)是一种由黑色素细胞引起的恶性肿瘤,是最具侵袭性的一种皮肤癌,近年来其发病率一直在上升。根据美国癌症协会的统计,黑色素瘤仍然是最致命的皮肤肿瘤类型,2021年估计有106,110例新病例和7,180例死亡。尽管开发和应用了更多的治疗方法,黑色素瘤的预后仍然很差,因此,迫切需要进一步研究,以确定更有效的治疗方案,以实现更好的临床效果。
发明内容
为了解决现有技术的上述不足,本发明提供一种,靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用,体内研究表明,靶向PBX1启动子区G-四链体的反义寡核苷酸的治疗几乎完全抑制了黑色素瘤的生长及肺转移,本发明为新一代治疗黑色素瘤药物的开发提供了一个全新的药物靶点以及新的治疗手段和思路。
本发明的目的是通过以下技术方案实现的:
作为本发明的第一方面,提供一种靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用。
具体地,所述治疗黑色素瘤药物具有如下任意一种或多种用途:
抑制黑色素瘤细胞增殖;
抑制黑色素瘤细胞迁移;
抑制黑色素瘤细胞侵袭;
抑制黑色素瘤生长;
抑制黑色素瘤肺转移。
可选地,所述靶向PBX1启动子区G-四链体的反义寡核苷酸包括如下至少一种:
特异性抑制PBX1-G4的化合物;
特异性干扰PBX1-G4表达的干扰分子;
特异性敲除PBX1的基因编辑试剂。
可选地,所述特异性抑制PBX1-G4的化合物为反义寡核苷酸。
优选地,所述药物中反义寡核苷酸的含量为15mg/ml~35mg/ml。
可选地,所述特异性干扰PBX1-G4表达的干扰分子为反义寡核苷酸。
优选地,所述反义寡核苷酸具有如SEQ ID NO:1~2所示的核苷酸序列。
作为本发明的第二方面,提供一种筛选药物的方法,所述药物用于预防或治疗黑色素瘤,所述方法包括:
将候选药物用于黑色素瘤模型;
定量检测用药前后黑色素瘤模型的PBX1蛋白;
用药后相比于用药前,所述黑色素瘤模型的PBX1蛋白的表达水平降低,表明所述候选药物为目标药物。
作为本发明的第三方面,提供一种试剂在制备试剂盒中的应用,所述试剂用于定量检测PBX1蛋白表达水平,所述试剂盒用于判断药物预防或治疗黑色素瘤的有效性。
本发明的有益效果:
本发明检测了PBX1-G4对黑色素瘤生长及肺转移的影响,发现PBX1的表达与黑色素瘤的发展正相关。PBX1的序列在进化上是保守的,在小鼠和人类中是相同的。人源(A375cell line)及鼠源(B16-F10 cell line)的体外模型结果表明PBX1对黑色素瘤细胞系的调节可能转化为人类病理生理学。本申请的研究表明,靶向PBX1启动子区G-四链体的反义寡核苷酸显著抑制黑色素瘤细胞系的增殖、迁移及侵袭能力。体内研究表明,靶向PBX1启动子区G-四链体的反义寡核苷酸的治疗几乎完全抑制了黑色素瘤的生长及肺转移。本发明对靶向PBX1启动子区G-四链体的反义寡核苷酸的治疗的鉴定为新一代治疗黑色素瘤药物的开发提供了一个全新的药物靶点以及新的治疗手段和思路。

下面将结合具体实施例对本发明的实施方案进行详细描述,但是本领域技术人员将会理解,下列实施例仅用于说明本发明,而不应视为限制本发明的范围。实施例中未注明具体条件者,按照常规条件或制造商建议的条件进行。所用试剂或仪器未注明生产厂商者,均为可以通过市售购买获得的常规产品。
本发明实施例中使用的动物和试剂如下:
实验用小鼠:本研究使用的4-6周无特异性病原体(specific pathogen free,SPF)C57BL/6小鼠和BALB/C雄性裸鼠均购自北京华福康实验动物科技有限公司(中国北京)。NCG(猫。#T001475)小鼠来自健药药业(中国江苏)。实验前在特定的无菌条件下饲养2周。所有体内实验均由吉林大学公共卫生学院机构动物护理与利用委员会批准并监督。所有的努力都是为了尽量减少动物的痛苦。
试剂:TMPyP4(美国莱克;猫。#P1202)和Pyridostatin(PDS;美国莱克;猫。#S7444)溶解,置于-80℃保存。实验前将TMPyP4和PDS在培养基中稀释到所需的工作浓度。所有的化学品都没有经过进一步的净化。pLVX-IRES-mcherry(货号:#VT1461),PBX1启动子pGL3-Basic(货号:#VT15541)和pFLAG-CMV 2(货号:#VT1068)来自YouBio(中国)。Anti-GAPDH(货号:#ab9485)和anti-ZIC2(Cat。#ab150404)来自英国的Abcam。Anti-PBX1(猫。#432s)和GAPDH(货号:#14793S)来自CST(美国)。Anti-PBX1(货号:#PA5-82118)来自Invitrogen公司(美国)。Anti-G4(货号:Ab00174-30.126)。
本发明实施例中的部分实验方法如下:
免疫组织化学(包含IHC)测定:将福尔马林固定的、石蜡包埋的黑色素瘤、肝脏、脾脏、肺和肾脏切除后加工成石蜡包埋的组织块。从免疫组化块上切下的组织切片被安装在玻片上,在二甲苯中脱蜡,在乙醇中再水合。用胰蛋白酶(0.1%)预处理,然后用过氧化物1阻断内源性过氧化物酶活性。5min后,背景惩罚器染色。封闭的载玻片是孵育的抗体。然后将载玻片与兔对犬辣根过氧化物酶二抗孵育。然后用NanoZoomer 2.0-RS数字病理(滨松,日本)拍摄图像。每个区域拍摄6张显微照片。
RNA提取及定量RT-PCR检测:使用TRIzol(Invitrogen,USA)从培养细胞中提取总rna,然后使用SuperScript III第一链合成系统(Invitrogen,USA)将其转化为cdna。qRT-PCR检测在ABI 7500(Invitrogen,USA)中使用SYBRMix(Invitrogen,USA)进行。用比较Ct法计算RNA的相对表达量。
免疫印迹分析:对于全细胞裂解液的蛋白质分析,用RIPA(CWBIO,北京,中国)缓冲液裂解细胞。对于肿瘤,液氮首先用于粉碎组织。然后用RIPA(CWBIO,北京,中国)缓冲液在冰上产生裂解液20分钟,然后在4℃,13000rpm离心15分钟。将含蛋白上清液转移至新鲜微离心管中,-20℃(短期保存)保存,以备下一步使用。BIO-RAD DC蛋白测定法(BIO-RAD,USA)对蛋白进行定量。总蛋白在sds-聚丙烯酰胺凝胶上电泳。然后,蛋白质被转移到聚偏二氟乙烯(PVDF)膜上。与一抗在4℃的摇板器上孵育一夜后,用TBS-T(Invitrogen,美国)冲洗膜6次(每次5分钟)。然后,印迹与辣根过氧化物酶(HRP)偶联的二抗在平板摇床上RT孵育60分钟。用TBS-T清洗膜(6×5分钟)。采用SuperSignalTMWest Pico化学发光底物试剂盒(ThermoFisher Scientific,USA)和Western blotting检测系统(BioRad,USA)检测免疫反应条带。GAPDH作为western blotting检测的加载对照。条带强度水平归一化至GAPDH。
集落形成实验:对于菌落形成试验,将A375和B16-F10细胞以500-1000个细胞的密度种在6孔板上。2周后,室温下用1%结晶紫染色,PBS和自来水冲洗,计数菌落数。
体外细胞侵袭实验:Transwell侵袭实验采用Matrigel作为侵入性上腔膜屏障(Corning,USA)。下transwell室充满含DMEM培养基的2.5%血清。细胞悬浮在上室。孵育24小时后,除去过滤器,用甲醇固定膜上的细胞。0.5%结晶紫染色细胞浸润周围基质。染料用水冲洗,细胞用显微镜检查(莱卡,德国)。对每个人进行了6张显微照片。
体内肿瘤生长试验:为评价PBX1对体内肿瘤生长的影响,在接种前将6周龄雄性裸鼠(BALB/C背景)随机分为指定组(5只/组)。将1×106A375黑色素瘤细胞皮下注射到小鼠背部上侧腹。注射后,给予PDS药物治疗21天(两天一次),在牺牲后测量肿瘤重量。
双荧光素酶报告基因检测:对于PBX1荧光素酶报告基因检测,将PBX1荧光素酶报告基因质粒和pRL-TK质粒在96孔板上通过Lipofectamine 2000(Invitrogen,USA)共转染HEK293T、A375和B16-F10细胞。转染48小时后,收集细胞并使用报告者分析系统(Promega,美国)进行分析。萤火虫的活性归一化为Renilla荧光素酶的活性。
EGFP报告载体构建:以pLV-EGFP-N为骨架基因的报告载体(Inovogen Tech.Co.,Beijing,China)对PBX1 5’UTR序列进行rG1位点编码。通过使用Strata基因快速改变位点定向诱变试剂盒,我们对rG1位点进行了点突变。
细胞增殖实验:根据制造商的说明(Abcam,英国),通过细胞计数试剂盒-8(CCK-8)测定细胞活力。细胞(每孔2000个细胞)在96孔板上孵育3个副本,然后在不同时间点以10%的终浓度添加到每孔中,在37℃下继续孵育。60分钟后,在450nm波长下测定样品的吸光度。使用GraphPad Prism软件对数据进行分析。
细胞划痕化验:将细胞接种于12孔板中,生长24小时至80%融合。一个裸露的区域是通过一个尖端在盘子的直径上创造的。PDS(2μM)和TMPyP4(5μM)处理24h后,PBS冲洗细胞,并在显微镜下(Leica,Germany)拍照评价细胞迁移情况。对每个人进行了6张显微照片。
体内肿瘤转移试验:尾静脉肿瘤注射2×105 B16-F10细胞入侧尾静脉。根据NCI动物护理和使用委员会制定的方案,每个实验通过最大原发肿瘤负荷和/或所有治疗组中濒死小鼠的出现来确定转移终点。PDS(5mg/kg)给药,每日2次,连用16天。
生物发光肿瘤细胞追踪:在活体全动物成像中,麻醉小鼠腹腔注射3mg d-荧光素(MCE,美国)。在IVIS系统(Perkin Elmer,美国)上收集发光数据。对于活体组织成像,组织灌注PBS,收获,并在PBS中孵育1μg/ml-1d-荧光素。使用IVIS Image软件包进行数据分析。
黑色素瘤的PDX模型:术后立即收集新鲜肿瘤组织,置于含有青霉素/链霉素的RPMI1640培养基(100U/ml;100μg/ml)、真菌带(1μg/ml)和庆大霉素(50μg/ml)(均来自美国Gibco公司)(4,5)。根据机构评审和吉林大学第二医院伦理委员会批准的本研究伦理指南,在知情同意的情况下,采集黑色素瘤患者的新鲜肿瘤样本进行移植。小鼠保留,所有程序均在吉林大学(中国长春)公共卫生机构动物护理和使用委员会(IACUC)的批准和监督下进行。移植前切除坏死组织,将肿瘤组织切碎,大小为2×2×2mm3,植入6周龄NCG雄性小鼠侧翼皮下,产生第一代(F1)PDX小鼠。成功移植的肿瘤模型用标准方法传代和储存。H&E染色用于评价患者肿瘤标本和建立的PDX模型的异种移植瘤的形态。当小鼠肿瘤体积达到220-280mm3时,开始用ASO Scr或ASO(静脉注射,20mg/kg)治疗。进行肿瘤生长和体重测定。治疗结束时肿瘤被解剖,用于RNA、蛋白质和组织学分析的进一步处理。
患者来源的肿瘤细胞的制备:PDXs的肿瘤组织用基本的RPMI1640培养基(Gibco,USA。Free-FBS介质)。患者来源的肿瘤细胞(PDC)由低温保存的异种移植碎片制备,使用肿瘤分离试剂盒(德国Miltenyi Biotec公司),遵循肿瘤治疗方案。按照Bruna和Shanker等人的描述进行PDCs培养(6,7)。PDCs用于菌落形成试验。
本发明实施例中所有统计学分析均用t检验进行,并表示为平均值±SEM。分析中未排除任何动物或样本。p值指定为:*,p<0.05;**,p<0.01;***,p<0.005;****,p<0.001;认为p<0.05时具有统计学意义。
实施例1 PBX1基因启动子和转录本中G4s基因的鉴定
鉴于PBX启动子和转录本中鸟嘌呤含量极其丰富,本发明首先决定采用整合策略筛选PBX1启动子和转录本中潜在的G4s(图1A)。通过结合2个独立的G4预测软件,我们在PBX1启动子区鉴定出2个潜在的DNA G4s(命名为dG1和dG2),在PBX1转录本中鉴定出3个潜在的RNA G4s(命名为rG1、rG2和rG3)。此外,通过连续G/C比值(cGcC)、G4Hunter(G4H)和G4神经网络(G4NN)评分来评价这些潜在G4的G4折叠能力。其中,dG1和rG1的cGcC、G4H和G4NN得分最高(图1B),说明dG1和rG1的G4形成能力最强。值得注意的是,在所有100种脊椎动物中,dG1区域高度保守,这表明dG1和rG1在PBX1调控中的进化作用(图1C)。因此,我们选择dG1和rG1作为进一步研究的典型靶点。
实施例2 PBX1 G4s体外和细胞内的形成
本实施例进一步采用了几种方法来验证dG1和rG1的形成。首先,利用圆二色谱(CD)检测dG1和rG1序列(33nt)及其突变体的摩尔椭圆度,该突变体旨在消除G4的形成。在100mM K+缓冲液中,野生型(WT)dG1和rG1序列的CD光谱显示典型的264nm正摩尔椭偏峰和240nm负摩尔椭偏峰,而突变体则没有(图2A)。通过凝胶迁移率测定,发现dG1和rG1比它们的突变体迁移更快,这表明dG1和rG1折叠成紧凑的二级结构(图2B)。n-甲基mesophorphyrin IX(NMM)是一种著名的荧光G4特异性配体,可用于研究G4的体外形成。发现dG1和rG1都能增强NMM的荧光(图2C)。此外,本实施例中使用一种众所周知的G4特异性抗体BG4在A375细胞中进行了染色质免疫沉淀(ChIP)和RNA免疫沉淀(RIP)检测。发现PBX1启动子和转录本可以被BG4拉下(图2D和2E),表明PBX1 G4s在细胞中形成。总的来说,dG1和rG1可以在体外和细胞内折叠稳定的G4结构。G4特异性配体,包括PDS和TMPyP4等,已被证明具有结合和稳定DNA和RNA G4s的能力。因此,接下来研究了这些G4特异性配体与PBX1 G4s的相互作用能力。发现PDS和TMPyP4均显著增强了dG1和rG1的热稳定性,表明PBX1 G4s与这两个g4特异性配体之间存在很强的相互作用。此外,ChIP和RIP分析表明,PDS和TMPyP4可以增加下拉PBX1启动子和BG4转录本(图2F和2G)。综上所述,PDS和TMPyP4能够结合并稳定PBX1 G4s。
实施例3 PBX1 G4s的形成抑制PBX1的转录和翻译
鉴于启动子和转录本中的G4s参与调控转录和翻译相关过程,我们推测PBX1启动子中的dG1可能调控PBX1的转录,而PBX1转录本5'UTR中的rG1可能调控PBX1的转录。为此,首先进行了荧光素酶报告基因检测。将PBX1的dG1启动子序列(指定为dG1-wt)和突变的dG1启动子序列(指定为dG1-mut)克隆到pGL3荧光素酶报告载体中。与dG1-MUT转染的细胞相比,转染dG1-WT的HEK293T、A375和B16-F10细胞的荧光素酶活性显著降低(图3A)。值得注意的是,PDS和TMPyP4处理显著抑制了dG1-wt转染的细胞的荧光素酶活性,但对dG1-mut转染的细胞没有影响(图3A),说明G4特异性配体通过靶向PBX1 dG1来抑制转录过程。此外,本实施例还构建了一个增强型绿色荧光蛋白(EGFP)报告基因系统来研究rG1对翻译的影响。将PBX1的rG1转录序列(指定rG1-wt)和突变rG1转录序列(指定rG1-mut)融合到EGFP的5'UTR上,将报告基因载体转染HEK293T细胞。通过共聚焦荧光实验,rG1-WT的EGFP荧光强度小于rG1-MUT,PDS或TMPyP4降低了HEK293T细胞中rG1-WT的荧光强度(图3B)。最后,本实施例研究了PBX1 G4s对A375和B16-F10细胞中PBX1表达的潜在影响。发现PDS和TMPyP4处理显著降低了PBX1的mRNA和蛋白水平(图3C-4J)。综上所述,PDS和TMPyP4诱导PBX1 G4s的形成抑制了PBX1的表达。
实施例4 PBX1 G4s的形成通过下调PBX1抑制黑色素瘤的进展
本实施例研究PBX1 G4s的形成是否通过下调PBX1抑制黑色素瘤的进展。首先检测了PDS和TMPyP4对黑色素瘤细胞系的影响。观察到PDS和TMPyP4处理显著降低了A375和B16-F10细胞的活力,且呈剂量依赖性(图4A和4B)。同样,PDS和TMPyP4处理抑制了A375和B16-F10细胞的集落形成(图4C和4D)。PDS和TMPyP4也能抑制A375和B16-F10细胞的迁移和侵袭(图4E-4H)。在小鼠中进一步测试了PDS对致瘤性的影响。与体外结果一致,PDS治疗通过下调PBX1表达显著抑制移植瘤生长和转移(图4I-4P)。综上所述,这些结果表明PDS和TMPyP4诱导的PBX1 G4s的形成通过下调PBX1抑制黑色素瘤的进展。
实施例5 PBX1 ASO通过特异性靶向rG1抑制黑色素瘤进展
虽然PDS和TMPyP4对黑素瘤表现出强大的抗肿瘤作用,但它们的抗肿瘤作用可能部分是由于其与其他细胞G4s结合,脱靶效应也可能导致副作用,限制了它们在黑素瘤治疗中的临床应用潜力。反义寡核苷酸(ASOs)具有良好的临床应用潜力,并已被用于诱导RNAG4s的形成。RNA G4结构是一个基序,它本身是一个更大的RNA上下文的一部分,其折叠受到邻近序列的影响,因为G4s和邻近序列可以参与一个基于Watson-Crick的稳定结构。因此,我们设计了特异性针对PBX1 5'UTR中rG1互补序列的ASO,诱导PBX1 rG1的形成(ASO PBX1-G4:

Control:mG*mT*mC*mC*mA*mC*mA*mA*mA*mC*mA*mC*mA*mA*mC*mT*mC*mC*mT*mG*)。事实上,RIP检测显示ASO治疗可以增加BG4拉下的PBX1转录本(图5A),表明ASO诱导rG1的形成。此外,ASO治疗A375和B16-F10黑色素瘤细胞株可有效降低PBX1蛋白和mRNA丰度(图5B和5C)。本实施例还建立了患者源性黑色素瘤异种移植(PDX)源性肿瘤细胞(PDCs)的短期培养(图5D)。因此,A375细胞和PDCs的菌落形成实验显示ASO处理后菌落形成能力下降(图5E)。为了进一步扩展这一分析,本实施例建立了黑色素瘤的PDX模型,每两天静脉注射(静脉注射)PBX1,持续21天。与PBX1 ASO scramble治疗的肿瘤相比,观察到ASO治疗的肿瘤生长和重量显著降低(图5F和5G)。同时,ASO治疗导致PBX1和增殖标志物Ki67表达显著下调(图5H和5I)。综上所述,这些结果表明ASO诱导PBX1 rG1形成是一种很有前途的抗黑色素瘤治疗策略。

Claims (9)
1.一种靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用。
2.如权利要求1所述的一种靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用,其特征在于,所述治疗黑色素瘤药物具有如下任意一种或多种用途:
抑制黑色素瘤细胞增殖;
抑制黑色素瘤细胞迁移;
抑制黑色素瘤细胞侵袭;
抑制黑色素瘤生长;
抑制黑色素瘤肺转移。
3.如权利要求2所述的一种靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用,其特征在于,所述靶向PBX1启动子区G-四链体的反义寡核苷酸包括如下至少一种:
特异性抑制PBX1-G4的化合物;
特异性干扰PBX1-G4表达的干扰分子;
特异性敲除PBX1的基因编辑试剂。
4.如权利要求3所述的一种靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用,其特征在于,所述特异性抑制PBX1-G4的化合物为反义寡核苷酸。
5.如权利要求4所述的一种靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用,其特征在于,所述药物中反义寡核苷酸的含量为15mg/ml~35mg/ml。
6.如权利要求3所述的一种靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用,其特征在于,所述特异性干扰PBX1-G4表达的干扰分子为反义寡核苷酸。
7.如权利要求6所述的一种靶向PBX1启动子区G-四链体的反义寡核苷酸在制备治疗黑色素瘤药物中的应用,其特征在于,所述反义寡核苷酸具有如SEQ ID NO:1~2所示的核苷酸序列。
8.一种筛选药物的方法,其特征在于,所述药物用于预防或治疗黑色素瘤,所述方法包括:
将候选药物用于黑色素瘤模型;
定量检测用药前后黑色素瘤模型的PBX1蛋白;
用药后相比于用药前,所述黑色素瘤模型的PBX1蛋白的表达水平降低,表明所述候选药物为目标药物。
9.一种试剂在制备试剂盒中的应用,其特征在于,所述试剂用于定量检测PBX1蛋白表达水平,所述试剂盒用于判断药物预防或治疗黑色素瘤的有效性。
